RESUMEN

The DFNA58 locus contains a genomic duplication involving three protein-coding genes (CNRIP1, PLEK, and PPP3R1's exon 1) and other uncharacterized lncRNA genes (LOC101927723, LOC107985892 and LOC102724389). To clarify the role of these genes in hearing and precisely determine their role in hearing loss, four iPSC lines were generated from two carriers and two noncarriers of the duplication.

Xia-Gibbs syndrome (XGS) is a syndromic form of intellectual disability caused by heterozygous AHDC1 variants, but the pathophysiological mechanisms underlying this syndrome are still unclear. In this manuscript, we describe the development of two different functional models: three induced pluripotent stem cell (iPSC) lines with different loss-of-function (LoF) AHDC1 variants, derived by reprogramming peripheral blood mononuclear cells from XGS patients, and a zebrafish strain with a LoF variant in the ortholog gene (ahdc1) obtained through CRISPR/Cas9-mediated editing. The three iPSC lines showed expression of pluripotency factors (SOX2, SSEA-4, OCT3/4, and NANOG). To verify the capacity of iPSC to differentiate into the three germ layers, we obtained embryoid bodies (EBs), induced their differentiation, and confirmed the mRNA expression of ectodermal, mesodermal, and endodermal markers using the TaqMan hPSC Scorecard. The iPSC lines were also approved for the following quality tests: chromosomal microarray analysis (CMA), mycoplasma testing, and short tandem repeat (STR) DNA profiling. The zebrafish model has an insertion of four base pairs in the ahdc1 gene, is fertile, and breeding between heterozygous and wild-type (WT) animals generated offspring in a genotypic proportion in agreement with Mendelian law. The established iPSC and zebrafish lines were deposited on the hpscreg.eu and zfin.org platforms, respectively. These biological models are the first for XGS and will be used in future studies that investigate the pathophysiology of this syndrome, unraveling its underlying molecular mechanisms.

Maintaining synaptic communication is required to preserve nervous system function as an organism ages. While much work has been accomplished to understand synapse formation and development, we understand relatively little regarding maintaining synaptic integrity throughout aging. To better understand the mechanisms responsible for maintaining synaptic structure and function, we performed an unbiased forward genetic screen to identify genes required for synapse maintenance of adult Drosophila neuromuscular junctions. Using flight behavior as a screening tool, we evaluated flight ability in 198 lines from the Drosophila Genetic Reference Panel to identify single nucleotide polymorphisms (SNPs) that are associated with a progressive loss of flight ability with age. Among the many candidate genes identified from this screen, we focus here on 10 genes with clear human homologs harboring SNPs that are most highly associated with synaptic maintenance. Functional validation of these genes using mutant alleles revealed a progressive loss of synaptic structural integrity. Tissue-specific knockdown of these genes using RNA interference (RNAi) uncovered important roles for these genes in either presynaptic motor neurons, postsynaptic muscles, or associated glial cells, highlighting the importance of each component of tripartite synapses. These results offer greater insight into the mechanisms responsible for maintaining structural and functional integrity of synapses with age.

RESUMO Objetivo: Descrever o IMPACTO-MR, um estudo brasileiro de plataforma nacional em unidades de terapia intensiva focado no impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Métodos: Descrevemos a plataforma IMPACTO-MR, seu desenvolvimento, critérios para seleção das unidades de terapia intensiva, caracterização da coleta de dados, objetivos e projetos de pesquisa futuros a serem realizados na plataforma. Resultados: Os dados principais foram coletados por meio do Epimed Monitor System® e consistiram em dados demográficos, dados de comorbidades, estado funcional, escores clínicos, diagnóstico de internação e diagnósticos secundários, dados laboratoriais, clínicos e microbiológicos e suporte de órgãos durante a internação na unidade de terapia intensiva, entre outros. De outubro de 2019 a dezembro de 2020, 33.983 pacientes de 51 unidades de terapia intensiva foram incluídos no banco de dados principal. Conclusão: A plataforma IMPACTO-MR é um banco de dados clínico brasileiro de unidades de terapia intensiva focado na pesquisa do impacto das infecções por bactérias multirresistentes relacionadas à assistência à saúde. Essa plataforma fornece dados para o desenvolvimento e pesquisa de unidades de terapia intensiva individuais e ensaios clínicos observacionais e prospectivos multicêntricos.


ABSTRACT Objective: To describe the IMPACTO-MR, a Brazilian nationwide intensive care unit platform study focused on the impact of health care-associated infections due to multidrug-resistant bacteria. Methods: We described the IMPACTO-MR platform, its development, criteria for intensive care unit selection, characterization of core data collection, objectives, and future research projects to be held within the platform. Results: The core data were collected using the Epimed Monitor System® and consisted of demographic data, comorbidity data, functional status, clinical scores, admission diagnosis and secondary diagnoses, laboratory, clinical, and microbiological data, and organ support during intensive care unit stay, among others. From October 2019 to December 2020, 33,983 patients from 51 intensive care units were included in the core database. Conclusion: The IMPACTO-MR platform is a nationwide Brazilian intensive care unit clinical database focused on researching the impact of health care-associated infections due to multidrug-resistant bacteria. This platform provides data for individual intensive care unit development and research and multicenter observational and prospective trials.

Autism Spectrum Disorder (ASD) is characterized by impaired social communication, restricted interests, and repetitive and stereotyped behaviors. The TRPC6 (transient receptor potential channel 6) represents an ASD candidate gene under an oligogenic/multifactorial model based on the initial description and cellular characterization of an individual with ASD bearing a de novo heterozygous mutation disrupting TRPC6, together with the enrichment of disruptive TRPC6 variants in ASD cases as compared to controls. Here, we perform a clinical re-evaluation of the initial non-verbal patient, and also present eight newly reported individuals ascertained for ASD and bearing predicted loss-of-function mutations in TRPC6. In order to understand the consequences of mutations in TRPC6 on nervous system function, we used the fruit fly, Drosophila melanogaster, to show that null mutations in transient receptor gamma (trpγ; the fly gene most similar to TRPC6), cause a number of behavioral defects that mirror features seen in ASD patients, including deficits in social interactions (based on courtship behavior), impaired sleep homeostasis (without affecting the circadian control of sleep), hyperactivity in both young and old flies, and defects in learning and memory. Some defects, most notably in sleep, differed in severity between males and females and became normal with age. Interestingly, hyperforin, a TRPC6 agonist and the primary active component of the St. John's wort antidepressant, attenuated many of the deficits expressed by trpγ mutant flies. In summary, our results provide further evidence that the TRPC6 gene is a risk factor for ASD. In addition, they show that the behavioral defects caused by mutations in TRPC6 can be modeled in Drosophila, thereby establishing a paradigm to examine the impact of mutations in other candidate genes.

BACKGROUND: Auriculocondylar syndrome (ARCND) is a rare genetic disease that affects structures derived from the first and second pharyngeal arches, mainly resulting in micrognathia and auricular malformations. To date, pathogenic variants have been identified in three genes involved in the EDN1-DLX5/6 pathway (PLCB4, GNAI3 and EDN1) and some cases remain unsolved. Here we studied a large unsolved four-generation family. METHODS: We performed linkage analysis, resequencing and Capture-C to investigate the causative variant of this family. To test the pathogenicity of the CNV found, we modelled the disease in patient craniofacial progenitor cells, including induced pluripotent cell (iPSC)-derived neural crest and mesenchymal cells. RESULTS: This study highlights a fourth locus causative of ARCND, represented by a tandem duplication of 430 kb in a candidate region on chromosome 7 defined by linkage analysis. This duplication segregates with the disease in the family (LOD score=2.88) and includes HDAC9, which is located over 200 kb telomeric to the top candidate gene TWIST1. Notably, Capture-C analysis revealed multiple cis interactions between the TWIST1 promoter and possible regulatory elements within the duplicated region. Modelling of the disease revealed an increased expression of HDAC9 and its neighbouring gene, TWIST1, in neural crest cells. We also identified decreased migration of iPSC-derived neural crest cells together with dysregulation of osteogenic differentiation in iPSC-affected mesenchymal stem cells. CONCLUSION: Our findings support the hypothesis that the 430 kb duplication is causative of the ARCND phenotype in this family and that deregulation of TWIST1 expression during craniofacial development can contribute to the phenotype.

Rapid diagnostics is pivotal to curb SARS-CoV-2 transmission, and saliva has emerged as a practical alternative to naso/oropharyngeal (NOP) specimens. We aimed to develop a direct RT-LAMP (reverse transcription loop-mediated isothermal amplification) workflow for viral detection in saliva, and to provide more information regarding its potential in curbing COVID-19 transmission. Clinical and contrived specimens were used to optimize formulations and sample processing protocols. Salivary viral load was determined in symptomatic patients to evaluate the clinical performance of the test and to characterize saliva based on age, gender and time from onset of symptoms. Our workflow achieved an overall sensitivity of 77.2% (n = 90), with 93.2% sensitivity, 97% specificity, and 0.895 Kappa for specimens containing >102 copies/µL (n = 77). Further analyses in saliva showed that viral load peaks in the first days of symptoms and decreases afterwards, and that viral load is ~10 times lower in females compared to males, and declines following symptom onset. NOP RT-PCR data did not yield relevant associations. This work suggests that saliva reflects the transmission dynamics better than NOP specimens, and reveals gender differences that may reflect higher transmission by males. This saliva RT-LAMP workflow can be applied to track viral spread and, to maximize detection, testing should be performed immediately after symptoms are presented, especially in females.

Biallelic pathogenic variants in TBCK cause encephaloneuropathy, infantile hypotonia with psychomotor retardation, and characteristic facies 3 (IHPRF3). The molecular mechanisms underlying its neuronal phenotype are largely unexplored. In this study, we reported two sisters, who harbored biallelic variants in TBCK and met diagnostic criteria for IHPRF3. We provided evidence that TBCK may play an important role in the early secretory pathway in neuroprogenitor cells (iNPC) differentiated from induced pluripotent stem cells (iPSC). Lack of functional TBCK protein in iNPC is associated with impaired endoplasmic reticulum-to-Golgi vesicle transport and autophagosome biogenesis, as well as altered cell cycle progression and severe impairment in the capacity of migration. Alteration in these processes, which are crucial for neurogenesis, neuronal migration, and cytoarchitecture organization, may represent an important causative mechanism of both neurodevelopmental and neurodegenerative phenotypes observed in IHPRF3. Whether reduced mechanistic target of rapamycin (mTOR) signaling is secondary to impaired TBCK function over other secretory transport regulators still needs further investigation.

A homozygous mutation in the inositol monophosphatase 1 (IMPA1) gene was recently identified in nine individuals with severe intellectual disability (ID) and disruptive behavior. These individuals belong to the same family from Northeastern Brazil, which has 28 consanguineous marriages and 59 genotyped family members. IMPA1 is responsible for the generation of free inositol from de novo biosynthesis and recycling from inositol polyphosphates and participates in the phosphatidylinositol signaling pathway. To understand the role of IMPA1 deficiency in ID, we generated induced pluripotent stem cells (iPSCs) from patients and neurotypical controls and differentiated these into hippocampal dentate gyrus-like neurons and astrocytes. IMPA1-deficient neuronal progenitor cells (NPCs) revealed substantial deficits in proliferation and neurogenic potential. At low passage NPCs (P1 to P3), we observed cell cycle arrest, apoptosis, progressive change to a glial morphology and reduction in neuronal differentiation. These observations were validated by rescuing the phenotype with myo-inositol supplemented media during differentiation of patient-derived iPSCs into neurons and by the reduction of neurogenic potential in control NPCs-expressing shIMPA1. Transcriptome analysis showed that NPCs and neurons derived from ID patients have extensive deregulation of gene expression affecting pathways necessary for neurogenesis and upregulation of gliogenic genes. IMPA1 deficiency did not affect cell cycle progression or survival in iPSCs and glial progenitor cells or astrocyte differentiation. Therefore, this study shows that the IMPA1 mutation specifically affects NPC survival and neuronal differentiation.

RESUMO Objetivo compreender o desenvolvimento da sensibilidade moral na prática de profissionais da Estratégia Saúde da Família. Métodos estudo de caso único, de abordagem qualitativa, realizado com trinta e cinco profissionais das equipes de saúde da família. A coleta de dados ocorreu por meio de entrevistas guiadas por roteiro semiestruturado e observação. Os dados foram analisados por meio de Análise de Conteúdo Temática. Resultados a análise revelou que os fatores relacionados à organização do trabalho, às relações interpessoais e aos valores pessoais influenciam o desenvolvimento da sensibilidade moral de profissionais das equipes de saúde da família. Conclusão o desenvolvimento da sensibilidade moral de profissionais da Estratégia Saúde da Família é influenciado por fatores potencializadores e dificultadores que se apresentam no cotidiano de trabalho.


ABSTRACT Objective to understand the development of moral sensitivity in the practice of Family Health Strategy professionals. Methods a unique case study, of qualitative approach, conducted with thirty-five professionals from the family health teams. Data collection occurred through interviews guided by semi-structured script and observation. The data was analyzed through Thematic Content Analysis. Results the analysis revealed that factors related to work organization, interpersonal relationships and personal values influence the development of the moral sensitivity of professionals in family health teams. Conclusion the development of the moral sensitivity of Family Health Strategy professionals is influenced by enabling and hindering factors that present themselves in daily work.

Resumo Objetivo: Compreender as expressões da ética e do distresse moral na prática do enfermeiro intensivista. Métodos: Pesquisa qualitativa, descritiva, interpretativa e analítica, desenvolvida com 12 enfermeiros lotados nos plantões diurnos do CTI de um hospital de ensino de Minas Gerais. Os dados foram coletados por meio de entrevistas, guiadas por roteiro semiestruturado e submetidos à análise de conteúdo. Resultados: Emergiram duas categorias temáticas: Expressões da ética: Influências dos valores, virtudes, relacionamentos e organização do trabalho; e, Distresse moral no contexto das expressões éticas. Conclusão: Na prática do enfermeiro intensivista, a ética está relacionada, principalmente, ao exercício da advocacia do paciente. No entanto, lacunas existentes nos relacionamentos com a equipe e na organização do trabalho, fomentam situações de conflitos geradoras de distresse moral.

Métodos: Investigación cualitativa, descriptiva, interpretativa y analítica, realizada con 12 enfermeros sobrecargados en las guardias diurnas de centros de terapia intensiva de un hospital universitario de Minas Gerais. Los datos fueron recopilados por medio de entrevista, con guion semiestructurado y sometidos al análisis de contenido. Resultados: Surgieron dos categorías temáticas. Expresiones de la ética: influencias de los valores, virtudes, relaciones y organización del trabajo, y Distrés moral en el contexto de las expresiones éticas. Conclusión: En la práctica del enfermero intensivista, la ética está relacionada principalmente con el ejercicio de la defensa del paciente. Sin embargo, los vacíos existentes en las relaciones con el equipo y en la organización del trabajo fomentan situaciones de conflicto que generan distrés moral.


Abstract Objective: To understand ethics and moral distress expressions in intensive care nursing practice. Methods: This is a qualitative, descriptive, interpretive and analytical research, developed with 12 nurses working on the day shifts in a ICU of a teaching hospital in Minas Gerais. Data were collected through interviews, guided by a semi-structured script and submitted to content analysis. Results: Two thematic categories emerged: Ethics expressions: influences of values, virtues, relationships, and work organization; Moral distress in the context of ethics expressions. Conclusion: In intensive care nursing practice, ethics is mainly related to the practice of patient advocacy. However, gaps in relationships with the team and in work organization encourage conflict situations that generate moral distress.

ABSTRACT Objective: to understand how the meanings attributed to work by professionals of the Family Health Strategy are linked to the mechanisms of risk and protection, influencing resilience. Method: a qualitative case study, conducted with professionals of the Family Health Strategy. 62 professionals participated: 14 doctors, 16 nurses, 16 nursing technicians and 16 community health agents. The data collected through interviews which were guided by semi-structured script and observation were submitted to thematic content analysis and Atlas.ti Software, version 7. Results: adversities in the work environment emerged from the analysis such as work overload, lack of articulation between health network services, insufficient autonomy and recognition and inadequate working conditions. Such experiences imply risks to the meanings and the development of work, limiting more resolute actions in the care of the population. Mechanisms that provide protection against risks were also highlighted, strengthening the positive meanings given to work that favor resilience. Conclusion: the study made it possible to understand how the meanings attributed to work are linked to risk and protection mechanisms, influencing resilience processes. Such reflections can support strategies to reduce adversity in the work environment, stimulate the professionals' potentialities and positive senses about work, aiming at reaching a healthy work environment, which affect the care actions to the population.


RESUMEN Objetivo: comprender cómo los significados atribuidos al trabajo por parte de los profesionales de la Estrategia de salud familiar se articulan con los mecanismos de riesgo y protección, influyendo en la resiliencia. Método: una investigación de enfoque cualitativo, un estudio de caso único, realizado con profesionales de la Estrategia de Salud Familiar. Participaron 62 profesionales: 14 médicos, 16 enfermeras, 16 técnicos de enfermería y 16 agentes de salud comunitaria. Los datos recopilados a través de entrevistas, orientas por guiones semiestructurados y observación, se enviaron a análisis de contenido temático y Atlas ti Software, versión 7. Resultados: del análisis surgieron adversidades en el entorno laboral relacionadas con la sobrecarga de trabajo, falta de articulación entre los servicios de salud, autonomía y reconocimiento insuficientes y condiciones de trabajo inadecuadas. Esas experiencias implican riesgos para los sentidos y el desarrollo del trabajo, limitando acciones más resueltas en el cuidado de la población. También se evidenciaron mecanismos que brindan protección contra los riesgos, fortaleciendo los significados positivos dados al trabajo que favorecen la resiliencia. Conclusión: el estudio permitió comprender cómo los significados atribuidos al trabajo están vinculados a los mecanismos de riesgo y protección, influyendo en los procesos de resiliencia. Esas reflexiones pueden apoyar estrategias para reducir la adversidad en el entorno laboral, estimular el potencial de los profesionales y los significados positivos sobre el trabajo, con el objetivo de lograr un entorno laboral saludable, que tenga un impacto en las acciones de atención para la población.

Método: pesquisa de abordagem qualitativa, do tipo estudo de caso único, realizada com profissionais da Estratégia Saúde da Família. Participaram 62 profissionais: 14 médicos, 16 enfermeiros, 16 técnicos em enfermagem e 16 agentes comunitários de saúde. Dados coletados por meio de entrevista, guiada por roteiro semiestruturado e observação, sendo submetidos à análise de conteúdo temática e ao Software Atlas ti, versão 7. Resultados: da análise, emergiram adversidades no ambiente laboral relacionadas à sobrecarga de trabalho, ausência de articulação entre os serviços da rede de saúde, autonomia e reconhecimento insuficientes e condições de trabalho inadequadas. Tais vivências implicam riscos aos sentidos e ao desenvolvimento do trabalho, limitando ações mais resolutivas no cuidado à população. Foram evidenciados, também, mecanismos que conferem proteção diante dos riscos, fortalecendo os sentidos positivos conferidos ao trabalho que favorecem à resiliência. Conclusão: o estudo possibilitou compreender como os sentidos atribuídos ao trabalho se articulam aos mecanismos de risco e proteção, influenciando processos de resiliência. Tais reflexões podem subsidiar estratégias de redução de adversidades no ambiente laboral, estimular as potencialidades dos profissionais e sentidos positivos sobre o trabalho, com vistas ao alcance de ambiente laboral saudável, que repercutam nas ações de cuidado à população.

Neural crest cells (NCCs) contribute to several tissues during embryonic development. NCC formation depends on activation of tightly regulated molecular programs at the neural plate border (NPB) region, which initiate NCC specification and epithelial-to-mesenchymal transition (EMT). Although several approaches to investigate NCCs have been devised, these early events of NCC formation remain largely unknown in humans, and currently available cellular models have not investigated EMT. Here, we report that the E6 neural induction protocol converts human induced pluripotent stem cells into NPB-like cells (NBCs), from which NCCs can be efficiently derived. NBC-to-NCC induction recapitulates gene expression dynamics associated with NCC specification and EMT, including downregulation of NPB factors and upregulation of NCC specifiers, coupled with other EMT-associated cell-state changes, such as cadherin modulation and activation of TWIST1 and other EMT inducers. This strategy will be useful in future basic or translational research focusing on these early steps of NCC formation.

OBJECTIVE: To understand the professional practice of nurses and their influence on the development of moral sensibility. METHODS: A qualitative and descriptive study, conducted between November 2015 and February 2016 in the hospitalization units of the medical clinic of two large hospitals located in Belo Horizonte, Minas Gerais. 14 nurses participated. Data collection was done through interviews, guided by semi-structured scripts. The data were submitted to content analysis. RESULTS: Two categories emerged: Professional practice of the nurse: internal and external goods and Moral sensitivity and the interface with the professional practice of nurses. The development of the moral sensitivity of nurses is influenced by factors related to professional practice, such as interpersonal relations, ethical education and management activities. Final considerations: In the professional practice, moral sensibility is an integral part of the ethical decision-making process in services, being essential for quality care.

OBJECTIVES: to understand the pleasure-suffering duality experienced by students in stricto sensu Graduate Program in Nursing. METHODS: qualitative, integrated single-case study based on the Psychodynamics of Work. The setting was the stricto sensu Graduate Program in Nursing in Brazil. Two Graduate Programs were chosen. Data collection took place between May 2017 and April 2018, with 23 students, through interviews guided by semi-structured script and Documentary Analysis. Content Analysis was carried out. RESULTS: two analysis units emerged: Situations generating pleasure: bridges built by the Graduate Program and Situations generating suffering: walls of the Graduate Program. They were discussed in two dimensions - organizational and socio-professional relations. FINAL CONSIDERATIONS: the results can contribute to the planning of actions capable of potentiating the creation of "bridges" that lead to reach of pleasure in the Graduate Program. It impels the confrontation of suffering-generating situations for the student.

RESUMO Objetivo: compreender a construção identitária do enfermeiro diante do processo de distresse moral, na perspectiva de enfermeiros intensivistas. Métodos: estudo qualitativo, descritivo, interpretativo e analítico, desenvolvido em 2016 em um centro de terapia intensiva de um hospital universitário. Participaram da pesquisa 12 enfermeiros do turno diurno. A coleta de dados se deu por meio de entrevistas orientadas por roteiro semiestruturado e os dados foram submetidos à análise temática de conteúdo. Resultados: na perspectiva dos enfermeiros, a relação entre a configuração identitária e o distresse moral se deu mediante vivências de situações no cotidiano do trabalho no CTI que os impedem de exercer a prática conforme acreditam ser o modo correto, tais como: o modelo assistencial de organização do trabalho; o desenvolvimento de atividades técnico-assistenciais em detrimento da gestão do cuidado; o trabalho mais mecânico do que intelectual; o não pertencimento à equipe de saúde; e aspectos organizacionais como as escalas de trabalho intensas, o relacionamento com a coordenação e as dificuldades em implantação de melhorias no processo de trabalho. Considerações Finais: concluiu-se que o trabalho do enfermeiro é permeado por vivências de problemas morais que influenciam na construção identitária e no seu compromisso ético em prestar o cuidado que julga ser o adequado ao paciente crítico. Isso porque a identidade constrói-se a partir da percepção de si e do seu trabalho e é influenciada pelos relacionamentos interpessoais, pela organização do trabalho e suas vivências no cotidiano.


RESUMEN Objetivo: comprender la construcción identitaria del enfermero ante el proceso de angustia moral, desde la perspectiva del enfermero de cuidados intensivos. Método: estudio cualitativo, descriptivo, interpretativo y analítico, llevado a cabo en 2016 en el centro de cuidados intensivos de un hospital universitario. En la investigación participaron 12 enfermeros del turno diurno. La recogida de datos se realizó mediante entrevistas semiestructuradas; los datos fueron sometidos a análisis de contenido temático. Resultados: desde la perspectiva de los enfermeros, la relación entre la configuración identitaria y la angustia moral se dio a través de vivencias de situaciones en el día a día de trabajo en la UCI pues sienten que no pueden ejercer la práctica de la forma que consideran ser más correcta, tales como: el modelo asistencial de organización del trabajo; el desarrollo de actividades técnicas y asistenciales en vez de gestión del cuidado; trabajo más mecánico que intelectual; no pertenecer al equipo de salud; y aspectos organizativos como horarios de trabajo intensos, la relación con la coordinación y las dificultades para implementar mejoras en el proceso de trabajo. Conclusión: el trabajo del enfermero está impregnado de vivencias de problemas morales que inciden en la construcción identitaria y en su compromiso ético para brindar los cuidados que estiman oportunos para el paciente crítico. Esto se debe a que la identidad se construye a partir de la percepción de uno mismo y de su trabajo y está influenciada por las relaciones interpersonales, la organización del trabajo y sus vivencias diarias.


ABSTRACT Objective: to understand nurses' identity construction in the face of the moral distress process, from the perspective of intensive care nurses. Methods: qualitative, descriptive, interpretive and analytical study, developed in 2016 in an intensive care center of a university hospital. Twelve nurses from the day shift participated in the research. Data collection took place through interviews guided by a semi-structured script and the data were submitted to thematic content analysis. Results: from the nurses' perspective, the relationship between the identity configuration and moral distress occurred through experiences of situations in the daily work at the ICU that prevent them from exercising the practice as they believe it is the correct way, such as: the model of care of work organization; the development of technical-assistance activities to the detriment of care management; work more mechanically than intellectually; not belonging to the health team; and organizational aspects such as intense work schedules, the relationship with coordination and difficulties in implementing improvements in the work process. Final Considerations: it was concluded that the nurse's work is permeated by experiences of moral problems that influence the construction of identity and their ethical commitment to provide the care that they deem appropriate for critical patients. This is because identity is built from the perception of oneself and one's work and is influenced by interpersonal relationships, the organization of work and their daily experiences.

Mammalian carnivores are considered a key group in maintaining ecological health and can indicate potential ecological integrity in landscapes where they occur. Carnivores also hold high conservation value and their habitat requirements can guide management and conservation plans. The order Carnivora has 84 species from 8 families in the Neotropical region: Canidae; Felidae; Mephitidae; Mustelidae; Otariidae; Phocidae; Procyonidae; and Ursidae. Herein, we include published and unpublished data on native terrestrial Neotropical carnivores (Canidae; Felidae; Mephitidae; Mustelidae; Procyonidae; and Ursidae). NEOTROPICAL CARNIVORES is a publicly available data set that includes 99,605 data entries from 35,511 unique georeferenced coordinates. Detection/non‐detection and quantitative data were obtained from 1818 to 2018 by researchers, governmental agencies, non‐governmental organizations, and private consultants. Data were collected using several methods including camera trapping, museum collections, roadkill, line transect, and opportunistic records. Literature (peerreviewed and grey literature) from Portuguese, Spanish and English were incorporated in this compilation. Most of the data set consists of detection data entries (n = 79,343; 79.7%) but also includes non‐detection data (n = 20,262; 20.3%). Of those, 43.3% also include count data (n = 43,151). The information available in NEOTROPICAL CARNIVORES will contribute to macroecological, ecological, and conservation questions in multiple spatio‐temporal perspectives. As carnivores play key roles in trophic interactions, a better understanding of their distribution and habitat requirements are essential to establish conservation management plans and safeguard the future ecological health of Neotropical ecosystems. Our data paper, combined with other largescale data sets, has great potential to clarify species distribution and related ecological processes within the Neotropics. There are no copyright restrictions and no restriction for using data from this data paper, as long as the data paper is cited as the source of the information used. We also request that users inform us of how they intend to use the data.

Como subunidades de análisis, se eligieron dos programas de Postgrado. La recolección de datos ocurrió entre mayo de 2017 y abril de 2018, con 23 estudiantes, a través de entrevistas guiadas por guión semiestructurado y Análisis Documental. Se realizó Análisis de Contenido Temático. Resultados: emergieron 2 unidades de análisis: Situaciones generadoras de placer: los puentes construidos por la Postgrado y Situaciones generadoras de sufrimiento: los muros del Postgrado, ambas discutidas em duas dimensões - organizacional e relações socioprofissionais. Consideraciones finales: los resultados pueden contribuir a la planificación de acciones capaces de potenciar la creación de "puentes" que lleven al alcance del placer en el Postgrado, impulsando el enfrentamiento de situaciones propulsoras del sufrimiento de lo estudiante.


RESUMO Objetivos: compreender a dualidade prazer-sofrimento vivenciada por discentes na Pós-Graduação stricto sensu em Enfermagem. Métodos: pesquisa qualitativa, do tipo estudo de caso único integrado, fundamentada na Psicodinâmica do Trabalho. O cenário foi a Pós-Graduação stricto sensu em Enfermagem do Brasil. Como subunidades de análise, elegeram-se dois programas de Pós-Graduação. A coleta de dados ocorreu entre maio de 2017 e abril de 2018, com 23 discentes, por meio de entrevistas guiadas por roteiro semiestruturado e Análise Documental. Realizou-se Análise de Conteúdo Temática. Resultados: emergiram duas unidades de análise: Situações geradoras de prazer: as pontes construídas pela Pós-Graduação e Situações geradoras de sofrimento: os muros da Pós-Graduação, ambas discutidas em duas dimensões - organizacional e relações socioprofissionais. Considerações finais: os resultados podem contribuir para o planejamento de ações capazes de potencializar a criação de "pontes" que levem ao alcance do prazer na Pós-Graduação, impulsionando o enfrentamento de situações propulsoras do sofrimento discente.
