BACKGROUND: Sargassum horneri came ashore after flowing from the South China Sea to Jeju Island a few years ago. This caused a significant environmental impact on coastal areas where S. horneri has accumulated because of decomposition and the release of toxic substances, such as hydrogen sulfide. AIMS: In this study, we evaluated a biological ingredient prepared from fucoidan-rich S. horneri and demonstrated its antiwrinkle effects on ultraviolet B (UVB)-induced fibroblast cells. MATERIALS AND METHODS: Fucoidan samples from S. horneri were prepared according to a previously published process with modifications. The compositional analysis of S. horneri fucoidan extract (SHFE) as well as its effects on antiaging were examined to determine its utility as a functional material. RESULTS: SHFE exhibited antioxidant properties using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Treatment of UVB-induced fibroblasts with SHFE significantly increased the synthesis of procollagen compared with adenosine treatment and inhibited MMP-1 and MMP-3 expression. In a clinical study, SHFE lotion improved skin barrier effects in forearms and transepidermal water loss (TEWL) values were reduced after 3 weeks of use compared with a placebo. CONCLUSION: SHFE has utility as an additive with functional antiaging effects for a range of cosmetic products as it restores skin hydration in the epidermal barrier.

Smilax sieboldii, a climbing tree belonging to Smilacaceae, has been used in traditional oriental medicine for treating arthritis, tumors, leprosy, psoriasis, and lumbago. To evaluate the anti-obesity effects of S. sieboldii (Smilacaceae), we screened methylene chloride (CH2Cl2), ethyl acetate (EtOAc), aqueous-saturated n-butanol, and ethanol (EtOH) extracts of the whole plant at various concentrations to inhibit adipogenesis in adipocytes. The 3T3-L1 cell line with Oil red O staining with the help of fluorometry was used as an indicator of anti-obesity activity. Bioactivity-guided fractionation of the EtOH extract and subsequent phytochemical investigation of the active CH2Cl2- and EtOAc-soluble fractions resulted in the isolation of 19 secondary metabolites (1-19), including a new α-hydroxy acid derivative (16) and two new lanostane-type triterpenoids (17 and 18). The structures of these compounds were characterized using various spectroscopic methods. All the isolated compounds were screened for adipogenesis inhibition at a concentration of 100 µM. Of these, compounds 1, 2, 4-9, 15, and 19 significantly reduced fat accumulation in 3T3-L1 adipocytes, especially compounds 4, 7, 9, and 19, showing 37.05 ± 0.95, 8.60 ± 0.41 15.82 ± 1.23, and 17.73 ± 1.28% lipid content, respectively, at a concentration of 100 µM. These findings provide experimental evidence that isolates from S. sieboldii extracts exert beneficial effects regarding the regulation of adipocyte differentiation.

This study analyzed the contents of critical reflective journals written by new nurses during their orientations using a text network. This study aimed to find ways to reduce turnover and improve clinical field adaptability among new nurses. The authors analyzed the content of reflective journals written by 143 new nurses from March 2020 to January 2021. Text network analysis was performed using the NetMiner 4.4.3 program. After data preprocessing, frequency of occurrence, degree centrality, closeness centrality, betweenness centrality, and eigenvector community were analyzed. In total, 453 words were extracted and refined, and words with high simple frequency and centrality were "incompetence," "preparation," "explanation," "injection," "time," "examination," and "first try." "Medication" had the highest frequency of occurrence, and "incompetence" was the most important keyword in the centrality analysis. In addition, component analysis and eigenvector community analysis revealed three sub-theme groups: (1) basic nursing skills required for new nurses, (2) insufficient competency, and (3) explanation of nursing work. Significantly, this study is the first to use the text network method to analyze the subjective experiences of the critical reflective journals of new nurses. In conclusion, changes are needed to improve the education system for new nurses and promote efficient sharing of nursing tasks.

BACKGROUND: During in-hospital cardiac arrest events, clinical nurses are often the first responders; therefore, nurses require sufficient advanced cardiac life support (ACLS) competency. This study aimed to verify the effects of a hybrid team-based ACLS simulation (HTAS) program (developed in this study) on nurses' ACLS performance, specifically ACLS knowledge, cardiopulmonary resuscitation (CPR) self-efficacy, and CPR-related stress. METHODS: The developed HTAS comprised four lecture videos, one team-based skills training video, and a team-based ACLS simulation. A quasi-experimental pretest-posttest design with a comparison group (CG) was used to evaluate the effectiveness of the HTAS. Of the 226 general ward nurses with more than 6 months of clinical experience, 117 were allocated to the intervention group (IG), which attended the HTAS, and 109 to the CG, which attended only basic ACLS training. RESULTS: The IG's ACLS performance significantly improved (t = 50.8, p < 0.001) after the training. Relative to the respective pretest conditions, posttest ACLS knowledge (t = 6.92, p < 0.001) and CPR self-efficacy (t = 6.97, p < 0.001) of the IG also significantly increased. However, when the mean difference values were compared, there was no significant difference between the two groups with respect to ACLS knowledge (t = 1.52, p = 0.130), CPR self-efficacy (t = -0.42, p = 0.673), and CPR stress (t = -0.88, p = 0.378). CONCLUSION: The HTAS for ward nurses was effective at enhancing the nurses' ACLS performance. It is necessary to develop effective training methods for team-based ACLS and verify the sustained effects of such training.

Studies on methods to share nursing and learning experiences with preceptors are lacking. This study was conducted to determine the preliminary effects of developing and applying a reflective practice program for preceptor nurses who experience stressful situations to convert negative emotions into positive ones. This study was conducted over 12 weeks from March to May 2022 on 47 participant nurses in South Korea. Preceptor Reflective Practice Program (PRPP) was conducted in parallel with writing a reflective journal and a reflective practice workshop. Data collection was integrated through quantitative and qualitative approaches. Quantitative data were collected through questionnaires on stress coping, the burden of preceptors, social support, and emotional intelligence, and analyzed by SPSS WIN 26.0 program. The questionnaire data were analyzed after the preceptor nurses had written a reflective journal. Stress coping, social support, and emotional intelligence significantly increased in preceptor nurses after participating in the PRPP. This study found that the PRPP helped nurses improve their emotional intelligence through reflective practice and convert stress into a more positive direction. Therefore, at the organizational and national levels, a reasonable compensation system to provide support workforce and to the work of preceptor nurses should be established.

Tetracera loureiri (T. loureiri) is a woody climber inhabiting open deciduous or evergreen forests in Southeast Asia. A decoction comprising its stem and other herbs is a traditional Thai remedy for fatigue and jaundice, as well as to promote overall health. Anti-inflammatory effects induced by T. loureiri extract have not been reported. In this study, we investigated the anti-inflammatory effect of an ethanol extract of T. loureiri (ETL) on lipopolysaccharide (LPS)-induced inflammatory response in RAW264.7 macrophages. We found that ETL treatment inhibited the production of nitric oxide (NO) in LPS-stimulated RAW264.7 cells, without affecting cell viability. The effect of ETL on the expression of various pro-inflammatory mediators was analyzed using reverse transcription-polymerase chain reaction (RT-PCR), Western blotting, and enzyme-linked immunosorbent assay (ELISA). We observed that ETL inhibited the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the mRNA and protein levels and decreased the production of prostaglandin E2 (PGE2) by COX-2 in RAW264.7 macrophages. ETL dose-dependently reduced the production of pro-inflammatory cytokines including tumor necrosis factor-α (TNF-α), interleukin-1ß (IL-1ß), and interleukin-6 (IL-6) in LPS-induced RAW264.7 cells, in a dose-dependent manner. Furthermore, ETL suppressed the LPS-induced nuclear translocation of the nuclear factor, NF-κB. Additionally, ETL was found to inhibit the activation of mitogen-activated protein kinases (MAPK), such as extracellular signal-regulated kinase, c-Jun-N-terminal kinase, and p38 MAPK. In conclusion, our findings demonstrate that ETL inhibits the expression of pro-inflammatory mediators and cytokines, thereby downregulating NF-κB and MAPK signaling pathways in LPS-stimulated macrophages, Consequently, ETL is a potential therapeutic agent for the treatment of inflammatory diseases.

The aim of this study was to establish one of the most efficient biocatalytic processes for turanose production by applying a robust Bifidobacterium thermophilum (BtAS) mutant developed through site-directed mutagenesis. A gene encoding the amylosucrase of B. thermophilum (BtAS) was cloned and used as a mutagenesis template. Among the BtAS variants generated by the site-directed point mutation, four different single-point mutants (P200R, V202I, Y265F, and Y414F) were selected to create double-point mutants, among which BtASY414F/P200R displayed the greatest turanose productivity without losing the thermostability of native BtAS. The turanose yield of BtASY414F/P200R reached 89.3% at 50 °C after 6 h with 1.0 M sucrose + 1.0 M fructose. BtASY414F/P200R produced significantly more turanose than BtAS-wild type (WT) by 2 times and completed the reaction faster by another 2 times. Thus, turanose productivity (82.0 g/(L h)) by BtASY414F/P200R was highly improved from 28.1 g/(L h) of BtAS-WT with 2.0 M sucrose + 0.75 M fructose.

This work introduces a facile geometry-controlled method for the fabrication of embossed and engraved polymeric moth-eye-inspired nanostructures in imprinting molds using anodic aluminum oxide (AAO) templates, resulting in a novel anti-reflective transparent coating. The moth-eye nanostructures are prepared directly on the surface of a flexible polyethylene terephthalate (PET) substrate. As a prerequisite procedure, a UV-curable polyurethane acrylate resin is spun on the PET. The shape of the moth-eye nanostructures can then be adjusted by controlling the size and shape of the nanopores in the AAO templates. Both embossed and concaved polymer moth-eye nanostructures were successfully mounted on a PET substrate. Embossed polymer replica molds were prepared using the AAO master templates in combination with an imprinting process. As revealed by field-emission electron microscope (FE-SEM) images, conical nanopatterns in the AAO template with a diameter of ~90 nm and a depth of ~100 nm, create a homogeneous embossed morphology in the polymer moth-eye nanostructure. The polymeric molds with the depths of 300 and 500 nm revealed the amalgamated structures in their apexes. In addition, a dip-imprinting process of the polymeric layers was implemented to yield a concaved mold by assembly on the surface of the 100 nm embossed polymer mold substrate. Considering that the embossed structures may be crumbled due to their protuberant shapes, the concaved geometries can have an advantage of stability in a certain application concerning physical degradation along with a higher transmission by ~2%, despite somewhat nonuniform structure. The experimental and theoretical results of this study indicate that this polymer layer has the potential for use in anti-reflective coating applications in transparent films.

In our search for novel plant-derived inhibitors of nitric oxide (NO) with potential for treating inflammatory diseases, the phytochemicals of Amomum tsao-ko fruits were investigated, leading to the isolation of one bicyclic nonane (1), three menthene skeleton monoterpenoids (2-4), and two acyclic monoterpenoids (5 and 6). Their structures were identified using one- and two-dimensional nuclear magnetic resonance spectroscopy, and mass spectrometry. To the best of our knowledge, compounds 2-5 were obtained from the genus Amomum for the first time. All isolates were tested for their ability to inhibit lipopolysaccharide-stimulated NO overproduction in RAW264.7 cells. Compound 4 was found to inhibit NO production. Western blotting analysis indicated that active compound 4 can regulate inducible NO synthase expression. In addition, lipopolysaccharide-induced interleukin 1 beta and interleukin-6 overproduction was reduced in a concentration-dependent manner.

High-k Y2O3 thin films were investigated as the gate dielectric for amorphous indium zinc tin oxide (IZTO) thin-film transistors (TFTs). Y2O3 gate dielectric was deposited by radio frequency magnetron sputtering (RF-MS) under various working pressures and annealing conditions. Amorphous IZTO TFTs with SiO2 as the gate dielectric showed a high field-effect mobility (µFE) of 19.6 cm²/Vs, threshold voltage (Vth) of 0.75 V, on/off current ratio (Ion/Ioff) of 2.0×106, and subthreshold swing (SS) value of 1.01 V/dec. The IZTO TFT sample device fabricated with the Y2O3 gate dielectric showed an improved subthreshold swing value compared to that of the IZTO TFT device with SiO2 gate dielectric. The IZTO TFT device using the Y2O3 gate dielectric deposited at a working pressure of 5 mtorr and annealed at 400 °C in 6 sccm O2 for 1 hour showed a high µFE of 51.8 cm²/Vs, Vth of -0.26 V, Ion/Ioff of 6.0×10³, and SS value of 0.19 V/dec. With the application of a Y2O3 gate dielectric, the Vth shift improved under a positive bias stress (PBS) but was relatively unaffected by negative bias stress (NBS). These shifts were attributed to charge traps within the gate dielectric and/or interfaces between the channel and gate dielectric layer.

Dielectric microstructures have been reported to have a negative influence on permselective ion transportation because ions do not migrate in areas where the structures are located. However, the structure can promote the transportation if the membrane is confined to a microscopic scale. In such a scale where the area to volume ratio is significantly large, the primary driving mechanisms of the ion transportation transition from electro-convective instability (EOI) to surface conduction (SC) and electroosmotic flow (EOF). Here, we provide rigorous evidence on how the SC and EOF around the dielectric microstructures can accelerate the ion transportation by multi-physics simulations and experimental visualizations. The microstructures further polarize the ion distribution by SC and EOF so that ion carriers can travel to the membrane more efficiently. Furthermore, we verified, for the first time, that the arrangements of microstructures have a critical impact on the ion transportation. While convective flows are isolated in the crystal pillar configuration, the flows show an elongated pattern and create an additional path for ion current in the aligned pillar configuration. Therefore, the fundamental findings of the electrokinetic effects on the dielectric microstructures suggest an innovative application in micro/nanofluidic devices with high mass transport efficiency.

Manipulating mechanism of particle's motion has been extensively studied for the sample preparation in microfluidic applications including diagnostics, food industries, biological analyses and environmental monitoring. However, most of conventional methods need additional external forces such as electric field or pressure and complicated channel designs, which demand highly complex fabrication processes and operation strategies. In addition, these methods have inherent limitations of dilution or mixing during separation or preconcentration step, respectively, so that a number of studies have reported an efficient selective preconcentration process, i.e. conducting the separation and preconcentration simultaneously. In this work, a power-free spontaneous selective preconcentration method was suggested based on leveraging convective flow over diffusiophoresis near the water-absorbing nanoporous ion exchange medium, which was verified both by simulation and experiment. Especially, the velocity of the convective flow by an imbibition deviated from the original tendency of t-1/2 due to non-uniformly patterned nanoporous medium that has multiple cross-sectional areas. As a result, the direction of particle's motion was controlled at one's discretion, which led to the spontaneous selective preconcentration of particles having different diffusiophoretic constant. Also, design rule for maximizing the efficiency was recommended. Thus, this selective preconcentration method would play as a key mechanism for power-free lab on a chip applications.

A unique thermostable amylosucrase from Bifidobacterium thermophilum was produced as a recombinant protein with the half-life of 577 h at 50 °C. By adding 1.0 M fructose, turanose yield was improved from 22.7% to 43.3% with 1.0 M sucrose, and from 23.7% to 39.4% with 1.5 M sucrose. Sucrose consumption rate was greatest at 55 °C, but the lowest amount of turanose was produced. Thus, turanose yield from sucrose biomass was inversely proportional to reaction temperature and was highly dependent on [fructose]. Meanwhile, insoluble α-glucan yield was clearly reduced as [fructose] increased. With 1.0 M fructose + 1.0 M sucrose, glucan byproduct yield significantly decreased from 29.4% to 1.1%. Molecular weights of linear glucans were almost identical among various [sucrose]s and were homogenous with very low polydispersity. This unique dual reaction patterns of amylosucrase enzyme would be very useful for massive productions of two different biomaterials simply by changing sucrose biomass concentration.

While the water impermeable constraint has been conventionally adopted for analyzing the transport phenomena at the interface of electrolyte/nanoporous medium, non-negligible water-permeance through the medium results in significant effect on ion and particle transportation. In this work, a rigorous theoretical and experimental analysis of the water-permeance effect were conducted based on a fully-coupled analytical/numerical method and micro/nanofluidic experiments. The regime diagram with three distinctive types of concentration boundary layers (ion depletion, ion accumulation, and intermediate) near the ion exchange nanoporous medium was proposed depending on the medium's permselectivity and the water-permeance represented by an absorbing parameter. Moreover, the critical absorbing parameters which divide the regimes were analytically obtained so that the bidirectional motion of particles were demonstrated only by altering the water-permeance without external stimuli. Conclusively, the presenting analysis of non-negligible water-permeance would be a substantial fundamental of transport phenomena at the interface of the ion exchange medium and electrolyte, especially useful for the tunable particle/ion manipulations in intermediate Peclet number environment.

IZTO20 (In0.6Zn0.2Sn0.2O1.5) ceramic target was prepared from oxide mixture of In2O3, ZnO, and SnO2 powders. IZTO20 thin films were then deposited onto glass substrate at 400 °C by DC magnetron sputtering. The average optical transmittance determined by ultraviolet-visible spectroscopy was higher than 85% for all films. The minimum resistivity of the annealed IZTO20 thin film was approximately 6.1×10-4 Ω·cm, which tended to increase with decreasing indium content. Substrate heating and annealing were found to be important parameters affecting the electrical and optical properties. An organic photovoltaic (OPV) cell was fabricated using the IZTO20 film deposited under the optimized condition as an anode electrode and the efficiency of up to 80% compared to that of a similar OPV cell using ITO film was observed. Reduction of surface roughness and electrical resistivity through annealing treatment was found to contribute to the improved efficiency of the OPV cell.

Clausena excavata (C. excavata) has been used as a traditional medicine for the treatment of abdominal pain, enteritis, dysentery, and malaria. The present study was designed to evaluate the effect of a 50% ethanol extract of C. excavata (ECE) on weight loss, adipocyte size, and obesity-related biochemical parameters in high-fat diet (HFD)-induced obese mice. After 6 weeks of HFD + ECE administration, HFD-induced total fat, subcutaneous fat, and visceral fat were evaluated by micro-computed tomography. The serum levels of triglyceride (TG), total cholesterol (TCH), high-density lipoprotein (HDL) cholesterol, and low-density lipoprotein (LDL) cholesterol were evaluated with a biochemical analyzer, and leptin and adiponectin levels in the serum were assessed via enzyme-linked immunoassay (ELISA). Moreover, adipocyte size and lipid formation in the liver were examined. We found that weight gain, epididymal fat pad weight, adipocyte size, and lipid formation were markedly attenuated in the livers of HFD-induced obese mice treated with ECE. Furthermore, TG, TCH, and leptin decreased in the serum, whereas adiponectin increased. In conclusion, our data show that ECE has potent anti-obesity activity in vivo and support the development of ECE as a potential therapeutic agent for the treatment of obesity.
