Aspartyl dipeptidase (dipeptidase E) can hydrolyze Asp-X dipeptides (where X is any amino acid), and the enzyme plays a key role in the degradation of peptides as nutrient sources. Dipeptidase E remains uncharacterized in Streptomyces. Orf2 from Streptomyces sp. 139 is located in the exopolysaccharide biosynthesis gene cluster, which may be a novel dipeptidase E with "S134-H170-D198" catalytic triad by sequence and structure comparison. Herein, recombinant Orf2 was expressed in E. coli and characterized dipeptidase E activity using the Asp-ρNA substrate. The optimal pH and temperature for Orf2 are 7.5 and 40 ℃; Vmax and Km of Orf2 are 0.0787 mM·min-1 and 1.709 mM, respectively. Orf2 exhibits significant degradation activities to Asp-Gly-Gly, Asp-Leu, Asp-His, and isoAsp-Leu and minimal activities to Asp-Pro and Asp-Ala. Orf2 contains a Ser-His-Asp catalytic triad characterized by point mutation. In addition, the Asp147 residue of Orf2 is also proven to be critical for the enzyme's activity through molecular docking and point mutation. Transcriptome analysis reveals the upregulation of genes associated with ribosomes, amino acid biosynthesis, and aminoacyl-tRNA biosynthesis in the orf2 mutant strain. Compared with the orf2 mutant strain and WT, the yield of crude polysaccharide does not change significantly. However, crude polysaccharides from the orf2 mutant strain exhibit a wider range of molecular weight distribution. The results indicate that the Orf2 links nutrient stress to secondary metabolism as a novel dipeptidase E. KEY POINTS: • A novel dipeptidase E with a Ser-His-Asp catalytic triad was characterized from Streptomyces sp. 139. • Orf2 was involved in peptide metabolism both in vitro and in vivo. • Orf2 linked nutrient stress to mycelia formation and secondary metabolism in Streptomyces.

The compost-derived humic acids (HA) and fulvic acids (FA) contain abundant active functional groups with strong redox capacity, which can function as an electron shuttles for promoting the reduction of heavy metals, thus changing the form of the pollutants in the environment and reducing their toxicity. Therefore, in this study, UV-Vis, FTIR, 3D-EEM, electrochemical analysis were applied to study the spectral characteristics and electron transfer capacity (ETC) of HA and FA. Upon analysis, the results showed an increasing trend of ETC and humification degree (SUVA254) for both HA and FA during composting. However, the aromatic degree (SUVA280) of HA was higher than FA. After 7 days of culture, 37.95% of Cr (Ⅵ) was reduced by Shewanella oneidensis MR-1 (MR-1) alone. Whereas, only if HA or FA existed, the diminution of Cr (Ⅵ) reached 37.43% and 40.55%, respectively. However, the removal rate of Cr (Ⅵ) by HA/MR-1 and FA/MR-1 increased to 95.82% and 93.84% respectively. It indicated that HA and FA acted as electron shuttles, mediating the transfer of electrons between MR-1 and the final electron acceptor, effectively facilitating the bioreduction of Cr (Ⅵ) to Cr (Ⅲ) and also determined via correlation analysis. This study suggested compost-derived HA and FA coupling with MR-1 exhibited excellent performance for the bioreduction of Cr (Ⅵ) to Cr (Ⅲ).

Extreme mining activities can risk human life and the environment via potentially toxic elements (PTEs) in road dust, thus making their quantification and assessment unavoidable. For this purpose, we collected 50 fine road dust samples from the Chehe mining area, China, to quantify the level of contamination and ecological and health risks of PTEs comprising As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Sb, and Zn, and their quantitative source apportionment using the positive matrix factorization model (PMF). Results indicated that the average values of Cd, Sb, As, Zn, Pb, and Cu in road dust were 1555.21, 586.78, 429.68, 429.43, 72.88, and 26.61 times higher than their background values. Pollution indices of PTEs revealed a strong level of contamination by Cd, Sb, As, Zn, and Pb, which were extremely polluted in the study area. The average values of the Nemerow integrated risk index (NIRI) and potential ecological risk index (RI) were 104.09 and 86.49 times the highest risk limit, respectively, which are extremely high ecological risks. Based on PMF for quantitative source identification, mining activities and fuel combustion were the main sources of PTEs in road dust contributing 57.25% and 35.95%, respectively. Furthermore, the health risk assessment indicated that Sb, As, Cr, Cd, and Pb in the Chehe road dust could lead to significantly serious carcinogenic and non-carcinogenic risks to both children and adults. The results of this study could be used to opt for strategies to mitigate the ecological and human health risk in the mining area of Hechi, China.

Multi-modal fusion can exploit complementary information from various modalities and improve the accuracy of prediction or classification tasks. In this paper, we propose a parallel, multi-modal, factorized, bilinear pooling method based on a semi-tensor product (STP) for information fusion in emotion recognition. Initially, we apply the STP to factorize a high-dimensional weight matrix into two low-rank factor matrices without dimension matching constraints. Next, we project the multi-modal features to the low-dimensional matrices and perform multiplication based on the STP to capture the rich interactions between the features. Finally, we utilize an STP-pooling method to reduce the dimensionality to get the final features. This method can achieve the information fusion between modalities of different scales and dimensions and avoids data redundancy due to dimension matching. Experimental verification of the proposed method on the emotion-recognition task using the IEMOCAP and CMU-MOSI datasets showed a significant reduction in storage space and recognition time. The results also validate that the proposed method improves the performance and reduces both the training time and the number of parameters.

Glycosylation inactivation is one of the important macrolide resistance mechanisms. The accumulated evidences attributed glycosylation inactivation to a glucosylation modification at the inactivation sites of macrolides. Whether other glycosylation modifications lead to macrolides inactivation is unclear. Herein, we demonstrated that varied glycosylation modifications could cause inactivation of midecamycin, a 16-membered macrolide antibiotic used clinically and agriculturally. Specifically, an actinomycetic glycosyltransferase (GT) OleD was selected for its glycodiversification capacity towards midecamycin. OleD was demonstrated to recognize UDP-D-glucose, UDP-D-xylose, UDP-galactose, UDP-rhamnose and UDP-N-acetylglucosamine to yield corresponding midecamycin 2'-O-glycosides, most of which displayed low yields. Protein engineering of OleD was thus performed to improve its conversions towards sugar donors. Q327F was the most favorable variant with seven times the conversion enhancement towards UDP-N-acetylglucosamine. Likewise, Q327A exhibited 30% conversion enhancement towards UDP-D-xylose. Potent biocatalysts for midecamycin glycosylation were thus obtained through protein engineering. Wild OleD, Q327F and Q327A were used as biocatalysts for scale-up preparation of midecamycin 2'-O-glucopyranoside, midecamycin 2'-O-GlcNAc and midecamycin 2'-O-xylopyranoside. In contrast to midecamycin, these midecamycin 2'-O-glycosides displayed no antimicrobial activities. These evidences suggested that besides glucosylation, other glycosylation patterns also could inactivate midecamycin, providing a new inactivation mechanism for midecamycin resistance. Cumulatively, glycosylation inactivation of midecamycin was independent of the type of attached sugar moieties at its inactivation site.

OBJECTIVE: To evaluate the adaptation of different lithium disilicate glass-ceramic crown and whether different fabrication processes affect the adaptation of crowns. METHODS: Thirty epoxy dies crowns were divided into 3 groups. They were cemented to domestic lithium disilicate glass-ceramic crowns in Group A, to IPS e.max CAD crowns in Group B and to IPS e.max Press crowns in Group C respectively. All crowns were cut by cutting machine. A confocal laser scanning microscope was used to measure the gap between crown tissue surface and die. RESULTS: There were significant differences in the three groups of measurement points(P<0.05). Within the groups, specimens in group A showed in the lowest marginal fit([46.8±9.1] µm). Those in group B showed the lowest shoulder suitability([59.3±7.9] µm), axial plane([50.5±3.6] µm) and occlusal surface([87.6±11.6 µm]. Those in group C demonstrated the lowest axial plane angle[(84.4±10.1) µm]. In addition to the axial plane angle, the CAD/CAM system exhibited good accuracy of fit. CONCLUSIONS: The three groups of lithium disilicate glass-ceramic crowns appeared to have clinically acceptable fit.

Quality assessment of 3D images encounters more challenges than its 2D counterparts. Directly applying 2D image quality metrics is not the solution. In this paper, we propose a new full-reference quality assessment for stereoscopic images by learning binocular receptive field properties to be more in line with human visual perception. To be more specific, in the training phase, we learn a multiscale dictionary from the training database, so that the latent structure of images can be represented as a set of basis vectors. In the quality estimation phase, we compute sparse feature similarity index based on the estimated sparse coefficient vectors by considering their phase difference and amplitude difference, and compute global luminance similarity index by considering luminance changes. The final quality score is obtained by incorporating binocular combination based on sparse energy and sparse complexity. Experimental results on five public 3D image quality assessment databases demonstrate that in comparison with the most related existing methods, the devised algorithm achieves high consistency with subjective assessment.

Perceptual three-dimensional image quality assessment (3D-IQA) aims to use computational models to measure image quality in a way that is consistent with human visual perception. Unlike previous studies that directly extended the two-dimensional metrics to the three-dimensional (3D) case, the major technical innovation of this paper is to simulate monocular and binocular visual perception and propose a monocular-binocular feature fidelity induced index for 3D-IQA. To be more specific, in the training stage, we train monocular and binocular dictionaries from the training database, so that the latent response properties can be represented as a set of basis vectors. In the quality estimation stage, we compute monocular feature fidelity and binocular feature fidelity indexes based on the estimated sparse coefficient vectors and compute a global energy response similarity index by considering global energy changes. The final quality score is obtained by incorporating them. Experimental results on four 3D-IQA databases demonstrate that in comparison with the most relevant existing methods, the devised algorithm achieves high consistency alignment with subjective assessment.

BACKGROUND: Caregiver productivity costs are an important component of the overall cost of care for individuals with birth defects and developmental disabilities, yet few studies provide estimates for use in economic evaluations. OBJECTIVE: This study estimates labor market productivity costs for caregivers of children and adolescents with spina bifida. METHODS: Case families were recruited from a state birth defects registry in Arkansas. Primary caregivers of children with spina bifida (N = 98) reported their employment status in the past year and demographic characteristics. Controls were abstracted from the Current Population Survey covering the state of Arkansas for the same time period (N = 416). Estimates from regression analyses of labor market outcomes were used to calculate differences in hours worked per week and lifetime costs. RESULTS: Caregivers of children with spina bifida worked an annual average of 7.5 to 11.3 hours less per week depending on the disability severity. Differences in work hours by caregivers of children with spina bifida translated into lifetime costs of $133,755 in 2002 dollars using a 3% discount rate and an age- and sex-adjusted earnings profile. Including caregivers' labor market productivity costs in prevention effectiveness estimates raises the net cost savings per averted case of spina bifida by 48% over the medical care costs alone. CONCLUSIONS: Information on labor market productivity costs for caregivers can be used to better inform economic evaluations of prevention and treatment strategies for spina bifida. Cost-effectiveness calculations that omit caregiver productivity costs substantially overstate the net costs of the intervention and underestimate societal value.

OBJECTIVE: To determine the relationships between BMI and workforce participation and the presence of work limitations in a U.S. working-age population. RESEARCH METHODS AND PROCEDURES: We used data from the Panel Study of Income Dynamics, a nationwide prospective cohort, to estimate the effect of obesity in 1986 on employment and work limitations in 1999. Individuals were classified into the following weight categories: underweight (BMI < 18.5), normal weight (18.5 < or = BMI < 25), overweight (25 < or = BMI < 30), and obese (BMI > or = 30). Using multivariable probit models, we estimated the relationships between obesity and both employment and work disability. All analyses were stratified by sex. RESULTS: After adjusting for baseline sociodemographic characteristics, smoking status, exercise, and self-reported health, obesity was associated with reduced employment at follow-up [men: marginal effect (ME) -4.8 percentage points (pp); p < 0.05; women: ME -5.8 pp; p < 0.10]. Among employed women, being either overweight or obese was associated with an increase in self-reported work limitations when compared with normal-weight individuals (overweight: ME +3.9 pp; p < 0.01; obese: ME +12.6 pp; p < 0.01). Among men, the relationship between obesity and work limitations was not statistically significant. DISCUSSION: Obesity appears to result in future productivity losses through reduced workforce participation and increased work limitations. These findings have important implications in the U.S., which is currently experiencing a rise in the prevalence of obesity.
