RESUMEN

Rational syntheses of trisubstituted sulfur-bearing closo-boranes are presented. In the development of these syntheses unusual cationic closo-boranes [1,7,9-(Me(2)S)(3)-B(12)H(9)](+) (3) and [1,2,10-(Me(2)S)(3)-B(10)H(7)](+) (4) have been identified. These were initially recognized to be intermediates in the formation of the neutral trisubstituted species 1,7-(Me(2)S)(2)-9-(MeS)-B(12)H(9) (1) and 1,10-(Me(2)S)(2)-2-(MeS)-B(10)H(7) (2), respectively. Stable tetrafluoroborate salts were prepared and isolated, and their structures are presented. They are believed to represent the first structural determinations of cationic borane clusters of any type.

RESUMEN

Alpha,beta-difluoromethylene deoxynucleoside 5'-triphosphates (dNTPs, N = A or C) are advantageously obtained via phosphorylation of corresponding dNDP analogues using catalytic ATP, PEP, nucleoside diphosphate kinase, and pyruvate kinase. DNA pol beta K(d) values for the alpha,beta-CF(2) and unmodified dNTPs, alpha,beta-NH dUTP, and the alpha,beta-CH(2) analogues of dATP and dGTP are discussed in relation to the conformations of alpha,beta-CF(2) dTTP versus alpha,beta-NH dUTP bound into the enzyme active site.

RESUMEN

The structural and functional analogy between difluoromethylene bisphosphonate (CF2PP) and pyrophosphate (PPi) is investigated in a reaction with V(V) in the form of vanadate. The reaction of CF2PP with vanadate was investigated using 1.00 M KCl as supporting electrolyte over the ranges 3 < or = [CF2PP] < or = 60 mM and 2.06 < or = pH < or = 11.80. 51V, 19F, and 31P NMR spectroscopic studies showed that a 1:1 species was formed with an H+-dependent formation constant of 110 M-1 at pH 7.22. Results of solution experiments and ab initio calculations are consistent with CF2PP coordinating V(V) in a bidentate manner, as previously reported for PPi. Below pH 4, a minor complex forms, which is consistent with a 1:2 stoichiometry. This complex was also observed with pyrophosphate. The X-ray crystal structure of the monoprotonated difluoromethylene bisphosphonate anion (H[CF2PP]3-)-toludine complex is presented. The H[CF2PP]3- anion crystallized in the triclinic space group P with a = 12.7629(7) A, b = 13.3992(7) A, c = 17.1002(9) A, and V = 2584.4(2) A3, and Z = 2. Sheets of the layers of anions are connected through a network of H-bonds and separated by a layer of toludine cations. The structural features are investigated, and the CF2PP anion was found to be longer and wider than the corresponding PPi. Given the larger size of this anion compared to PPi, the chelation affinity upon CF2 substitution was found to be 4-5-fold reduced at neutral pH.

Cyanosilylation of aldehydes and aliphatic ketones can be carried out in dimethylformamide even without the use of any catalyst. In the presence of nucleophilic catalysts such as carbonate and phosphate salts, the reaction rate is significantly enhanced.

New cobalt-containing secondary phosphine oxides [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(4){mu,eta-PhC[triple chemical bond]CP(==O)(H)(R)}] (8 a: R=tBu; 8 b: R=Ph) were prepared by reaction of secondary phosphine oxides PhC[triple chemical bond]CP- (==O)(H)(R) (6 a: R=tBu; 6 b: R=Ph) with dppm-bridged dicobalt complex [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(6)] (2). The molecular structures of 8 a and 8 b were determined by single-crystal X-ray diffraction. Although palladium-catalyzed Heck reactions employing 8 b as ligand gave satisfying results, 8 a performed poorly in the same reaction. Judging from these results, a tautomeric equilibrium between 8 b and its isomeric form [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(4){mu,eta-PhC[triple chemical bond]CP(OH)(Ph)}] 8 b' indeed takes place, but it is unlikely between 8 a and [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(4){mu,eta-PhC[triple chemical bond]CP(OH)(tBu)}] (8 a'). The DFT studies demonstrated that reasonable activation energies for the tautomeric conversions can be achieved only via a bimolecular pathway. Since a tBu group is much larger than a Ph group, the conversion is presumably only feasible in the case of 8 bright harpoon over left harpoon8 b', but not in the case of 8 aright harpoon over left harpoon8 a'. Another cobalt-containing phosphine, namely, [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(4){mu,eta-PhC[triple chemical bond]CP(NEt(2))(tBu)}] (7 a), and its oxidation product [(mu-PPh(2)CH(2)PPh(2))Co(2)(CO)(4){mu,eta-PhC[triple chemical bond]CP(==O)(NEt(2))(tBu)}] 7 a' were prepared from the reaction of PhC[triple chemical bond]CP(NEt(2))(tBu) (5 a) with 2. The molecular structures of 7 a and 7 a' were determined by single-crystal X-ray diffraction. The phosphorus atom is surrounded by substituents in a tetrahedral environment. A P--N single bond (1.676(3) A) is observed in the molecular structure of 7 a. Heck reactions employing 7 a/Pd(OAc)(2) as catalyst system exhibited efficiency comparable to that of 8 a/Pd(OAc)(2).

Organofluorine compounds are becoming increasingly important in different fields, such as material science, agro chemistry, and the pharmaceutical industry. Nucleophilic trifluoromethylation is one of the widely used methods to incorporate a trifluoromethyl moiety into organic molecules. We have carried out extensive studies to develop varieties of easily accessible nucleophilic catalysts to promote such reactions. TMS-protected trifluoromethylated alcohols were prepared from both aldehydes and ketones in excellent yields using catalytic amount of amine N-oxide. Carbonate and phosphate salts also showed efficient catalytic activity toward this reaction. These reactions were highly solvent dependent, and DMF was found to be the most suitable one among the various solvents studied. All these reactions proceeded under very mild conditions, giving clean products and avoiding the use of any fluoride initiators or expensive catalysts, and extremely water-free conditions. The mechanism for the reaction is discussed in detail. DFT calculations were performed on the possible reaction intermediates using the Gaussian 03 program at B3LYP/6-311+G* level to support the proposed mechanism.

The structures of n-B18H22 and of n-B18H22 x C6H6 were determined by single-crystal X-ray analysis at -60 degrees C. The geometry of the boron cluster itself does not seem to be appreciably affected by solvation. There does, however, appear to be an unusual interaction of a polyborane bridging hydrogen atom with the benzene pi system, giving rise to an extended stacked structure. The 1H{11B} spectrum of n-B18H22 in [D6]benzene differs from that in [D12]cyclohexane most noticeably in the bridging proton region. Upon moving from the aliphatic to the aromatic solvent, the greatest increase in shielding was for the signal corresponding to the bridge hydrogen atom that interacts with the pi system of benzene; the signal was shifted upfield by 0.49 ppm. Density functional theory calculations were performed on 1:1 and 2:1 complexes of the n-B18H22 unit with benzene.

Two series of compounds, 9-X-1,7-(Me(2)S)(2)B(12)H(9) and 9,10-X(2)-1,7-(Me(2)S)(2)B(12)H(8) (X = Cl, Br, I), have been synthesized from reactions of 1,7-(Me(2)S)(2)B(12)H(10) with various halogenating reagents. In addition, reactions of 1,7-(Me(2)S)(2)B(12)H(10) with 2,4-(NO(2))(2)C(6)H(3)SCl and PhSeBr resulted in 9-(2',4'-(NO(2))(2)C(6)H(3)S)-1,7-(Me(2)S)(2)B(12)H(9) and 9,10-(PhSe)(2)-1,7-(Me(2)S)(2)B(12)H(8), respectively. X-ray studies of the dibromo, monoiodo, and aryl thioether derivatives show that electrophilic substitution in 1,7-(Me(2)S)(2)B(12)H(10) takes place at positions 9 and 10, as in the case of the meta-carborane 1,7-C(2)B(10)H(12). From 1,12-(Me(2)S)(2)B(12)H(10) the halides 2-X-1,12-(Me(2)S)(2)B(12)H(9) (X = Br, I) were prepared. For both 1,7- and 1,12-(Me(2)S)(2)B(12)H(10) the best iodination results were obtained using iodine monochloride in refluxing acetonitrile. In the presence of 5 mol % (PPh(3))(2)PdCl(2) the iodides 9-I-1,7-(Me(2)S)(2)B(12)H(9), 2-I-1,12-(Me(2)S)(2)B(12)H(9), and 9,10-I(2)-1,7-(Me(2)S)(2)B(12)H(8) react with RMgX (R = Me, Ph, Bn; X = Cl, Br) in THF to yield the corresponding B-alkyl- and B-aryl-substituted products in good yields without using CuI as a cocatalyst. The bromo derivative 9-Br-1,7-(Me(2)S)(2)B(12)H(9) did not react under similar conditions. No interference from the nearby Me(2)S substituent was observed in palladium-catalyzed substitution of iodide in 2-I-1,12-(Me(2)S)(2)B(12)H(9). Presumably due to the intramolecular activation of an aryl C-H bond of the benzyl substituent in the intermediate palladium complex, the yield of 9,10-Bn(2)-1,7-(Me(2)S)(2)B(12)H(8) was significantly lower than those of the dimethyl and diphenyl derivatives. The molecular structures of 9-R-1,7-(Me(2)S)(2)B(12)H(9) (R = Ph, Bn) and 2-Bn-1,12-(Me(2)S)(2)B(12)H(9) were obtained by single-crystal X-ray analysis.

A variety of S-alkylated products was prepared by alkylation of methyl thioethers [MeSB(12)H(11)](2-) (5), [1-(MeS)-2(7,12)-(Me(2)S)B(12)H(10)](-) (6-8), and [1,2(7,12)-(MeS)(2)B(12)H(10)](2-) (9-11) with alkyl halides and tosylates in acetonitrile. Since these methyl thioethers can be prepared easily in B-10-enriched form on a large scale and due to their chemical versatility, they are potentially very attractive boron entities for the design and synthesis of therapeutics for boron neutron capture therapy of cancer. It was found that alkylation of 6-8 can be complicated by an equilibrium which establishes between, on the one hand, one of the former species and, on the other hand, 1,2(7,12)-(Me(2)S)(2)B(12)H(10) (2-4) and [1,2(7,12)-(MeS)(2)B(12)H(10)](2-) (9-11). A boronated phosphonate 1-(MeS(CH(2))(4)P(O)(OEt)(2))-7-(Me(2)S)B(12)H(10) (14g) and a gem-bisphosphonate 1-(MeS(CH(2))(3)CH[P(O)(OEt)(2)](2))-7-(Me(2)S)B(12)H(10) (14h) were prepared from thioether 7 and the corresponding iodide and tosylate, respectively, and subsequently converted to their sodium salts. The propargyl sulfonium salts obtained by alkylation of thioethers 7, 8, 10, and 11 with propargyl bromide have been further converted to two- and three-cage oligomers containing both ortho-carborane and dodecaborane moieties. Methyl thioethers derived from closo-[B(12)H(12)](2-) are excellent participants in Michael addition reactions in the presence of a strong acid. The sulfonium salts with tertiary alkyl and vinyl substituents have been prepared by this method. Methyl thioethers 5-11 react with hydroxylamine-O-sulfonate yielding the corresponding aminosulfonium salts, albeit in lower yields as compared to those in the alkylation reactions. Several derivatives of methyl thioethers 5-11 have been characterized by single-crystal X-ray diffraction.
