RESUMEN

Osteoporosis is a systemic skeletal disease caused by an imbalance between bone resorption and formation. Current treatments primarily involve systemic medication and hormone therapy. However, these systemic treatments lack directionality and are often ineffective for locally severe osteoporosis, with the potential for complex adverse reactions. Consequently, treatment strategies using bioactive materials or external interventions have emerged as the most promising approaches. This review proposes twelve microenvironmental treatment targets for osteoporosis-related pathological changes, including local accumulation of inflammatory factors and reactive oxygen species (ROS), imbalance of mitochondrial dynamics, insulin resistance, disruption of bone cell autophagy, imbalance of bone cell apoptosis, changes in neural secretions, aging of bone cells, increased local bone tissue vascular destruction, and decreased regeneration. Additionally, this review examines the current research status of effective or potential biophysical and biochemical stimuli based on these microenvironmental treatment targets and summarizes the advantages and optimal parameters of different bioengineering stimuli to support preclinical and clinical research on osteoporosis treatment and bone regeneration. Finally, the review addresses ongoing challenges and future research prospects.

The chemical safety of poly (butylene adipate-co-terephthalate) (PBAT) based food contact articles (FCAs) has aroused increasing toxicological concerns in recent years, but the chemical characterization and associated risk assessment still remain inadequate as it fails to elucidate the distribution pattern and discern the potential genotoxic and carcinogenic hazards of the identified substances. Herein, the volatile organic compounds (VOCs) in 50 batches of PBAT-based FCAs of representative categories and 10 batches of PLA and PBAT pellets were characterized, by which 237 VOCs of 10 chemical categories were identified and exhibited characteristic distribution patterns in the chemical spaces derived from their molecular descriptors. Chemical hazards associated with the identified VOCs were discerned by a hazard-driven classification scheme integrating hazard-related knowledge from multiple publicly available sources, and 34 VOCs were found to bear genotoxic or carcinogenic hazards and to feature higher average molecular weight than the other VOCs. Finally, the Risk and hazard quotient (HQ) calculated as the metrics of risk suggested that all identified VOCs posed acceptable risks (Risk<10-4 or HQ < 1), whereas oxolane, butyrolactone, N,N-dimethylacetamide, 2-butoxyethanol, benzyl alcohol, and 1,2,3-trichloropropane posed non-negligible (Risk>10-6) genotoxic or carcinogenic risk and thus should be of prioritized concern to promote the chemical safety of PBAT-based FCAs.

OBJECTIVE: This study aimed to estimate and predict the burden of osteoarthritis (OA) and site-specific OA (hip, knee, hand, and others) from 1990 to 2030 and their attributable risk factors in China. METHOD: Data were obtained from the Global Burden of Diseases 2019. The burden was estimated by analyzing the trends of prevalence, incidence, and disability-adjusted life years (DALY). Population attributable risk (PAR) was calculated to assess the impact of high body mass index (BMI). The prediction from 2020 to 2030 was implemented by Bayesian age-period-cohort analysis. RESULTS: In China, prevalent cases, DALY, and incident cases of OA increased to 132.81 million, 4.72 million, and 10.68 million, respectively. Age-standardized rates (ASRs) of prevalence, DALYs, and incidence increased for OA and site-specific OA, especially for hip OA. Site-specific OA showed different susceptible peaking ages, and the burden for those over 50 years old became serious. Female preference existed in the trends for knee OA but not in those for hip, hand, and other OA. PARs of high BMI continued to increase, impacting knee OA more than hip OA and showing female preference. In the next decade, incident cases for OA and site-specific OA will continue to increase, despite that the ASR of OA incidence will decrease. CONCLUSIONS: OA and site-specific OA remain huge public health challenges in China. The burden of OA and site-specific OA is increasing, especially among people over 50 years old. Health education, exercise, and removing modifiable risk factors contribute to alleviate the growing burden. Key Points • In China, the burden of osteoarthritis and site-specific osteoarthritis (hip, knee, hand, and others) as well as the Risk Factor (high body mass index) increased greatly from 1990 to 2019. • It is estimated that incident cases for OA and site-specific OA will continue to increase, despite that the ASR of OA incidence will decrease.

The recycling of polyethylene terephthalate (PET) stands as an effective strategy for mitigating plastic pollution and reducing resource waste. The study aimed to investigate the characterization and elimination efficiency of volatile organic compounds (VOCs) present in rPET at various recycling stages using comprehensive two-dimensional gas chromatography-quadrupole-time-of-flight-mass spectrometry coupled with chemometrics. The results revealed that 52, 135, 95, 44, and 33 VOCs, mostly classified into three chemical groups, were tentatively identified in virgin - PET (v-PET), cold water washed - rPET (C-rPET), decontaminated - rPET (D-rPET), melt-extruded - rPET (M-rPET), and solid-state polycondensation - rPET (S-rPET), respectively. Regarding the VOCs with high and median detection frequencies, fatty acyls showed the highest elimination efficiency (100 % and 92 %), followed by organooxygen compounds (81 % and 99 %), others (97 % and 95 %), and benzene and substituted derivatives (82 % and 95 %) in term of HS-SPME. Following the recycling process, there was a general decrease in the concentration of almost all VOCs, as evidenced by the substantial reduction of o-Xylene, hexanoic acid, octanal, and D-limonene from 18.11, 22.43, 30.74, and 7.41 mg/kg to 0, 0, 3.97, and 0 mg/kg, respectively. However, it was noteworthy that the VOCs identified in the samples were not completely extracted, owing to the limitations of HS-SPME. Furthermore, chemometrics analysis indicated significant discrimination among VOCs from vPET, C-rPET, D-rPET, and M-rPET, while indistinct differences were observed between M-rPET and S-rPET. This study contributes to the enhancement of the recycling process and emphasizes the importance of safeguarding consumer health in terms of elimination of VOCs.

Incorporating spent coffee grounds into single-use drinking straws for enhanced biodegradability also raises safety concerns due to increased chemical complexity. Here, volatile organic compounds (VOCs) present in coffee ground straws (CGS), polylactic acid straws (PLAS), and polypropylene straws (PPS) were characterized using headspace - solid-phase microextraction and migration assays, by which 430 and 153 VOCs of 10 chemical categories were identified by gas chromatography - mass spectrometry, respectively. Further, the VOCs were assessed for potential genetic toxicity by quantitative structure-activity relationship profiling and estimated daily intake (EDI) calculation, revealing that the VOCs identified in the CGS generally triggered the most structural alerts of genetic toxicity, and the EDIs of 37.9% of which exceeded the threshold of 0.15 µg person-1 d-1, also outnumbering that of the PLAS and PPS. Finally, 14 VOCs were prioritized due to their definite hazards, and generally higher EDIs or detection frequencies in the CGS. Meanwhile, the probability of producing safer CGS was also illustrated. Moreover, it was uncovered by chemical space that the VOCs with higher risk potentials tended to gather in the region defined by the molecular descriptor related to electronegativity or octanol/water partition coefficient. Our results provided valuable references to improve the chemical safety of the CGS, to promote consumer health, and to advance the sustainable development of food contact materials.

Recycled PET (rPET) is gaining popularity for use in the production of new food contact materials (FCMs) under the context of circular economy. However, the limited information on contaminants in rPET from China and concerns about their potential risk are major obstacles to their use in FCM in China. Fifty-five non-volatile compounds were tentatively identified in 126 batches of hot-washed rPET flakes aimed for food packaging applications in China. Although the 55 substances are not necessarily migratable and may not end up in the contacting media, their presence indicates a need for proper management and control across the value chain. For this reason, the 55 substances prioritized on the basis of level of concerns and in-silico genotoxicity profiler. Among them, dimethoxyethyl phthalate, dibutyl phthalate, bis(2-ethylhexyl) phthalate were classified as level V substances, and Michler's ketone and 4-nitrophenol were both categorized as level V substances and had the genotoxic structure alert, while 2,4,5-trimethylaniline was specified with genotoxic structure alert. The above substances have high priority and may pose a potential risk to human health, therefore special attention should be paid to their migration from rPET. Aside from providing valuable information on non-volatile contaminants present in hot-washed rPET flakes coming from China, this article proposed a prioritization workflow that can be of great help to identify priority substances deserving special attention across the value chain.

Supramolecular peptide nanofiber hydrogels are emerging biomaterials for tissue engineering, but it is difficult to fabricate multi-functional systems by simply mixing several short-motif-modified supramolecular peptides because relatively abundant motifs generally hinder nanofiber cross-linking or the formation of long nanofiber. Coupling bioactive factors to the assembling backbone is an ideal strategy to design multi-functional supramolecular peptides in spite of challenging synthesis and purification. Herein, a multi-functional supramolecular peptide, P1R16, is developed by coupling a bioactive factor, parathyroid hormone related peptide 1 (PTHrP-1), to the basic supramolecular peptide RADA16-Ⅰ via solid-phase synthesis. It is found that P1R16 self-assembles into long nanofibers and co-assembles with RADA16-Ⅰ to form nanofiber hydrogels, thus coupling PTHrP-1 to hydrogel matrix. P1R16 nanofiber retains osteoinductive activity in a dose-dependent manner, and P1R16/RADA16-Ⅰ nanofiber hydrogels promote osteogenesis, angiogenesis and osteoclastogenesis in vitro and induce multi-functionalized osteoregeneration by intramembranous ossification and bone remodeling in vivo when loaded to collagen (Col) scaffolds. Abundant red blood marrow formation, ideal osteointegration and adapted degradation are observed in the 50% P1R16/Col scaffold group. Therefore, this study provides a promising strategy to develop multi-functional supramolecular peptides and a new method to topically administrate parathyroid hormone or parathyroid hormone related peptides for non-healing bone defects.

The objective was to establish a robust and reliable approach for the characterisation of volatile organic compounds (VOCs) present in food contact paperboard. This was achieved through the utilisation of headspace solid-phase microextraction in tandem with comprehensive two-dimensional (2D) gas chromatography (GC) and quadrupole time-of-flight mass spectrometry (HS-SPME-GC × GC-QTOF-MS). The experimental parameters were optimised, involving the use of a DVB/C-WR/PDMS fibre at a temperature of 80 °C for a duration of 30 min. A total of 344 VOCs comprising aldehydes, ketones, alcohols, ethers, esters, alkanes and aromatic compounds, were tentatively identified in the samples. Twelve compounds believed to be from biogenic sources had a high odour impact making them major contributors to potential taint from the paperboard samples. The catalytic conversion of nitrogen to ammonia is one of the most significant processes in nature and the chemical industry. However, the traditional Haber-Bosch process of ammonia synthesis consumes substantial energy and emits a large amount of carbon dioxide. The efficiency of photocatalytic N2 activation is severely limited by the lack of N2 adsorption sites and poor carrier utilization. Herein, an efficient α-Bi2O3/Bi3O4Br heterojunction is proposed with a photocatalytic nitrogen fixation activity of 238.67 µmol·g-1·h-1. Compared with the BiOBr precursor, α-Bi2O3 and Bi3O4Br, the α-Bi2O3/Bi3O4Br heterojunction with oxygen vacancies can improve the adsorption and activation capacity of N2 and promote the separation efficiency of charge carrier pairs by accommodating photogenerated electrons under visible light through the mechanism of N-type semiconductors. Therefore, oxygen vacancies and heterojunction engineering of semiconductive nanomaterials provide a promising method for the rational design of photocatalysts to enhance the rate of ammonia synthesis under mild conditions.

Bone grafting, as the current gold-standard for large scaled bone damage of various causes, has faced challenges from both the source and appliance. Emerging new tissue engineering substitutes are demonstrating more options and possibilities, with their improved biocompatibility, accessibility, and customizable function. Amongst them, injectable gels (IGs) are a class of gel material displaying astonishing non-invasive properties and surgical viability. While possessing responsiveness toward specific stimuli, they change their physical form in vivo, thus serving as wonderful biomaterials and drug delivery systems. In this review, the mechanics of stimuli-responsive IGs developed during the past decade are illustrated. Two branches of crosslinked gels - co-valent and non-covalent crosslinked IGs and their composition and customization are introduced. In conclusion, the present trend in bone tissue engineering research is summarized and made an outlook for future. It is hoped that this comprehensive review can provide a proper reference for the development of new IGs.

A comparison was performed on various methods detecting the volatile contaminants (VCs) in recycled poly(ethylene terephthalate) (rPET) flakes, the results demonstrated that head-space solid phase micro-extraction combined with comprehensive two-dimensional gas chromatograph-tandem quadrupole-time-of-flight mass spectrometry (HS-SPME-GC×GC-QTOF-MS) was a sensitive, effective, accurate method, and successfully applied to analyze 57 rPET flakes collected from different recycling plants in China. A total of 212 VCs were tentatively identified, and the possible source were associated with plastic, food, and cosmetics. 45 VCs are classified as high-priority compounds with toxicity level IV or V and may pose a risk to human health. Combined chemometrics for further analysis revealed that significant differences among these three geographical recycling regions. 6, 7, and 6 volatile markers were chosen based on VIP values and S-plot among plant1 plant 2 and plant 3, respectively. The markers differed significantly between recycled rPET samples in three geographical recycling regions based on chemometrics analysis. The initial classification rate and cross-validation accuracy were 100% on the identified VCs. These significant differences demonstrate that a systematic study is needed to obtain a comprehensive data on the contamination of rPET for food contact applications in China.

Bone tissue engineering has been becoming a promising strategy for surgical bone repair, but the risk of infection during trauma repair remains a problematic health concern worldwide, especially for fracture and infection-caused bone defects. Conventional antibiotics fail to effectively prevent or treat bone infections during bone defect repair because of drug-resistance and recurrence, so novel antibacterial agents with limited resistance are highly needed for bone tissue engineering. Antimicrobial peptides (AMPs) characterized by cationic, hydrophobic and amphipathic properties show great promise to be used as next-generation antibiotics which rarely induce resistance and show potent antibacterial efficacy. In this review, four common structures of AMPs (helix-based, sheet-based, coil-based and composite) and related modifications are presented to identify AMPs and design novel analogs. Then, potential effects of AMPs for bone infection during bone repair are explored, including bactericidal activity, anti-biofilm, immunomodulation and regenerative properties. Moreover, we present distinctive applications of AMPs for topical bone repair, which can be either used by delivery system (surface immobilization, nanoparticles and hydrogels) or used in gene therapy. Finally, future prospects and ongoing challenges are discussed.

A non-targeted method was developed for screening the semi-volatile compounds of different mechanically recycled PET intended for food contact materials. The data was further analyzed by multiple chemometrics methods to obtain the difference level, and the potential influence factors were investigated. The results showed that total dissolution with comprehensive two-dimensional gas chromatography coupled with time-of-flight mass spectrometry was more effective than other reported methods. Based on the difference level, 97 compounds were characterized into 4 levels. 1-Methyl-2-pyrrolidinone originating from organic solvent was recognized as level IV and could be determined as the primary difference indicator. The contaminant is mainly attributed to the residuum derived from non-food consumer products. The specific types of contaminants and process parameters of the recycling, such as moisture content, properties of rPET, and temperature, were the potential key factors affecting the presence of semi-volatile compounds of mechanically recycled rPET.

The use of non-decontaminated recycled poly(ethylene terephthalate) (PET) in food packages arouses consumer safety concerns, and thus is a major obstacle hindering PET bottle-to-bottle recycling in many developing regions. Herein, machine learning (ML) algorithms were employed for the discrimination of 127 batches of virgin PET and recycled PET (rPET) samples based on 1247 volatile organic compounds (VOCs) tentatively identified by headspace solid-phase microextraction comprehensive two-dimensional gas chromatography quadrupole-time-of-flight mass spectrometry. 100% prediction accuracy was achieved for PET discrimination using random forest (RF) and support vector machine (SVM) algorithms. The features of VOCs bearing high variable contributions to the RF prediction performance characterized by mean decrease Gini and variable importance were summarized as high occurrence rate, dominant appearance and distinct instrument response. Further, RF and SVM were employed for PET discrimination using the simplified input datasets composed of 62 VOCs with the highest contributions to the RF prediction performance derived by the AUCRF algorithm, by which over 99% prediction accuracy was achieved. Our results demonstrated ML algorithms were reliable and powerful to address PET adulteration and were beneficial to boost food-contact applications of rPET bottles.

Bone tissue engineering is becoming an ideal strategy to replace autologous bone grafts for surgical bone repair, but the multihierarchical complexity of natural bone is still difficult to emulate due to the lack of suitable biomaterials. Supramolecular peptide nanofiber hydrogels (SPNHs) are emerging biomaterials because of their inherent biocompatibility, satisfied biodegradability, high purity, facile functionalization, and tunable mechanical properties. This review initially focuses on the multihierarchical fabrications by SPNHs to emulate natural bony extracellular matrix. Structurally, supramolecular peptides based on distinctive building blocks can assemble into nanofiber hydrogels, which can be used as nanomorphology-mimetic scaffolds for tissue engineering. Biochemically, bioactive motifs and bioactive factors can be covalently tethered or physically absorbed to SPNHs to endow various functions depending on physiological and pharmacological requirements. Mechanically, four strategies are summarized to optimize the biophysical microenvironment of SPNHs for bone regeneration. Furthermore, comprehensive applications about SPNHs for bone tissue engineering are reviewed. The biomaterials can be directly used in the form of injectable hydrogels or composite nanoscaffolds, or they can be used to construct engineered bone grafts by bioprinting or bioreactors. Finally, continuing challenges and outlook are discussed.

The repair of critical bone defects remains challenging worldwide. Three canonical pillars (biomaterial scaffolds, bioactive molecules, and stem cells) of bone tissue engineering have been widely used for bone regeneration in separate or combined strategies, but the delivery of bioactive molecules has several obvious drawbacks. Biophysical stimuli have great potential to become the fourth pillar of bone tissue engineering, which can be categorized into three groups depending on their physical properties: internal structural stimuli, external mechanical stimuli, and electromagnetic stimuli. In this review, distinctive biophysical stimuli coupled with their osteoinductive windows or parameters are initially presented to induce the osteogenesis of mesenchymal stem cells (MSCs). Then, osteoinductive mechanisms of biophysical transduction (a combination of mechanotransduction and electrocoupling) are reviewed to direct the osteogenic differentiation of MSCs. These mechanisms include biophysical sensing, transmission, and regulation. Furthermore, distinctive application strategies of biophysical stimuli are presented for bone tissue engineering, including predesigned biomaterials, tissue-engineered bone grafts, and postoperative biophysical stimuli loading strategies. Finally, ongoing challenges and future perspectives are discussed.

BACKGROUND: Organic tin compounds (OTCs), a group of high-risk hazardous substances, are highly concerned in safety regulation of consumer products especially for toys because they can cause serious damage to organs after prolonged or repeated exposure. Gastric juice migration is nowadays widely implemented to assess organic tin intake from toys or food-contact materials; however, the followed up detection method using sodium tetraethylborate [NaB(Et4)] as a derivatization agent may directly lead to false positive of monomethyltins (MMT). OBJECTIVE: In order to avoid the phenomena of false positive of MMT in the course of laboratory testing of toys, it is necessary and important to perform relative experimental and theoretical studies to reveal the cause of false positive of MMT. METHOD: With metal tin powder as a representative of inorganic tin which existed in real samples, it was treated with artificial gastric juice (0.07 mol/L·HCl), followed by ethyl derivatization using sodium tetraethylborate [NaB(Et)4] and then analyzed by gas chromatography-mass spectrometry (GC-MS) according to the procedure specified in the standard of EN 71-3:2013+A3:2018 issued by the European Committee for Standardization (CEN). RESULTS: Without any OTCs in the starting materials, MMT false positive can be reproduced by detecting 0.56 mg/L of triethylmethyltin (TEMT) together with approximately 1000 mg/L of tetraethyltin (TeET), which is similar to real samples. Further, it is demonstrated that the detected amount of TEMT is linearly related with the added amount of NaB(Et)4, and that the formation of TEMT (methyl derivative) is easier than TeET (ethyl derivative) even though the ethyl group is present in a larger amount than the methyl group. CONCLUSIONS: The phenomena of MMT false positive which occurred in the laboratory testing of toys is mainly because that TEMT is highly likely to be obtained from the reaction of inorganic tin and trace level of methylation agent impurities contained in the derivatization reagent-NaB(Et)4. To avoid MMT false positive, it is concluded that the maximum acceptable mole ratio of methylation agent impurities contained in NaB(Et)4 is approximately 0.028%. This research is helpful to be aware of methylation impurities and is favorable to avoid false judgment caused by MMT false positive in routine analysis of toys.

Large amounts of jarosites are produced during zinc hydrometallurgy and bioleaching, as well as in acid sulfate soils and acid mine drainage environments. As such, understanding the behavior of jarosite dissolution is important for analyzing the iron cycle process and promoting the control and treatment of jarosites. In general, soluble ferric ions and jarosites coexist in acid environments; however, the relationship between soluble ferric ions and jarosites under anaerobic reductive conditions is still not well understood. In this study, the effect of adding Fe3+ on the promotion of the bio-dissolution of jarosites using Acidithiobacillus ferrooxidans is investigated. With the addition of 12 mM Fe3+, the efficiency and maximum rate of jarosite bio-dissolution were found to reach 84.1% and 2.66 mmol/(L·d), respectively. The addition of Fe3+ at concentrations higher than 12 mM did not further improve the jarosite bio-dissolution. These results indicate that the mechanisms underlying these improvements include: (i) the reduction of the zeta potential due to the compression of the diffusion layer of the electric double layer by Fe3+; (ii) bacteria growth enhancement and the stabilization of the pH of cultures via the reduction of soluble Fe3+. Based on these observations, this study serves to promote the development of jarosite bio-dissolution using Acidithiobacillus ferrooxidans and challenges the idea that soluble Fe3+ suppresses the bio-dissolution reaction of solid Fe3+ substances such as jarosite when soluble ferric ions and jarosite coexist.

Based on migration procedures using simulated gastric juice specified in the EU toy Safety Standard EN 71-3:2013/A2:2017, a method was developed for the determination of monomethyltin (MMT) migration from toys by gas chromatography-mass spectrometry (GC-MS). Further, a reliable confirmation method for judgement of a false positive for MMT obtained by GC-MS, was established. After optimizing the migration conditions, derivatization steps and chromatographic parameters, the method yielded a linear range from 0.02 to 1.0 mg/L with a correlation coefficient (R2) of 0.9992. The limits of detection and quantification of the method were 0.11 and 0.32 mg/kg, respectively. The recoveries were 86.2%-104.2% under different spiked levels (0.5, 1.0 and 1.5 µg), and the relative standard deviations were 3.1%-8.2% (n=6). MMT migration ranging from 0.44 to 0.67 mg/kg was detected in the surface coating of tin-plating (Sn-Fe alloy) toy materials, which was subsequently confirmed to be false positive. Therefore, a novel confirmative approach using methanol or acetone as the migration solvent was proposed, aiming at verifying the false positive of MMT. The results showed that the previously positive MMT detected by GC-MS could no longer be detected when treated by these migration solvents.Hence, this approach can be used to confirm false positive detected for MMT after GC-MS detection.

Background: Bath-type ultrasonic-assisted extraction (UAE) has been developed as one of the most important sample pretreatment methods, especially for batch-sample pretreatment. So far, however, requirements for the performance of bath-type UAE equipment have not been standardized, nor has a suitable evaluation method that can be used to judge the feasibility of ultrasonic equipment for extraction been presented in the available regulations or standards. Objective: A simple and efficient method that can be used to evaluate the performance of bath-type UAE equipment is necessary to be proposed and established. Methods: First, distribution of a sound field in ultrasonic equipment was measured by acoustimeter and the dyeing method, through which influencing factors including frequency, preheating time, and output power of the equipment, as well as the horizontal and vertical position for locating the sample in the equipment, were investigated, and optimized parameters for extraction were achieved. Then, through the aluminum foil lattice method, by calculating the perforated rate of the aluminum foil, cavitation intensity of the ultrasonic equipment can be quantitatively determined. Results: With the optimized working conditions and by selecting appropriate parameters for the aluminum foil, perforated holes formed on the foil displayed a good pattern. Further validation experiments indicated conformity between the established method and the actual extraction effect of the ultrasonic equipment, proposing a suitable requirement for the cavitation effect of the bath-type UAE equipment. Conclusions: The aluminum foil lattice method has been proved to be simple, convenient, inexpensive, and reliable for quickly evaluating the extraction performance of bath-type UAE equipment.
