RESUMEN

Rats were irradiated by 8Gr in a doze power of 2.33 R per second. A portion of animals were injected subcutaneously with a cow colostrum polypeptide (CP) in a doze of 1 mg per g of body weight before the irradiation and daily after the irradiation during 4 days. In irradiated rat liver nuclei injected with CP the lipid peroxide oxidation was restored. At 90th day after the irradiation the production of dienes and dieneketones in liver nuclei oscillated in normal limits. The structure and cytochrome-c-oxidase activity of the nuclei was restored to 60-90th day after the irradiation.

RESUMEN

A single exposure of pregnant rats to 1 and 2 Gy radiation on days 3, 13 and 17 of reception changes the phospholipid content in membranes of liver mitochondria of fetus and maternal body. The changes are particularly pronounced on day 3 of reception: as the embryo grows the decline in the phospholipid content from the control level decreases.

RESUMEN

A study was made of the incorporation of 35S-methionine into nuclear matrix proteins of hepatic cells of pregnant rats and their embryos subjected to single gamma-irradiation (60Co, 1 and 2 Gy, 0.0233 Gy/s) on days 3, 13 and 17 of pregnancy and embryogenesis. On day 21 of pregnancy and embryogenesis a decrease in the rate of incorporation of 35S-methionine into nuclear matrix proteins was shown to be a function of radiation dose and time of pregnancy and embryogenesis on the moment of exposure.

RESUMEN

The data on the effect of synthetic antioxidant Paginol C-2000 on respiration and oxidative phosphorylation in liver mitochondria of rats primed with butylcaptax are given. It is shown that intoxication of organism with butylcaptax in the dose of 1/10 LD50 during 5 days resulted in suppression of respiration and oxidative phosphorylation in rat liver mitochondria. It is found, that administration of Raginol C-2000 into organisms of primed animals leads to the reduction of succinate pathway of oxidation, that, in its turn, results in the normalization of functioning level of ATP-synthesizing system of mitochondria.

RESUMEN

The experiments on the investigation of pesticide fluometuron (cotoran) influence on nuclease sensitivity and template activity of rat liver chromatin were carried out. Cotoran was found to bind specifically with non-histone proteins of chromatin. It was shown that this pesticide considerably decreases template activity of chromatin and its sensitivity to the action of nucleases. It suggests, that certain conformation changes occur in chromatin upon the action of cotoran.

RESUMEN

Morphological and biochemical investigations of pregnant rats and embryo liver cell nuclei after in vivo irradiation in the doses of 1 and 2 Gr revealed their high radiosensitivity at all stages of gestation and embryonal development. At damaging effect of radiation, we managed to observe sharp accumulation of products of lipid peroxide oxidation and suppression of the activities of such enzymes as cytochrome-c-oxidase, NAD.N-cytochrome-c-reductase, ATPase and RNAase in liver nuclei of pregnant rats and embryos. The changes of such a kind are shown to intensify with the increasing of irradiation doses. The most profound inhibition of the activities of these enzymes in liver nuclei of embryos irradiated in utero was observed during the period of organogenesis (the 13th day of the development) and in fetal period of embryogenesis (the 17th day of the development), as well as at the 13th and 17th day of gestation. The morphological data also demonstrate the high level of cell nucleus sensitivity to the action of radiation during gestation and embryogenesis.

RESUMEN

Intracellular distribution of labeled cotoran was studied. 3H-cotoran was shown to penetrate through the nuclear membrane to accumulate uneventfully in the intranuclear components. An insignificant amount of 3H-cotoran was associated with the nucleoplasm and the outer nuclear membrane. At the same time, essential radioactivity was observed in the proteins of the nuclear matrix (up to 30%) and in non-histone proteins of chromatin (up to 60%). Acception of 3H-cotoran on metaphase chromosomes of cultured cells as well as specificity of cotoran binding with non-histone proteins of chromatin in vivo and in vitro was studied by radioautography. It was revealed that cotoran was translocated into the interphase nuclei to be accepted by metaphase chromosomes of the HeLa line cells and fibroblasts in human embryo, and specifically, in receptor-like manner, bound to chromatin proteins.

RESUMEN

The oxidative phosphorylation and the activity of polyenzymatic systems in the rat liver mitochondrial membranes were studied upon a 5 day administration to rats of butylcaptax in the dose of 1/10 LD50. It is established that butylcaptax led to significant disturbances in the system of oxidative phosphorylation and in the transfer of electrons in the rat liver mitochondrial respiratory chain.

RESUMEN

Butiphose (Tributyltritiophosphate, (C4H9S)3PO) a commonly used defoliant in cotton growing regions of USSR, caused extensive alterations in morphological features of erythrocyte and nuclear membranes and affected the permeability properties of rat liver mitochondrial membrane. It disrupted Ca2+ transport system and other energy dependent processes in mitochondria. A reduction in the activity of cytochrome-c-oxidase and NAD.H-oxidase was also observed.
