Lymphatic dissemination is one of the most important pathways for metastasis in many solid tumors, including head and neck carcinomas. The lymphatic growth of cancer has been used as a significant independent adverse prognostic factor and provides information about tumor progression. Salivary gland tumors present different prognoses and have the ability to develop metastases; however, this information regarding the lymphatic spread is scarce. This paper quantifies the lymphatic microvessel density (LMD) in benign and malignant salivary gland tumors and analyzes the relationship between LMD and tumor expression of vascular endothelial growth factors C (VEGF-C) and the proliferative index. The results show that there is no correlation between LMD, VEGF-C and the proliferative index in the majority of salivary gland tumors analyzed, apart from polymorphous low-grade carcinoma which exhibits statistical correlation between LMD and the proliferative index (p < 0.05). This correlation probably does not indicate a poor prognosis for this PLGA, since this is a low metastasizing carcinoma of the salivary glands. Different from other solid tumors, such as breast or prostatic carcinomas, there is no correlation between VEGF-C and LMD in salivary gland tumors, and so these traits are not able to estimate the metastatic risk or the prognosis of these tumors.

Spindle cell lipoma is a benign lipomatous neoplasm, which rarely occurs in the oral cavity. The aims of this paper are to report a case of spindle cell lipoma located in buccal mucosa and discuss the main clinical, histological, and immunohistochemical findings of this entity. Thus, we report a 4-year history of an asymptomatic smooth surface nodule in an elderly Caucasian man with clinical hypothesis of fibroma. The histopathological examination showed spindle cells, mature adipose tissue, and many mast cells in a stroma of connective tissue presenting ropey collagen fibers bundles. After immunohistochemical analysis, the final diagnosis was spindle cell lipoma.

Oral verruciform xanthoma represents an uncommon entity, which affects mainly oral mucosa. This paper presents the major clinical and histological features of oral verruciform xanthoma and reports a case on the tongue. The differential diagnosis and a literature review are also provided in light of recent information.

The calcifying cystic odontogenic tumour (CCOT) is a rare benign cystic neoplasm not infrequently associatedwith odontoma. This report documents a case of CCOT associated with compound odontoma arising in theanterior maxilla in a 25-year-old woman. Conventional radiographs showed a large calcified mass with poorlyvisualized radiolucent margins. The extent and condition of the internal structure of the CCOT associated withodontoma was able to be determined based on radiographic findings from cone beam computed tomography.This advanced image technique proved to be extremely useful in the radiographic assessment of this particularneoplasm of the jawbones (AU)

A proteína Akt modula a função de numerosos substratos envolvidos na regulação da sobrevivência celular, progressão do ciclo celular e crescimento celular. Estudos prévios realizados em nosso laboratório demonstraram a superexpressão de Akt em adenoma pleomórfico, mioepitelioma e carcinoma adenóide cístico. O objetivo deste estudo foi analisar a via da proteína Akt através da avaliação da expressão das proteínas NFkB e PTEN em neoplasias benignas e malignas de glândulas salivares através das técnicas de imunohistoquímica, western blotting e imunofluorescência, e a possível interação protéica direta entre p-Akt/Mdm2 e p-Akt/PTEN em linhagem de carcinoma adenóide cístico. A superexpressão nuclear na proteína PTEN foi encontrada nas duas neoplasias malignas estudadas. Além disso, não foi observada interação direta entre as proteínas p-Akt/Mdm2 e p-Akt/PTEN, as quais apresentam localização nuclear em neoplasias de glândulas salivares.


The Akt protein modulates the function of numerous substrates involved in the regulation of cell survival, cell cycle progression and cell growth. Previous studies from our laboratories showed overexpression of Akt and Mdm2 followed by the lack of p53 expression in pleomorphic adenoma, myoepithelioma and adenoid cystic carcinoma. The aim of this study was to analyze the Akt pathway through evaluation of expression of NFkB and PTEN proteins in pleomorphic adenoma, carcinoma ex pleomophic adenoma and adenoid cystic carcinoma by western blotting, immunofluorescence and immunohistochemical techniques, and we have intended to analyse a possible direct interaction between p-Akt/Mdm2 and p-Akt/ PTEN protein in salivary gland tumours. Overexpression of nuclear PTEN was present in both carcinomas studied. In addition, there was no direct interaction between p-Akt/Mdm2 and p-Akt/ PTEN protein, which presents a nuclear localization in salivary gland tumours.

BACKGROUND: MDM2, P53, P21(WAF1) and pAKT are proteins associated with the balance between cell death and survival. There are many hypotheses regarding the role of these proteins in salivary gland tumours. However, many molecular events that activate or inactivate regulatory genes remain unknown. The aim of this study was to evaluate and to correlate MDM2, P53, P21(WAF1) and pAKT protein expressions in adenoid cystic carcinomas (ACC). METHODS: Twenty-two cases of ACC were evaluated by immunohistochemistry and one cell line derived from ACC was analyzed by Western Blotting and immunofluorescence techniques. RESULTS: Strong MDM2 and pAKT, variable P53 and null P21 expressions were found in the cases analyzed, but no statistical correlation was established when comparing MDM2 and pAKT expressions in the 3 different ACC subtypes. The ACC cell line showed intense nuclear and cytoplasmatic MDM2 and pAKT expressions and null P53 and P21 expressions. CONCLUSIONS: Results indicate that MDM2 and pAKT are related to the tumorigenesis of ACC, but they might not be directly connected to tumour progression. We also demonstrate that the pAKT pathway is active in ACC and it seems to be activating the MDM2 shuttle from the cytoplasm to the nucleus, where it phosphorylates P53 and carries it to the cytoplasm for degradation.

RESUMEN

Chondrosarcoma is a malignant cartilaginous tumor that rarely occurs in the maxillofacial bones. A 44-year-old woman complained about swelling and mild pain during mastication in the right parasymphysis region. Clinical and radiographic examinations revealed characteristics of osteosarcoma. A microscopic examination revealed an abundant proliferation of malignant neoplastic cartilage cells of varying sizes arranged as immature tissue and the absence of an osteoid matrix. This article presents a case of chondrosarcoma of the jaw and discusses the differences between osteosarcoma and chondrosarcoma.

The p53 protein can be altered virtually in all human cancers. In the absence of p53 mutations, p53 inactivation is possible via complex formation with other proteins, such as Mdm2. Previous studies have shown an overexpression of Mdm2 and lack of p53 expression in pleomorphic adenomas. The pAkt protein is closely related to Mdm2, and has not been previously reported in salivary gland tumors. The aim of this study was to analyze the expression of Mdm2, p53, p21 and pAkt proteins in pleomorphic adenomas and myoepitheliomas by immunohistochemistry, Western blotting and immunofluorescence techniques. Overexpression of Mdm2 and pAkt was present in all the cell lines and tumors studied, whereas the expression of p53 and p21 proteins was considered absent. In conclusion, the signaling pathway in benign salivary gland neoplasm showed an important participation of Mdm2 overexpression protein in tumor formation, progression through inactivation of p53 action, or both, and of pAkt overexpression through increased translocation of Mdm2 protein to cellular nuclei.

Goldenhar syndrome is a well-known condition featuring the following triad of anomalies: ocular abnormalities, microtia and vertebral anomalies. This syndrome involves structures arising from the first and second branchial arches. Craniofacial anomalies, including mandibular, zygomatic and/or maxillary hypoplasias are found in 50% of patients with Goldenhar syndrome. Patients with this syndrome may present unilateral or bilateral underdevelopment of the mandible. Several treatments for the correction of the dento-facial deformity have been described, among them distraction osteogenesis is one that shows promising results. Distraction osteogenesis is the process of bone formation that occurs during slow separation of the segments of bone after an osteotomy and it has been used to alleviate facial asymmetry. Mandibular distraction osteogenesis has been applied for many years, but long-term reports present controversial results. The purpose of the case report is to describe the immediate and long-term effects of distraction osteogenesis used to treat mandible asymmetry in a 5-year-old boy with Goldenhar syndrome.
