The effects of early supplementation with oat ß-glucan during the suckling period on piglet gut microbiota composition, concentrations of short-chain fatty acids, and gut physiological markers were assessed. Fifty piglets from five litters, balanced for sex and birth weight, were divided within litters into two treatment groups: ß-glucan and control. Piglets in the ß-glucan group received the supplement three times/week from day 7 of age until weaning. Rectal swab samples were collected from 10 piglets per treatment group (balanced across litters) from week 1 to week 4, and plasma samples were collected at 1, 3, and 4 weeks of age. Additional samples of intestinal tissues and jugular and portal vein plasma were collected from 10 animals at weaning (one per treatment group and litter). The concentrations of short-chain fatty acids in plasma and the microbiota composition in rectal swabs were mainly influenced by piglet age, rather than the supplement. There were significant differences in microbiota composition between litters and several correlations between concentrations of short-chain fatty acids in plasma and specific microbial taxa in rectal swabs. Overall, ß-glucan supplementation did not have any clear impact on the gut environment in suckling piglets, whereas a clear age-related pattern emerged.

Tail biting is an abnormal behaviour that causes stress, injury and pain. Given the critical role of the gut-microbiota in the development of behavioural problems in humans and animals, the aim of this study was to determine whether pigs that are biters, victims of tail biting or controls (nine matched sets of pigs) have a different microbiota composition, diversity and microbial metabolite profile. We collected faecal and blood samples from each individual for analysis. The gut microbiota composition was most different between the biter and the control pigs, with a higher relative abundance of Firmicutes in tail biter pigs than the controls. Furthermore, we detected differences in faecal and plasma short chain fatty acids (SCFA) profiles between the biter and victim pigs, suggesting physiological differences even though they are kept in the same pen. Thus, in addition to supporting an association between the gut microbiota and tail biting in pigs, this study also provides the first evidence of an association between tail biting and SCFA. Therefore, further research is needed to confirm these associations, to determine causality and to study how the SCFA profiles of an individual play a role in the development of tail biting behaviour.

Free faecal liquid (FFL) in horses is characterised by the excretion of faeces in two phases (one solid and one liquid), which may cause dermatitis on the hindlegs. The causes of FFL are not known. Results from previous studies have indicated that feed ration composition and management factors may play important roles in the occurrence of FFL. A case-control study was therefore performed in which data on feed rations, feeding practices and management factors were compared between horses with (case) and without (control) FFL on 50 private farms in Sweden and Norway. The comparisons show that case and control horses were reported to be fed similar average amounts of wrapped forage (p = 0.97) and to be subject to similar management practices, but case horses were fed higher proportions of concentrates in their diet (p < 0.001) and lower average amounts of straw and lucerne (p < 0.05) compared to control horses. Case horses were reported to be fed twice as much concentrate per 100 kg BW and day as control horses and a higher daily intake of starch and water-soluble carbohydrates (p < 0.05). Case horses also had a lower daily intake of digestible crude protein and neutral detergent fibre compared to control horses (p < 0.05). These differences were small but are of interest for further studies of factors causing FFL.

Free faecal liquid (FFL) is a condition in horses where faeces are voided in one solid and one liquid phase. The liquid phase contaminates the tail, hindlegs and area around the anus of the horse, resulting in management problems and potentially contributing to impaired equine welfare. The underlying causes are not known, but anecdotal suggestions include feeding wrapped forages or other feed- or management-related factors. Individual horse factors may also be associated with the presence of FFL. This study, therefore, aimed to characterize horses showing FFL particularly when fed wrapped forages, and to map the management and feeding strategies of these horses. Data were retrieved by a web-based survey, including 339 horses with FFL. A large variety of different breeds, ages, disciplines, coat colours, housing systems and feeding strategies were represented among the horses in the study, meaning that any type of horse could be affected. Respondents were asked to indicate if their horse had diminished signs of FFL with different changes in forage feeding. Fifty-eight percent (n = 197) of the horse owners reported diminished signs of FFL in their horses when changing from wrapped forages to hay; 46 (n = 156) of the horse owners reported diminished signs of FFL in their horses when changing from wrapped forages to pasture; 17% (n = 58) reported diminished signs of FFL when changing from any type of forage batch to any other forage. This indicated that feeding strategy may be of importance, but cannot solely explain the presence of FFL. The results also showed that the horses in this study had a comparably high incidence of previous colic (23%, n = 78) compared to published data from other horse populations. In conclusion, the results showed that FFL may affect a large variety of horse types and that further studies should include detailed data on individual horse factors including gastrointestinal diseases as well as feeding strategies, in order to increase the chance of finding causes of FFL.

The mechanism-based risk for hyperkalemia has limited the use of mineralocorticoid receptor antagonists (MRAs) like eplerenone in cardio-renal diseases. Here, we describe the structure and property-driven lead generation and optimization, which resulted in identification of MR modulators ( S)-1 and ( S)-33. Both compounds were partial MRAs but still demonstrated equally efficacious organ protection as eplerenone after 4 weeks of treatment in uni-nephrectomized rats on high-salt diet and aldosterone infusion. Importantly, and in sharp contrast to eplerenone, this was achieved without substantial changes to the urine Na+/K+ ratio after acute treatment in rat, which predicts a reduced risk for hyperkalemia. This work led to selection of ( S)-1 (AZD9977) as the clinical candidate for treating MR-mediated cardio-renal diseases, including chronic kidney disease and heart failure. On the basis of our findings, we propose an empirical model for prediction of compounds with low risk of affecting the urinary Na+/K+ ratio in vivo.

This study investigated the effect of providing extra water and nesting material to Moo Lath sows on piglet survival and growth. Three treatments were evaluated in a randomized block design with six sows/treatment. In the Control treatment, sows were not provided with nesting material or extra water apart from that included in the feed (conventional smallholder practice). In treatment NM, nesting material was provided 1-2 days before expected farrowing. In treatment NMW, nesting material as in NM and extra water were provided ad libitum throughout the study. Data on sow feed and water intake, plasma protein concentration (TPP), body weight, and re-mating period, and on litter size, body weight, and survival of piglets, were collected for two reproduction cycles. NMW sows had higher water intake than Control and NM sows (14.7, 4.5, and 4.5 L/day, respectively, SE = 0.2). The weight loss from 2 weeks prior to farrowing until weaning was smaller in NMW than in NM and Control sows (16.0, 23.8, and 22.9 kg, respectively, SE = 0.9). TPP dropped from farrowing until 21 days of lactation in NMW sows, whereas it increased or was unchanged in NM and Control sows. The re-mating period was shorter and the number of litters/year was higher in NMW than in Control and NM sows (2.2, 2.0, and 2.0, respectively, SE = 0.01). Piglet mortality was lower in NMW than in Control and NM (9.5, 43.9, and 26.7%, respectively, SE = 4.9). Piglets in NMW were heavier at weaning and had higher daily weight gain than Control and NM piglets. It was concluded that providing water ad libitum and nesting material improved piglet survival and growth, and that providing water ad libitum improved sow physiological and reproductive fitness. However, provision of nesting material without access to ad libitum water might increase susceptibility to heat stress in sows.

BACKGROUND: The purpose of this study was to investigate how inclusion of chicory forage or ribwort forage in a cereal-based diet influenced the fecal microbial community (microbiome) in newly weaned (35 days of age) piglets. The piglets were fed a cereal-based diet without (B) and with inclusion (80 and 160 g/kg air-dry forage) of vegetative shoots of chicory (C) and leaves of ribwort (R) forage in a 35-day growth trial. Fecal samples were collected at the start (D0), 17 (D17) and 35 (D35) days after weaning and profiles of the microbial consortia were generated using terminal restriction fragment length polymorphism (T-RFLP). 454-FLX pyrosequencing of 16S rRNA gene amplicons was used to analyze the microbial composition in a subset of the samples already analyzed with T-RFLP. RESULTS: The microbial clustering pattern was primarily dependent on age of the pigs, but diet effects could also be observed. Lactobacilli and enterobacteria were more abundant at D0, whereas the genera Streptococcus, Treponema, Clostridium, Clostridiaceae1 and Coprococcus were present in higher abundances at D35. Pigs fed ribwort had an increased abundance of sequences classified as Treponema and a reduction in lactobacilli. However, the abundance of Prevotellaceae increased with age in on both the chicory and the ribwort diet. Moreover, there were significant correlations between the abundance of Bacteroides and the digested amount of galactose, uronic acids and total non-starch polysaccharides, and between the abundance of Bacteroidales and the digested amount of xylose. CONCLUSION: This study demonstrated that both chicory and ribwort inclusion in the diet of newly weaned pigs influenced the composition of the fecal microbiota and that digestion of specific dietary components was correlated with species composition of the microbiota. Moreover, this study showed that the gut will be exposed to a dramatic shift in the microbial community structure several weeks after weaning.

AIM: To describe the implementation of a novel first-responder programme in which home care providers equipped with automated external defibrillators (AEDs) were dispatched in parallel with existing emergency medical services in the event of a suspected out-of-hospital cardiac arrest (OHCA). METHODS: We evaluated a one-year prospective study that trained home care providers in performing cardiopulmonary resuscitation (CPR) and using an AED in cases of suspected OHCA. Data were collected from cardiac arrest case files, case files from each provider dispatch and a survey among dispatched providers. The study was conducted in a rural district in Denmark. RESULTS: Home care providers were dispatched to 28 of the 60 OHCAs that occurred in the study period. In ten cases the providers arrived before the ambulance service and subsequently performed CPR. AED analysis was executed in three cases and shock was delivered in one case. For 26 of the 28 cases, the cardiac arrest occurred in a private home. Ninety-five per cent of the providers who had been dispatched to a cardiac arrest reported feeling prepared for managing the initial resuscitation, including use of AED. CONCLUSION: Home care providers are suited to act as first-responders in predominantly rural and residential districts. Future follow-up will allow further evaluation of home care provider arrivals and patient survival.

Heat shock proteins (HSPs) are a set of highly conserved proteins that can serve as intestinal gate keepers in gut homeostasis. Here, effects of a probiotic, Lactobacillus rhamnosus GG (LGG), and two novel porcine isolates, Lactobacillus johnsonii strain P47-HY and Lactobacillus reuteri strain P43-HUV, on cytoprotective HSP expression and gut barrier function, were investigated in a porcine IPEC-J2 intestinal epithelial cell line model. The IPEC-J2 cells polarized on a permeable filter exhibited villus-like cell phenotype with development of apical microvilli. Western blot analysis detected HSP expression in IPEC-J2 and revealed that L. johnsonii and L. reuteri strains were able to significantly induce HSP27, despite high basal expression in IPEC-J2, whereas LGG did not. For HSP72, only the supernatant of L. reuteri induced the expression, which was comparable to the heat shock treatment, which indicated that HSP72 expression was more stimulus specific. The protective effect of lactobacilli was further studied in IPEC-J2 under an enterotoxigenic Escherichia coli (ETEC) challenge. ETEC caused intestinal barrier destruction, as reflected by loss of cell-cell contact, reduced IPEC-J2 cell viability and transepithelial electrical resistance, and disruption of tight junction protein zonula occludens-1. In contrast, the L. reuteri treatment substantially counteracted these detrimental effects and preserved the barrier function. L. johnsonii and LGG also achieved barrier protection, partly by directly inhibiting ETEC attachment. Together, the results indicate that specific strains of Lactobacillus can enhance gut barrier function through cytoprotective HSP induction and fortify the cell protection against ETEC challenge through tight junction protein modulation and direct interaction with pathogens.

Trillions of microorganisms that inhabit the intestinal tract form a diverse and intricate ecosystem with a deeply embedded symbiotic relationship with their hosts. As more detailed information on gut microbiota complexity and functional diversity accumulates, we are learning more about how diet-microbiota interactions can influence the immune system within and outside the gut and host health in general. Heat shock proteins are a set of highly conserved proteins that are present in all types of cells, from microbes to mammals. These proteins carry out crucial intracellular housekeeping functions and unexpected extracellular immuno-regulatory features in order to maintain the mucosal barrier integrity and gut homeostasis. It is becoming evident that the enteric microbiota is one of the major determinants of heat shock protein production in intestinal epithelial cells. This review will focus on the interactions between diet, gut microbiota and their role for regulating heat shock protein production and, furthermore, how these interactions influence the immune system and the integrity of the mucosal barrier.

Optimization of AZD6482 (2), the first antiplatelet PI3Kß inhibitor evaluated in man, focused on improving the pharmacokinetic profile to a level compatible with once daily oral dosing as well as achieving adequate selectivity towards PI3Kα to minimize the risk for insulin resistance. Structure-based design and optimization of DMPK properties resulted in (R)-16, a novel, orally bioavailable PI3Kß inhibitor with potent in vivo anti-thrombotic effect with excellent separation to bleeding risk and insulin resistance.

Chitinolytic activity, nutrient uptake and intestinal barrier functions were investigated in gut tissues of Arctic charr (Salvelinus alpinus) fed iso-nitrogenous diets based on fish meal, with or without inclusion of zygomycetes (Rhizopus oryzae). We found that gut tissue of Arctic charr had significant chitinase activity, of both endo- and exo-chitinase iso-forms. Moreover, the distribution pattern along the GI tract of Arctic charr differed between endo-chitinase and exo-chitinase. The endo-chitinase activity in stomach tissue and in the distal intestine was several hundred-fold higher than the exo-chitinase activity in stomach tissue. The greatest exo-chitinase activity was found in the distal intestine. The zygomycete-based diet resulted in higher chitinolytic activity in gut tissue compared to the fish meal-based diet. Disturbed intestinal integrity and increased uptake rate of the amino acid lysine were observed in the distal, but not proximal, intestine of fish fed the zygomycete-based feed.

The gastrointestinal (GI) tract of mammals is inhabited by trillions of microorganisms, resulting in exceedingly complex networking. The interaction between distinct bacterial species and the host immune system is essential in maintaining homeostasis in the gut ecosystem. For instance, the gut commensal microbiota dictates intestinal mucosa maturation and its abundant immune components, such as cytoprotective heat shock proteins (HSP). Here we examined physiological expression of HSP in the normal porcine GI tract and found it to be gut region- and cell type-specific in response to dietary components, microbes, and microbial metabolites to which the mucosa surface is exposed. Correlations between HSP72 expression and ileal Lactobacillus spp. and colonic clostridia species, and between HSP27 expression and uronic acid ingestion, were important interplays identified here. Thus this study provides novel insights into host-microbe interactions shaping the immune system that are modifiable by dietary regime.

Dietary fiber is associated with impaired nutrient utilization and reduced net energy values. However, fiber has to be included in the diet to maintain normal physiological functions in the digestive tract. Moreover, the negative impact of dietary fiber will be determined by the fiber properties and may differ considerably between fiber sources. Various techniques can be applied to enhance nutritional value and utilization of available feed resources. In addition, the extent of fiber utilization is affected by the age of the pig and the pig breed. The use of potential prebiotic effects of dietary fiber is an attractive way to stimulate gut health and thereby minimize the use of anti-microbial growth promoters. Inclusion of soluble non-starch polysaccharides (NSP) in the diet can stimulate the growth of commensal gut microbes. Inclusion of NSP from chicory results in changes in gut micro-environment and gut morphology of pigs, while growth performance remains unaffected and digestibility was only marginally reduced. The fermentation products and pH in digesta responded to diet type and were correlated with shifts in the microbiota. Interestingly, fiber intake will have an impact on the expression of intestinal epithelial heat-shock proteins in the pig. Heat-shock proteins have an important physiological role in the gut and carry out crucial housekeeping functions in order to maintain the mucosal barrier integrity. Thus, there are increasing evidence showing that fiber can have prebiotic effects in pigs due to interactions with the gut micro-environment and the gut associated immune system.

Dietary fiber, resistant to host-mediated digestion in the small intestine due to lack of endogenous enzymes, impacts many facets of animal health and is associated with gut development especially in young monogastrics. Furthermore, it can be used as in-feed antibiotic alternative. Chicory (Cichorium intybus L.) forage with high content of pectin (uronic acids as building blocks) is a novel class of dietary fiber that is chemically different from cereal grains (with high content of arabinoxylans). In the present study, we investigated effects of dietary inclusion of chicory forage on digestibility, gut morphology and microbiota in broilers and young pigs. In the chicken experiment, 160 1-d old broiler chicks were fed 3 nutritionally balanced diets for 30 d including a cereal-based diet and 2 diets with part of the cereals substituted with 60 and 120 g/kg chicory forage (CF60 and CF120), whereas in the pig experiment, 18 seven-wk old Yorkshire pigs were fed 3 diets for 18 d including a cereal-based diet and 2 diets with 80 and 160 g/kg chicory forage inclusion (CF80 and CF160). Our results showed that young pigs were capable to utilize chicory forage well with higher total tract apparent digestibility (TTAD) of all fiber fractions, particularly uronic acid, compared with the control (P < 0.01). In contrast, a decreased TTAD of all fiber fractions was observed in chickens fed on diet CF120 (P < 0.05). Moreover, diet induced changes in gut morphology were observed in the large intestine of chickens. The alteration of cecal mucosal thickness was further positively correlated with TTAD of non-starch polysaccharides (NSP) and its constituent sugars (P < 0.05). In addition, in pigs, terminal restriction fragment length polymorphism (T-RFLP) analysis of intestinal microbiota revealed substantial dietary effects (cereal control diet vs. chicory forage inclusion) on the relative abundance of 2 dominant bacterial phylotypes (Prevotella sp. vs. Roseburia sp.) respectively (P < 0.05). In conclusion, our data showed that chicory forage (Cichorium intybus L.), a novel dietary fiber source in animal nutrition, have potential beneficial properties as fiber ingredient in diets for both pigs and chickens.

The effect of replacing soybean crude protein (CP) with legume leaf meal (LLM) CP on feed intake, growth performance and carcass traits was studied in native female Moo Lath Lao pigs. The diets comprised one traditional diet (T) without soybean meal, one control diet (C) with soybean meal and six diets iso-nitrogenous with diet C in which soybean protein was replaced (33, 66 and 100 % of CP) with LLM CP from porcupine joint vetch (PLM) or Stylosanthes (SLM). Feed and water were offered ad libitum. Replacing soybean CP with LLM CP reduced (P < 0.05) intake of dry matter (DMI), CP (CPI), metabolisable energy (MEI), final body weight (BW) and average daily gain (ADG), but had no effect (P > 0.05) on feed conversion ratio (FCR). Increasing the replacement rate of soybean CP with LLM CP had a negative linear effect (P < 0.001) on DMI, CPI, MEI, final BW and ADG, and on all carcass traits except lean meat percentage. There were no differences in dressing percentage or organ weight and length between treatments. Supplementing diet T with soybean meal resulted in higher (P < 0.05) DMI, CPI and MEI, lower (P < 0.05) FCR, and higher (P < 0.05) final BW and ADG. Slaughter weight, hot carcass weight, eye muscle thickness, back fat, fat in carcass and lean meat were lower (P < 0.05) with diet T than diet C. In conclusion, LLM from stylo and porcupine joint vetch can be used as a CP source to partially replace soybean meal CP in the diet of growing native female Moo Lath Lao pigs.

This study identified the indigenous criteria used by livestock farmers in urban and peri-urban areas of Kampala to assess the nutritional quality of available feed resources. Focus group discussions and questionnaire interviews (with a total of 120 livestock farming households) were conducted. The findings showed that banana peels, leftover food and own-mixed feeds were the most commonly used feed resources for cattle, pigs and chickens, respectively. Farmers use several indigenous criteria to judge the nutritional quality of the available feed resources. These included perceived effects on disease resistance, feed intake, growth/body condition, hair coat appearance, faecal output, faecal texture and level of production, among others. According to farmers, animals offered with a feed resource of good nutritional quality are more resistant to diseases, ingest much of the feed, gain weight with well-filled bodies, have smooth hair coats, produce large quantities of faeces that are not too firm or watery and exhibit good performance (lactating cows produce more milk, sows produce piglets of good body size, hens lay more eggs of normal size, etc.). Although this indigenous knowledge exists, farmers put more importance on availability and cost as opposed to nutritional quality when choosing feed resources. This explains why banana peels were among the feed resources perceived to be of low nutritional quality but, at the same time, were found to be the most commonly used. Hence, there is a need to sensitise farmers on the importance of nutritional quality in ensuring better and efficient utilisation of the available feed resources.

Thirty castrated male Moo Lath pigs (6-8 weeks of age) were used in a 15-week growth trial to study the effect of high dietary sodium chloride (NaCl) content on feed and water intake, performance, sodium (Na) and potassium (K) balance, and plasma aldosterone concentration. The pigs were randomly allocated (ten per treatment) to diets containing 0.24 % Na (Na0.24), 0.28 % Na (Na0.28), and 0.32 % Na (Na0.32) per kg diet. Feed and water was provided ad libitum, and water consumption, feed offered, and feed residues were recorded daily. Every third week, the pigs were weighed, blood samples were collected, and a 3-day total collection of urine and feces was performed. Water intake was higher (P = 0.001) in pigs fed with diets Na0.28 (3.7 L/day) and Na0.32 (3.9 L/day) than in pigs fed with diet Na0.24 (3.4 L/day), and dry matter (DM) intake was higher on diet Na0.32 (P = 0.041) than on the other diets. The average daily body weight (BW) gain was higher on diet Na0.32 than on the other diets (P = 0.031). The feed conversion ratio (in kilogram feed DM to kilogram BW gain) was 4.6, 4.6, and 4.1 on treatments Na0.24, Na0.28, and Na0.32, respectively (P = 0.14). The highest Na balance was observed on diet Na0.32 followed by diets Na0.28 and Na0.24 (P < 0.001), while there was no treatment-related pattern for the K balance. The Na/K ratio in feces and urine increased (P < 0.001), and the K/Na ratio in feces (P < 0.001) decreased with increasing Na content in the diet. Plasma aldosterone concentration decreased (P < 0.001) with increasing dietary content of Na. These results indicate that high NaCl intake and free access to water will increase Na balance but do not negatively influence feed intake and performance of growing local pigs.

The present study examined the impact of replacing crude protein (CP) from soybean with CP from ensiled stylo (ES) on growth performance, carcass traits and organ weights in Landrace × Yorkshire (LY) and Moo Lath (ML) native Lao pigs. Twenty-four castrated male pigs, 12 of each breed, were allocated to the diet treatments according to a completely randomised 3 × 2 factorial (three ES levels × two breeds) arrangement, with four pigs per diet treatment. Pigs were kept in individual pens and fed at 4 % dry matter of body weight for 98 days. The control diet was formulated with soybean meal as the main CP source, and in the other two experimental diets, CP from soybean was replaced at 25 % (ES25) and 50 % (ES50) by CP from ES. Calculated metabolisable energy intake decreased with ES50 diet, while dry matter intake (DMI) and CP intake (CPI) were the highest in ES25 diet (P < 0.001). Overall, average daily gain (ADG) and feed conversion ratio (FCR) were unaffected by diet treatments. Carcass weight, backfat thickness and dressing percentage were unaffected by soybean CP replacement, while the weights of lung, large intestine and stomach were higher (P < 0.001) when 25 % of soybean CP was replaced by CP from ES. LY pigs had higher (P < 0.001) DMI, CPI and ADG and poorer (P < 0.001) FCR than ML pigs. LY pigs had higher carcass weight (P < 0.001), lower backfat thickness (P < 0.001) and higher organ weight (P < 0.001) than ML pigs, except for small intestine weight, where there was no difference between the LY and ML pigs (P > 0.05). In conclusion, ES can replace up to 50 % of soybean CP in the diet of growing Lao LY and ML pigs without negative effects on performance and carcass traits.

The aim of the present study was to investigate feed allowances, body weight (BW), haematocrit, haemoglobin, plasma ionised calcium (iCa), sodium, potassium, pH and glucose concentration and faecal K/Na ratio in local growing pigs, sows and piglets kept by small-holder farmers in Laos. Starting hypotheses were that (1) local pigs are under fed, (2) BW is higher in pigs receiving supplementary feed and (3) the blood profile of pigs subjected to very poor nutrition is outside the normal range. On 54 pig-keeping smallholdings in Borikhamxay province, Lao PDR, daily feed allowances were weighed and BW recorded for 27 lactating sows, 54 piglets and 27 growing pigs. Blood samples were collected from the vena jugularis in all pigs. Feed supplementation did not affect BW, but plasma iCa concentration was outside the normal range for all pigs. There was a tendency for lower faecal K/Na ratio in Na-supplemented sows. The results confirm that local pigs in small-scale production systems in Laos suffer from poor nutrition. The most important challenge for farmers appears to be provision of more feed, particularly feed with a high Ca content.
