RESUMEN

Harmful algal blooms kill fish populations worldwide, as exemplified by the haptophyte microalga Prymnesium parvum. The suspected causative agents are prymnesins, categorized as A-, B-, and C-types based on backbone carbon atoms. Impacts of P. parvum extracts and purified prymnesins were tested on the epithelial rainbow trout fish gill cell line RTgill-W1 and on the human colon epithelial cells HCEC-1CT. Cytotoxic potencies ranked A > C > B-type with concentrations spanning from low (A- and C-type) to middle (B-type) nM ranges. Although RTgill-W1 cells were about twofold more sensitive than HCEC-1CT, the cytotoxicity of prymnesins is not limited to fish gills. Both cell lines responded rapidly to prymnesins; with EC50 values for B-types in RTgill-W1 cells of 110 ± 11 nM and 41.5 ± 0.6 nM after incubations times of 3 and 24 h. Results of fluorescence imaging and measured lytic effects suggest plasma membrane interactions. Postulating an osmotic imbalance as mechanisms of toxicity, incubations with prymnesins in media lacking either Cl-, Na+, or Ca2+ were performed. Cl- removal reduced morphometric rearrangements observed in RTgill-W1 and cytotoxicity in HCEC-1CT cells. Ca2+-free medium in RTgill-W1 cells exacerbated effects on the cell nuclei. Prymnesin composition of different P. parvum strains showed that analog composition within one type scarcely influenced the cytotoxic potential, while analog type potentially dictate potency. Overall, A-type prymnesins were the most potent ones in both cell lines followed by the C-types, and lastly B-types. Disturbance of Ca2+ and Cl- ionoregulation may be integral to prymnesin toxicity.

Prymnesium parvum causes harmful algal blooms worldwide that are often associated with massive fish-kills and subsequent economic losses. Most of our knowledge of the toxicity of P. parvum derives from bioassays since methods for the identification and quantification of their toxins have been lacking. Recently, a quantitation method was developed for the causative lytic toxins, the prymnesins. Here, we for the first time present data on the influence of irradiance on cellular content and production of prymnesins under nutrient replete conditions in two P. parvum strains, which both produce B-type prymnesins. Large differences were observed between the two strains with regard to the influence of irradiance on prymnesin cell quota and production rates. At the highest irradiance level (550 µmol photons m-2 s-1), the cellular prymnesin quota was thirty times higher in strain K-0081 strain than in K-0374. The cellular prymnesin quota and production rates were closely linked to rates of growth and photosynthesis in strain K-0081, while this was not the case for K-0374. Yet, growth rate did explain the differences in prymnesin quota in the two strains. Consequently, the maximum prymnesin production rate (414 attomol cell-1 d-1) was only about three times higher in strain K-0081 than in K-0374, and revealed an optimum at the same irradiance of 200 µmol photons m-2 s-1 in both strains. At low irradiance levels, the difference in production rates between both strains became smaller, with 41 and 49 attomol cell-1 d-1 for K-0081 and K-0374, respectively. The carbon content of prymnesins made up for ∼3% and <1% of the total cellular carbon content in strains K-0081 and K-0374, respectively. The fraction of extracellular dissolved prymnesins was measured for strain K-0081, where it accounted for 14-30% of total prymnesin concentration in the cultures, irrespective of irradiance. The concentrations of prymnesins released to the water by the K-0081 strain were not significantly influenced by irradiance. Overall, we observed comparable responses in growth and photosynthesis of both tested strains toward changes in irradiance. However, the effects of irradiance on prymnesin quota and production rates were remarkably different between the two strains.

It remains unclear as to how mixoplankton (coupled phototrophy and phagotrophy in one cell) affects the estimation of grazing rates obtained from the widely used dilution grazing technique. To address this issue, we prepared laboratory-controlled dilution experiments with known mixtures of phyto-, protozoo-, and mixoplankton, operated under different light regimes and species combinations. Our results evidenced that chlorophyll is an inadequate proxy for phytoplankton when mixoplankton are present. Conversely, species-specific cellular counts could assist (although not fully solve) in the integration of mixoplanktonic activity in a dilution experiment. Moreover, cell counts can expose prey selectivity patterns and intraguild interactions among grazers. Our results also demonstrated that whole community approaches mimic reality better than single-species laboratory experiments. We also confirmed that light is required for protozoo- and mixoplankton to correctly express their feeding activity, and that overall diurnal grazing is higher than nocturnal. Thus, we recommend that a detailed examination of initial and final plankton communities should become routine in dilution experiments, and that incubations should preferably be started at the beginning of both day and night periods. Finally, we hypothesize that in silico approaches may help disentangle the contribution of mixoplankton to the community grazing of a given system.

BACKGROUND: An important functional trait of organisms is their trophic mode. It determines their position within food webs, as well as their function within an ecosystem. For the better part of the 20th century, aquatic protist communities were thought to consist mainly of producers (phytoplankton) and consumers (protozooplankton). Phytoplankton cover their energy requirements through photosynthesis (phototrophy), while protozooplankton graze on prey and organic particles (phagotrophy). However, over the past decades, it was shown that another trophic group (mixoplankton) comprise a notable part of aquatic protist communities. Mixoplankton employ a third trophic mode by combining phototrophy and phagotrophy (mixotrophy). Due to the historical dichotomy, it is not straightforward to gain adequate and correct information on the trophic mode of aquatic protists. Long hours of literature research or expert knowledge are needed to correctly assign trophic modes. Additionally, aquatic protists also have a long history of undergoing taxonomic changes which make it difficult to compare past and present literature. While WoRMS, the World Register of Marine Species, keeps track of the taxonomic changes and assigns each species a unique AphiaID that can be linked to its various historic and present taxonomic hierarchy, there is currently no machine-readable database to query aquatic protists for their trophic modes. NEW INFORMATION: This paper describes a dataset that was submitted to WoRMS and links aquatic protist taxa, with a focus on marine taxa, to their AphiaID and their trophic mode. The bulk of the data used for this dataset stems from (routine) monitoring stations in the North Sea and the Baltic Sea. The data were augmented and checked against state-of-the-art knowledge on mixoplankton taxa by consulting literature and experts. Thus, this dataset provides a first attempt to make the trophic mode of aquatic protists easily accessible in both a human- and machine-readable format.

Inequalities in relative age distribution have previously been demonstrated to influence participation and performance achievements in Masters athletes. The purpose of the present study was to examine the participation- and performance-related constituent year effect among Masters athletes (N = 2,474) from the European Masters Track and Field Championships across subdisciplines and age. The results indicated that a participation-related constituent year effect was observed. The likelihood of participation was significantly higher for athletes in their first year of any 5-year age category (χ2 = 149.8, p < .001) and decreased significantly when they were in the fourth or fifth year. The results also indicated a performance-related constituent year effect. Masters athletes in their first year won significantly more medals than expected based on observed participation rate (χ2 = 23.39, p < .001). We compare our results with the existing literature and discuss potential mechanisms for this constituent year effect.

A participation-related constituent year effect, has been found to exist in masters sports in that relatively younger masters athletes (i.e., those in the first or second year of a 5-year age category) participate in competitions significantly more often than relatively older masters athletes (i.e., those in the fourth or fifth year of a 5-year age category). The main purpose of this study was to examine if the participation-related constituent year effect in US masters swimming always existed or if it has developed over time at different historical time periods. Using archived data, participation in the US Masters national short course swimming championships at each of the historical time periods in years 1972, 1982, 1992, 2002, 2012 and 2016 were examined as a function of an individual's constituent year within any 5-year age category and across gender and age. The results indicated the existence of a participation-related constituent year effect for each of the six time periods. In particular, a participation-related constituent year effect seemed to have existed from the inception of organized masters swimming competitions in the US but has developed more strongly over the years especially for males and older-aged masters swimmers. Generally, the tendency to participate at National swimming competitions during the first year of an age category was significantly more pronounced, whereas the tendency of participating during the fifth year of an age category was lower. Findings suggest that the 5-year age categories may not provide an equal competitive opportunity especially for relatively older athletes as for those who are relatively younger, but may encourage more strategic periodized training and participation.

Effect of altering body posture and barbell position on the within-session reliability and magnitude of force-time curve characteristics in the isometric midthigh pull. J Strength Cond Res 33(12): 3252-3262, 2019-A large degree of variation in the position used during isometric midthigh pull (IMTP) testing and conflicting results of the effects of these changes can be found in the literature. This study investigated the effect of altering body posture and barbell position on the reliability and magnitude of force-time characteristics generated during the IMTP. Seventeen strength-power athletes (n = 11 males, height: 177.5 ± 7.0 cm, body mass: 90.0 ± 14.1 kg, age: 30.6 ± 10.4 years; n = 6 females, height: 165.8 ± 11.4 cm; body mass: 66.4 ± 13.9 kg, age: 30.8 ± 8.7 years) with greater than 6 months of training experience in the clean (1 repetition maximum: 118.5 ± 20.6 kg, 77.5 ± 10.4 kg) volunteered to undertake the experimental protocol. Subjects performed the IMTP using 4 combinations of hip and knee angles, and 2 different barbell positions. The first barbell position corresponded to the second pull of the clean, while the second rested at the midpoint between the iliac crest and the patella. Peak force (PF), time-specific force (F50, F90, F150, F200, and F250), peak rate of force development (pRFD), and impulse (IMP) time bands were reliable in all 4 testing positions examined. Statistically greater PF, F50, F90, F150, F200, F250, pRFD, and IMP0-50, IMP0-90, IMP0-150, and IMP0-200 were generated in a testing position corresponding to the second pull of the clean when compared with a bent over torso angle, regardless of the barbell position used. Moderate to large effect sizes favoring a testing position corresponding to the second pull were also found. Overall, when performing the IMTP, an upright torso and a barbell position that matches the second pull of the clean should be used.

Harmful algal blooms of Prymnesium parvum have recurrently been associated with the killing of fish. The causative ichthyotoxic agents of this haptophyte are believed to be prymnesins, a group of supersized ladder-frame polyether compounds currently divided into three types. Here, the development of a quantitative method to assess the molar sum of prymnesins in water samples and in algal biomass is reported. The method is based on the derivatization of the primary amine group and subsequent fluorescence detection using external calibrants. The presence of prymnesins in the underivatized sample should be confirmed by liquid chromatography mass spectrometry. The method is currently only partly applicable to water samples due to the low amounts that are present. The growth and cellular toxin content of two B-type producing strains were monitored in batch cultures eventually limited by an elevated pH. The cellular toxin contents varied by a factor of ~2.5 throughout the growth cycle, with the highest amounts found in the exponential growth phase and the lowest in the stationary growth/death phases. The strain K-0081 contained ~5 times more toxin than K-0374. Further investigations showed that the majority of prymnesins were associated with the biomass (89% ± 7%). This study provides the basis for further investigations into the toxicity and production of prymnesins.

Earth temperature is rising and oligotrophication is becoming apparent even in coastal seas. In this changing environment, phytoplankton use carbon and nutrients to form important biomolecules, including lipids. However, the link between lipid production and changing environment is still unexplored. Therefore, we investigated the phytoplankton lipid production in the diatom Chaetoceros pseudocurvisetus cultures under controlled temperatures ranging from 10 to 30 °C and nutrient regimes mimicking oligotrophic and eutrophic conditions. Results were compared to plankton community's lipid production in the northern Adriatic at two stations considered as oligotrophic and mesotrophic during an annual monthly sampling. In order to gain detailed information on the investigated system, we supplemented lipid data with chlorophyll a concentrations, phytoplankton taxonomy, cell abundances and nutrient concentration along with hydrographic parameters. We found enhanced particulate lipid production at higher temperatures, and substantially higher lipid production in oligotrophic conditions. Enhanced lipid production has two opposing roles in carbon sequestration; it can act as a retainer or a sinker. Lipid remodeling, including change in ratio of phospholipids and glycolipids, is more affected by the nutrient status, than the temperature increase. Triacylglycerol accumulation was observed under the nitrogen starvation.

Seventeen strength and power athletes (n = 11 males, 6 females; height: 177.5 ± 7.0 cm, 165.8 ± 11.4 cm; body mass: 90.0 ± 14.1 kg, 66.4 ± 13.9 kg; age: 30.6 ± 10.4 years, 30.8 ± 8.7 years), who regularly performed weightlifting movements during their resistance training programs, were recruited to examine the effect of altering body posture and barbell position on the between-session reliability of force-time characteristics generated in the isometric mid-thigh pull (IMTP). After participants were familiarised with the testing protocol, they undertook two testing sessions which were separated by seven days. In each session, the participants performed three maximal IMTP trials in each of the four testing positions examined, with the order of testing randomized. In each position, no significant differences were found between sessions for all force-time characteristics (p = >0.05). Peak force (PF), time-specific force (F50, F90, F150, F200, F250) and IMP time-bands (0⁻50, 0⁻90, 0⁻150, 0⁻200, 0⁻250 ms) were reliable across each of the four testing positions (ICC ≥ 0.7, CV ≤ 15%). Time to peak force, peak RFD, RFD time-bands (0⁻50, 0⁻90, 0⁻150, 0⁻200, 0⁻250 ms) and peak IMP were unreliable regardless of the testing position used (ICC = <0.7, CV = >15%). Overall, the use of body postures and barbell positions during the IMTP that do not correspond to the second pull of the clean have no adverse effect on the reliability of the force-time characteristics generated.

The constituent year effect, a source of relative age disparities, in masters sport has been demonstrated mainly amongst North American samples. Thus, the purpose of this study was to examine whether a participation-related constituent year effect exists among athletes (n = 6492) competing in Australian Masters Athletics competitions between 2000 and 2014. The results indicated that a participation-related constituent year effect was observed as the likelihood of participating was significantly higher for masters athletes in their first and second constituent year of any five-year age category (p < 0.0001) and was lower when they were in the fourth or fifth constituent year. The results also indicated this effect is influenced by gender and age. Specifically, the effect was significant for both male (p < 0.0001) and female (p < 0.001) masters athletes; as well during the third, sixth, seventh, and eighth + decades of life (all ps < 0.001). These data demonstrate that despite masters sport being an avenue for promotion of participation and overall health, there is potential for improving how competitive organizational strategies are implemented given the recurring intermittent patterns of participation associated with five-year age brackets which are likely to compromise benefits.

Chaetoceros peruvianus is a marine diatom species with circumglobal distribution. While frequently observed, it appears never to dominate the marine phytoplankton community hence it can be characterized as a rather opportunistic, generalistic species. Here we present ecological interpretations from a long-term data set on marine microphytoplankton in the northern Adriatic Sea, where the abundancies and relative contributions of C. peruvianus were observed along a set of steep ecological gradients. Limited supply of dissolved inorganic phosphate was identified as the driving ecological factor for this ecosystem. In parallel C. peruvianus was cultivated in monoclonal cultures and its morphological and physiological reaction to replete and phosphorus depleted medium was analysed. C. peruvianus reacted to phosphorus depletion by an increase in cell height and length as well as thickness and length of setae. This morphological reaction included an increase in cellular volume and calculated carbon content. Additionally, it represents the transition between two described morphological varieties, C. peruvianus and C. peruvianus var. robusta. C. peruvianus showed a significant induction of extracellular alkaline phosphatase activity if grown in phosphate depleted medium. Microscopical analysis demonstrated this activity to be located exclusively on the setae of the cells.

Physical activity in children and adolescents is on a decline trend. To this end, we conducted a matched-pair randomized controlled trial to examine the effects of a 4-week STAR (School-based; Train-the-trainer; Accessibility of resources; Recreational) skipping programme. 1,386 schoolchildren from 20 primary and secondary schools were recruited. Schools were randomized into the experimental or wait-list control group. Participants self-reported their health-related quality of life using the KIDSCREEN-27. Accelerometers were used to measure the time a subgroup of participants (n = 480) spent in moderate-to-vigorous physical activity during school hours on five consecutive days. Measures were taken at pre- and post-test. At post-test, students in the experimental group, compared to those in the control group, engaged in less moderate-to-vigorous physical activity during school hours. Health-related quality of life from two groups of students was similar, but the experimental group reported higher levels of autonomy and parent relationships. Results suggested that although the intervention did not increase students' physical activity levels, it slightly improved their health-related quality of life. Future studies should explore personal factors that might mediate the effect of the intervention.

Purpose: Habitual action is an important aspect of health behaviour, but the relevance of various habit strength indicators continues to be debated. This study focused specifically on moderate-to-vigorous physical activity (MVPA) and evaluated the construct validity of a framework emphasizing patterned action, stimulus-response bonding, automaticity, and negative consequences for nonperformance as indicators of habit strength for this form of exercise. Methods: Upper-level undergraduates (N = 124) provided demographic information and responded to questionnaire items assessing historical MVPA involvement, current MVPA involvement, and the four proposed habit strength dimensions. Factor analyses were used to examine the latent structure of the habit strength indicators, and the model's construct validity was evaluated via an examination of relationships with repetition history and current behaviour. Results: At a measurement level, findings indicated that the proposed four-component model possessed psychometric integrity as a coherent set of factors. Criterion-related validity was also demonstrated via significant changes in three of the four factors as a function of past involvement in MVPA and significant correlations with the frequency, duration, and intensity of current MVPA. Conclusions: These findings support the construct validity of this exercise habit strength model and suggest that it could provide a template for future research on how MVPA habits are developed and maintained.

The purpose of this study was to examine the contrasting perceptions of masters swimmers related to the first and fifth constituent years of a 5-year age category. Swimmers aged between 35 and 93 years (154 male, 184 female) were surveyed at the 2008 FINA World Masters Championships. Exploratory factor analysis indicated the existence of the following five factors considered important for preparation, attendance, and success at masters competitions: awareness of advantages, expectancy, motivation, training, and physiological capacity. One sample t-tests showed that masters swimmers are conscious of advantages that 5-year age categories afford to relatively-younger cohorts (i.e., those who are in the first year of any age category). They also perceive that, in the first compared to the fifth year of an age category, they have greater physiological capacity, engage in more training, have higher expectations to perform well, and are more motivated (all ps < .001). Findings point to perceived psycho-social and physical factors that potentially explain why relatively younger masters athletes are more likely to perform better and to participate in masters competitions than relatively older masters athletes. Key PointsThere are at least five psycho-social and physical factors (i.e., awareness of advantages, expectancy, motivation, training, and physiological capacity) that may explain why relatively younger masters athletes are more likely to perform better and to participate more in masters competitions than relatively older masters athletes.Masters athletes are conscious of the advantage that 5-year age categories afford to relatively younger cohorts of athletes.Differential perceptions associated with the 5-year age categories might compromise masters athletes' continuity of sport competitiveness and underlying training.

To overcome the weakness of the contextual interference (CI) effect within applied settings, Brady, 2008 recommended that the amount of interference be manipulated. This study investigated the effect of five practice schedules on the learning of three field hockey skills. Fifty-five pre-university students performed a total of 90 trials for each skill under blocked, mixed or random practice orders. Results showed a significant time effect with all five practice conditions leading to improvements in acquisition and learning of the skills. No significant differences were found between the groups. The findings of the present study did not support the CI effect and suggest that either blocked, mixed, or random practice schedules can be used effectively when structuring practice for beginners. Key pointsThe contextual interference effect did not surface when using sport skills.There appears to be no difference between blocked and random practice schedules in the learning of field hockey skills.Low (blocked), moderate (mixed) or high (random) interference practice schedules can be used effectively when conducting a multiple skill practice session for beginners.

Medic and colleagues (Medic, Starkes, & Young, 2007) found that relatively younger cohorts of Masters athletes had higher participation rates and achieved higher performances compared with a relatively older cohort. Considering that Medic and colleagues' (2007) study was cross-sectional in nature, the purpose of this investigation was to employ a retrospective longitudinal study design to examine the participation rates of Masters swimmers as a function of an individual's constituent year within any 5-year age category over a period of 6 years. Using archived data from the 2003 to 2009 US Masters Short Course National Championships, swimmers' attendance was followed for a period of six consecutive years. Results indicated that a participation-related relative age effect was observed among swimmers who, over a period of 6 years, competed in either at least one championship (N = 2596; Cochran's Q4 = 64.16, r(s) = -0.92, both P < 0.0001) or at least three championships (N = 441; Cochran's Q4 = 47.51, r(s) = -0.91, both P < 0.0001). Overall, effect size analyses indicated that the odds of a Masters swimmer participating in the championship during the first constituent year of any 5-year age category was more than two times greater than the odds of that athlete participating during the fifth constituent year.

RESUMEN

A relative age effect has been identified in Masters sports (Medic, Starkes, & Young, 2007). Since gender, age, and type of sport have been found to influence the relative age effect in youth sports (Musch & Grondin, 2001), we examined how these three variables influenced possible relative age effects among Masters swimmers and track and field athletes. Using archived data between 1996 and 2006, frequency of participation entries and record-setting performances at the US Masters championships were examined as a function of an individual's constituent year within any 5-year age category. Study 1 investigated the frequency of Master athletes who participated; Study 2 examined the frequency of performance records that were set across constituent years within an age category, while accounting for the distribution of participation frequencies. Results showed that a participation-related relative age effect in Masters sports is stronger for males, that it becomes progressively stronger with each successive decade of life, and that it does not differ across track and field and swimming. In addition, a performance-related relative age effect in Masters sport seems to be stronger for swimming than track and field, but it does not differ across gender and decades of life.
