RESUMEN

AIMS: Citrus canker caused by Xanthomonas citri subsp. citri (X. citri) is a disease of economic importance. Control of this disease includes the use of metallic copper, which is harmful to the environment and human health. Previous studies showed that the crude extract from the fungus Pseudogymnoascus sp. LAMAI 2784 isolated from Antarctic soil had in vitro antibacterial action against X. citri. The aim of the present study was to expand the applications of this extract. METHODS AND RESULTS: In greenhouse assays, the crude extract was able to reduce bacterial infection on citrus leaves from 1.55 lesions/cm2 (untreated plants) to 0.04 lesions/cm2. Bisdechlorogeodin was identified as the main compound of the bioactive fraction produced by Pseudogymnoascus sp. LAMAI 2784, which inhibited bacterial growth in vitro (IC90 ≈ 156 µg ml-1) and permeated 80% of X. citri cells, indicating that the membrane is the primary target. CONCLUSION: The present results showed that the bioactive fraction of the extract is mainly composed of the compound bisdechlorogeodin, which is likely responsible for the biological activity against X. citri, and the main mechanism of action is the targeting of the cell membrane. This study indicates that bisdechlorogeodin has valuable potential for the control of X. citri.

INTRODUCTION: It has been hypothesized that cognitive and memory-related brain function in transgender during cross-sex hormonal treatment might be activated towards that of the subjective gender. However, research on this topic has produced inconsistent results, and to the best of our knowledge no studies have investigated neurocognitive changes in androgen-treated female-to-male (FM) transgender adolescents. SUBJECTS AND METHODS: A total of 15 FM transgender adolescents (14-17 years) underwent neuropsychological testing in order to examine the effects of androgen on visuo-spacial abilities, verbal memory language, processing speed and executive functions. We used a longitudinal design in which 10 participants were tested twice, before and after receiving 12 months of testosterone treatment. This group was also compared with 5 FM transgender adolescents off-androgen treatment. RESULTS: Participants tested before and after 12 months of androgen treatment improved significantly on processing speed in a visuo-spatial (Rey-Osterrieth complex figure test) and in a visuo-oral task (Stroop), their performance on a verbal memory task (TAVEC) and on interference (Stroop) and they exhibited lower impulsivity control (CARAS-R). On-androgen treatment adolescents exhibited worse cognitive impulsivity control than off-androgen treatment adolescents. CONCLUSIONS: The results indicate that androgen has an influence on immediate verbal memory, cognitive interference, impulsivity control and processing speed.


TITLE: Efectos del tratamiento con andrógenos sobre la neurocognición en adolescentes transgénero de mujer a hombre.Introducción. Se ha planteado la hipótesis de que la neurocognición en personas transgénero durante el tratamiento hormonal cruzado podría aproximarse a la del género subjetivo. Sin embargo, la investigación sobre este tema ha producido resultados inconsistentes y, hasta donde sabemos, ningún estudio ha investigado los cambios neurocognitivos en adolescentes transgénero de mujer a hombre (FM) tratados con andrógenos. Sujetos y métodos. Quince adolescentes transgénero FM (14-17 años) se sometieron a pruebas neuropsicológicas para examinar los efectos de los andrógenos en sus habilidades visuoespaciales, memoria verbal, velocidad de procesamiento y funciones ejecutivas. Utilizamos un diseño longitudinal en el que se evaluó a 10 participantes dos veces, antes y después de recibir, durante 12 meses, tratamiento con testosterona. Este grupo también se comparó con cinco adolescentes transgénero FM sin tratamiento con andrógenos. Resultados. Los participantes evaluados antes y después de 12 meses de tratamiento con andrógenos mejoraron significativamente en velocidad de procesamiento en una tarea visuoespacial (prueba de la figura compleja de Rey-Osterrieth) y en una tarea visual (Stroop), en una tarea de memoria verbal (test de aprendizaje verbal España-Complutense) y en interferencia (Stroop), y exhibieron un menor control de la impulsividad (test de percepción de diferencias revisado). Los adolescentes que recibieron tratamiento con andrógenos mostraron un peor control de la impulsividad cognitiva que los adolescentes que no recibieron tratamiento con andrógenos. Conclusiones. Los resultados indican que los andrógenos influyen en la memoria verbal, la interferencia cognitiva, el control de la impulsividad y la velocidad de procesamiento.

RESUMEN

The purpose of this article was to describe a simple graphical driven method for determining the necessary two-vector mechanics (TVM) as an alternative solution to the ideal or single force. In orthodontics, some tooth movements are difficult to achieve, mainly because of the impossibility of getting the point of application for the necessary force system. Given two points in biomechanically accessible areas, adding together two vectors applied to those points will result in an equivalent force system. The method was developed based on previous research, 3,880 hypothetical cases were analyzed to determine the trends between the resulting forces and their relative locations. A graphical driven TVM method (GTVM) for establishing the necessary TVM is presented in a manner that combine different procedures from the traditional method that simplify the analysis and may allow the user to perform more complex orthodontic cases making easier to compare systems of force equivalents.

Harsh and extreme environments, such as Antarctica, offer unique opportunities to explore new microbial taxa and biomolecules. Given the limited knowledge on microbial diversity, this study aimed to compile, analyze and compare a subset of the biobank of Antarctic fungi maintained at the UNESP's Central of Microbial Resources (CRM-UNESP). A total of 711 isolates (240 yeasts and 471 filamentous fungi) from marine and terrestrial samples collected at King George Island (South Shetland Islands, Antarctica) were used with the primary objective of investigating their presence in both marine and terrestrial environments. Among the yeasts, 13 genera were found, predominantly belonging to the phylum Basidiomycota. Among the filamentous fungi, 34 genera were represented, predominantly from the phylum Ascomycota. The most abundant genera in the marine samples were Metschnikowia, Mrakia, and Pseudogymnoascus, while in the terrestrial samples, they were Pseudogymnoascus, Leucosporidium, and Mortierella. Most of the genera and species of the CRM-UNESP biobank of Antarctic fungi are being reported as an important target for biotechnological applications. This study showed the relevance of the CRM-UNESP biobank, highlighting the importance of applying standard methods for the preservation of the biological material and associated data (BMaD), as recommended in national and international standards.

Stress experienced during ontogeny can have profound effects on the adult phenotype. However, stress can also be experienced intergenerationally, where an offspring's phenotype can be moulded by stress experienced by the parents. Although early-life and intergenerational stress can alter anatomy, physiology, and behaviour, nothing is known about how these stress contexts interact to affect the neural phenotype. Here, we examined how early-life and intergenerational stress affect the brain in eastern fence lizards (Sceloporus undulatus). Some lizard populations co-occur with predatory fire ants, and stress from fire ant attacks exerts intergenerational physiological and behavioural changes in lizards. However, it is unclear if intergenerational stress, or the interaction between intergenerational and early-life stress, modulates the brain. To test this, we captured gravid females from fire ant invaded and uninvaded populations, and subjected offspring to three early-life stress treatments: (1) fire ant attack, (2) corticosterone, or (3) a control. Corticosterone and fire ant attack decreased some aspects of the neural phenotype while population of origin and the interaction of early-life stress and population had no effects on the brain. These results suggest that early-life stressors may better predict adult brain variation than intergenerational stress in this species.

PURPOSE: To explore the experiences of uncertainty in the clinical reasoning of nurses in the postanesthesia care unit (PACU). DESIGN: A phenomenological descriptive design, following Colaizzi's analysis. METHODS: Semistructured interviews were conducted with 14 nurses from a PACU on their experience of uncertainty in clinical reasoning. The interviews were digitally audio-recorded and transcribed verbatim. Two researchers conducted data analysis independently and followed seven phases: (re)reading the transcripts, extracting significant statements, formulating meanings from significant statements, aggregating formulated meanings into themes, developing a description of the phenomenon's essential structure, generating of the fundamental structure of the phenomenon, validating of the findings through participant feedback. The process employed MAXQDA analytics Pro 2022 software. Consolidated Criteria for Reporting A Qualitative Research checklist was used for reporting. FINDINGS: From uncertainty experiences in nurses' clinical reasoning, 10 themes emerged: ambiguity and decision latitude, communication, work ethic, difficulty interpreting and predicting outcomes, cognitive performance impairment, incivility, core competence vagueness of postanesthesia nurses, high-tech care, (in)security and risk, and occupational stress. CONCLUSIONS: The experiences of uncertainty in clinical reasoning of nurses in postanesthesia care units are highly focused on patient safety. Exploring these experiences has made uncertainty more tangible and explicit, which will enable nurses in postanesthesia care units to prepare for adaptive responses to deal with uncertainty when it occurs in clinical practice.

Khellin and visnagin furanochromones were recently reported as potential new bioherbicides with phytotoxic activities comparable to those of some commercially available herbicides. In this study, we examined the effect of O-alkylation and O-arylalkylation of both khellin and visnagin on its effect on herbicidal and antifungal activity. Synthetic analogues included O-demethyl khellin and visnagin, acetylated O-demethyl khellin and visnagin, O-benzylated demethyl khellin and visnagin, four O-demethyl alkylated khellin analogues, and six O-demethyl alkylated visnagin analogues, many of which are reported here for the first time. Both acetate analogues of khellin and visnagin indicated more activity as herbicides on Lemna pausicostata than visnagin, with IC50 values of 71.7 and 77.6 µM, respectively. Complete loss of activity for all O-alkyl analogues with a carbon chain length of greater than 14 carbons was observed. The O-demethyl butylated visnagin analogue was the most active compound with an IC50 of 47.2 µM against L. pausicostata. O-Demethyl ethylated analogues of both khellin and visnagin were as effective as khellin. In the antifungal bioautography bioassay against Colletotrichum fragariae at 100 µg, the only active O-alkyl and O-arylalkyl analogues were O-ethylated, O-butylated, and O-benzylated visnagin analogues with zones of inhibition of 10, 9, and 9 mm, respectively, an effect comparable to that of visnagin and khellin.

The aim of this study was to evaluate the stress distribution in cortical and cancellous bones and within the screw when a stainless steel extra-alveolar screw (EA-S) is inserted into the retromolar region of the mandible using the finite element method (FEM). The stress values in the EA-S were highest at 60° with horizontal (shear) force and lowest at 90° with axial force. In the bones (cortical and cancellous), the stress value was greatest at 90° with horizontal force. Greater stability of the screw may be achieved by tilting it to the force when horizontal loads are applied.

OBJECTIVES: Rituximab (RTX) is a safe and effective treatment for rheumatoid arthritis (RA). However, there are some concerns about infection risk and preliminary data suggest dose and time dependency. This study aims to determine the infection incidence in a large real-life population of RA patients using RTX, with special focus on (ultra-)low dosing and time since last infusion. METHODS: RA patients treated with 1000, 500 or 200 mg RTX per cycle between 2012 and 2021 at the Sint Maartenskliniek were included in a retrospective cohort study. Patient-, disease-, treatment- and infection characteristics were retrieved from electronic health records. Infection incidence rates, dose and time relations with RTX infusion were analysed using mixed-effects Poisson regression. RESULTS: Among 490 patients, we identified 819 infections in 1254 patient years. Most infections were mild and respiratory tract infections were most common. Infection incidence rates were 41, 54 and 71 per 100 patient years for doses of 200, 500 and 1000 mg. Incidence rate ratio (IRR) was significantly lower for 200 mg compared to 1000 mg (adjusted IRR 0.35, 95% CI 0.17-0.72, p = 0.004). In patients receiving 1000 or 500 mg RTX, infections seemed to occur more frequently within the first two months after infusion compared to later on in the treatment cycle, suggesting an association with peak concentration. CONCLUSION: Ultra-low dosing (200 mg) of RTX is associated with a lower risk of infections in RA. Future interventions focusing on ultra-low dosing and slow release of RTX (e.g. by subcutaneous administration) may lower infection risk.

The demand for fluorescent organic dyes across a broad range of applications has led to investigation into tuneable emission dyes. The tuneable nature of these dyes makes them desirable for applications in a variety of fields, including organic light-emitting diodes (OLEDs), optical sensing devices, and fluorescence imaging. In recent investigations, there have only been a handful of mechanisms used to tune emission. Herein, we present four novel perylene-acene dyads that undergo solvent tuneable emission, and propose a novel mechanism for this tuneability based on the presence of a charge transfer state. These dyes were shown to reach photoluminescence quantum efficiencies (PLQEs) as high as 45%, depending on the solvent, showing the ability for this mechanism to be used to access higher PLQE tuneable emission.

RESUMEN

Professional societies play a unique role in our personal and professional lives as spaces for connection and as regulatory entities. Often composed of volunteers from a variety of backgrounds, professional societies can guide the cultural norms and expectations of an industry by acting as critical leaders for supporting diversity, equity, and inclusion initiatives. The present manuscript explains how professional societies, like the Organization of Biological Field Stations (OBFS), can embrace transdisciplinary approaches to create more inclusive (in)tangible spaces to address serious problems facing industries today. Climate change, sexism, and racism, are examples of "wicked" problems that cannot be solved using existing modes of inquiry and decision making because of their complex and interrelated nature. As members of OBFS, a professional society dedicated to the advancement of field-based research, we explain how transdisciplinarity can and has been used to begin addressing serious issues like racism and sexism in field-based research and provide steps for future professional societies to engage in transdisciplinary thinking. We close with examples of our own transdisciplinary work developed through our membership within OBFS.

Luminescent solar concentrators (LSCs) concentrate light via luminescence within a planar-waveguide and have potential use for building-integrated photovoltaics. However, their commercialization and potential applications are currently hindered greatly by photon reabsorption, where emitted waveguided light is parasitically reabsorbed by a luminophore. Nanotetrapod semiconductor materials have been theorized to be excellent luminophores for LSCs owing to their inherently large Stokes shifts. Here we present the first nanotetrapod-based LSCs (5 × 5 × 0.3 cm3) reported in the literature. External quantum efficiencies as high as 4.9 ± 0.5% were achieved under AM1.5G conditions. We also perform an in-depth investigation by optical characterization of the different operational metrics of our nanotetrapod-based LSCs and show reabsorption to be eliminated (mean number of average reabsorption events per photon equal to 0.00) in our most extended nanotetrapod devices.

High Content Screening (HCS) combines high throughput techniques with the ability to generate cellular images of biological systems. The objective of this work is to evaluate the performance of predictive models using CNN to identify the number of cells present in digital contrast microscopy images obtained by HCS. One way to evaluate the algorithm was through the Mean Squared Error metric. The MSE was 4,335.99 in the A549 cell line, 25,295.23 in the Huh7 and 36,897.03 in the 3T3. After obtaining these values, different parameters of the models were changed to verify how they behave. By reducing the number of images, the MSE increased considerably, with the A549 cell line changing to 49,973.52, Huh7 to 79,473.88 and 3T3 to 52,977.05. Correlation analyzes were performed for the different models. In lineage A549, the best model showed a positive correlation with R = 0.953. In Huh7, the best correlation of the model was R = 0.821, it was also a positive correlation. In 3T3, the models showed no correlation, with the best model having R = 0.100. The models performed well in quantifying the number of cells, and the number and quality of the images interfered with this predictive ability.

The papilionoid legume genus Sophora (Fabaceae) exhibits a worldwide distribution, but a phylogenetic framework to understand the evolution of this group is lacking to date. Previous studies have demonstrated that Sophora is not monophyletic and might include Ammodendron, Ammothamnus, and Echinosophora, but the relationships among these four genera (defined as Sophora s.l.) are unclear. Here we used a nuclear DNA dataset (ETS, ITS, SQD1) and a plastid DNA dataset (matK, rbcL, rpl32-trnL, trnL-F) of 654 accession sequences to reconstruct the phylogenetic relationships, estimate the divergence times and ancestral range of Sophora s.l., and infer the evolution of chromosome number and morphological characteristics. Our major aim was to reconstruct phylogenetic relationships to test monophyly and elucidate relationships within the genus. Our results indicated that Ammodendron, Ammothamnus, and Echinosophora are embedded within Sophora s.s. and that nine well-supported clades can be recognized within comprise Sophora s.l. Ancestral character state estimation revealed that the most recent common ancestor of Sophora s.l. was a deciduous shrub that lacks rhizome spines and has unwinged legumes. Divergence times estimation and ancestral area reconstruction showed that Sophora s.l. originated in Central Asia and/or adjacent Southeast China in the early Oligocene (ca. 31 Mya) and dispersed from these regions into East and South Asia's adjacent areas and North America via the Bering land bridge. The analyses also supported a South American origin for S. sect. Edwardsia, which experienced rapid radiation with its major lineages diversifying over a relatively narrow timescale (8 Mya).

BACKGROUND: Several studies have demonstrated the high frequency of medication errors in pediatric patients by prehospital providers during both patient care and simulation. In 2015, our hospital-based urban EMS system introduced the HandtevyTM Field Guide that provides precalculated pediatric doses in milliliters (mL) by patient age. We hypothesized that implementation of the Field Guide would increase the percentage of correct pediatric medication doses to greater than 85%. METHODS: We performed a single center retrospective cohort study of medications administered to patients < 13 years of age from August 2017 to July 2019 compared to 2014 baseline data through electronic medical record review. We excluded nebulized medications and online medication direction cases. Our primary outcome was the percentage of correct doses defined as a dose within 80-120% of the Field Guide dose recommendation. Each dosing error was reviewed by two investigators. RESULTS: We analyzed 483 drug administrations in 375 patients for the Field Guide study period. Doses were correct in 89.4% of medication administrations with 68.5% reportedly administered exactly as dictated by the Field Guide compared to 51.1% in the baseline period (p < 0.001). During the Field Guide study period, the following medications had 100% appropriate dosing: adenosine, dextrose 10%, diphenhydramine, epinephrine 1:10,000, glucagon, naloxone and oral ondansetron. Overdoses accounted for 4.4% of medication errors and underdoses accounted for 6.2% of medications errors. The most overdosed medications were intranasal (IN) midazolam (11.8%) and intravenous fentanyl (9.4%). The most underdosed medications were IN midazolam (23.5%) and intramuscular epinephrine 1:1000 (12.5%). The highest percentage of errors (20%) were seen in the zero to one-year-old age group. CONCLUSION: After implementation of a precalculated mL dose system by patient age for EMS providers, most pediatric medications were reportedly administered within the appropriate dose range. A field guide with precalculated doses (in mL) may be an effective tool for reducing pediatric medication dosing errors by EMS providers.

The dissolved oxygen (DO) level in the hypolimnion of lakes and reservoirs can reach anoxic conditions, which favor the release of phosphorus from the sediment bed to the water column. However, to estimate nutrient release from sediment is extremely important to quantify the duration of anoxia. In low latitude regions, the water-sediment layer is warmer than in temperate regions and eutrophication is usually more severe, potentially accelerating oxygen depletion and extending the anoxia period. Considering that the available equations to quantify the duration of anoxia were developed for temperate lakes, there is a need to effectively quantify this period in lakes and reservoirs located in other climate regions, such as the semiarid. In this study, the dynamics of thermal stratification was analyzed as a function of the Relative Water Column Stability coefficient (RWCS) and then correlated with DO dynamics for nineteen tropical semiarid reservoirs. RWCS values were higher during the rainy season, when anoxia duration was longer and the hypolimnion was thicker with respect to total water depth. Then, two new equations for quantification of anoxia duration, based on the equation originally developed for temperate climate, were adapted for the wet and dry seasons of the tropical semiarid region. The results showed that the proposed equations presented a better performance compared to the original one, which tends to underestimate anoxia in tropical semiarid reservoirs. This work intended to provide simple and locally adjusted tools to better quantify anoxic events and support the water quality and internal phosphorus load modeling for tropical semiarid reservoirs.

Probing for spatial cognitive processes in model rodent species has a long history in the psychological literature, with well-established protocols and paradigms successfully revealing the mechanisms underlying spatial learning and memory. There has also been much interest in examining the ecological and evolutionary context of spatial cognition, with a focus on how selection has molded spatial cognitive abilities in nonmodel species, how spatial cognitive traits vary across species, the neural mechanisms underlying spatial cognitive abilities, and the fitness outcomes of spatial cognition. Behavioral ecologists have been able to take advantage of paradigms from experimental psychology's rich history of spatial cognitive testing for use in nonmodel species. However, as the field advances, it is important to highlight noncognitive factors that can impact performance on spatial cognitive tasks (e.g., motivation to perform the task, switching navigational strategies, variation across protocols, ecological relevance of the task), as these factors may explain discrepancies in findings among some studies. This review highlights how these noncognitive factors can differentially modulate performance on spatial cognitive tests in different nonmodel species. Accounting for these factors when creating protocols and paradigms allows for a more nuanced approach with more explanatory power when probing for spatial cognitive abilities in nonmodel species. Women's roles in the US military have progressively changed over the past several decades. Previously women were barred from combat roles. Recent change in policy allow women into combat roles in the Marine Corps, and this has led to women being trained for combat specialties. Objectives: This observational cross-sectional study describes body composition and performance values for modern Marine Corps women. Methods: Volunteers were 736 Marine women who were assessed for body composition and physical performance; (age 29.5±7.3 (18-56) years; height 163.6±6.8 (131.0-186.1) cm; body mass 68.3±9.2 (42.0-105.3) kg; years in the military 8.9±6.8 (0.5-37) years-in-service). Body composition measures were obtained using dual-energy X-ray absorptiometry and single-frequency bioelectrical impedance analyses. Performance measures were obtained from official physical and combat fitness test scores (PFT; CFT) as well as from data on measured countermovement jumps (CMJ) on a calibrated force platform. Results: Mean body composition metrics for Marine women were: 47.5±5.7 fat free mass (FFM) (kg), 30.1%±6.4% body fat (%BF), 2.6±0.3 bone mineral content (kg), and 25.5±2.8 body mass index (kg/m2); performance metrics included 43.4±3.2 maximal oxygen uptake (VO2max; mL.kg.min), 22.4±7.1 CMJ height (cm) and 2575±565.2 CMJ peak power (W). Data showed strong correlations (r) (≥0.70) between PFT and VO2max scores (0.75), and moderate correlations (≥0.50) between CFT and VO2max scores (0.57), CFT and PFT scores (0.60), FFM and CMJ peak power (W) (0.68), and %BF to VO2max (-0.52), PFT (-0.54), CMJ-Ht (-0.52) and CMJ relative power (W/kg) (-0.54). Conclusion: Modern Marine women are both lean and physically high performing. Body composition is a poor predictor of general physical performance.

The magnetic properties and thermodynamical description of the Fe-Mn-Al ternary alloy are studied using the spin 1 antiferromagnetic Blume-Capel (BC) model by the pair approximation based on the Gibbs-Feymann-Bogoliubov inequality for the free energy. The values of the spin operator are +1 for ferromagnetic interaction (Fe-Fe), -1 for the antiferromagnetic one (Mn-Mn, Mn-Fe), and 0 for the magnetic diluter (corresponding to Al). The BC model with antiferromagnetic (AF) next-nearest-neighbor coupling better accurately fit the mean hyperfine field experimental data obtained by Mössbauer spectroscopy. Considering the crystalline field, the predicted temperature as a function of the Al concentration phase diagram, for the fcc lattice, from the numerical solution is remarkably good and significantly improves the traditional Ising and random-site Ising models.
