RESUMEN

T follicular helper (Tfh) cells help direct the production of antibodies by B cells. In addition to promoting antibody responses to vaccination and infection, Tfh cells have been found to mediate antibody production to food antigens. Work over the past decade has delineated the specific phenotypes of Tfh cells that induce antibodies to food while also clarifying the divergent Tfh cell requirement for different food-specific antibody isotypes. Furthermore, Tfh and antibody responses to food can occur at multiple barrier sites - namely, skin, airway, and gut. Depending on the context of food antigen exposure, the immune response to food at these sites can be protective, as in the case of tolerance or immunotherapy, or pathogenic, as in the case of allergy. This review will highlight recent advances in our understanding of how Tfh cells promote antibodies to food as well as future avenues for continued discovery.

BACKGROUND: Vertebroplasty (VP) and balloon kyphoplasty (KP) are effective means with which to improve pain and function in osteoporotic vertebral compression fractures. However, the risk of complications after these procedures is poorly understood, with concerns regarding adjacent vertebral fractures. This study retrospectively investigated the clinical and radiological outcomes of these procedures. METHODS: A total of 115 patients who experienced their first vertebral fracture were treated with VP (N=63) or KP (N=52) at the Dankook University Hospital between January 2013 and December 2022. The clinical outcomes were evaluated using the visual analog scale (VAS) preoperative and at 1-year follow-up. Radiological comparisons were performed for kyphosis correction, vertebral height restoration, and postoperative cement leakage. RESULTS: KP was more effective than VP, especially for vertebral body height restoration and kyphotic angle reduction (P<0.05). However, the incidence of cement leakage, new adjacent vertebral fractures, and improvement in pain assessed by VAS did not differ statistically between the 2 groups (P>0.05). CONCLUSIONS: Considering that KP was performed on fractures with severe deformity, no differences were observed in the clinical outcomes and incidence of adjacent vertebral fractures compared Considering that KP was performed for fractures with severe deformity, there was no difference in clinical outcomes and incidence of adjacent vertebral fractures compared to VP. Improvements in radiological measurements were demonstrated. Therefore, KP may be a good treatment option for pain relief and long-term prognosis in patients with high-compressive-rate vertebral fractures.

The development of efficient methods for evaluating pesticide residues is essential in order to ensure the safety and quality of agricultural products since the Republic of Korea implemented the Positive List System (PLS). The objective of this research was to establish a method for the simultaneous analysis of 322 pesticide residues in fruits and vegetables (such as coffee, potato, corn, and chili pepper), using the quick, easy, cheap, effective, rugged, and safe (QuEChERS) approach in combination with gas chromatography-tandem mass spectrometry (GC-MS/MS). This study introduces a robust, high-throughput GC-MS/MS method for screening the target pesticide residues in agricultural products, achieving the PLS criterion of 0.01 mg/kg LOQ. Despite some compounds not aligning with the CODEX recovery guideline, sufficient reproducibility was confirmed, attesting to the method's applicability in qualitative analyses. A health risk assessment conducted using estimated daily intake/acceptable daily intake ratios indicated low risks associated with product consumption (<0.035391%), thereby confirming their safety. This efficient method holds significant implications for the safe distribution of agricultural products, including during import inspections.

Previous studies have shown that female adolescents are at a higher risk of low self-esteem than male adolescents, and self-esteem in adolescents is critical for academic performance, adult health, and economic status. Depression, social withdrawal, and grit are predicted to be internal factors that affect self-esteem, and an integrated exploration of the relationship between them is required for a proper approach to enhance self-esteem in female adolescents. Therefore, this study investigated the influence of social withdrawal and depression on self-esteem among female adolescents and explored the mediating effect of grit on self-esteem. Data collected from 1,106 girls in the third year of middle school of the third-year survey (2020) of the Korean Children and Youth Panel Survey 2018 were analyzed in this study. For data analysis, partial least square-structural equation modeling was performed using SmartPLS 3.0. Social withdrawal was negatively related to grit, but not related to self-esteem. Depression was negatively related to grit and self-esteem. Grit was positively related to self-esteem. In addition, grit showed mediating effects in the associations between social withdrawal and self-esteem, and between depression and self-esteem in female adolescents. In conclusion, in female adolescents, the mediating effects of grit attenuated the negative effects of social withdrawal and depression on self-esteem. To enhance self-esteem in female adolescents, it is important to develop and implement strategies to strengthen grit and control negative emotional states, such as depression.

PURPOSE: Studies in lower mammals demonstrate enhancement of the medial olivocochlear reflex (MOCR) following noise exposure. A similar effect may occur in humans, and there is some evidence of an individual's acoustic history affecting the MOCR. The current work evaluates the relationship between an individual's annual noise exposure history and their MOCR strength. Given the potential role of the MOCR as a biological hearing protector, it is important to identify factors associated with MOCR strength. METHOD: Data were collected from 98 normal-hearing young adults. Annual noise exposure history was estimated using the Noise Exposure Questionnaire. MOCR strength was assayed using click-evoked otoacoustic emissions (CEOAEs) measured with and without noise presented to the contralateral ear. MOCR metrics included the MOCR-induced otoacoustic emission (OAE) magnitude shift and phase shift. A CEOAE signal-to-noise ratio (SNR) of at least 12 dB was required for estimation of the MOCR metrics. Linear regression was applied to evaluate the relationship between MOCR metrics and annual noise exposure. RESULTS: Annual noise exposure was not a statistically significant predictor of the MOCR-induced CEOAE magnitude shift. However, annual noise exposure was a statistically significant predictor of the MOCR-induced CEOAE phase shift-the MOCR-induced phase shift decreased with increasing noise exposure. Additionally, annual noise exposure was a statistically significant predictor of OAE level. CONCLUSIONS: Findings contrast with recent work that suggests MOCR strength increases with annual noise exposure. Compared with previous work, data for this study were collected using more stringent SNR criteria, which is expected to increase the precision of the MOCR metrics. Additionally, data were collected for a larger subject population with a wider range of noise exposures. Whether findings generalize to other exposure durations and levels is unknown and requires future study.

OBJECTIVES: The objective of the study was to quantify inter-subject variability in the dependence of the medial-olivocochlear reflex (MOCR) on noise bandwidth. Of specific interest was whether inter-subject variability in MOCR dependence on bandwidth explained variability in the MOCR response elicited by wideband noise. DESIGN: Thirty-two young adults with normal hearing participated in the study. Click-evoked otoacoustic emissions were measured in the ipsilateral ear with and without noise presented in the contralateral ear. Presentation of contralateral noise served to activate the MOCR. The MOCR was activated using five different noise stimuli with bandwidths ranging from 1- to 5-octaves wide (center frequency of 2 kHz; bandwidth incremented in 1-octave steps). Noise spectral levels (19.6 dB SPL/Hz) were held constant across all bandwidths. MOCR metrics included the normalized-percent change in the otoacoustic emission (OAE), the MOCR-induced OAE magnitude shift, and the MOCR-induced OAE phase shift. Linear mixed-effect models were fit to model the dependence of MOCR-induced OAE magnitude and phase changes on noise bandwidth. The use of a mixed-effect modeling approach allowed for the estimation of subject-specific model parameters that capture on- and off-frequency contributions to the MOCR effects. Regression analysis was performed to evaluate the predictive capacity of subject-specific model parameters on the MOCR response elicited by wideband noise. RESULTS: All OAE-based MOCR metrics increased as the noise bandwidth increased from 1- to 5-octaves wide. The dependence of MOCR-induced OAE magnitude and phase shifts on activator bandwidth was well approximated using a linear model with intercept and slope terms. On average, MOCR-induced magnitude and phase shifts increased at a rate of 0.3 dB/octave and 0.01 cycles/octave, respectively, as bandwidth extended beyond the predicted region of OAE generation. A statistically significant random effect of subject was found for both the intercept and slope parameter of each model. Subject-specific slope estimates were statistically significant predictors of a repeated measure of the wideband MOCR response. A higher slope was predictive of larger wideband MOCR effects. CONCLUSIONS: MOCR-induced changes to the OAE are greatest when the MOCR is elicited using wideband noise. Variability in the process of spectral integration within the MOCR pathway appears to explain, in part, inter-subject variability in OAE-based estimates of the MOCR response elicited by wideband noise.

Porcine circovirus type 2 (PCV2) is an economically important swine pathogen that causes porcine circovirus-associated diseases (PCVADs). The objective of this study was to evaluate the use of specific pathogen-free Yucatan miniature pigs (YMPs) as an experimental model for PCV2d challenge and vaccine assessment because PCV2-negative pigs are extremely rare in conventional swine herds in Korea. In the first experiment, every three pigs were subjected to PCV2d field isolate or mock challenge. During three weeks of experiments, the PCV2d infection group exhibited clinical outcomes of PCVAD with high viral loads, lymphoid depletion, and detection of PCV2d antigens in lymphoid organs by immunohistochemistry. In the second experiment, three groups of pigs were challenged with PCV2d after immunization for three weeks: a nonvaccinated group (three pigs), a PCV2b-Vac group vaccinated with a commercial PCV2b-based inactivated vaccine SuiShot® Circo-ONE (five pigs), and a PCV2d-Vac group vaccinated with an experimental PCV2d-based inactivated vaccine (five pigs). During the three weeks of the challenge period, nonvaccinated pigs showed similar clinical outcomes to those observed in the PCV2d infection group from the first experiment. In contrast, both the PCV2b and PCV2d vaccinations produced good levels of protection against PCV2d challenge, as evidenced by reduced viral loads, improved growth performance, high virus-neutralizing antibody titers, and less development of PCV2-associated pathological lesions. Taken together, these data suggest that YMPs could be an alternative model for PCV2 challenge experiments, and these animals displayed typical clinical and pathological features and characteristics of protective immunity induced by the vaccines that were consistent with those resulting from PCV2 infections in conventional pigs.

Oscillatory neural network (ONN)-based classification of clustered data relies on frequency synchronization to injected signals representing input data, showing a more efficient structure than a conventional deep neural network. A frequency tunable oscillator is a core component of the network, requiring energy-efficient, and area-scalable characteristics for large-scale hardware implementation. From a hardware viewpoint, insulator-metal transition (IMT) device-based oscillators are attractive owing to their simple structure and low power consumption. Furthermore, by introducing non-volatile analog memory, non-volatile frequency programmability can be obtained. However, the required device characteristics of the oscillator for high performance of coupled oscillator have not been identified. In this article, we investigated the effect of device parameters of IMT oscillator with non-volatile analog memory on coupled oscillators network for classification of clustered data. We confirmed that linear conductance response with identical pulses is crucial to accurate training. In addition, considering dispersed clustered inputs, a wide synchronization window achieved by controlling the hold voltage of the IMT shows resilient classification. As an oscillator that satisfies the requirements, we evaluated the NbO2-based IMT oscillator with non-volatile Li-based electrochemical random access memory (Li-ECRAM). Finally, we demonstrated a coupled oscillator network for classifying spoken vowels, achieving an accuracy of 85%, higher than that of a ring oscillator-based system. Our results show that an NbO2-based oscillator with Li-ECRAM has the potential for an area-scalable and energy-efficient network with high performance.

To date, few studies related to the evaluation of the pathogenicity of different PRRSV isolates using a reproductive model have been undertaken, and the main focus has remained on respiratory models using young pigs. This study aimed to evaluate the pathogenicity of two PRRSV-1 isolates (D40 and CBNU0495) and two PRRSV-2 isolates (K07-2273 and K08-1054) in a reproductive model. Pregnant sows were experimentally infected with PRRSV at gestational day 93 or used as an uninfected negative control. Sera were collected at 0, 3, 7, 14, and 19 days post-challenge (dpc) for virological and serological assays. At 19 dpc, all sows were euthanized, and their fetuses were recovered by performing cesarean section and immediately euthanized for sample collection. Here, compared to the other isolates, the CBNU0495 isolate replicated most efficiently in the pregnant sows, and K07-2273 produced the highest rate of reproductive failure even though it did not replicate as efficiently as the other isolates in sows and fetuses, indicating that vertical transmission and reproductive failure due to PRRSV infection do not have any significant correlation with the viral loads in samples from sows and fetuses. Similarly, the viral loads and the histopathological lesions did not show any correlation with each other, as the PRRSV-2-infected groups displayed more prominent and frequent histopathological lesions with lower viral loads than the PRRSV-1-infected groups. However, viral loads in the myometrium/endometrium might be related to the spreading of PRRSV in the fetuses, which affected the birth weight of live fetuses. This study contributes to a better understanding of the pathogenicity of the most prevalent Korean PRRSVs in a reproductive model.

OBJECTIVES: The objective of this study was to evaluate the influence of music versus speech on a listener's signal-to-noise ratio (SNR) preferences when listening in a noise background. SNR preferences were quantified using acceptable noise level (ANL) and preferred SNR metrics. The measurement paradigm for ANL allows the listener to adjust the level of background noise while listening to the target at their most comfortable loudness level. A higher ANL indicates less tolerance for noise and a lower ANL indicates high tolerance for noise. The preferred SNR is simply the SNR the listener prefers when attending to a target in a fixed-amount (level) of background noise. In contrast to the ANL, the listener does not have control over the noise. Rather, they are only able to manipulate the target level. The first aim of the study was to determine if listeners' tolerances for noise, quantified using the ANL, when listening to music is different from that when listening to speech. The second aim of the study was to determine if listeners' tolerances for noise, quantified using their preferred SNR, when listening to music is different from that when listening to speech. The third aim of the study was to quantify the relationship between ANL and preferred SNR. DESIGN: Ninety-nine normal-hearing, native-English speakers participated in this study. The ANL and preferred SNR were measured for speech and music targets. Music targets included two variations (with lyrics and without lyrics) of the song "Rocky Top." Measurements were made in the sound field at 0° azimuth, 1.5 m from a loud speaker. For both ANL and preferred SNR, targets were presented in 12-talker babble noise. The level of the noise was adjusted by the listener during ANL measurement but was fixed in level during the preferred SNR measurement (75 dB A). Repeated-measures analysis of variance was performed to identify any significant effect of target on the ANL and preferred SNR. Correlation analysis was performed to evaluate the relationship between ANL and preferred SNR. RESULTS: Findings demonstrate a significant effect of target on ANL and preferred SNR. ANLs were highest for speech (mean = 7.2 dB), followed by music with lyrics (6.1 dB), and music without lyrics (2.5 dB). Preferred SNRs were highest for music with lyrics (mean = 2.3 dB), followed by speech (1.2 dB), and music without lyrics (-0.1 dB). A listener's ANL for a given target was strongly correlated with their ANL for a different target (the same was true for preferred SNR); however, ANL for a given target was not a statistically significant predictor of preferred SNR for the same target. CONCLUSIONS: When listening in a background of noise, the listener's tolerance for noise depends on the target to which they are attending, whether music or speech. This dependence is especially evident for ANL measures, and less so for preferred SNR measures. Despite differences in ANL and preferred SNR across targets, a listener's ANL and preferred SNR for one target predicts their ANL and preferred SNR, respectively, for a different target. The lack of correlation between ANL and preferred SNR suggests different mechanisms underly these listener-preference metrics.

T follicular helper (Tfh) cells help orchestrate optimal humoral immunity by helping B cells to produce affinity-matured, class-switched antibodies in germinal centers. Recent studies have unveiled the complexity and heterogeneity in Tfh cell populations, particularly with respect to their cytokine production. These distinct Tfh cell subsets help tune the class, magnitude and quality of the immunoglobulins produced by B cells, thus shaping the course of humoral responses. The Tfh cell-B cell axis-dependent antibody production is mostly beneficial, but at times can go awry and result in the generation of pathologic antibodies that can harm the host. While IgE class of antibodies are infamous for their detrimental role in allergic diseases, emerging evidence is indicative of their pathologic roles in other dysregulated immune states. Here, we discuss the role of Tfh cell subsets in the regulation of pathologic IgE production in allergies and other immunopathologic conditions.

Despite the routine use of porcine reproductive and respiratory syndrome (PRRS)-modified live vaccines, serious concerns are currently being raised due to their quick reversion to virulence and limited cross-protection against divergent PRRS virus (PRRSV) strains circulating in the field. Therefore, a PRRS chimeric vaccine (JB1) was produced using a DNA-launched infectious clone by replacing open reading frames (ORFs) 3-6 with those from a mixture of two genetically different PRRSV2 strains (K07-2273 and K08-1054) and ORF1a with that from a mutation-resistant PRRSV strain (RVRp22) exhibiting an attenuated phenotype. To evaluate the safety and cross-protective efficacy of JB1 in a reproductive model, eight PRRS-negative pregnant sows were purchased and divided into four groups. Four sows in two of the groups were vaccinated with JB1, and the other 4 sows were untreated at gestational day 60. At gestational day 93, one vaccinated group and one nonvaccinated group each were challenged with either K07-2273 or K08-1054. All of the sows aborted or delivered until gestation day 115 (24 days post challenge), and the newborn piglets were observed up to the 28th day after birth, which was the end of the experiment. Overall, pregnant sows of the JB1-vaccinated groups showed no meaningful viremia after vaccination and significant reductions in viremia with K07-2273 and K08-1054, exhibiting significantly higher levels of serum virus-neutralizing antibodies than non-vaccinated sows. Moreover, the JB1-vaccinated groups did not exhibit any abortion due to vaccination and showed improved piglet viability and birth weight. The piglets from JB1-vaccinated sows displayed lower viral concentrations in serum and fewer lung lesions compared with those of the piglets from the nonvaccinated sows. Therefore, JB1 is a safe and effective vaccine candidate that confers simultaneous protection against two genetically different PRRSV strains.

In this paper, the N,N-dimethylformamide (DMF)-assisted shape evolution of highly uniform and shape-pure copper nanocrystals (Cu NCs) is presented for the first time. Colloidal Cu NCs are synthesized via the disproportionation reaction of copper (I) bromide in the presence of a non-polar solvent mixture. It is observed that the shape of Cu NCs is systematically controlled by the addition of different amounts of DMF to the reaction mixture in high-temperature reaction conditions while maintaining a high size uniformity and shape purity. With increasing amount of DMF in the reaction mixture, the morphology of the Cu NCs change from a cube enclosed by six {100} facets, to a sphere with mixed surface facets, and finally, to an octahedron enclosed by eight {111} facets. The origin of this shape evolution is understood via first-principles density functional theory calculations, which allows the study of the change in the relative surface stability according to surface-coordinating adsorbates. Further, the shape-dependent plasmonic properties are systematically investigated with highly uniform and ligand-exchanged colloidal Cu NCs dispersed in acetonitrile. Finally, the facet-dependent electrocatalytic activities of the shape-controlled Cu NCs are investigated to reveal the activities of the highly uniform and shape-pure Cu NCs in the methanol oxidation reaction.

In this paper, we present the imaging formation process of the piecewise mirror eyes of the deep-sea spookfish, which has a strange combination of refractive and reflective eyes. The biological reflective eye structure is formulated to the curved surface's flat mirror array. Zemax is utilized to evaluate optical features such as the modulation transfer function, distortion, and imaging performances. However, the natural images are highly distorted, and the resolution is lower than expected. Therefore, we increase the number of piecewise mirrors of the fisheye to see higher quality images, which can be improved entirely by the mirror shapes. Finally, the fisheye's imaging analysis reveals the deep-sea creature's resolution limit and also shows the possibility of artificial and biomimetic camera applications.

In this study, cholesterol (CH), ß-sitosterol (SI), and stigmasterol (ST) were explored to improve the stability of retinol in the liposome bilayer. Retinol was incorporated into liposomes composed of soybean-derived L-α-phosphatidylcholine (PC) and 10% sterol (w/w), which were prepared as multilamellar vesicles. Under all conditions, the efficiency of retinol incorporation into liposomes was higher than 99%, and the average particle size of liposomes was similar to that of PC alone. Liposomes were stored at 4 and 25 °C, with and without light, respectively, for 10 days. It was found that during the storage, CH and SI were effective in stabilizing the retinol in liposomes. These results indicate that an appropriate phytosterol could improve the stability of retinol in liposomes.

RESUMEN

Synchronized-spontaneous otoacoustic emissions (SSOAEs) are slow-decaying otoacoustic emissions (OAEs) that persist up to several hundred milliseconds following presentation of a transient stimulus. If the inter-stimulus interval is sufficiently short, SSOAEs will contaminate the stimulus window of the adjacent epoch. In medial-olivocochlear reflex (MOCR) assays, SSOAE contamination can present as a change in the stimulus between quiet and noise conditions, since SSOAEs are sensitive to MOCR activation. Traditionally, a change in the stimulus between MOCR conditions implicates acoustic reflex activation by the contralateral noise; however, this interpretation is potentially confounded by SSOAEs. This study examined the utility of jittering stimulus onset to desynchronize and cancel short-latency SSOAE energy. Transient-evoked (TE) OAEs and SSOAEs were measured from 39 subjects in contralateral-quiet and -noise conditions. Clicks were presented at fixed and quasi-random intervals (by introducing up to 8 ms of jitter). For the fixed-interval condition, spectral differences in the stimulus window between quiet and noise conditions mirrored those in the SSOAE analysis window, consistent with SSOAE contamination. In contrast, spectral differences stemming from SSOAEs were attenuated and/or absent in the stimulus window for the jitter conditions. The use of jitter did not have a statistically significant effect on either TEOAE level or the estimated MOCR.

This study represents the heating-up synthesis of lead-free cesium bismuth bromide perovskite nanocrystals (NCs). CsBr and BiBr3 precursors are used to synthesize uniform and phase-pure cesium bismuth bromide NCs, and the reaction is performed via an injection-free, heating-up method in the presence of a solvent mixture with a high boiling point. The size and composition of cesium bismuth bromide NCs are readily controlled by changing the reaction time, temperature, and amount of surfactant added to the reaction mixture. Upon heating, sequential phase evolution occurs, resulting in the formation of kinetically stable BiOBr in the early reaction stages, which transformed into the thermodynamically stable Cs3BiBr6 and Cs3Bi2Br9 with an increase in either the reaction time or the reaction temperature. Furthermore, the absorption and photoluminescence properties of Cs3BiBr6 and Cs3Bi2Br9 NCs are characterized to investigate their composition-dependent optical properties. This work provides the potential to synthesize various types of lead-free perovskite NCs by tailoring the size and compositions.

Since the 2013-2014 incursion of the virulent G2b porcine epidemic diarrhoea virus (PEDV) pandemic strains in South Korea, frequent moderate-scale regional outbreaks have recurred. In particular, areas of Jeju Island with extensive swine production have faced repeated epidemics since the re-emergence in 2014. The current study reports the complete genome sequences and molecular characterization of the representative PEDV strains responsible for the 2018 endemic outbreaks on Jeju Island. All isolates were determined to belong genetically to the highly pathogenic pandemic G2b group. Full-length genome sizes of four isolates differed from that of the G2b epidemic field strain due to insertion or deletion (DEL) mutations in the non-structural protein (nsp)- or spike (S) protein-coding regions. The 2018 Jeju isolates shared 96.7%-98.7% and 98.5%-99.4% identity at the S gene and whole-genome levels, respectively, compared to global G2b PEDV strains. Genetic and phylogenetic analyses indicated that the 2018 isolates were closest to the 2014 G2b re-emergent Jeju strains, but appeared to have undergone substantial rapid independent evolution. Among the isolates, a notable nsp3 DEL variant strain, KOR/KNU-1807/2018, was isolated and propagated by continuous passages in Vero cells, and displayed typical PEDV-induced syncytia formation. Genomic sequencing identified a unique 8-nt DEL in the extreme C-terminal region of the S gene at the 4th passage (KNU-1807-P4) compared to its original sample. This DEL resulted in the premature termination of S by nine amino acid residues (EVFEKVHVQ), which contained a KxHxx motif that is a potential endoplasmic reticulum retrieval signal. In vivo animal studies showed that variant strain KNU-1807 had decreased virulence in suckling piglets. These results advance our knowledge regarding the genetic variation and pathogenicity of the G2b PEDV endemic strains prevalent in Jeju swine herds in South Korea.
