RESUMEN

The aging of parents results in changes in the filial relationship. The increasing vulnerability of parents leads adult children to realize that they have individual needs and cannot fully function as sources of security and protection, as they did before. Simultaneously, the evidence of losses and disability imposes the need for care, which tends to be assumed by adult children. Therefore, there is a progressive change in the volume of support exchanges between parents and children, with more support from adult children to parents. The way adult children adapt to these transitions is influenced by several internal and relational factors. Filial maturity has been associated with filial caregiving towards aging parents. The concept of filial maturity describes a developmental stage in which the adult child overcomes the filial crisis, realizing and accepting that the parent also needs support and comfort and starting to relate to him/her beyond the strictly parental role. Thus, this study aims to explore the role of attachment and mental representation of caregiving in filial maturity. A total of 304 children aged between 35 and 64 years old participated in this study, with at least one of the living parents aged 65 years or older, not institutionalized. Attachment was assessed with the Adult Attachment Scale, mental representation of caregiving with the Mental Representations of Caregiving Scale and filial maturity with the Filial Maturity Measure. The results suggest that attachment, mental representation of caregiving and the interaction between the two explain 24.5% (p < 0.01) of variability in Comprehending and 11.1% (p < 0.05) of variability in Distance, two dimensions of filial maturity. These findings suggest that it is important to consider mental representation of caregiving and attachment when adult children must adapt to changes in the filial relationship and to the need to care for parents.

Pseudomonas aeruginosa is a Gram-negative opportunistic bacterium that presents resistance to several antibiotics, thus, representing a major threat to human and animal health. Phage-derived products, namely lysins, or peptidoglycan-hydrolyzing enzymes, can be an effective weapon against antibiotic-resistant bacteria. Whereas in Gram-positive bacteria, lysis from without is facilitated by the exposed peptidoglycan layer, this is not possible in the outer membrane-protected peptidoglycan of Gram-negative bacteria. Here, we suggest the encapsulation of lysins in liposomes as a delivery system against Gram-negative bacteria, using the model of P. aeruginosa. Bioinformatic analysis allowed for the identification of 38 distinct complete prophages within 66 P. aeruginosa genomes (16 of which newly sequenced) and led to the identification of 19 lysins of diverse sequence and function, 5 of which proceeded to wet lab analysis. The four purifiable lysins showed hydrolytic activity against Gram-positive bacterial lawns and, on zymogram assays, constituted of autoclaved P. aeruginosa cells. Additionally, lysins Pa7 and Pa119 combined with an outer membrane permeabilizer showed activity against P. aeruginosa cells. These two lysins were successfully encapsulated in DPPC:DOPE:CHEMS (molar ratio 4:4:2) liposomes with an average encapsulation efficiency of 33.33% and 32.30%, respectively. The application of the encapsulated lysins to the model P. aeruginosa led to a reduction in cell viability and resulted in cell lysis as observed in MTT cell viability assays and electron microscopy. In sum, we report here that prophages may be important sources of new enzybiotics, with prophage lysins showing high diversity and activity. In addition, these enzybiotics following their incorporation in liposomes were able to potentiate their antibacterial effect against the Gram-negative bacteria P. aeruginosa, used as the model.

To modulate the physicochemical features of poly(ethylene terephthalate) (PET) multifilaments surface composing a complex textile structure (core and shell system), intended to improve upon current implants for high extension injuries of the Achilles tendon or even for its total replacement, two surface treatments with different purposes (bioactive and biopassive) were studied. The first treatment is based on amino groups grafting using ethylenediamine molecules to be applied in the structure core to improve cell adhesion and proliferation. The other treatment relates to a polytetrafluoroethylene (PTFE) coating to be applied in the structure shell to decrease its coefficient of friction and avoid adhesions. Both treatments were optimized to reach their purposed goals without harming the tensile properties of PET yarns, which were evaluated by static tensile tests. The resazurin assay and scanning electron microscopy analysis showed that the purposed goals related to fibroblast adhesion were achieved for both treatments and in the case of PTFE coating, the abrasion resistance was also improved according to the yarn-on-yarn abrasion tests.

RESUMEN

The large surface area and ability to retain moisture of textile structures enable microorganisms' growth, which causes a range of undesirable effects, not only on the textile itself, but also on the user. Due to the public health awareness of the pathogenic effects on personal hygiene and associated health risks, over the last few years, intensive research has been promoted in order to minimize microbes' growth on textiles. Therefore, to impart an antimicrobial ability to textiles, different approaches have been studied, being mainly divided into the inclusion of antimicrobial agents in the textile polymeric fibers or their grafting onto the polymer surface. Regarding the antimicrobial agents, different types have been used, such as quaternary ammonium compounds, triclosan, metal salts, polybiguanides or even natural polymers. Any antimicrobial treatment performed on a textile, besides being efficient against microorganisms, must be non-toxic to the consumer and to the environment. This review mainly intends to provide an overview of antimicrobial agents and treatments that can be performed to produce antimicrobial textiles, using chemical or physical approaches, which are under development or already commercially available in the form of isolated agents or textile fibers or fabrics.

Human development is a bidirectional, person-context relational process, but scarce evidence is available about the relation between the individual variability across the life-span and the neighborhood ecological assets. Therefore, it is important that research focus not only on personal characteristics but on ecological assets as well. This way this study aims to analyze the association between neighborhood ecological assets categorized into four dimensions: human, physical or institutional, social or collective activity, accessibility, and the individual functioning. A 3% sample of residents aged 65 years and older in two downtown and three uptown parishes stratified by age and sex was interviewed at home using a protocol that included the Portuguese version of the Barthel Index in basic activities of daily living (BADL), the Lawton Instrumental Activities of Daily Living Scale (IADL), the Mini Mental State Examination (MMSE), and the Geriatric Depression Scale-15 items (GDS) for evaluating functionality, cognitive performance, and depression. The 162 participants were aged on average 75 years (sd = 7.0), 54% were women and 90% had less than 7 years of education. The majority of participants were independent in BADL (M = 90; sd = 17.7) and moderately dependent in IADL (M = 13, sd = 6.0), 20% showed cognitive impairment and a mean score of 8 (sd = 2.1) in GDS-15. After controlling for the effect of socio-demographic characteristics, functionality, and cognitive performance decreases in persons with worst outdoor mobility. On the other hand depressive symptoms are less common as the number of recreation opportunities, namely associative groups (cultural, educative, professional), increases. These results suggest that aging policies and practices must be ecologically embedded.

A novel bioactive bone substitute with improved osteoblastic performance and effective antibacterial activity was developed, using a completely new approach based on samarium (Sm3+) doped P2O5 glass-reinforced hydroxyapatite composites (GR-HA). The composites were prepared by adding 2.5% (w/w) of the P2O5 glass to 97.5% (w/w) of HA. Four composites were developed, i.e. one non-doped composite, and three Sm3+ doped composites prepared with the P2O5 glass containing 0.5, 1 and 2 (mol%) of Sm2O3. The composites were labeled as GR-HA_control, GR-HA_0.5Sm, GR-HA_1Sm and GR-HA_2Sm. The composites were physicochemical and mechanically characterized, namely performing SEM, EDS and XRD analysis and flexural bending strength (FBS) assessment. The incorporation of Sm3+ in the GR-HA matrix resulted in the presence of a residual Sm3+ containing phase besides HA, ß-TCP and α-TCP phases, increased surface hydrophilicity and slightly higher FBS. Sm3+ doped composites exhibited improved osteoblastic cell response, as evidenced by a better F-actin cytoskeleton organization and higher cell proliferation and expression of relevant osteoblastic genes. In addition, adhesion of Staphylococcus aureus and Staphylococcus epidermidis was greatly reduced on these composites. The improved osteoblastic behavior and the antibacterial effects were dependent on the amount of Samarium in the composite, this being particularly evident in the composite with a higher Sm3+ content. Therefore, the developed composite GR-HA_2Sm appears as a successful bone substitute with osteoconductive and antibacterial properties.
