RESUMEN

Healthy sleep of sufficient duration preserves mood and disturbed sleep is a risk factor for a range of psychiatric disorders. As adults commonly experience chronic sleep restriction (SR), an enhanced understanding of the dynamic relationship between sleep and mood is needed, including whether susceptibility to SR-induced mood disturbance differs between sexes. To address these gaps, data from N = 221 healthy adults who completed one of the two multi-day laboratory studies with identical 9-day SR protocols were analyzed. Participants randomized to the SR (n = 205) condition underwent 5 nights of SR to 4 hours of time-in-bed and were then randomized to one of the seven sleep doses that ranged from 0 to 12 hours in 2 hours increments; participants randomized to the control (n = 16) condition received 10 hours time-in-bed on all study nights. The Profile of Mood States (POMS) was used to assess mood every 2 hours during wakefulness and markers of sleep homeostasis (EEG slow-wave activity (SWA)) were derived via polysomnography. Mood progressively deteriorated across SR with marked disturbances in somatic mood components. Altered sleep physiology contributed to mood disturbance whereby increased EEG SWA was associated with increased POMS Total Mood Disturbance scores, a finding specific to males. The mood was restored in a dose-response fashion where improvements were greater with longer sleep doses. These findings suggest that when lifestyle and environmental factors are inhibited in the laboratory, the affective consequences of chronic sleep loss are primarily somatic mood disturbances. Altered sleep homeostasis may contribute to mood disturbance, yet sleep-dependent mechanisms may be sex-specific.

Sleep plays an important role in memory processing and is disrupted in individuals with post-traumatic stress disorder (PTSD). A growing body of research has experimentally investigated how sleep - or lack thereof - in the early aftermath of a traumatic experience contributes to intrusive memory formation. The aim of this meta-analytic review was to examine the effects of various experimental sleep manipulations (e.g., sleep deprivation, daytime naps) on intrusive memories following exposure to an experimentally induced analogue traumatic event. Eight eligible studies were systematically identified through PsycInfo and PubMed and provided sufficient data to contribute to a meta-analysis of the effects of sleep versus wakefulness on intrusive memory frequency. Sleep was found to reduce intrusive memory frequency when compared to wakefulness at a small but significant effect size (Hedge's g = 0.29). There was no evidence of publication bias and heterogeneity of effect sizes across studies was moderate. Results suggest that sleep plays a protective role in the aftermath of exposure to a traumatic event with implications for early post-trauma intervention efforts.

STUDY OBJECTIVES: Converging evidence from neuroimaging, sleep, and genetic studies suggest that dysregulation of thalamocortical interactions mediated by the thalamic reticular nucleus (TRN) contribute to autism spectrum disorder (ASD). Sleep spindles assay TRN function, and their coordination with cortical slow oscillations (SOs) indexes thalamocortical communication. These oscillations mediate memory consolidation during sleep. In the present study, we comprehensively characterized spindles and their coordination with SOs in relation to memory and age in children with ASD. METHODS: Nineteen children and adolescents with ASD, without intellectual disability, and 18 typically developing (TD) peers, aged 9-17, completed a home polysomnography study with testing on a spatial memory task before and after sleep. Spindles, SOs, and their coordination were characterized during stages 2 (N2) and 3 (N3) non-rapid eye movement sleep. RESULTS: ASD participants showed disrupted SO-spindle coordination during N2 sleep. Spindles peaked later in SO upstates and their timing was less consistent. They also showed a spindle density (#/min) deficit during N3 sleep. Both groups showed significant sleep-dependent memory consolidation, but their relations with spindle density differed. While TD participants showed the expected positive correlations, ASD participants showed the opposite. CONCLUSIONS: The disrupted SO-spindle coordination and spindle deficit provide further evidence of abnormal thalamocortical interactions and TRN dysfunction in ASD. The inverse relations of spindle density with memory suggest a different function for spindles in ASD than TD. We propose that abnormal sleep oscillations reflect genetically mediated disruptions of TRN-dependent thalamocortical circuit development that contribute to the manifestations of ASD and are potentially treatable.

Interindividual differences in the neurobehavioral response to sleep loss are largely unexplained and phenotypic in nature. Numerous factors have been examined as predictors of differential response to sleep loss, but none have yielded a comprehensive view of the phenomenon. The present study examines the impact of baseline factors, habitual sleep-wake patterns, and homeostatic response to sleep loss on accrued deficits in psychomotor vigilance during chronic partial sleep restriction (SR), in a total of 306 healthy adults that participated in one of three independent laboratory studies. Findings indicate no significant impact of personality, academic intelligence, subjective reports of chronotype, sleepiness and fatigue, performance on working memory, and demographic factors such as sex, ethnicity, and body mass index, on neurobehavioral vulnerability to the negative effects of sleep loss. Only superior baseline performance on the psychomotor vigilance test and ability to sustain wakefulness on the maintenance of wakefulness test were associated with relative resilience to decrements in vigilant attention during SR. Interindividual differences in vulnerability to the effects of sleep loss were not accounted for by prior sleep history, habitual sleep patterns outside of the laboratory, baseline sleep architecture, or homeostatic sleep response during chronic partial SR. A recent theoretical model proposed that sleep-wake modulation may be influenced by competing internal and external demands which may promote wakefulness despite homeostatic and circadian signals for sleep under the right circumstances. Further research is warranted to examine the possibility of interindividual differences in the ability to prioritize external demands for wakefulness in the face of mounting pressure to sleep.

Eye-tracking studies on social attention have consistently shown that humans prefer to attend to other human beings. Much less is known about whether a similar preference is also evident in covert attentional processes. To enable a direct comparison, this study examined covert and overt attentional guidance within two different experimental setups using complex naturalistic scenes instead of isolated single features. In the first experiment, a modified version of the dot-probe paradigm served as a measure of covert reflexive attention toward briefly presented scenes containing a social feature in one half of the visual field compared to nonsocial elements in the other while controlling for low-level visual saliency. Participants showed a stable congruency effect with faster reaction times and fewer errors for probes presented on the social side of the scene. In a second experiment, we tracked eye movements for the same set of stimuli while manipulating the presentation time to allow for differentiating reflexive and more sustained aspects of overt attention. Supportive of the first results, analyses revealed a robust preference for social features concerning initial saccade direction as well as fixation allocation. Collectively, these experiments imply preferential processing of social features over visually salient aspects for automatic allocation of covert as well as overt attention.
