BACKGROUND: The traditional herbal medicine Atractylodes macrocephala Koidz. (A. macrocephala) is commonly utilized for alleviating symptoms associated with spleen deficiency, abdominal distension, diarrhea, and constipation. These pharmacological effects are attributed to a variety of active constituents. However, the specific bioactive compounds responsible for promoting defecation and gastrointestinal transit in A. macrocephala remain unidentified. METHODS: The primary polysaccharide characteristics of PAMK was elucidated by HPLC, FT-IR, and HGPGC. Efficacy of PAMK (0.07, 0.14, and 0.28 mg/g) on mice was evaluated in a spleen deficiency constipation mouse model by analyzing stool parameters, constipation-related physiological indexes, and SCFAs. The expression levels of 5-HT3R, 5-HT4R, and related receptor genes were examined by RT-qPCR, and neurotransmitters were examined using ELISA. Finally, the diversity of gut microbiota was analyzed with 16S rDNA sequencing. KEY RESULTS: The results showed that PAMK significantly reduced the gastrointestinal transport time and increased the number of fecal pellets and fecal water content in spleen deficiency constipation model mice. PAMK kept the balance of 5-HT, SCFAs, TPH-1, SERT, CgA, and neurotransmitter levels (VIP, SP, MTL) in mice colon. In addition, PAMK could regulate the abundance of gut microbiota such as Alistopes, Bacteroides, and Odoribacter in spleen deficiency constipation model mice gut. CONCLUSIONS AND INFERENCES: It can be concluded that PAMK effectively ameliorated the symptoms of spleen deficiency constipation in mice by modulating the expression of 5-HT and its associated receptors. The underlying mechanism was elucidated, providing a solid theoretical foundation for the therapeutic application of A. macrocephala in treating spleen deficiency constipation and offering potential for developing novel approaches to address this condition.

Application of elicitors can greatly enhance plant immune resistance against pathogens. However, it is still obscure whether elicitor activity is influenced by diverse sources. This study investigated the effect of foliar spraying of 19 batches of Chrysanthemum indicum polysaccharides (CIPs) on the disease resistance of Atractylodes macrocephala Koidz. (A. macrocephala) and explored the main reasons for the differences of inducing activity of CIP elicitors. PCA, OPLS-DA, grey relational analysis and entropy weight method had good predictability for the activity evaluation of CIP elicitors and other plant-derived elicitors. The results showed that 19 batches of CIPs had definite regional differences in inducing activity and monosaccharide content. CIP elicitors with high inducing activity could significantly increase the accumulation of Atractylenolide Ⅱ and Atractylenolide Ⅲ, the mRNA relative transcription level of CAT, POD, PAL genes, the amount of pH change in the medium and effectively reduce the disease index of A. macrocephala. Furthermore, CIP with high inducing activity exhibited the high contents of Rha, Ara and GalA, which might be the main contributor to their high activity. The evaluation procedure developed in this work can be applied for screening CIP elicitors with high inducing activity, and it lays a foundation for identifying more functional elicitors related to plant immune resistance.

Atractylodes macrocephala polysaccharide (AC1) is extracted from the root of the Chinese herb Atractylodes Macrocephala and is used in the treatment of constipation due to its effects on strengthening cellular immunity and regulating intestinal function. In this study, Metagenomics and Metabolomic are used to analyze the effects of AC1 on the gut microbiota and host metabolites in mice models of constipation. The results show that the abundance of Lachnospiraceae_bacterium_A4, Bact-oides_vulgatus and Prevotella_sp_CAG:891 increased significantly, indicating that AC1-targeted strain modulation effectively alleviated the dysbiosis of the gut microbiota. Besides, the microbial alterations also influenced the metabolic pathways of the mice, including tryptophan metabolism, unsaturated fatty acid synthesis and bile acid metabolism. The physiological parameters of the mice treated with AC1 are improved, such as tryptophan in the colon, 5-hydroxytryptamine (5-HT) and short-chain fatty acids (SCFAs). In conclusion, AC1 as a probiotic can regulate intestinal flora to normal levels and achieve the effect of treating constipation.

During the actual operation of subways, high-speed railway, and other infrastructure, subgrade soil will be subjected to periodic vibration and intermittent loading, i.e., intermittent cyclic load. To study the deformation characteristics of subgrade soil under intermittent cyclic loading, the Newtonian dashpot in the generalized Kelvin model is replaced by the fractional Abel dashpot, a fractional generalized Kelvin model is established. Then, a nonlinear viscoplastic body model considering damage is proposed, which is connected in series with the improved fractional generalized Kelvin model, and the deformation constitutive model of soil under intermittent cyclic loading is obtained. Finally, the effectiveness and applicability of the model are analyzed, and the model is compared with other constitutive models. The results show that the constitutive model established in the paper can reflect the combined effect of confining pressure, frequency, dynamic stress amplitude and other factors on the cumulative plastic strain of soil, and can well characterize the development law of three types of cumulative plastic strain curves of soil: stable, critical, and failure. The fitting parameters [Formula: see text] and [Formula: see text] tend to decay and rise, respectively, with the increase in loading stage N, which reflects the rationality and accuracy of the model.

Transparent nano-ceramics have an important high-transmittance, material-integrating structure and function and a variety of potential applications, such as use in infrared windows, optical isolators, composite armors, intelligent terminal screens, and key materials of solid-state lasers. Transparent ceramics were originally developed to replace single crystals because of their low fabricating cost, controllable shape, and variable composition. Therefore, this study reviews and summarizes the development trends in transparent nano-ceramics and their potential applications. First, we review the research progress and application of laser nano-ceramic materials, focusing on the influence of controllable doping of rare earth ions on thermal conductivity and the realization of large-scale fabrication technology. Second, the latest research progress on magneto-optical transparent nano-ceramics, mainly including terbium gallium garnet (Tb3Ga5O12, TGG) ceramics and terbium aluminum garnet (Tb3Al5O12, TAG) ceramics, are summarized, and their performance is compared. Third, the research progress of transparent armor nano-ceramic materials, represented by MgAl2O3 and Aluminum oxynitride (AlON), are reviewed. Lastly, the progress in electro-optical transparent nano-ceramics and scintillation transparent nano-ceramics is reported, and the influence of the material-fabrication process on electro-optic effect or luminous intensity is compared. Moreover, the effect of particle diameter on fabrication, the relationship between nano powder and performance, and different sintering methods are discussed. In summary, this study provides a meaningful reference for low-cost and sustainable production in the future.

Laser interaction with nanoparticles in liquid is the fundamental theoretical basis for many applications but it is still challenging to observe this nanoscale phenomenon within a few nanoseconds in liquid by experiment. The successful implementation of the two-temperature method integrated with molecular dynamics (TTM-MD) in laser interaction with bulk material has shown great potential in providing a panoramic view of the laser interaction with the nanoparticles. However, the current TTM-MD model has to divide the system into cubic cells, which leads to mistakes near the nanoparticle's surface. We introduce the latest model, which performs the TTM-MD on each individual cluster instead of the cubic cells, and its high-performance parallel cluster analysis algorithm to update the cluster size. The cluster-based TTM-MD revealed the nanoparticle formation mechanism of laser fragmentation in liquid (LFL) and facilitated the study of laser fluence's effect on the size distribution. In addition to LFL, this model is promising to be implemented in the laser thermal therapy of tumors, laser melting in liquid (LML), etc. Although cluster-based TTM-MD has proven to be a powerful tool for studying laser interaction with nanoparticles, a few challenges and future developments for the cluster-based TTM-MD, especially the ionization induced by femtosecond, are also discussed.

Yam is an important food staple for millions of people globally, particularly those in the developing countries of West Africa and the Pacific Islands. To sustain the growing population, yam production must be increased amidst the many biotic and abiotic stresses. Plant viruses are among the most detrimental of plant pathogens and have caused great losses of crop yield and quality, including those of yam. Knowledge and understanding of virus biology and ecology are important for the development of diagnostic tools and disease management strategies to combat the spread of yam-infecting viruses. This review aims to highlight current knowledge on key yam-infecting viruses by examining their characteristics, genetic diversity, disease symptoms, diagnostics, and elimination to provide a synopsis for consideration in developing diagnostic strategy and disease management for yam.

This research evaluates the elicitors activity and structure characterization of four Chrysanthemum indicum polysaccharides (CIPs) which were isolated from C. indicum, obtained CIP1, CIP2, CIP3, CIP4. Results demonstrated that there was a distinct difference in inducibility and CIP3 was significantly stronger than other CIPs through bioactivity-tests. Taking CIP3 with total carbohydrate content 91.93 % as a representative, its structure was elucidated as a relative molecular weight of 8. 741 × 103 g/mol and mainly composed of xylose, galacturonic acid, galactose and glucuronic acid. Through GC, IR and NMR, CIP3 was determined to possess a backbone comprised of T-α-d-GalpA, 1,4-α-d-GlcpA, 1,2-α-d-Xylp, 1,3-α-l-Rhap, 1,2,4-α-l-Rhap and sidechains comprised of 1,3-ß-d-Galp, 1,6-α-d-Galp, T-α-Glcp, 1,3-ß-d-Glcp, 1,4-α-d-Glcp, 1,3,4-α-d-Manp, T-α-l-Fucp. Further results indicated that CIP3 with active sidechains could significantly increase the expression of defense genes in Atractylodes macrocephala Koidz (AM). It is believed that the sidechains of CIP3 were necessary to its elicitor activity via bioactivity tests.

OBJECTIVE: To study the mechanism and clinical effect of using the manipulatin of move, adduction, rotation method for the treatment of shoulder dislocation. METHODS: from January 2010 to March 2012,120 patients with shoulder dislocation admitted were randomly divided into treatment group and control group, 60 cases in each group. In the treatment group, there were 31 males and 29 females; In control group, there were 30 males and 30 females. In treatment group, 60 patients create their own "on the move, adduction, rotation manipulative treatment,the other 60 cases in control group used traditional restoration methods in wearing the traction treatment. The efficacy of two methods of treatment of shoulder dislocation were compared on reset process once successfully rate, the patient's level of pain (VAS). RESULTS: In treatment group, restoration once successfully were in 59 cases, improvement of the pain (VAS) values was 3.76 +/- 1.05, the results were excellent in 57 cases, good in 2, poor in 1,without concurrent fractures and nerve injury, without re-dislocation after 3 months. In control group, restoration once successfully were in 50 cases, improvement of pain (VAS) values was 5.67 +/- 1.15, the results were excellent in 45 cases, good in 7, poor in 8. The reset once successfully rate,improvement of VAS values and clinical effect in treatment group were better than that of control group. CONCLUSION: On the move, adduction, rotation method has advantages of more wearing traction with uniform force, work together to focus, saving time and effort, easy and convenient to avoid iatrogenic injury.
