RESUMEN

Objective@#To investigate the effect and mechanism of phosphoprotein phosphatase 5 catalytic(PPP5C) on the migration , invasion and tumor stemness of human lung adenocarcinoma H1299 cells.@*Methods@#The PPP5C⁃pcDNA3. 1 overexpression vector was constructed. PPP5C⁃pcDNA3. 1 and pcDNA3. 1 were transfected into H1299 cells , and H1299 stable cell lines were screened with G418. The mRNA and protein expression levels of PPP5C were identified by qRT⁃PCR and Western blot. The proliferation activity of H1299 cells was detected by drawing cell growth curve and cell clonal formation assay. The wound⁃healing assay and transwell assay were used to test the migration and invasion abilities of H1299 cells , respectively. The stemness of H1299 cells was evaluated by sphere formation assay.@*Results@#The PPP5C⁃pcDNA3. 1 eukaryotic expression vector was successfully constructed and the expression levels of PPP5C significantly increased after transfection into H1299 cells. After overexpression of PPP5C in H1299 cells , the cell growth curve and clonal formation assay displayed that the proliferation ability was not affected , the migration and invasion of cells were significantly enhanced through wound⁃healing assay and transwell assay , accompanied by an increase in the expression of MMP9 , stem cell spheroid assay showed a significant increase in stemness of cells , accompanied by increased expression of SOX2. @*Conclusion@#The proliferation ability of cells is not affected , the migration and invasion and the stemness of cells are enhanced by regulating MMP9 and SOX2 respectively , after overexpression of PPP5C in human lung adenocarcinoma H1299 cells.

RESUMEN

Klotho gene was originally discovered as an anti-aging gene, Klotho protein encoded by Klotho gene is expressed in multiple human tissues, and its most prominent function is the regulation of phosphate homeostasis. Klotho protein possesses various activities, including inhibition of multiple signaling pathways, reducing oxidative stress and suppressing inflammation, and these activities are associated with cancer. Klotho protein is discovered as a universal tumor suppressor, and its expression is associated with tumorigenesis and prognosis of patients. Lung cancer is the most common malignancy tumor, and it is the leading cause of cancer deaths worldwide because of its high incidence and mortality. This article summarizes the research progress of the role of Klotho on pathogenesis, therapeutic effect and prognosis in lung cancer, in order to provide new biomarker and target for diagnosis, treatment and prognosis of lung cancer.

RESUMEN

Objective To analyze the influence of single nucleotide polymorphisms (SNPs) of focal adhesion kinase (FAK) and integrin β1 (ITG β1) in epilepsy seizures.Methods The clinical data were collected from 330 participants (163 epilepsy patientss and 167 normal controls) from January 2017 to May 2018.Peripheral blood was extracted from these patients.The SNPs of FA K genotypes (rs7460,rs10100025,rs7843014,and rs306954) and ITGβ1 genotypes (rs2230395,rs2298141,rs2230394,and rs35016806) were detected by Sequenom MassArray.Logistic regression analysis was used to determine the influence of genotype at each locus in onset of epilepsy.Results The distribution of the 8 genotypes mentioned above accorded with Hardy-Weinberg Equilibrium.There was significant difference in the frequency distribution of FAK genotype rs7843014 between the epilepsy group and normal group (P<0.05).Logistic regression analysis of FAK genotype rs7843014 could cause epilepsy (OR=1.676,L95=1.100,and P=0.012).Conclusion The mutation of FAK genotype rs7843014 may be one of the risk factors of epilepsy.

RESUMEN

Objective To study the prevalence of thrombocytosis in patients with non-small-cell lung cancer (NSCLC) and its correlation with clinicopathological features.Methods 156 patients with advanced NSCLC were retrospectively analyzed.The platelets degree between the groups with different sex,age,smoking,histological type of advanced NSCLC was compared and analyzed statistically.The relationship between the platelet count and chemotherapy effects was analyzed.Single analysis and Cox regression analysis were used for TTP and OS.Results Compared with the healthy persons,Plt significantly elevated in group with advanced NSCLC (36.5 %,57/156 vs 5.0 %,5/100) (P < 0.01),and thrombocytosis group responded poorly to chemotherapy (22.8 %,13/57 vs 39.4 %,39/99) (P < 0.05).The TTP (3.0 months vs 5.2 months) and OS (11.2 months vs 14.2 months) of Plt elevated group were significantly shorter than those of normal group.Conclusion Thrombocytosis is closely related to progress and metastasis of advanced NSCLC.Platelet count can be used as an assistant index in the prognosis judgment of patients with advanced NSCLC.

RESUMEN

Purpose To investigate the expression of androgen receptor ( AR) in molecular subtypes of breast cancer and its relation-ship with clinicopathological features. Methods The expression of estrogen receptor (ER), progesterone receptor (PR), HER-2, CK5/6, CK14 in breast cancer was detected by EnVision immunohistochemical staining, and they further be divided into 84 Luminal A subtypes, 27 Luminal B subtypes, 14 HER-2 overexpression subtypes, 11 Basal-like subtypes, and 17 Normal-like subtypes, respec-tively. The distribution of AR in 5 molecular subtypes and its relationship with clinicopathological factors, such as age, histopathologi-cal grading, tumor size, lymph node status and clinical staging, were analyzed. Results In 153 cases of breast cancer, the positive rates of AR expression were 73. 20%, Luminal A was the majority ( 62. 50%) in 5 molecular subtypes. In the HER-2 negative and positive breast cancer, the positive rates of AR expression were 75. 00% and 68. 29% and the difference between the two groups was no statistical significant (P=0. 196). Of the 153 breast cancers, 23 (15. 03%) were triple-negative breast cancers (ER-, PR- and HER-2-), 15 (9. 80%) breast cancers were negative for ER, PR and AR, 8(5. 23%) breast cancers were quadruple negative (ER-, PR-, AR- and HER-2 -) . There was significant difference of the expression of AR in ER/PR positive and negative breast cancer (P=0. 001),it was positively correlated with ER/PR protein expression (rs =0. 874, P=0. 024). The difference of AR ex-pression between histological grading was statistically significance ( P=0. 022 ) , the expression of AR was negatively correlated with histopathologic grading (rs = -0. 201, P=0. 013). The positive expression of AR was related to tumor size, but not correlated with lymph node metastasis ( P=0. 211 ) and clinical staging ( P=0. 065 ) . Conclusion The expression of AR in breast cancer tissues and its relationship with clinicopathological features may provide a basis for the further improvement molecular typing of breast cancer, and also provide the theoretical basis for clinical treatment and judging prognosis.

RESUMEN

Though all the marketed drugs of dipeptidyl peptidase IV inhibitors are structurally different, their inherent correlation is worthy of further investigation. Herein we rapidly discovered a novel DPP-IV inhibitor 8g (IC50 = 4.9 nmol.L-1) which exhibits as good activity and selectivity as the market drugs through scaffold hopping and drug splicing strategies based on alogliptin and linagliptin. This study demonstrated that the employment of classic medicinal chemistry strategy to the marketed drugs with specific target is an efficient approach to discover novel bioactive molecules.

RESUMEN

Objective To research early diagnosis and treatment of acute spontaneous spinal epidural hematoma (SSEH). Methods Medical records of 13 SSEH patients adimtted in Timone Ste Marguerite Hospital,France and Ruijin Hospital,China from 1985 to 2000 were retrospectively reviewed.The etiology, neurological symptoms, neuroradiology therapy, as well as the prognosis of this rare disease are discussed in comparison with the literaturs.Results Six of thirteen SSEH wee relatd with innormal coagulation. All patients had the same original symptom which was radicular pain. Eleven cases were diagnosed by MRI. After decompressive surgery, recovery occurred in 3 of 5 patients. Five of 8 patients had a favorable outcome after medical treatment. Conclusions The majority of spontaneous hematomas results from a rupture of the venous plexus. It could be diagnosed according typical symptoms and MRI. Decompressive surgery is urgent but not unique. A idiosyncratic type which has a spontaneous complete recovery could be found.
