RESUMEN

Wetland plant diversity reveals key aspects of the environmental and ecological status of wetlands and plays an important role in the maintenance of ecosystem functions and services. The present study surveyed the plant species diversity of 22 wetlands in Beijing and, combining field data and remote sensing data, evaluated ecological qualities of the wetlands based on indicators of habitat status, plant species diversity, typical wetland plant community, and status of alien plant invasion. A total of 338 species (including varieties and subspecies) belonging to 220 genera of 74 families of wetland plants were recorded in Beijing. The wetlands could be divided into four types according to plant species composition, which showed a spatial gradient pattern from urban core to ecological conservation areas. Wetlands located in ecological conservation areas were of better ecological quality than those located in urban core and suburban plain areas. The value of ecological quality index (EQI) for Baihe River, Huaishahe-Huaijiuhe River, Jinniu Lake, Hanshiqiao Wetland, and Yongdinghe River (Mentougou Section) were in the top five, whereas the value of EQI for river-type wetlands located in suburban plain areas were relatively lower. The results of the canonical correspondence analysis indicated that the most critical factors affecting wetland plant species composition were the distance to the nearest road, water total nitrogen, and area percentage of ecological green land. Furthermore, the results of the canonical correlation analysis indicated that the most critical factors affecting wetland plant species diversity were the area percentage of construction land and water total organic carbon. Human activity intensity and water quality have a strong impact on the plant diversity and ecological quality of wetlands in Beijing. It is suggested that efforts should be made to strengthen the ecological protection and restoration of the river-type wetlands located in suburban plain areas.

In mammals, Interleukin-17 cytokine family plays critical roles in both acute and chronic inflammatory responses. In fish species, three Interleukin-17A/F (IL-17A/F) genes have been identified to be homologous to mammalian IL-17A and IL-17F, but little is known about their functional activity. In this study, Pf_IL-17A/F1, 2 and 3 genes were cloned from yellow catfish (Pelteobagrus fulvidraco) and they differed in protein structure and exon length, implying that they may have divergent bioactivity. Real-time quantitative PCR analyses revealed that three Pf_IL-17A/F genes were highly expressed in blood and mucosal tissues (skin+mucus and gill) from healthy adult fish. The mRNA expressions of Pf_IL-17A/F1, 2 and 3 genes were significantly up-regulated in the gill, skin+mucus, head kidney and spleen after challenge with Edwardsiella ictaluri and in the isolated peripheral blood leucocytes (PBLs) of yellow catfish after stimulation with phytohaemagglutinin (PHA), lipopolysaccharides (LPS), peptidoglycan (PGN) and polyinosinic-polycytidylic acid (Poly I:C). These results indicate that Pf_IL-17A/F1, 2 and 3 genes may play a vital role in the regulation of immune against pathogens. Additionally, the recombinant (r) Pf_IL-17A/F1, 2 and 3 proteins significantly induced the mRNA expressions of proinflammatory cytokines, chemokines and antibacterial peptides genes, and the rPf_IL-17A/F 2 and 3 proteins promoted phagocytosis of PBLs more powerfully than the rPf_IL-17A/F1. Furthermore, the rPf_IL-17A/F1, 2 and 3 proteins might activate the NF-κB and MAPK signal pathways by IL-17RA, ACT1, TRAF6, TRAF2, TRAF5 and TAK1, indicating that the three Pf_IL-17A/F proteins may play different roles in promoting inflammatory response.

Inflammation is a common manifestation of body immunity and mediates a cascade of cytokines. Tumor necrosis factor-α (TNF-α), as a multi-effect cytokine, plays an important role in the inflammatory response by interacting with its receptor (TNFR). In this study, Pf_TNF-α, Pf_TNFR1 and Pf_TNFR2 genes were cloned from yellow catfish (Pelteobagrus fulvidraco), and bioinformatics analyses showed that the three genes were conserved and possessed similar sequence characteristics as those of other vertebrates. The qPCR results showed that Pf_TNF-α, Pf_TNFR1 and Pf_TNFR2 mRNAs were constitutively expressed in 14 tissues and the lymphocytes of four tissues from healthy adults. The mRNA expression levels of Pf_TNF-α and Pf_TNFR1 genes were significantly up-regulated in the spleen, liver, trunk kidney, head kidney and gill after Edwardsiella ictaluri infection, while the mRNA expression of Pf_TNFR2 was significantly up-regulated in the spleen, and down-regulated in the liver and gill. In the isolated peripheral blood leukocytes (PBLs) of yellow catfish, the expression of Pf_TNF-α mRNA was notably up-regulated and the two Pf_TNFR transcripts were distinctly down-regulated after stimulation with lipopolysaccharides (LPS), peptidoglycan (PGN), polyinosinic-polycytidylic acid (Poly I:C) and phytohaemagglutinin (PHA). After stimulated by recombinant (r) Pf_sTNF protein, the mRNA expressions of various inflammatory factors genes were up-regulated in the PBLs. Meanwhile, rPf_sTNF promoted the phagocytic activity of leukocytes, whereas the activity mediated by rPf_sTNF could be inhibited by rPf_TNFR1CRD2/3 and rPf_TNFR2CRD2/3. The up-regulation of TNF-α and IL-1ß mRNAs expression triggered by rPf_sTNF could be inhibited by MAPK inhibitor (VX-702) and NF-κB inhibitor (PDTC). rPf_sTNF induced the expression of FADD mRNA in PBLs and increased the apoptotic rate of PBLs, and inhibiting the NF-κB and MAPK signal pathways could enhance the apoptosis of PBLs. The results indicate that Pf_TNF-α, Pf_TNFR1 and Pf_TNFR2 play important roles in the immune response of yellow catfish to bacterial invasion.

Edwardsiella ictaluri is a highly destructive pathogen in cultured yellow catfish, thus it was very necessary to study the immune response of yellow catfish against bacterial infection. In this study, RNA-Seq technology was used to study the immune response in two distinct tissues of yellow catfish at eight different time points (h) after E. ictaluri infection. The number of differentially expressed genes (DEGs) in the spleen and liver was low at 3 h and 6 h post-infection, respectively. Afterwards, the most number of DEGs in the spleen was detected at 72 h, while the number of DEGs in the liver maintained a high level from 24 h to 120 h. The GO and KEGG enrichment analyses of DEGs at different time points uncovered that cytokines were continuously transcribed at 6 h to 120 h; whereas the liver is the main organ that secretes the components of the complement system, and metabolic regulation was activated from 12 h to 120 h. Moreover, an overview of the inflammation response of yellow catfish was exhibited including pattern-recognition receptors, inflammatory cytokines, chemokines, complements, and inflammation-related signal pathways. The similar expression tendency of nine genes by qRT-PCR validated the accuracy of transcriptome analyses. The different transcriptomic profiles obtained from the spleen and liver will help to better understand the dynamic immune response of fish against bacterial infection, and will provide basic information for establishing effective measures to prevent and control diseases in fish.

Human disturbances have altered global stream ecosystems in multiple ways. Understan-ding spatio-temporal dynamics of aquatic organism composition and their relationships with environmental factors play an important role in revealing changes in the structure and function of river ecosystems. The macroinvertebrates and environmental parameters were sampled in the Yongding River Basin at spring (March), summer (July), and autumn (November) in 2017. The results showed that chironomids were the dominant group in the Yongding River Basin, including Orthocladius, Chironomus, Glyptotendipes, and Polypedilum. Results of the cluster analysis showed significant spatial variations of macroinvertebrate community. Sampling sites were clustered into two groups based on the similarities of macroinvertebrate composition in the Yongding River. Sampling sites belonging to group 1 mainly located in the Sanggan River and Dongyang River, with Orthocladius sp.being the dominant species. Sampling sites belonging to group 2 mainly located in the Yang River, Guishui River and main section of the Yongding River, with Glyptotendipes sp. being the dominant species. The biomass, Shannon index, Margalef index, Pielou evenness index and taxa richness of group 2 were significantly higher than those of group 1. Higher values of biological indices indicated better ecological condition of the stream ecosystem. The density, biomass and diversity indices of summer were significantly lower than those of both spring and autumn. Results of Canonical correspondence analysis (CCA) showed that the spatio-temporal distribution of macroinvertebrate in the Yongding River was mainly influenced by water temperature, ammonium, pH, dissolved oxygen, and total phosphorus. From the perspective of hydrological characteristics, flow velocity and flow capacity were significantly correlated. The significant positive correlations between hydrological parameters of velocity and capacity and biological indices of density and EPT% in group 2 indicated the preference of fast flow to sensitive taxa of EPT (Ephemeroptera, Plecoptera, and Trichoptera). The negative correlations between flow capacity and biodiversity indices implied the adverse effects of high flow on macroinvertebrate biodiversity. Our results confirmed that the composition and biodiversity of macroinvertebrate were affected by both water environment and hydrology, which reflected the multiple human activities in aquatic ecosystems of the Yongding River. It is important to control water contamination and sustain environmental flow at the specific reaches for the restoration of Yongding River.

Aiming at the fact that the current approaches of evaluating the efficacy of ecosystem provisioning service were lack of spatial information and did not take the accessibility of products into account, this paper established an evaluation model to simulate the spatial distribution of ecosystem provisioning service and its economic value, based on ArcGIS 9. 2 and taking the supply and demand factors of ecosystem products into account. The provision of timber product in Laojunshan in 2000 was analyzed with the model. In 2000, the total physical quantity of the timber' s provisioning service in Laojunshan was 11.12 x 10(4) m3 x a(-1), occupying 3.2% of the total increment of timber stock volume. The total provisioning service value of timber was 6669.27 x 10(4) yuan, among which, coniferous forest contributed most (90.41%). Due to the denser distribution of populations and roads in the eastern area of Laojunshan, some parts of the area being located outside of conservancy district, and forests being in scattered distribution, the spatial distribution pattern of the physical quantity of timber's provisioning service was higher in the eastern than in the western area.

By using software ArcGIS 9.2, an evaluation model was established to simulate the ecosystem service of Ertan Reservoir watershed in mitigating the sand sedimentation in the reservoir. In the meantime, sediment delivery ratio and universal soil loss equation were used to simulate the spatial patterns of the annual sediment yield and sediment retention in the watershed as well as the value during the service life period. In 2000, the total quantity of soil retention in the watershed was 12. 1 x 10(8) t x a(-1). The region with higher soil retention was near the main and branch streams of Yalong River, and that with higher sediment delivery ratio was near the streams and the Ertan Reservoir. The region with higher sediment yield and sediment retention was around the reservoir. The actual sediment yield in the study area was 629.3 x 10(4) t x a(-1), occupying 12.7% of the actual soil erosion volume. Farmland was the most important source of sediment yield, with its sediment yield occupying 62.9% of the total. The contribution of forestland to the mitigation of reservoir sand sedimentation was higher than that of the other lands on a per unit area basis. For the reservoir's designed operating life (100 a), the total value of the watershed in the service of mitigating Ertan Reservoir sand sedimentation was 2.75 billion yuan.
