RESUMEN

A potential cause of amphibian population declines are the impacts of environmental degradation on tadpole development. We conducted RNA sequencing on developing northern leopard frog tadpoles and through de novo transcriptome assembly we annotated a large number of open reading frames comparable in number and extent to genes identified in Xenopus. Using our transcriptome, we found transcript level changes between early (Gosner 26-31) and late (Gosner 36-41) stage tadpoles were the greatest in the tail, which is reabsorbed throughout development. There was an up-regulation of immunity genes in both the head and tail of the late tadpoles and a down-regulation of genes associated with the energy pathways of the mitochondria and the production of myosin. Overall, transcript level changes across development were consistent with studies on Xenopus and our findings highlight the broader utility of using RNA-seq to identify genes differentially expressed throughout development and in response to environmental pressures.

Anuran larvae exhibit behavioral and morphological plasticity in response to perceived predation risk, although response type and magnitude varies through ontogeny. Increased baseline corticosterone is related to morphological response to predation risk, whereas the mechanism behind behavioral plasticity remains enigmatic. Since tadpoles alter behavioral responses to risk immediately upon exposure to predator cues, we characterized changes in whole body corticosterone at an acute (<1h post-exposure) timescale. Tadpoles (Lithobates sylvaticus) at Gosner stage (GS) 25 (free-swimming, feeding larvae) increased corticosterone levels to a peak at 10-20min post-exposure to predator cues, paralleling the acute stress response observed among other taxa. Tadpoles reared for 3weeks (mean GS29) with predation risk (caged, fed Aeshnid dragonfly nymph) had lower corticosterone levels at 10-20min post-exposure to dragonfly cues than predator-naïve controls, suggesting habituation, although the magnitude of increase was markedly diminished when compared to younger tadpoles (GS25). These experiments represent the first assessment of tadpole hormonal responses to predation risk at the acute timescale. Further research is required to establish causality between hormonal responses and behavioral changes, and to examine how and why responsiveness changes over ontogeny and with chronic exposure to risk.

RESUMEN

Peer review is pivotal to science and academia, as it represents a widely accepted strategy for ensuring quality control in scientific research. Yet, the peer-review system is poorly adapted to recent changes in the discipline and current societal needs. We provide historical context for the cultural lag that governs peer review that has eventually led to the system's current structural weaknesses (voluntary review, unstandardized review criteria, decentralized process). We argue that some current attempts to upgrade or otherwise modify the peer-review system are merely sticking-plaster solutions to these fundamental flaws, and therefore are unlikely to resolve them in the long term. We claim that for peer review to be relevant, effective, and contemporary with today's publishing demands across scientific disciplines, its main components need to be redesigned. We propose directional changes that are likely to improve the quality, rigour, and timeliness of peer review, and thereby ensure that this critical process serves the community it was created for.
