RESUMEN

Approximately 90% of beef cattle on feed in the United States receive at least one anabolic implant, which results in increased growth, efficiency, and economic return to producers. However, the complete molecular mechanism through which anabolic implants function to improve skeletal muscle growth remains unknown. This study had 2 objectives: (1) determine the effect of polyamines and their precursors on proliferation rate in bovine satellite cells (BSC); and (2) understand whether trenbolone acetate (TBA), a testosterone analog, has an impact on the polyamine biosynthetic pathway. To address these, BSC were isolated from 3 finished steers and cultured. Once cultures reached 75% confluency, they were treated in 1% fetal bovine serum (FBS) and/or 10 nM TBA, 10 mM methionine (Met), 8 mM ornithine (Orn), 2 mM putrescine (Put), 1.5 mM spermidine (Spd), or 0.5 mM spermine (Spe). Initially, a range of physiologically relevant concentrations of Met, Orn, Put, Spd, and Spe were tested to determine experimental doses to implement the aforementioned experiments. One, 12, or 24 h after treatment, mRNA was isolated from cultures and abundance of paired box transcription factor 7 (Pax7), Sprouty 1 (Spry), mitogen-activated protein kinase-1 (Mapk), ornithine decarboxylase (Odc), and S adenosylmethionine (Amd1) were determined, and normalized to 18S. No treatment × time interactions were observed (P ≥ 0.05). Treatment with TBA, Met, Orn, Put, Spd, or Spe increased (P ≤ 0.05) BSC proliferation when compared with control cultures. Treatment of cultures with Orn or Met increased (P ≤ 0.01) expression of Odc 1 h after treatment when compared with control cultures. Abundance of Amd1 was increased (P < 0.01) 1 h after treatment in cultures treated with Spd or Spe when compared with 1% FBS controls. Cultures treated with TBA had increased (P < 0.01) abundance of Spry mRNA 12 h after treatment, as well as increased mRNA abundance of Mapk (P < 0.01) 12 h and 24 h after treatment when compared with 1% FBS control cultures. Treatment with Met increased (P < 0.01) mRNA abundance of Pax7 1 h after treatment as compared with 1% FBS controls. These results indicate that treatments of BSC cultures with polyamines and their precursors increase BSC proliferation rate, as well as abundance of mRNA involved in cell proliferation. In addition, treatment of BSC cultures with TBA, polyamines, or polyamine precursors impacts expression of genes related to the polyamine biosynthetic pathway and proliferation.

Our objectives were to determine the effect of post rigor calcium chloride injection or freezing on 1) sarcoplasmic calcium concentration and calpain-2 activity of beef longissimus lumborum (LL) and semimembranosus (SM) steaks aged 1, 4, and 14days post-treatment and on 2) Warner-Bratzler shear force, water holding capacity, and consumer acceptability of LL and SM steaks aged 4 and 14days post-treatment. Free calcium levels in the calcium, frozen, and control steaks averaged 1256, 127, and 121µM for the LL and 1520, 120, and 111µM for the SM, respectively. Measurable LL native calpain-2 activity was lower in calcium and frozen steaks than control steaks (P<0.01), while SM native calpain-2 activity was lowest in calcium steaks and intermediate in frozen steaks (P<0.01). LL calcium steaks were more tender (P=0.04) than control steaks. In conclusion, calcium chloride injection and freezing activate calpain-2 earlier postmortem in both muscles and calcium injection improves LL tenderness.

We hypothesized that variable composition in finishing rations, more specifically; the proportion of potato-by-product (PBP) and rumen protected histidine (His) supplementation may influence growth and meat quality attributes. Two different diets were fed (1) finishing ration with corn and barley as grains (CB, n = 20) and (2) substitution of 10% corn, DM basis, with PBP (PBP, n = 20). Additionally, half of each dietary treatment received 50 g/hd/d rumen protected His (HS, n= 20) while the other half received no supplement (NS, n = 20). Inclusion of 10% PBP or HS did not affect growth or carcass traits. Color stability was analyzed using Hunter color values as well as AMSA visual appraisal in both longissimus thoracis (LT) and gluteus medius (GM) muscles. The LT, but not the GM, of CB steers was more color stable over a 9 d simulated retail display compared to those fed a PB diet. Steers receiving HS produced significantly (P < 0.05) more color stable LT and GM steaks.

Investigating the genetic and physiological drivers of postweaning residual feed intake (RFI) and finishing phase feed efficiency (FE) may identify underlying mechanisms that are responsible for the variation in these complex FE traits. The objectives were 1) to evaluate the relationship of serum IGF-I concentration and muscle gene expression with postweaning RFI and sire maintenance energy (MEM) EPD and 2) to determine fiber type composition as it relates to postweaning RFI and finishing phase FE. Results indicate that RFI and serum IGF-I concentration were not associated (P > 0.05); however, negative correlations (P < 0.05) between sire MEM EPD and serum IGF-I concentration were observed. Gene expression differences between high- and low-RFI animals were observed in cohort 1, where IGFBP5 expression was greater (P < 0.05) in high-RFI animals. When animals were grouped according to sire MEM EPD, the low MEM EPD group of cohort 1 showed greater muscle mRNA expression (P < 0.01) of fatty acid synthase (FASN) and marginally (P < 0.10) greater expression of IGFBP5 and C/EBP alpha (C/EBPα) whereas the high MEM EPD group of cohort 2 had greater muscle mRNA expression of IGFBP2 (P < 0.05) and C/EBPα (P ≤ 0.01) and marginally (P < 0.10) greater expression of IGFBP3. Biopsy tissue samples collected at harvest revealed that the percentage of type IIa fibers was lower (P ≤ 0.05) in high-RFI steers, with a similar trend (P < 0.10) being observed in high finishing phase FE steers. The percentage of type IIb fibers was higher (P < 0.05) in high-RFI (and finishing phase FE) steers than in low-RFI (and finishing phase FE) steers. There was a marginal, negative correlation between RFI and type I (r = -0.36, P = 0.08) and IIa (r = -0.37, P = 0.07) fiber percentages and a positive correlation (r = 0.48, P = 0.01) between RFI and type IIb fiber percentage whereas finishing phase FE was negatively correlated (r = -0.43, P = 0.03) with type I fiber percentage and positively correlated (r = 0.44, P = 0.03) with type IIb fiber percentage. Therefore, our data indicate that 1) serum IGF-I (collected at weaning) is not an indicator of postweaning RFI, 2) the GH-IGF axis appears to have some involvement with RFI at the molecular level; however, muscle gene expression results were not consistent across cohorts, and 3) low-RFI animals may have the ability to more efficiently maintain and accrete muscle mass due to their fiber type composition, specifically a greater proportion of type I fibers.

Myoblast differentiation is mediated by a cascade of changes in gene expression including transcription factors such as myogenin. Subsequent to myoblast differentiation, there is an increase in expression of the transmembrane protein NADPH oxidase (Nox). Nox is one of the primary factors for the generation of reactive oxygen species (ROS) in myogenic (C2C12) cells. Recently, ROS have been shown to be important regulators of several intracellular signaling pathways, and the full extent of their regulatory roles is yet to be discovered. In the present study, qRT PCR analysis demonstrated that Nox4 isoform is primarily expressed in differentiating C2C12 cells and contributes to the generation of ROS in C2C12 myoblast during differentiation. Over-expression and silencing of Nox4 expression during myoblast differentiation was accompanied by a reduction in intracellular ROS concentrations and an alteration in the expression patterns of Myf5, Pax7, MyoD1, and myogenin. This modulation was found to be associated with ERK1/2 phosphorylation. In both over-expression and reduced expression of Nox4, we found significant reductions in ERK1/2 phosphorylation. This indicates that cellular differentiation may be affected by Nox4-mediated endogenous ROS generation. These data suggest a new opportunity to study the temporal expression of Nox4 in the generation of ROS accompanying changes in myogenic differentiation.

Energy expenditure is a physiological process that may be closely associated with residual feed intake (RFI). The maintenance energy (ME(M)) EPD was developed by the Red Angus Association of America (RAAA) and is used as an indicator of energy expenditure. The objectives of this study were to evaluate and quantify the following relationships using progeny of Red Angus (RA) sires divergent for ME(M) EPD: 1) postweaning RFI and finishing phase feed efficiency (FE), 2) postweaning RFI and end-product quality, and 3) postweaning RFI and sire ME(M) EPD. A total of 12 RA sires divergent for ME(M) EPD were chosen using the RAAA-generated ME(M) EPD values and were partitioned into 2 groups: high ME(M) EPD (≥4 Mcal/mo) and low ME(M) EPD (<4 Mcal/mo), based on the breed average of 4 Mcal/mo. Commercial crossbred cows were inseminated to produce 3 cohorts of progeny, which were tested for postweaning RFI (cohorts 1, 2, and 3) and finishing phase FE (cohorts 1 and 3). Results indicate that postweaning RFI and finishing phase FE of steer progeny tended to be positively correlated (r = 0.38; P = 0.06) in cohort 1 and were positively correlated (r = 0.50; P = 0.001) in cohort 3. In addition, postweaning RFI was not phenotypically correlated (P > 0.05) with any carcass traits or end-product quality measurements. Sire ME(M) EPD was phenotypically correlated (P < 0.05) with carcass traits in cohort 1 (HCW, LM area, KPH, fat thickness, and yield grade) and cohort 2 (KPH and fat thickness). Since variation in measured LM area was not explained by the genetic potential of rib eye area EPD, and therefore, the observed correlation between sire ME(M) EPD and measured LM area may suggest an association between ME(M) EPD and LM area. A correlation (r = 0.24; P = 0.02) was observed between postweaning RFI and ultrasound intramuscular fat percentage in cohort 2 but was not detected in cohorts 1 or 3. In addition, no phenotypic relationship was observed (P > 0.05) between progeny postweaning RFI and sire ME(M) EPD. Therefore, results suggest 1) RFI measured during the postweaning growth phase is indicative of FE status in the finishing phase, 2) neither RFI nor sire ME(M) EPD negatively affected carcass or end-product quality, and 3) RFI and sire ME(M) EPD are not phenotypically associated.

A total of 42 F(1) Red Angus progeny from sires divergent in maintenance energy (ME(M)) EPD were analyzed to determine whether selecting for sire ME(M) would alter end-product meat quality. Data from animals were grouped based on the divergence of the ME(M) EPD of their sire from the Red Angus Association-reported breed average and defined as either high or low, the assumption being that high-ME(M) cattle are less efficient because their maintenance requirements represent a larger proportion of their dietary intake. Steer progeny (n = 7) from the high group produced bottom round steaks with a greater a* (redness) color value (P = 0.02) after 5 d in a simulated retail display when compared with bottom round steaks from the low group (n = 18). Bottom round steaks from the high group had a greater b* (yellowness) color value at d 1 (P = 0.03) and d 5 (P = 0.01) of retail display. Samples from the biceps femoris were taken at 12 mo (from both steers and heifers) and 15 mo (from steers only) of age for fiber type proportion analysis. At 12 mo of age, steers from the low group had more type I fibers (P = 0.02), whereas steers from the high group had more type IIb fibers (P = 0.01). Furthermore, samples from steers in the low group at 15 mo had more type I fibers (P = 0.02), and steers from the high group maintained more type IIb fibers (P = 0.02). No changes in fiber type proportions were observed between the high- and low-ME(M) EPD heifers (n = 17). Relative mRNA abundance of genes involved in the synthesis, storage, and breakdown of glycogen were analyzed as a variable important for meat quality, but no statistical differences were observed. At 12 mo age, glycogenin (glyc) was negatively correlated with the proportion of type IIa fibers (r = -0.32 and P = 0.12) as well as with the proportion of type IIb fibers (r = -0.42 and P = 0.03) in the biceps femoris of the steers. In samples taken from the biceps femoris at 15 mo age, glyc was negatively correlated with the proportion of type IIa fibers (r = -0.42 and P = 0.03) in the steers. This indicates that relative mRNA expression of glyc may serve as a marker of muscle glycogen storage capacity in steers. Thus, selection for efficient Red Angus beef cattle based on sire ME(M) EPD does not adversely affect meat quality in F(1) progeny, based on the variables assessed in this study. Furthermore, selection for progeny from low-ME(M) EPD sires may improve fresh meat quality within Red Angus beef cattle.

An experiment was conducted in market dairy cows to determine the effect of feeding time and ractopamine hydrochloride (RAC) on growth performance, carcass characteristics, and end product quality. In 3 replicates, 9 Holstein cows per replicate (n = 27; 659 +/- 25.3 kg initial BW) culled from 3 dairies were randomly assigned to 3 treatments: 1) slaughter immediately (control), 2) feed for 90 d (NoR), or 3) feed for 90 d with RAC (312 mg.cow(-1).d(-1)) for the final 32 d (RAC). On d 0, NoR and RAC cows were placed in individual pens and fed a high concentrate diet (86% concentrate, DM basis) for 90 d before slaughter. All cows were subjectively scored for BCS and locomotion score on d 0, and NoR and RAC cows were evaluated again after 90 d. Individual DMI was recorded daily throughout the trial, and BW was collected every 14 d. Age and age x treatment did not affect (P > 0.05) any of the traits evaluated in this study. When cows fed for 90 d (NoR and RAC combined) were compared with nonfed controls, fed cows had greater (P < 0.001) final BCS, BW and HCW, lower (P < 0.001) final locomotion score, and greater (P < 0.03) dressing percentage, external fat thickness, and marbling score. Fed cows also tended to have more desirable yield grade (P = 0.08), ribeye area (P = 0.11), fat color (P = 0.09), lean maturity (P = 0.06), and quality grade (P = 0.09) compared with control cows. Warner-Bratzler shear force was not affected (P = 0.23) by feeding. However, a 12-member trained sensory panel revealed that fed cow carcasses had more desirable (P < 0.04) tenderness, juiciness, and overall acceptability than control cow carcasses. Flavor intensity also tended (P = 0.10) to be more desirable for fed vs. control cows. No difference (P > 0.10) in off-flavor was detected among treatments. Finally, there was no effect (P > 0.10) of RAC on growth performance, carcass characteristics, or end product quality. In conclusion, feeding a high concentrate diet for 90 d improved important live animal, carcass, and end product characteristics related to the quality and palatability of beef from market dairy cows; however, no effect of RAC supplementation was observed.

Canada is committed to reducing its greenhouse gas emissions to 6% below 1990 amounts between 2008 and 2012, and methane is one of several greenhouse gases being targeted for reduction. Methane production from ruminants is one area in which the agriculture sector can contribute to reducing our global impact. Through mathematical modeling, we can further our understanding of factors that control methane production, improve national or global greenhouse gas inventories, and investigate mitigation strategies to reduce overall emissions. The purpose of this study was to compile an extensive database of methane production values measured on beef cattle, and to generate linear and nonlinear equations to predict methane production from variables that describe the diet. Extant methane prediction equations were also evaluated. The linear equation developed with the smallest root mean square prediction error (RMSPE, % observed mean) and residual variance (RV) was Eq. I: CH(4), MJ/d=2.72 (+/-0.543) + [0.0937 (+/-0.0117) x ME intake, MJ/d] + [4.31 (+/-0.215) x Cellulose, kg/d] - [6.49 (+/-0.800) x Hemicellulose, kg/d] - [7.44 (+/-0.521) x Fat, kg/d] [RMSPE=26.9%, with 94% of mean square prediction error (MSPE) being random error; RV=1.13]. Equations based on ratios of one diet variable to another were also generated, and Eq. P, CH(4), MJ/d=2.50 (+/-0.649) - [0.367 (+/-0.0191) x (Starch:ADF)] + [0.766 (+/-0.116) x DMI, kg/d], resulted in the smallest RMSPE values among these equations (RMSPE=28.6%, with 93.6% of MSPE from random error; RV=1.35). Among the nonlinear equations developed, Eq. W, CH(4), MJ/d=10.8 (+/-1.45) x (1-e([-0.141 (+/-0.0381) x DMI, kg/d])), performed well (RMSPE=29.0%, with 93.6% of MSPE from random error; RV=3.06), as did Eq. W(3), CH(4), MJ/d=10.8 (+/-1.45) x [1-e({-[-0.034 x (NFC/NDF)+0.228] x DMI, kg/d})] (RMSPE=28.0%, with 95% of MSPE from random error). Extant equations from a previous publication by the authors performed comparably with, if not better than in some cases, the newly developed equations. Equation selection by users should be based on RV and RMSPE analysis, input variables available to the user, and the diet fed, because the equation selected must account for divergence from a "normal" diet (e.g., high-concentrate diets, high-fat diets).

Uterine crowding in the pig results in intrauterine growth restriction (IUGR), and permanently affects fetal muscle fibre development, representing production losses for the commercial pig herd. The present study sought to understand how different levels of uterine crowding in sows affects muscle fibre development in the early embryo at the time of muscle fibre differentiation and proliferation. Sows either underwent surgical, unilateral oviduct ligation (LIG; n = 10) to reduce the number of embryos in the uterus, or remained as intact, relatively-crowded controls (CTR; n = 10). Embryos and placentae were collected at Day 30 of gestation, and myogenic regulatory factor (MRF) transcript abundance was determined using real-time PCR for both myogenin (MYOG) and myoblast differentiation 1 (MYOD1). Unilateral tubal ligation resulted in lower numbers of embryos in utero, higher placental weights and a higher male : female sex ratio (P < 0.05). Relative MYOD1 expression was not different, but MYOG expression was higher (P < 0.05) in the LIG group embryos; predominantly due to effects on the male embryos. Relatively modest uterine crowding therefore affects MRF expression, even at very early stages of embryonic development, and could contribute to reported differences in fetal muscle fibre development, birthweight and thus post-natal growth performance in swine.

Feed restriction of primiparous sows during the last week of lactation has been shown to decrease embryonic growth and female embryo survival to Day 30 of gestation. This study sought to determine whether global DNA methylation and epigenetic gene expression of the candidate genes Igf2, Igf2r, and Xist were associated with these treatment effects. Given that these epigenetic traits are expected to be important for embryo viability, changes in variance for these traits at Day 30 were predicted to be reflected in the loss of abnormal embryos at this time. Consistent with this prediction, variance in DNA methylation was reduced (P < 0.001) in Restrict male embryo, and there was a tendency for reduced variance (P < 0.06) in Restrict female embryos. Variation in DNA methylation tended to be correlated (R = 0.42, P < 0.1) with the difference in variance of embryo weights between treatments (P < 0.01), suggesting a relationship between epigenetic changes and embryonic development. Variance in Igf2r expression tended to decrease (P < 0.07) in Restrict female embryos while variance in Xist expression tended to decrease in Restrict male embryos (P < 0.08), suggesting that maternally inherited epigenetic defects may cause female embryonic loss and reduced growth before Day 30 of gestation.

RESUMEN

The objective of this study was to investigate whether administration of L-Gln would affect mediators of acute phase response in postparturient dairy cows. Twenty-four multiparous Holstein cows were blocked by the expected day of calving and randomly assigned to 1 of the 3 treatment groups (n = 8/group): 1) i.v. infusion of 10 L of 0.85% NaCl (control), 2) i.v. infusion of 106, or 3) 212 g/d of L-Gln mixed with 10 L of 0.85% NaCl solution; each treatment was given 8 h/d for each of 7 consecutive days starting on d 1 after calving. Blood samples were collected 1 wk before the expected day of parturition as well as on d 0, 7, 14, and 21 after parturition; plasma concentrations of serum amyloid A (SAA), haptoglobin, and lipopolysaccharide-binding protein were measured by ELISA, and alpha(1)-acid glycoprotein was assessed by radial immunodiffusion. Concentrations of SAA, haptoglobin, and alpha(1)-acid glycoprotein increased in control cows after parturition, reaching peak values on d 0 or 7 postpartum (60, 1,093, and 963 microg/mL, respectively). Cows infused with 106 g/d of L-Gln had greater concentrations of SAA in plasma on d 14 and 21 compared with controls (62.8 vs. 30.2 and 71.1 vs. 34.5 microg/mL, respectively). Cows infused with 212 g/d of L-Gln had greater concentrations of SAA on d 7 (82.5 vs. 53.9 microg/mL) and lower concentrations of haptoglobin on d 14 and 21 postpartum compared with controls (264 vs. 621 and 175 vs. 587 microg/mL, respectively). Cows treated with 106 and 212 g/d of L-Gln had greater plasma lipopolysaccharide-binding protein concentrations on d 7 compared with control group (50.0 and 35.6 vs. 10.8 microg/mL, respectively). There were no treatment differences with respect to milk yield and DM intake during the experimental period. In conclusion, our data indicate that i.v. administration of L-Gln modulated acute phase mediators in dairy cows after parturition and warrants further research into the mechanisms behind these effects.
