RESUMEN

The binding of 125I-labeled lipoproteins to subcellular structures of rat hepatocytes was studied. The protein component of HDL, LDL, and VLDL was found in large and small membranes, nuclei, mitochondria, lysosomes, microsomes, and cell cytosol. The presence in liver nuclear chromatin of proteins immunochemically related to apoA-1 was demonstrated by solid phase immunoenzymatic analysis, dot immunoanalysis, and immunoelectroblotting. The apoprotein A-1 immunoreactivity was detected in transcriptionally inactive total chromatin, transcriptionally active chromatin, and nuclear matrix. The specific apoA-1 immunoreactivity of transcriptionally active chromatin and nuclear matrix was two times as low as that of total and transcriptionally inactive chromatin. Immunoelectroblotting revealed two proteins possessing an apoA-1 immunoreactivity (M(r) 28 and 14 kDa). The 28 kDa protein was found in transcriptionally active chromatin and nuclear matrix, while the 14 kDa one--only in transcriptionally inactive chromatin. the roles of apoproteins in lipid transport to nuclei and the maintenance of chromatin in a transcriptionally active state are discussed.

RESUMEN

The processing of the recombinant analogue of beta-lipotropin (beta-LHP) having 11 additional N-terminal amino acid residues and separated from the hormone by the processing signal, was investigated using rat adrenal secretory granule lysate as a test system of processing "in vitro". It was found that incubation of the beta-LPH analogue with secretory granule enzymes leads to its limited specific degradation with a release of native beta-endorphin. It is concluded that the additional N-terminal amino acids induced no qualitative changes in beta-LPH processing.

RESUMEN

A study of 70 patients with polyps and polyposis of the stomach indicates that the use of microwave electromagnetic radiation of low intensity associated with polypectomy increases significantly the treatment and improves the immediate and long-term results. Microwave resonance therapy possesses a regulating action, normalizing the bioenergy of blood cells and restoring the content of T-lymphocytes.

RESUMEN

The antitumour efficacy of a single cryosurgery use in combination with hyperthermia was studied in 165 mice with the Lewis carcinoma, sarcoma 180 and 169 rats with Guérin's carcinoma and sarcoma 45. A considerable decrease in the recurrence frequency was observed irrespective of the histological type of the primary tumour. The haemostasis-associated expressed hypoxia has been established to arise in tumour tissues after cryodestruction. A day after the cryodestruction a shift to alkaline pH in the tumour tissue has been observed along with hypoxia. The above conditions possibly specify the high antitumour efficacy of a combined use of the cryosurgery and hyperthermia.

Using the analytic microscope "Parmoquant-2" (GDR), histograms were obtained demonstrating electrophoretic mobility (EPM) of lymphoid cells of C57BL/6 mice in the course of growth of the Lewis carcinoma (3LL) and melanoma B16 administered under the skin of the femur. Changes in the average values of EPM of thymic, splenic and lymph nodal cells in the process of tumor growth appeared similar. It is shown that the medium thymocyte EMP is growing towards the terminal stage of tumor growth, at the expense of the decrease in the share of PNA+ cells. Splenic cell bimodal distribution according to EPM became, in the course of tumor growth in intact mice, unimodal with some insignificant decrease in the median EPM values. The median EPM of regional and distant lymph nodes in the process of tumor growth is of phase character. It is supposed that investigation of lymph node EPM could be used for studying tumor growth kinetics.

RESUMEN

Antitumour efficacy of a combined use of cryosurgery and solcoseryl in mice with Lewis carcinoma was studied. Solcoseryl improves results of cryosurgery. An increase in antitumour effect with a combined use of cryodestruction and solcoseryl is supposed to be followed by the rise of oxygen tension in tissues. Solcoseryl injection followed by cryosurgery of cancer of mouth cavity mucosa in patients considerably accelerates the wound surface regeneration. Thus solcoseryl increases the treatment efficacy of the cryosurgical method.

RESUMEN

Antitumor efficacy of cryosurgery and standard surgical procedures for Lewis carcinoma, melanoma B16, Guerin's carcinoma and Walker's carcinosarcoma was compared in the treatment of 846 experimental animals. The study established the superiority or, at least, similar efficacy of cryosurgery in terms of cure, survival and radicality of primary tumor treatment. Unlike standard surgery, cryosurgery was followed by distant metastases being inhibited in mice. Although there was no significant difference in the incidence of metastases to regional lymph nodes in rats, it was higher than in untreated animals in both treatment groups. Cryonecrosis of tumor tissue is a factor of distant and regional metastases development following cryosurgery.

RESUMEN

Using a differential non-detergent phenol-salt deproteinization technique, the DNA fractions corresponding to functionally different parts of the genome, i.e., DNA I (transcriptionally active DNA), DNA II (stably repressed genome DNA), and DNA III (potentially active genome DNA) were obtained. Under conditions of cortisol-induced activation of transcription in rat liver cells, the number or repetitive sequences in DNA I (represented by DNA III in intact animals) increased. In this way, the hormone-induced transcription appeared to be associated with a transition to a transcriptionally active state of chromatin containing repetitive sequences that exist in intact liver in potentially active parts of the genome. It was shown that these inducible repetitive sequences contain binding sites for glucocorticoid-receptor complexes. In DNA III, these sites are present in large amounts in the sequences within the composition of stable DNA-protein complexes, which suggests the specificity of DNA sequences firmly bound to chromatin proteins.

RESUMEN

The influence of cryodestruction and surgical treatment of the primary tumour in rats with the transplanted Guerin's carcinoma was studied including their effect on the degree of metastatic spreading to the regional lymph nodes. The rate of regional metastases under the cryotherapy influence is shown to be somewhat higher than resulting from surgical removal of a tumor and is positively lower than in the case of amputation of tumor affected extremity.

RESUMEN

The effect of the primary tumour cryodestruction on a degree of metastatic spreading in the lungs was investigated in experiments with C57B1/6 mice with Lewis carcinoma of the lungs. It is found that after the tumour cryosurgery there was a less quantity of metastases as compared with operated animals (surgical removal of the tumour or amputation of the limb with the tumour) or with untreated animals. A regular correlation is observed between the degree of metastatic spreading and the efficiency of the primary tumour treatment. The relapse after cryosurgery or surgical removal of the tumour led to an increase in the quantity of pulmonary metastases.

The m5C content was determined in three functionally different fractions of rat liver DNA isolated by a modified phenol fractionation. The transcriptionally active DNA-I contains up to 12-14% of hybrid RNA not hydrolyzed by RNAase, is enriched with unique sequences and makes up to about 20% of total DNA. In its total nucleotide content (43 mol.% of GC-pairs) DNA-I does not differ from the major fraction of DNA-II making up to about 70% of the genome, and from hardly extractable DNA-III, whose content in total DNA does not exceed 10%. However, the degree of methylation of the transcriptionally active DNA-I (1.28 mol.% of m5C) is higher than that of DNA-II and DNA-III (1.0 mol.% of m5C). Under cortisol-induced activation of transcription the number of repeating sequences in DNA-I is increased by about 10% concomitant with a decrease of these sequences in DNA-III. The hormone-induced changes in the transcriptionally active fraction of DNA are reversible. 4 hrs after cortisol injection a demethylation of highly repeating and supermethylation of normally repeating sequences in DNA-I are observed. The level of methylation of the unique sequences in DNA-I (Cot greater than 200) remains unchanged. The degree of methylation of various subfractions of nucleotide sequences of DNA-II and DNA-III afer cortisol injection does not differ from normal. Presumably the hormone-induced supermethylation of normally repeating sequences may control the transcription.
