RESUMEN

Immune checkpoint inhibitors have revolutionized cancer therapy leading to exceptional success. However, there is still the need to improve their efficacy in non-responder patients. Natural killer (NK) cells represent the first line of defence against tumours, due to their ability to release immunomodulatory cytokines and kill target cells that have undergone malignant transformation. Harnessing NK cell response will open new possibilities to improve control of tumour growth. In this respect inhibitory checkpoints expressed on these innate lymphocytes represents a promising target for next-generation immunotherapy. In this review, we will summarize recent evidences on the expression of NK cells receptors in cancer, with a focus on the inhibitory checkpoint programmed cell death protein 1 (PD-1). We will also highlight the strength and limitations of the blockade of PD-1 inhibitory pathway and suggest new combination strategies that may help to unleash more efficiently NK cell anti-tumour response.

The epidemiological research benefits from an accurate characterization of both spatial and temporal variability of exposure to air pollution. This work aims at proposing a method to combine the high spatial resolution of Land Use Regression (LUR) models with the high temporal resolution of fixed site monitoring data, to model spatiotemporal variability of NO2 over a wide geographical area in Northern Italy. We developed seasonal LUR models to reconstruct the spatial distribution of a scaling factor that relates local concentrations to those measured at two reference central sites, one for the northern flat area and one for the southern mountain area. We calculated the daily average concentrations at 19 locations spread over the study areas as the product of the local scaling factor and the reference central site concentrations. We evaluated model performance comparing modeled and measured NO2 data. LUR model's R2 ranges from 0.76 to 0.92. The main predictors refers substantially to traffic, industrial land use, buildings volume and altitude a.s.l. The model's performance in reproducing measured concentrations was satisfactory. The temporal variability of concentrations was well captured: Spearman correlation between model and measures was >0.7 for almost all sites. Model's average absolute errors were in the order of 10µgm-3. The model for the southern area tends to overestimate measured concentrations. Our modeling framework was able to reproduce spatiotemporal differences in NO2 concentrations. This kind of model is less data-intensive than usual regional atmospheric models and it may be very helpful to assess population exposure within studies in which individual relevant exposure occurs along periods of days or months.

Innate lymphoid cells (ILCs) have a central role in innate defenses against pathogens, lymphoid organogenesis, and tissue remodeling. They have been detected in human decidua, however, their role in this tissue remains unclear. Successful pregnancy requires an early inflammatory phase favoring implantation and tissue remodeling as well as a subsequent regulatory phase to prevent fetal rejection and supporting neoangiogenesis. Here, we show that, during the first trimester of pregnancy, neutrophils infiltrate decidua basalis and are more abundant in normal pregnancy than in spontaneous miscarriages. Decidual neutrophils localize in proximity of NCR+ILC3, which may influence neutrophil migration and survival given their production of CXCL8 and granulocyte macrophage colony-stimulating factor (GM-CSF). Moreover, NCR+ILC3-derived GM-CSF was found to induce the expression of heparin-binding EGF-like growth factor and IL1ra in neutrophils, two proteins/cytokines involved in tissue remodeling and maintenance of pregnancy. Our data suggest that the simultaneous presence of NCR+ILC3 and neutrophils in decidual tissues and their possible cross talk, may have a role in the early phases of pregnancy.

OBJECTIVE: We describe an emerging entity, recently recognized as a pitfall in the diagnostic practice among eosinophilic renal cell tumours. METHODS: A 60-year-old male underwent enucleation of a 1.2 cm nodule. Immunohistochemistry and FISH analysis were performed. RESULTS: Histology revealed a neoplasm composed of large cells with eosinophilic cytoplasm, Fuhrman grade 3, arranged in papillae. At the immunohistochemical level, cells showed positivity for AMACR and CD10. Fluorescence in situ hybridization (FISH) demonstrated gains of chromosomes 7 and 17 and loss of Y. A diagnosis of oncocytic papillary renal cell carcinoma was made. CONCLUSIONS: The distinction between renal oncocytoma and oncocytic papillary renal cell carcinoma is of substantial importance because of their different behaviour and prognosis, since the latter has malignant potential. Although the available evidence supporting tumour enucleation as the surgical treatment for renal cortical tumours < or = 4 cm, due to aforementioned clinicopathological features such tumours need to be evaluated using appropriate immunophenotypical and cytogenetic analyses.

To assess the differences in the granulometry of milk fat globules between swine and bovine species, milk samples from 30 lactating sows were analyzed for fat globule dimensions and compared with cow milk samples. Results showed differences between the fat globules: sow milk presents reduced globule diameters compared with cow milk (volume-weighted diameter 2.62 vs. 3.27 microm, p < 0.001) and reduced interglobular distance. A positive relationship was observed between milk fat content and globule diameter, while a slight, insignificant inverse trend was detected between the day of lactation and fat globule diameter. These complex interactions between milk lipids, globule membrane proteins, and globule dimensions provide a better understanding of digestion/absorption phenomena in the design of milk replacers.

Granulosa cells from bovine and porcine ovaries were cultured either in monolayer or in follicle-like barium alginate capsules for 6 days. Morphological investigation by electron scanning microscopy indicated that culture in a three-dimensional (3D) system allows self-organization of spherical-polyhedral shape cells. The luteinization index (progesterone:17beta-estradiol ratio) was significantly higher for monolayer cells than for the 3D cell culture system, confirming the results of morphological analysis and indicating more physiological growth. The encapsulated 3D culture system appears to be a promising way of obtaining in vitro maturation and development of follicles and oocytes.

Platelet count (PLT) mean platelet volume (MPV) and plateletcrit (PCT) were determined for 117 Landrace x Large White piglets aged 3-21 days; counts were performed with an automated blood cell counter (ABX Pentra 120). Reference values were estimated following the guidelines of the International Federation of Clinical Chemistry (IFCC) and the International Committee for Standardization in Haematology (ICSH). The calculated central 95% reference limits for PLT was 49.9-516.2 x 10(9)/1, for MPV 6.71-9.91 fl and for PCT 0.009-0.395%. When observations are divided into three age groups (about 1 week each) there is an increase in mean PLT count and PCT in 2-week piglets, and a decrease in MPV from the first to the third week of life. These reference values provide guidelines for interpreting for experimental and clinical observations, as well as for monitoring of the health status of similar aged piglets determined using automated impedance-light focusing methodology.

Blood samples were taken between February and April from 105 healthy Stamboek pre-pubertal gilts, aged 1-3 months, which were housed at a modern pig farm in northern Italy. The blood was analysed for nine haematological and nine selected haematochemical variables by means of automated and semiautomated blood analysis apparatus. After detection and rejection of outliers, the data were submitted to reference limits evaluation, also taking into account the limits for the red blood cell volume histogram as the anisocytosis index. Some haematological reference values deal with previously published data; in the haematochemical parameters, several discrepancies between evaluated limits and existing reference limits were noted, mainly for aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase levels and total protein concentration. The results confirm the relevance of age in determining blood reference intervals and that 'normal' values should be determined by each laboratory, taking into account the age of subjects, the sample size and methods of analysis.

BACKGROUND: Nitric oxide (NO) plays an important role as an inflammatory mediator in the airways. Since chronic obstructive pulmonary disease (COPD) is characterised by airway inflammation, a study was undertaken to determine NO levels in the exhaled air of patients with COPD. METHODS: Two groups of patients with clinically stable COPD were studied, 10 current smokers and 10 ex-smokers. Two control groups of healthy subjects consisting of 10 current smokers and 20 non-smokers were also studied. Exhaled NO levels were measured by the collection bag technique and NO chemiluminescence analyser. RESULTS: Mean (SE) levels of exhaled NO in ex-smokers and current smokers with COPD (25.7 (3.0) ppb and 10.2 (1.4) ppb, respectively) were significantly higher than in non-smoker and current smoker control subjects (9.4 (0.8) ppb and 4.6 (0.4) ppb, respectively). In current smokers with COPD exhaled levels of NO were significantly lower than in ex-smokers. In this latter group of patients there was a significant negative correlation between smoking history (pack years) and levels of exhaled NO (r = -0.8, p = 0.002). A positive correlation was seen between forced expiratory volume in one second (FEV1) and levels of exhaled NO (r = 0.65, p = 0.001) in patients with COPD. CONCLUSIONS: These data show that exhaled NO is increased in patients with stable COPD, both current and ex-smokers, compared with healthy control subjects.

BACKGROUND: Measurement of nitric oxide (NO) in exhaled air shows promise as a non-invasive method of detecting lung inflammation. However, variable concentrations of NO are measured in environmental air. The aim of this study was to verify a possible relationship between exhaled NO and atmospheric NO values during high atmospheric NO days. METHOD: Exhaled air from 78 healthy non-smokers of mean age 35.3 years was examined for the presence of NO using a chemiluminescence NO analyser and NO levels were expressed as part per billion (ppb). The exhaled air from all the subjects was collected into a single bag and into two sequential bags. Before each test atmospheric NO was measured. RESULTS: The mean (SE) concentration of exhaled NO collected into the single bag was 17.1 (0.6) ppb while the mean values of exhaled NO in bags 1 and 2 were 16.7 (1.3) ppb and 13.8 (1.2) ppb, respectively. The atmospheric NO concentrations registered before each test varied from 0.4 to 71 ppb. There was a significant correlation between exhaled NO in the single bag and atmospheric NO (r = 0.38, p = 0.001). The atmospheric NO concentration also correlated with exhaled NO both in bag 1 (r = 0.44, p = 0.0001) and in bag 2 (r = 0.42, p = 0.0001). These correlations disappeared with atmospheric NO concentrations lower than 35 ppb. CONCLUSIONS: These results indicate a relationship between atmospheric NO and NO levels measured in exhaled air, therefore exhaled NO should not be measured on very high atmospheric NO days.

RESUMEN

We describe two children with Leri-Weill syndrome (Dyschondrosteosis), one of which showed the clinical features at very early age. Stature was moderately reduced in both, due to shortening of the bones of the legs. Furthermore were evident in them the clinical and radiological features of the propositus and of their affected parents are described. The problems concerning the early recognizing of the disease and the genetic inheritance are discussed.
