RESUMEN

The size and condition of the peripheral CD4 T cell population determine the capacity of the immune response. Under homeostatic conditions, the size of the peripheral CD4 T cell population is maintained through turnover and survival. However, the underlying mechanisms remain inadequately understood. Here, we observed a significant decrease in the percentage of CD4 T cells in the periphery following the targeted deletion of the Paxbp1 gene in mouse T cells. In the absence of Paxbp1, naïve CD4 T cells displayed reduced surface interleukin-7 receptor levels and a decreased capacity to respond to survival signals mediated by interleukin-7. In addition, naïve CD4 T cells deficient in Paxbp1 demonstrated impaired T cell antigen receptor signalling, compromised cell cycle entry, decreased proliferation, and increased apoptosis following stimulation, all of which contributed to the reduction in the number of peripheral CD4 T cells. Therefore, our study highlights the indispensable role of Paxbp1 in maintaining peripheral CD4 T cell homeostasis.

Accurate and robust medical image segmentation is crucial for assisting disease diagnosis, making treatment plan, and monitoring disease progression. Adaptive to different scale variations and regions of interest is essential for high accuracy in automatic segmentation methods. Existing methods based on the U-shaped architecture respectively tackling intra- and inter-scale problem with a hierarchical encoder, however, are restricted by the scope of multi-scale modeling. In addition, global attention and scaling attention in regions of interest have not been appropriately adopted, especially for the salient features. To address these two issues, we propose a ConvNet-Transformer hybrid framework named SSCFormer for accurate and versatile medical image segmentation. The intra-scale ResInception and inter-scale transformer bridge are designed to collaboratively capture the intra- and inter-scale features, facilitating the interaction of small-scale disparity information at a single stage with large-scale from multiple stages. Global attention and scaling attention are cleverly integrated from a spatial-channel-aware perspective. The proposed SSCFormer is tested on four different medical image segmentation tasks. Comprehensive experimental results show that SSCFormer outperforms the current state-of-the-art methods.

Population genomics analysis holds great potential for informing conservation of endangered populations. We focused on a controversial case of European whitefish (Coregonus spp.) populations. The endangered North Sea houting is the only coregonid fish that tolerates oceanic salinities and was previously considered a species (C. oxyrhinchus) distinct from European lake whitefish (C. lavaretus). However, no firm evidence for genetic-based salinity adaptation has been available. Also, studies based on microsatellite and mitogenome data suggested surprisingly recent divergence (c. 2500 years bp) between houting and lake whitefish. These data types furthermore have provided no evidence for possible inbreeding. Finally, a controversial taxonomic revision recently classified all whitefish in the region as C. maraena, calling conservation priorities of houting into question. We used whole-genome and ddRAD sequencing to analyse six lake whitefish populations and the only extant indigenous houting population. Demographic inference indicated post-glacial expansion and divergence between lake whitefish and houting occurring not long after the Last Glaciation, implying deeper population histories than previous analyses. Runs of homozygosity analysis suggested not only high inbreeding (FROH up to 30.6%) in some freshwater populations but also FROH up to 10.6% in the houting prompting conservation concerns. Finally, outlier scans provided evidence for adaptation to high salinities in the houting. Applying a framework for defining conservation units based on current and historical reproductive isolation and adaptive divergence led us to recommend that the houting be treated as a separate conservation unit regardless of species status. In total, the results underscore the potential of genomics to inform conservation practices, in this case clarifying conservation units and highlighting populations of concern.

The key feature of non-contact temperature measurement provided by infrared (IR) cameras underpins their versatility. However, the accuracy of temperature measurements with IR cameras depends on imaging quality due to their non-contact nature, such as the lens, body temperature, and measurement environment. This paper addresses the correction of radial distortion and nonlinear response issues in IR cameras. To address radial distortion, we have designed a passive checkerboard calibration board specifically for infrared cameras. This board is used to calibrate the IR camera and derive the necessary camera parameters. Subsequently, these parameters are applied during the actual measurement process to rectify radial distortion effectively. Building on the radial distortion correction method mentioned above, we propose a multi-point segmented calibration approach that considers different temperature ranges and imaging regions. This method alleviates the issue of reduced temperature measurement accuracy due to variations in camera responses by computing gain and offset coefficient matrices for each temperature range. Experimental results demonstrate the effectiveness of the calibration board in correcting radial distortion in IR cameras, with a mean reprojection error of less than 0.16 pixels. Regarding the nonlinear response problem, the introduced method significantly reduces the relative error in temperature measurement. In the verification phase, spanning from 100 to 500 °C, the average relative error in temperature measurement decreases by 0.49% from 1.61% before and after correction, which highlights a substantial improvement in temperature measurement accuracy. This work gives a useful reference to improve the imaging quality and temperature measurement accuracy using infrared cameras.

Naked-eye 3D display technology has excellent 3D visual effects and does not require wearable devices assistance. It can present the depth, position and complex structure information of 3D medical images, allowing viewers to obtain information about tissues and organs from different points, reducing cognitive load, contributing to medical teaching and opening up innovative methods for planning and diagnosis. Naked-eye 3D augmented reality display can display medical images in real 3D space, achieving virtual and real vision. It helps a lot to medical research. The applications of naked-eye 3D display technology in three major aspects of medical diagnosis, clinical surgery and rehabilitation training is reviewed in the study. It provides the direction for the subsequent research in medical field, thus assisting medical research and improving medical practice.

OBJECTIVES: In vitro fenestration of stent-graft (IVFS) demands high-precision navigation methods to achieve optimal surgical outcomes. This study aims to propose an augmented reality (AR) navigation method for IVFS, which can provide in situ overlay display to locate fenestration positions. METHODS: We propose an AR navigation method to assist doctors in performing IVFS. A deep learning-based aorta segmentation algorithm is used to achieve automatic and rapid aorta segmentation. The Vuforia-based virtual-real registration and marker recognition algorithm are integrated to ensure accurate in situ AR image. RESULTS: The proposed method can provide three-dimensional in situ AR image, and the fiducial registration error after virtual-real registration is 2.070 mm. The aorta segmentation experiment obtains dice similarity coefficient of 91.12% and Hausdorff distance of 2.59, better than conventional algorithms before improvement. CONCLUSIONS: The proposed method can intuitively and accurately locate fenestration positions, and therefore can assist doctors in performing IVFS.

C-C bond forming reaction by alkylation of aryl rings is a main pillar of chemistry in the production of broad portfolios of chemical products. The dominant mechanism proceeds via electrophilic substitution of secondary and tertiary carbocations over acid catalysts, forming multiple aryl alkylation products non-selectively through all secondary and tertiary carbons in the alkyl chains but producing little α-C alkylation products because primary carbocations are poorly stable. Herein, we report that anatase TiO2 (TiO2-A) catalyzes nucleophilic α-C alkylation of phenols with alcohols in high selectivity to simply linear alkylphenols. Experimental and computational studies reveal the formation of Ti=C- bond with the α-carbon of the alkyl group at oxygen vacancies of the TiO2-A surface. The subsequent α-C alkylation by selective substitution of phenol ortho-C-H bond is verified by deuterium exchanged substrate and DFT calculations.

Phenols are important building blocks widely applied in many fields. The pronounced orientational effect of the phenolic hydroxyl group makes achieving selective synthesis of meta-phenols challenging. Accessing meta-phenols needs lengthy synthetic sequences. Herein, we first developed a heterogeneous CO2-mediated CeO2-5CuO catalyst for decarboxylative oxidation of benzoic acids with a more than 80% selectivity to meta-phenols. This technology is based on a traceless directing group relay method. The CeO2-CuO catalysts with different Ce/Cu ratios exhibited controllable reaction selectivity between decarboxylation and decarboxylative oxidation. Spectroscopy experiments and computational studies showed the adsorption state of benzoic acid was found to be crucial for subsequent reaction pathways. The moderate adsorption on CO2-mediated CeO2-5CuO catalyst contributes to the distinct selectivity of phenol. Furthermore, the paddlewheel intermediate facilitates the synthesis of meta-phenols from benzoic acids. This traceless directing group method would promote the development of useful one-pot meta-substituted phenols from bio-based benzoic acids.

The lifespan of double-positive (DP) thymocytes is critical for intrathymic development and shaping the peripheral T cell repertoire. However, the molecular mechanisms that control DP thymocyte survival remain poorly understood. Paxbp1 is a conserved nuclear protein that has been reported to play important roles in cell growth and development. Its high expression in T cells suggests a possible role in T cell development. Here, we observed that deletion of Paxbp1 resulted in thymic atrophy in mice lacking Paxbp1 in the early stages of T cell development. Conditional loss of Paxbp1 resulted in fewer CD4+CD8+ DP T cells, CD4 and CD8 single positive (SP) T cells in the thymus, and fewer T cells in the periphery. Meanwhile, Paxbp1 deficiency had limited effects on the CD4-CD8- double negative (DN) or immature single-positive (ISP) cell populations. Instead, we observed a significant increase in the susceptibility of Paxbp1-deficient DP thymocytes to apoptosis. Consistent with this, RNA-Seq analysis revealed a significant enrichment of the apoptotic pathway within differentially expressed genes in Paxbp1-deficient DP cells compared to control DP cells. Together, our results suggest a new function for Paxbp1, which is an important mediator of DP thymocyte survival and critical for proper thymic development.

Anthropogenic biological invasions represent major concerns but enable us to investigate rapid evolutionary changes and adaptation to novel environments. The goldfish Carassius auratus with sexual diploids and unisexual triploids coexisting in natural waters is one of the most widespread invasive fishes in Tibet, providing an ideal model to study evolutionary processes during invasion in different reproductive forms from the same vertebrate. Here, using whole-genome resequencing data of 151 C. auratus individuals from invasive and native ranges, we found different patterns of genomic responses between diploid and triploid populations during their invasion of Tibet. For diploids, although invasive individuals derived from two different genetically distinct sources had a relative higher diversity (π) at the population level, their individual genetic diversity (genome-wide observed heterozygosity) was significantly lower (21.4%) than that of source individuals. Population structure analysis revealed that the invasive individuals formed a specific genetic cluster distinct from the source populations. Runs of homozygosity analysis showed low inbreeding only in invasive individuals, and only the invasive population experienced a recent decline in effective population size reflecting founder events. For triploids, however, invasive populations showed no loss of individual genetic diversity and no genetic differentiation relative to source populations. Regions of putative selective sweeps between invasive and source populations of diploids mainly involved genes associated with mannosidase activity and embryo development. Our results suggest that invasive diploids deriving from distinct sources still lost individual genetic diversity resulting from recent inbreeding and founder events and selective sweeps, and invasive triploids experienced no change in genetic diversity owing to their reproduction mode of gynogenesis that precludes inbreeding and founder effects and may make them more powerful invaders.

OBJECTIVE: Among various assessment paradigms, the cardiopulmonary exercise test (CPET) provides rich evidence as part of the cardiopulmonary endurance (CPE) assessment. However, methods and strategies for interpreting CPET results are not in agreement. The purpose of this study is to validate the possibility of using machine learning to evaluate CPET data for automatically classifying the CPE level of workers in high-latitude areas. METHODS: A total of 120 eligible workers were selected for this cardiopulmonary exercise experiment, and the physiological data and completion of the experiment were recorded in the simulated high-latitude workplace, within which 84 sets of data were used for XGBOOST model training and36 were used for the model validation. The model performance was compared with Support Vector Machine and Random Forest. Furthermore, hyperparameter optimization was applied to the XGBOOST model by using a genetic algorithm. RESULTS: The model was verified by the method of tenfold cross validation; the correct rate was 0.861, with a Micro-F1 Score of 0.864. Compared with RF and SVM, all data achieved a better performance. CONCLUSION: With a relatively small number of training samples, the GA-XGBOOST model fits well with the training set data, which can effectively evaluate the CPE level of subjects, and is expected to provide automatic CPE evaluation for selecting, training, and protecting the working population in plateau areas.

Understanding the role of genetic and nongenetic variants in modulating phenotypes is central to our knowledge of adaptive responses to local conditions and environmental change, particularly in species with such low population genetic diversity that it is likely to limit their evolutionary potential. A first step towards uncovering the molecular mechanisms underlying population-specific responses to the environment is to carry out environmental association studies. We associated climatic variation with genetic, epigenetic and microbiome variation in populations of a social spider with extremely low standing genetic diversity. We identified genetic variants that are associated strongly with environmental variation, particularly with average temperature, a pattern consistent with local adaptation. Variation in DNA methylation in many genes was strongly correlated with a wide set of climate parameters, thereby revealing a different pattern of associations than that of genetic variants, which show strong correlations to a more restricted range of climate parameters. DNA methylation levels were largely independent of cis-genetic variation and of overall genetic population structure, suggesting that DNA methylation can work as an independent mechanism. Microbiome composition also correlated with environmental variation, but most strong associations were with precipitation-related climatic factors. Our results suggest a role for both genetic and nongenetic mechanisms in shaping phenotypic responses to local environments.

The selective activation of C-O ether bonds is an essential tool in organic synthesis and natural polymer depolymerization. However, the direct cleavage of the ether bond is still challenging work, especially breaking this inert and redox-neutral bond to provide one active carbon radical and another oxygen-centered fragment with oxidation capacity that can participate in the controllable radical reaction. We herein report that commercial 2H-MoS2 with negligible acidity can efficiently catalyze the benzylation of arenes with benzyl ethers, and a new Radical-Friedel-Crafts mechanism is proposed, which is quite different from the strong acid-catalyzed Friedel-Crafts mechanism. With dibenzyl ether as the model benzylation reagent, 2H-MoS2 can achieve the homolytic cleavage of the Bn-OR bond to generate the benzyl carbon radical and RO˙ species, identified by EPR measurement and radical trap experiments. The following radical-involved benzylation is confirmed by the Hammett results and a plausible pathway is proposed to clarify the Radical-Friedel-Crafts process. Heterogeneous 2H-MoS2 can be consecutively used four times without regeneration and it offers 94-95% yields of 2-benzyl-1,4-dimethylbenzene from dibenzyl ether and p-xylene in 30 min at 140 °C. Furthermore, this mechanism can provide some inspiration to activate the ether bond and to utilize ether as an oxidant in C-H bond activation.

Objective: The objectives of this systematic review are to describe features and specific application scenarios for current cancer survivorship care services of Artificial intelligence (AI)-driven digital health technologies (DHTs) and to explore the acceptance and briefly evaluate its feasibility in the application process. Methods: Search for literatures published from 2010 to 2022 on sites MEDLINE, IEEE-Xplor, PubMed, Embase, Cochrane Central Register of Controlled Trials and Scopus systematically. The types of literatures include original research, descriptive study, randomized controlled trial, pilot study, and feasible or acceptable study. The literatures above described current status and effectiveness of digital medical technologies based on AI and used in cancer survivorship care services. Additionally, we use QuADS quality assessment tool to evaluate the quality of literatures included in this review. Results: 43 studies that met the inclusion criteria were analyzed and qualitatively synthesized. The current status and results related to the application of AI-driven DHTs in cancer survivorship care were reviewed. Most of these studies were designed specifically for breast cancer survivors' care and focused on the areas of recurrence or secondary cancer prediction, clinical decision support, cancer survivability prediction, population or treatment stratified, anti-cancer treatment-induced adverse reaction prediction, and so on. Applying AI-based DHTs to cancer survivors actually has shown some positive outcomes, including increased motivation of patient-reported outcomes (PROs), reduce fatigue and pain levels, improved quality of life, and physical function. However, current research mostly explored the technology development and formation (testing) phases, with limited-scale population, and single-center trial. Therefore, it is not suitable to draw conclusions that the effectiveness of AI-based DHTs in supportive cancer care, as most of applications are still in the early stage of development and feasibility testing. Conclusions: While digital therapies are promising in the care of cancer patients, more high-quality studies are still needed in the future to demonstrate the effectiveness of digital therapies in cancer care. Studies should explore how to develop uniform standards for measuring patient-related outcomes, ensure the scientific validity of research methods, and emphasize patient and health practitioner involvement in the development and use of technology.

Natural organic matter (NOM) refers to the dissolved organic matter in natural water that can pass through 0.45 µm filter membrane. As a pivotal role in the surface water body, it has a significant effect on the efficiency of AOPs. In this study, Excitation emission matrix - parallel factor (EEM-PARAFAC) analysis is used to elucidate the changes of NOM fluorescence peaks after electrochemical oxidation process, two-dimensional correlation spectroscopy (2D-FTIR-COS) and Fourier transform ion cyclotron resonance mass spectrometry (FTICR-MS) are utilized to clarify the molecular characteristics of NOM in surface water and the effects of electrochemical oxidation on NOM molecules. The results indicate that parts of NOM molecules are mineralized into simple compounds and precursors of refractory organic matters produced by some NOM molecules after AOPs. It is concluded that the precursors of these refractory organic matters may belong to terrestrial humus (C2). Therefore, for the purpose of avoiding more refractory organic pollutants produced by NOM which can reduce the performance of AOPs in the water treatment process, factories should choose water sources with less humus as industrial water supply, or degrade humus by physical or chemical methods before industrial water supply.

The role of methylation in adaptive, developmental and speciation processes has attracted considerable interest, but interpretation of results is complicated by diffuse boundaries between genetic and non-genetic variation. We studied whole genome genetic and methylation variation in the European eel, distributed from subarctic to subtropical environments, but with panmixia precluding genetically based local adaptation beyond single-generation responses. Overall methylation was 70.9%, with hypomethylation predominantly found in promoters and first exons. Redundancy analyses involving juvenile glass eels showed 0.06% and 0.03% of the variance at SNPs to be explained by localities and environmental variables, respectively, with GO terms of genes associated with outliers primarily involving neural system functioning. For CpGs 2.98% and 1.36% of variance was explained by localities and environmental variables. Differentially methylated regions particularly included genes involved in developmental processes, with Hox clusters featuring prominently. Life stage (adult versus glass eels) was the most important source of inter-individual variation in methylation, probably reflecting both ageing and developmental processes. Demethylation of transposable elements relative to pure European eel was observed in European X American eel hybrids, possibly representing postzygotic barriers in this system characterized by prolonged speciation and ongoing gene flow. Whereas the genetic data are consistent with a role of single-generation selective responses, the methylation results underpin the importance of epigenetics in the life cycle of eels and suggest interactions between local environments, development and phenotypic variation mediated by methylation variation. Eels are remarkable by having retained eight Hox clusters, and the results suggest important roles of methylation at Hox genes for adaptive processes.

With the increasing problem of fluoride pollution, it is urgent to find an efficient method to remove fluoride (F-). In this study, a new material goethite-montmorillonite-sorbent (GMS) was prepared and added into the electrocoagulation (EC) reaction to form a new pathway (EC/GMS) for the removal of fluoride. Scanning electron microscopy (SEM), Brunauer-Emmett-Teller (BET), Fourier-transform infrared (FT-IR), X-ray photoelectron spectroscopy (XPS) and other characterization methods were used to analyze the properties of GMS. The fluoride removal performance and mechanism of EC/GMS was studied. The results showed that GMS could provide numerous adsorption sites. EC/GMS could achieve a high removal efficiency of 95.98% and lower energy consumption of 0.58 kW h m-3 for 60 min. EC/GMS could achieve a removal efficiency of 99.47% after optimization by single-factor experiments and RSM-BBD optimal experiments. Meantime, the removal rate of the EC/GMS still reached over 87% after six cycles. The kinetic analysis indicated that the degradation pathways could also achieve a high removal rate for high fluoride-containing concentration solutions within a short time. The stretching vibration of C-F and C-O and the existence of F- revealed that the electrophoresis of the electrodes, adsorption of GMS, and co-precipitation of flocs were the main removal pathways, and the accelerating effect between the electrocoagulation and adsorption process was addressed. This study provides a new pathway for removing fluoride from aqueous environments.

The persulfate advanced oxidation is an emerging and efficient pollutant treatment method, but usually requires the help of other materials or energy to catalyze and produce highly oxidizing active substances. In this paper, electrochemical-assisted ultraviolet light coupled peroxodisulfate system (E-UV-PDS) was used to degrade ciprofloxacin (CIP), and it was determined that electrolysis and ultraviolet photolysis were synergistic by calculation. The effects of initial pH, voltage, peroxodisulfate dosage, CIP concentration and coexisting anions on the degradation process were explored. The quenching experiments showed that 1O2, ⋅OH and SO4-⋅ were the main active oxygen species. Under the following conditions, ultraviolet light = 6 W, voltage = 4 V, [peroxodisulfate] = 20 mM, [pH]0 = 7 and [CIP] = 100 mgL-1, the degradation rate of CIP reached about 100% after 120 min, and the influence of inorganic anions was also discussed. Several intermediate products were identified by LC-MS, and three degradation pathways were speculated for CIP degradation. Finally, economic evaluation of the E-UV-PDS system was made, and it was useful to construct environmentally friendly and low-cost catalytic processes for the efficient degradation of organic pollutants.

Virtual reality (VR) presents opportunities for innovative patient educational methods. This study used a combination of subjective questionnaires and objective physiological measures to investigate the impact of a VR radiotherapy (RT) educational system on patients' understanding and anxiety prior to commencing RT. Sixty patients were randomized to control (n = 30) and intervention (n = 30) groups prior to initiating RT. The control group received the standard nursing care process. The intervention group additionally participated in a detailed introduction to RT positioning, procedures, treatments, and other RT-related information via VR education. All patients completed a data collection from pre- and postintervention, which included questions on RT comprehension, anxiety-related scales, and objective physiological data reflecting the patient's psychological state, such as blood pressure, heart rate, and respiration. Both groups had high anxiety levels before the intervention, and there was no significant difference between the questionnaire and physiological data of the two groups. Following the intervention, anxiety scores (state-trait anxiety scale and visual analog scale) of the intervention group decreased significantly compared with those of the control group, and there was a significant decrease in systolic blood pressure (p < 0.05) and increase in cognitive score (p < 0.05). This study reports the positive impact of a virtual reality radiotherapy (VRRT) patient educational system on increasing patient RT comprehension and reducing anxiety. Further work is needed to improve the acceptability of the system to patients and to explore further the impact of VR education on patients' psychological and physical needs.

Epstein-Barr virus (EBV) infection is prevalent and associated with distinct diseases including infectious mononucleosis (IM), chronic active EBV infection (CAEBV) and NK/T-cell lymphoma (NKTL). However, the specific roles of EBV in these diseases remain unclear. Here, the whole miRNA expression datasets derived from 7 IM, 6 CAEBV, and 3 NKTL biopsies were obtained. Homo sapiens microRNA-372-5p (Hsa-miR-372-5p) was upregulated in both CAEBV and NKTL patients. Overexpression of hsa-miR-372-5p altered the expression of over 100 proteins. In addition, hsa-miR-372-5p may target NIMA related kinase 7 to regulate NLRP3 inflammasome activation in host cell. Taken together, we reported different miRNA expression profiles in distinct EBV associated diseases, which provided novel insights to understand how host miRNAs contribute to the mechanism of EBV associated diseases. Hsa-miR-372-5p, as well as other differential expressed miRNA, might serve as potential targets in the therapy of various EBV associated diseases.
