RESUMEN

Oxaliplatin (L-OHP) is one of the effective chemotherapeutic drugs for colorectal cancer (CRC). Further investigation into the molecular mechanism of chemoresistance could improve outcomes for patients with colorectal cancer. Recently, microRNAs have been reported as a key in drug resistance of tumors. In this study, we aimed to investigate the effects of miR-153-5p in L-OHP-resistant CRC cells, and its underlying mechanism. Downregulation of miR-153-5p was observed in CRC cells, while upregulation of miR-153-5p enhances the chemosensitivity of CRC/L-OHP cells. The autophagy of CRC/L-OHP cells was markedly increased after exposure to L-OHP but abolished by the upregulation of miR-153-5p. Dual-luciferase reporter assays validated that Bcl-2 was a direct target of miR-153-5p. In conclusion, our data suggested that miR-153-5p was a mediator of cisplatin resistance in colorectal cancer by affecting Bcl-2-mediated autophagy, indicating a new therapeutic target for CRC treatment.

RESUMEN

BACKGROUND: Emerging data has suggested that Tregs, Th17, Th1 and Th2 are correlated with early immune mechanisms by controlling Plasmodium infection. Plasmodium infection appeared to impair the antigen presentation and maturation of DCs, leading to attenuation of specific cellular immune response ultimately. Hence, in this study, we aim to evaluate the relevance between DCs and Tregs/Th17 populations in the process and outcomes of infection with Plasmodium yoelii 17XL (P.y17XL). METHODS: DCs detection/analysis dynamically was performed by Tregs depletion or Th17 neutralization in P.y17XL infected BALB/c mice via flow cytometry. Then the levels of cytokines production were detected using enzyme-linked mmunosorbent assay (ELISA). RESULTS: Our results indicated that Tregs depletion or Th17 neutralization in BALB/c mice infected with P.y17XL significantly up-regulated the percentages of mDC and pDC, increased the expressions of major histocompatibility complex (MHC) class II, CD80, CD86 on DCs and the levels of IL-10/IL-12 secreted by DCs, indicating that abnormal amplification of Tregs or Th17 may damage the maturation and function of DCs during the early stage of malaria infection. Interestingly, we also found that the abnormal amplification of Th17, as well as Tregs, could inhibit the maturation of DCs. CONCLUSIONS: Tregs skewing or Th17 amplification during the early stage of malaria infection may inhibit the maturation and function of DCs by modifying the subsets of DCs, expressions of surface molecules on DCs and secretion mode of cytokines.

RESUMEN

Batch equilibrium experiments were used to reveal cyromazine adsorption on five kinds of soils, namely Ali-Perudic Ferrosols collected from Yingtan of Jiangxi, Udic Argosols collected from Nanjing and Gleyic-Stagnic Anthrosols collected from Changshu of Jiangsu, Ustic Cambosols collected from Fengqiu of Henan, and Udic Isohumosols collected from Hailun of Heilongjiang. Results show that the experimental data are best described by the Freundlich and Langmuir model, while fitted successfully by the linear model. Different adsorption behaviors of Cyromazine are observed in the five tested soils, with the lgK(f) values varying from 1.6505 (cambosols), 1.6715 (argosols) and 1.7153 (ferrosols) to 2.4579 (anthrosols) and 2.6557 (isohumosols). Moreover, the Kf values are in a positive correlation to the OM of the soil (r = 0.989) but significantly negative correlated to soil pH (r = -0.938). The free energy of sorption ranged from -20.8 to -23.0 kJ/mol, indicated that the adsorption could be largely attributed to the physical adsorption.

RESUMEN

OBJECTIVE: To study the mechanism of microelement including Cu and Zn on the accumulation of three danshinones in Salviae miltiorrhizae root and build a theory basis for its good quality and high yield. METHOD: Sand culture experiments were conducted to study the effect of Cu and Zn on the accumulation of three danshinones and oxidase including peroxidase and polyphenol oxidase activity in the plant root. The correlation between available Cu and Zn contents in matrix and oxidase activity in the plant root and, the correlation between available Cu and Zn contents in matrix and contents of tanshinones in the root were discussed. RESULT: Contents of danshinones in the root increased with the increasing of Cu and Zn concentration. Dynamic monitoring on contents of dan-shinones of the plant roots growing in the pots with different Cu and Zn concentration in the whole growing season showed that the contents of danshinones for 60 days were the lowest, for 120 days the highest and then dropped for 150 days. In addition, among available Cu and Zn contents of matrix, oxidase including peroxidase and polyphenol oxidase activity and contents of tanshinones in the root, the correlation between two factors were significant difference (P < 0.05 or P < 0.01). CONCLUSION: The mechanism of Cu and Zn on the accumulation of danshinones may be that Cu and Zn improve the activity of peroxidase and polyphenol oxidase, which promote transformation of phenolic compounds to terpenes and therefore to increase contents of danshinones.

RESUMEN

OBJECTIVE: To observe the preventive protection of Kangdu Bufei Decoction (KBD) on acute severe respiratory syndrome (SARS) in medical personnel of Hong Kong at the epidemic period. METHODS: At the epidemic period of SARS in Hong Kong, there were 2,601 medical staffs administered with KBD distributed by the Center. It was confirmed that 1,063 persons had taken it for successive two weeks according to the request and returned the effective questionnaire, they were regarded as the TCM group. The control group was consisted of 15,374 subjects who didn't take Chinese herbal medicine. Contents of the questionnaire including the condition of testee in terms of quality of life (QOL), changes of influenza-like symptoms and Warm disease symptoms. Serum immunological examination had been conducted in 37 of the persons in the TCM group. RESULTS: No one in the TCM group got SARS infection while in the control group, 64 (0.4%) had got. Self-control before and after treatment examination showed that significant improvement appeared in the TCM in aspects of influenza-like and Warm disease symptoms and QOL. The results of serum immunological examination showed that after administration of KBD, the immunity of organism was improved and elevated. CONCLUSION: KBD could prevent the occurrence of SARS, it is possibly realized through improving symptoms, elevating QOL and enhancing immunity of organism.

RESUMEN

OBJECTIVE: To study the changes of soil fertility in Sheyang county where Chrysanthemum morifolium has been cultivated for more than 30 years and to develop the special fertilizer for cultivation of C. morifolium. METHOD: The pH values, organic matter and the contents of total and available N, P, K and Zn in the soil layer of 0 to 40 cm, as well as the total N, P, K and Zn contents in the flowers, roots, stems and leaves of the plants, were analysed. The balanced fertilization plan for cultivation of C. morifolium was put forward. In addition, the formula of special fertilizer for cultivation of C. morifolium was determined according to flower yield and utilization rate of N, P, and K. RESULTS AND CONCLUSION: The soil had high pH values and high soil salt contents, with unbalanced application of N, P, and K fertilizers and a shortage of available Zn after cultivation of C. morifolium. The contents of soil organic carbon, N and P declined with increasing cultivation time of C. morifolium, which resulted from the improper rotations and fertilization. The balance fertilization practice and the special fertilizer utilization are effective ways to improve soil fertility for C. morifolium.
