Under ideal conditions, Escherichia coli cells divide after adding a fixed cell size, a strategy known as the adder. This concept applies to various microbes and is often explained as the division that occurs after a certain number of stages, associated with the accumulation of precursor proteins at a rate proportional to cell size. However, under poor media conditions, E. coli cells exhibit a different size regulation. They are smaller and follow a sizer-like division strategy where the added size is inversely proportional to the size at birth. We explore three potential causes for this deviation: degradation of the precursor protein and two models where the propensity for accumulation depends on the cell size: a nonlinear accumulation rate, and accumulation starting at a threshold size termed the commitment size. These models fit the mean trends but predict different distributions given the birth size. To quantify the precision of the models to explain the data, we used the Akaike information criterion and compared them to open datasets of slow-growing E. coli cells in different media. We found that none of the models alone can consistently explain the data. However, the degradation model better explains the division strategy when cells are larger, whereas size-related models (power-law and commitment size) account for smaller cells. Our methodology proposes a data-based method in which different mechanisms can be tested systematically.

Gestational diet manipulation can lead to inadequate fetal nutrient supply resulting in low birth weight, limited postnatal growth, and consequently, reduced reproductive performance in the progeny. However, effects of short-term maternal pre-conceptional dietary manipulation on postnatal growth and reproductive parameters of male offspring in large animals remains unexplored. To determine these consequences, female crossbred (Polypay x Dorset) sheep were allocated to three groups (n = 33/group) of dietary manipulation for 21 days prior to mating under the following conditions: (1) control at 100 % of maintenance energy requirements (40 Kcal of metabolizable energy/kg body weight [BW]), (2) undernutrition (UN) at 50 % of Control intake, and (3) overnutrition (ON) at 200 % of maintenance energy. Singleton ram lambs (UN:9; C:12; ON:6) were monitored from birth until 8 months of age, including birth weight, weekly weights, weight gain, body mass index (BMI), and circulating testosterone. After weaning, monthly scrotal circumference and subcutaneous fat depth were measured. Semen morphology and motility were evaluated at 7 and 8 months of age. Birth weight, weight gain, and BMI at birth and weaning were not significantly different among nutritional treatments. None of the pre-conceptional diets affected body weight change from weaning until 36 weeks of age, BMI, fat depth, or scrotal circumference across the experiment. A sustained rise in plasma testosterone concentrations was detected when ram lambs were, on average, 82 days old and 37 kg. Both testosterone concentrations and scrotal circumference were positively correlated to body weight regardless of treatment group. In addition, seminal parameters did not differ among treatments, but a transient increase in plasma testosterone at 18 weeks of age was observed in ON ram lambs compared to control rams. In conclusion, birth weight, growth indices, and seminal parameters in singleton rams are resilient features in the progeny upon maternal pre-conceptional dietary manipulation in sheep.

Ewe lambs that are heavier due to improved nutrition pre- and post-weaning achieve puberty at a younger age, are more fertile, and have a higher reproductive rate. Fatness is intimately linked to reproduction, and we hypothesised that higher body condition scores at breeding would have positive effects on the reproductive rate of ewe lambs over and above liveweight. We also expected that if only a proportion of ewe lambs were presented for breeding, then it would be more effective to select them based on both liveweight and body condition score. To test these hypotheses, we analysed data from over 17,000 records from Merino and non-Merino ewe lambs from 22 different flocks across Australia. Non-Merino ewe lambs were more fertile (69.4% vs. 48.7%) and achieved a higher reproductive rate than Merino ewe lambs (96.9% vs. 60.7%). There were significant curvilinear relationships between liveweight (p < 0.001) or body condition score (p < 0.001) prior to breeding and reproductive rate for both Merino and non-Merino ewe lambs. For both breeds, there was a significant (p < 0.001) quadratic effect of body condition score prior to breeding on reproductive rate, independent of the correlated changes in liveweight, and at the same liveweight, an extra 0.5 of a body condition score up to 3.3 improved reproductive rate by about 20%. Nevertheless, the results indicated that if only a proportion of ewe lambs were selected for breeding, then selection based on both liveweight and body condition scores may only improve the overall reproductive rate by 1 to 2% compared to selection based on liveweight alone. We conclude that liveweight is a more effective method than body condition score for selecting ewe lambs for breeding.

We tested whether utilising the male effect to stimulate ewes before the mating period can reduce the time to conception following the introduction of entire rams, and increase fertility, prolificacy, and reproductive rate (number of fetuses per 100 ewes exposed to fertile rams). A retrospective analysis was used to analyse records from 59,716 ewes collected over 34 years (1986-2020) from seven genotypes: Border Leicester, Composite (crossbred), Dorset, Merino, Dorset x Polypay, Rambouillet, White Suffolk. The dataset also included nulliparous young ewes (mated at age 8 months) and adult parous ewes. Vasectomized rams were used to stimulate 20,632 ewes before a mating period that lasted 2 or 3 estrous cycles, and the outcomes were compared with those from 39,084 ewes that had not been stimulated. Independently of genotype, utilising the male stimulus advanced the average conception date by 8 days for young ewes (P < 0.0001) and by 1 day for adult ewes (P < 0.0001). The male stimulus also increased the proportion of ewes that conceived in their first cycle by 33 % for young ewes and by 6 % for adult ewes (P < 0.0001). For the cycle of conception, there were significant (P < 0.0001) effects of two interactions: male stimulus x age at mating and male stimulus x live weight at mating. The male stimulus improved fertility in both adult ewes (99.8 % vs 89 %; P < 0.001) and young ewes (77.7 % vs 81.3 %; P < 0.001). The male stimulus increased the number of young ewes (41.9 % vs 11.1 %; P < 0.001) and adult ewes (16.6 % vs 2.7 %; P < 0.001) that conceived multiple fetuses in the first 17 days of the mating period. The reproductive rate was improved by the male stimulus in young ewes (129 % vs 135 %; P < 0.001) but not in adult ewes (120 % vs 122 %; P = 0.12). When all animals for all breeds were included in the analyses, there were improvements in fertility, prolificacy, and reproductive rate as age and live weight increased at mating (P < 0.0001). We conclude that, independently of genotype, utilising the male stimulus before the mating period reduces the time to conception and improves reproductive performance in both young and adult ewes.

The pygmy hippopotamus (Choeropsis liberiensis) is an endangered species endemic to the Upper Guinea Forest ecosystem in West Africa. We have limited information concerning the species' reproduction and well-being under managed care. We therefore developed non-invasive methods for characterizing gonadal androgen and adrenal hormone profiles in pygmy hippos using fecal samples collected from 12 males and 12 females housed in North American zoological institutions. We aimed to: 1) identify and validate enzyme immunoassays (EIAs) for measuring metabolites of corticosteroids and testosterone in feces; and 2) test whether gonadal activity is correlated with previous breeding history, season or type of housing. For glucocorticoids, several EIAs for measuring metabolites were investigated. A group-specific EIA exhibiting cross-reactivity with 11,17-dioxoandrostane (DOA) metabolites of cortisol most clearly reflected adrenocortical activity in response to pharmocological challenge with adrenocorticotropic hormone (ACTH) in both males and females. However, day-to-day concentrations of this metabolite in the feces of pygmy hippos that did not undergo ACTH challenge were near the detection limits of the assay, making this EIA impractical for assessing glucocorticoid activity in this species. Another group-specific EIA, exhibiting cross-reactivity with 5α-pregnane-3ß,11ß,21-triol-20-one, produced biologically relevant data and evidence of an appropriate response to pharmacological challenge with exogenous ACTH. The testosterone metabolite assay C196 (Arbor Assays, Ann Arbor, Michigan, USA) also produced biologically coherent data: adult males exhibited the highest mean androgen metabolite concentrations (477 ng/g), followed by adult females (259 ng/g) and juvenile males (160 ng/g). Proven breeding males had higher, but not significantly different, mean concentrations (472 ng/g) to unproven males (352 ng/g; P = 0.400). Similarly, adult males housed outdoors year-round in subtropical climates exhibited higher, but not statistically different mean concentrations (554 ng/g) to males in temperate climates that were housed indoors at least part of the year (412 ng/g; P = 0.208). There were, however, significant differences in mean concentrations among seasons for adult males, with higher values in spring (546 ng/g) and summer (542 ng/g) than in autumn (426 ng/g) and winter (388 ng/g, P = 0.003). In conclusion, we identified EIAs for the measurement of fecal metabolites of androgens and glucocorticoids that can be used for further studies to monitor gonadal activity in male pygmy hippos and adrenocortical activity in both sexes. We also identified a seasonal trend in male gonadal activity in this species under managed care in North America. Finally, our findings highlight an important consideration when using non-invasive methods for evaluating fecal cortisol metabolites: ACTH used for pharmacological validation of an EIA does not necessarily equate to biological relevance.

Recently, there has been an increasing need for tools to simulate cell size regulation due to important applications in cell proliferation and gene expression. However, implementing the simulation usually presents some difficulties, as the division has a cycle-dependent occurrence rate. In this article, we gather a recent theoretical framework inPyEcoLib, a python-based library to simulate the stochastic dynamics of the size of bacterial cells. This library can simulate cell size trajectories with an arbitrarily small sampling period. In addition, this simulator can include stochastic variables, such as the cell size at the beginning of the experiment, the cycle duration timing, the growth rate, and the splitting position. Furthermore, from a population perspective, the user can choose between tracking a single lineage or all cells in a colony. They can also simulate the most common division strategies (adder, timer, and sizer) using the division rate formalism and numerical methods. As an example of PyecoLib applications, we explain how to couple size dynamics with gene expression predicting, from simulations, how the noise in protein levels increases by increasing the noise in division timing, the noise in growth rate and the noise in cell splitting position. The simplicity of this library and its transparency about the underlying theoretical framework yield the inclusion of cell size stochasticity in complex models of gene expression.

Single cells actively coordinate growth and division to regulate their size, yet how this size homeostasis at the single-cell level propagates over multiple generations to impact clonal expansion remains fundamentally unexplored. Classical timer models for cell proliferation (where the duration of the cell cycle is an independent variable) predict that the stochastic variation in colony size will increase monotonically over time. In stark contrast, implementing size control according to adder strategy (where on average a fixed size added from cell birth to division) leads to colony size variations that eventually decay to zero. While these results assume a fixed size of the colony-initiating progenitor cell, further analysis reveals that the magnitude of the intercolony variation in population number is sensitive to heterogeneity in the initial cell size. We validate these predictions by tracking the growth of isogenic microcolonies of Corynebacterium glutamicum in microfluidic chambers. Approximating their cell shape to a capsule, we observe that the degree of random variability in cell size is different depending on whether the cell size is quantified as per length, surface area, or volume, but size control remains an adder regardless of these size metrics. A comparison of the observed variability in the colony population with the predictions suggests that proliferation matches better with a cell division based on the cell surface. In summary, our integrated mathematical-experimental approach bridges the paradigms of single-cell size regulation and clonal expansion at the population levels. This innovative approach provides elucidation of the mechanisms of size homeostasis from the stochastic dynamics of colony size for rod-shaped microbes.

The potential effect of intravenous administration of glutamate on the ovarian activity and the LH secretion pattern, considering the anestrous yearling goat as an animal model, were assessed. In late April, yearling goats (n = 20) were randomly assigned to either (1) Glutamate supplemented (GLUT; n = 10, Live Weight (LW) = 29.6 ± 1.02 kg, Body Condition (BCS) = 3.4 ± 0.2 units; i.v. supplemented with 7 mg GLUT kg−1 LW) or (2) Non-supplemented (CONT; n = 10; LW = 29.2 ± 1.07 kg, BCS = 3.5 ± 0.2 units; i.v. saline). The oats were estrus-synchronized; blood sampling (6 h × 15 min) was carried out for LH quantification. Response variables included pulsatility (PULSE), time to first pulse (TTFP), amplitude (AMPL), nadir (NAD), and area under the curve (AUC) of LH. Ovaries were ultra-sonographically scanned to assess ovulation rate (OR), number of antral follicles (AF), and total ovarian activity (TOA = OR + AF). LH-PULSE was quantified with the Munro algorithm; significant treatment x time interactions were evaluated across time. The variables LW and BCS did not differ (p > 0.05) between the experimental groups. Nevertheless, OR (1.77 vs. 0.87 ± 0.20 units), TOA (4.11 vs. 1.87 ± 0.47 units) and LH-PULSE (5.0 vs. 2.2 pulses 6 h-1) favored (p < 0.05) to the GLUT group. Our results reveal that targeted glutamate supplementation, the main central nervous system neurotransmitter, arose as an interesting strategy to enhance the hypothalamic−hypophyseal−ovarian response considering the anestrous-yearling goat as an animal model, with thought-provoking while promising translational applications.

The last third of gestation is a period of high energy and protein demand for the dam to support fetal growth and the following onset of lactation. Methionine is an essential amino acid that contributes to protein formation, fetal development, and milk synthesis; thus, is likely to have positive effects on the weight and size of the newborn and, afterward, milk yield and milk composition, which may improve growth patterns of the progeny. To test these hypotheses, we used 60 pregnant multiparous Alpine goats with similar live weights and gestational ages (~Day 100 of pregnancy; Mean ± SD; 1410 ± 14 days old and 50.4 ± 6.6 kg) and were separated into two groups: control and supplemented with the delivery. Treatments were T-MET (n = 30; received 1% herbal methionine Optimethione® dry matter based on from Day 100 of the pregnancy to delivery) or T-CTL (n = 30; served as the control and did not receive methionine). The methionine powder provided individual supplementation and was adjusted every week as the live weight and dry matter intake changed. At birth, the weight, body mass index (BMI), birth type, and sex of the kids were determined. Subsequently, the progeny was weighed weekly up to weaning. Two weeks after parturition, the milk composition was recorded weekly, and the milk yield was recorded monthly. The maternal live weight at the start (Mean ± SEM; T-CTL: 50.5 ± 1.1 vs. T-MET: 50.3 ± 1.3 kg) and end (T-CTL: 54.2 ± 1.3 vs. T-MET: 52.8 ± 1.4 kg) of the experiment did not differ statistically among treatments (p > 0.05); however, daily live weight changes tended to differ between groups (T-CTL: 73 ± 10 vs. T-MET: 51 ± 7 g day−1; p = 0.06). The birth weight (T-CTL: 3.1 ± 0.1 vs. T-MET: 3.5 ± 0.1 kg; p < 0.001), daily live weight change (T-CTL: 121 ± 6 vs. T-MET: 141 ± 6 g day−1; p < 0.01), and weaning weight (T-CTL: 8.3 ± 0.2 vs. T-MET: 9.3 ± 0.3 kg; p < 0.01) differed between treatments. The BMI at birth (T-CTL: 0.28 ± 0.01 vs. T-MET: 0.3 ± 0.01 units kg m−2; p < 0.01) and at weaning (T-CTL: 0.85 ± 0.1 kg vs. T-MET: 1.00 ± 0.06 units kg m−2; p < 0.05) differed between treatments. Milk components (protein, fat, lactose, and solids non-fat) and milk yield were similar between treatments (p > 0.05). It is concluded that the inclusion of methionine in the maternal goat diet during the last third of gestation increases the birth and growth variables of the progeny but without significant influence on the milk yield and composition.

The traditional goat production systems on arid and semi-arid rangelands of northern Mexico are characterized by minimum external inputs; therefore, little is known about goat response to technological improvements. This observational study aimed to ascertain the effect of using improved goat production technologies and breed on milk yield and reproductive performance of goats on rangeland. The study was conducted from January 2017 to June 2018. Thirty-seven goat herds comprising 6393 animals were used. Goats supplemented with 250 g of concentrate (S-goats; 14% crude protein) 30 days before breeding produced significantly more daily milk yield (218 ± 61 vs. 200 ± 60 g) than the unsupplemented (UNS-goats) goats. Milk production per lactation was 12 percentage points greater in goats with access to plain salt throughout the year than those without salt access. The absence of deworming significantly depressed daily milk yield (189 ± 55 vs. 221 ± 61 for non-treated and treated goats). In 5 months, the lowest total milk yield was for Boer goats (26.9 ± 8.3 kg) and the highest for Nubian goats (36.1 ± 9.2 kg). The kidding rate was significantly higher in the S-goats than in the UNS-goats group (66.9 vs. 62.8%). Salt-supplemented goats had a significantly greater kidding rate than goats not receiving salt (68.5 vs. 61.2%). The lowest kidding rate was for Saanen goats and the highest for Nubian goats (66.2%). These findings indicate that milk yield and reproductive performance were primarily influenced by supplementing grazing goats with concentrates and salt in this semi-arid rangeland.

Objetivos: estudiar la evolución de la cohorte a la largo de 11 años (2007-2018). Mientras que, los objetivos específicos han sido: analizar las diferencias entre casos y controles; conocer la tasa de mortalidad de la cohorte y su relación con el síndrome de piernas inquietas; estimar el impacto del síndrome de piernas inquietas en la calidad de vida relacionada con la salud y el sueño. Métodos: el estudio ha seguido una metodología observacional, retrospectiva y longitudinal de una cohorte de casos y controles, formada por 232 pacientes. En ambas evaluaciones se recogieron datos sociodemográficos, clínicos (se administraron las siguientes escalas: EuroQol, Índice de Comorbilidad de Charlson, escala de ansiedad y depresión de Goldberg, escala de somnolencia de Epworth y el cuestionario de Calidad de Vida del síndrome de piernas inquietas) y analíticos, y en la segunda evaluación también se recolectaron datos de actigrafía (actividad física y sueño). Methods: the study has followed an observational, retrospective and longitudinal methodology of a cohort of cases and controls, made up of 232 patients. Sociodemographic and clinical data were collected in both evaluations (the following scales were administered: EuroQol, Charlson's Comorbidity Index, Goldberg's anxiety and depression scale, Epworth's sleepiness scale and the Quality-of-Life questionnaire for restless legs syndrome) and analytical, and in the second evaluation actigraphy data (physical activity and sleep) were also collected. Tillandsia recurvata is an epiphyte that grows on the canopy of many trees in tropical and subtropical areas of America. The objective of this study was to evaluate the effect of partial or complete substitution of sorghum stubble with T. recurvata on liveweight change, metabolic profile, and complete blood count of goats fed increasing levels (0, 30, and 60%, dry matter basis) of T. recurvata. Thirty non-pregnant three-year-old, non-lactating, healthy mixed-breed goats, ten animals per treatment (T0, T30, and T60), were adapted to diets and facilities for 14 days (d-14). Blood samples were collected at d-15, 28, and 56. At the last phase of the trial (from days 67 on), control goats tended to gain more (p = 0.09) weight than their counterparts consuming T. recurvata. Plasma protein, glucose, triglycerides, calcium, and phosphorus concentrations did not differ among dietary treatments (p > 0.05). Dietary treatment influenced red blood cells (higher for T60; p < 0.01), white blood cells (higher for T30; p < 0.05), mean corpuscular volume (higher for T0; p < 0.001), and mean corpuscular hemoglobin concentration (higher for T0; p < 0.01), although not the rest of the blood variables (p > 0.05). The hematocrit percentage tended to be higher (p = 0.06) in T30 than T0 and T60. It was concluded that replacing sorghum stubble with T. recurvata did not modify the metabolic status and maintained live weight of goats. Nevertheless, the use of T. recurvata as feed for goats would improve the nutrition of these animals in the dry season compared to the current diet obtained from an arid rangeland, reducing production costs, and would alleviate the damage caused by this aggressive epiphyte to host trees.

Bacterial division is an inherently stochastic process with effects on fluctuations of protein concentration and phenotype variability. Current modeling tools for the stochastic short-term cell-size dynamics are scarce and mainly phenomenological. Here we present a general theoretical approach based on the Chapman-Kolmogorov equation incorporating continuous growth and division events as jump processes. This approach allows us to include different division strategies, noisy growth, and noisy cell splitting. Considering bacteria synchronized from their last division, we predict oscillations in both the central moments of the size distribution and its autocorrelation function. These oscillations, barely discussed in past studies, can arise as a consequence of the discrete time displacement invariance of the system with a period of one doubling time, and they do not disappear when including stochasticity on either division times or size heterogeneity on the starting population but only after inclusion of noise in either growth rate or septum position. This result illustrates the usefulness of having a solid mathematical description that explicitly incorporates the inherent stochasticity in various biological processes, both to understand the process in detail and to evaluate the effect of various sources of variability when creating simplified descriptions.

In this paper, we tested the hypothesis that ewe lambs that are heavier and older at breeding will wean more offspring, due to increased reproductive rate and offspring survival and lower maternal mortality. To test this hypothesis, we analyzed data from more than 11,500 maternal composite ewe lambs collected over eight years. The ewe lambs had full pedigree records including birth type, age and liveweight at breeding plus records of the birthweight and survival of their offspring and the dam. The average liveweight and age at breeding was 40.2 kg and 228 days. The reproductive rate and weaning rate responses to liveweight at breeding were curvilinear (p < 0.001), and if ewe lambs achieved 45 kg by the start of breeding, their reproductive rate and weaning rate were within 5% of their maximum. By contrast, the effects of age at breeding on weaning rate was linear and increased by 0.4% per day, despite a quadratic (p < 0.01) effect of age at breeding on reproductive rate which increased only marginally when ewe lambs were older than 8 months at breeding. Increasing liveweight (p < 0.05) or age (p < 0.001) at breeding increased survival of their offspring, however an extra 10 kg of liveweight or 30 days of age at breeding increased offspring survival by less than 5%. Both liveweight (p < 0.001) and age (p < 0.01) at breeding also influenced survival of the ewe lamb dam but survival rates exceeded 95% across the range in liveweights from 30 to 55 kg and ages from 6 to 9 months. This understanding of the trade-off between age and liveweight at breeding will assist farmers to optimize the management of their ewe lambs, given the earlier they can be bred successfully the easier they can be integrated with the breeding of the adult ewe flock the following year.

Ample nitrogen (N) is required for potato production, but its use efficiency is low. N supply strongly interacts with maturity type of the cultivar grown. We assessed whether variation among 189 cultivars grown with 75 or 185 kg available N/ha in 2 years would allow detecting quantitative trait loci (QTLs) for relevant traits. Using phenotypic data, we estimated various traits and carried out a genome-wide association study (GWAS) with kinship correction. Twenty-four traits and 10,747 markers based on single-nucleotide polymorphisms from a 20K Infinium array for 169 cultivars were combined in the analysis. N level affected most traits and their interrelations and influenced the detection of marker-trait associations; some were N-dependent, others were detected at both N levels. Ninety percent of the latter accumulated on a hotspot on Chromosome 5. Chromosomes 2 and 4 also contained regions with multiple associations. After correcting for maturity, the number of QTLs detected was much lower, especially of those common to both N levels; however, interestingly, the region on Chromosome 2 accumulated several QTLs. There is scope for marker-assisted selection for maturity, with the main purpose of improving characteristics within a narrow range of maturity types, in order to break the strong links between maturity type and traits like N use efficiency.

We tested whether the milk yield capacity of mixed-breed goats on a Chihuahuan desert rangeland in northern Mexico during the dry season affects milk composition, body weight gain, and weaning weight of their progeny. Milk yield and composition, and progeny postnatal growth performance, were recorded weekly. One week after kidding, mixed-breed goats (a mixture of Criollo × dairy breeds; n = 40) were allotted into medium (MP) or low (LP) milk yielding groups (20 goats per group). Mean 105-d total milk yield for MP and LP goats was 45.2 ± 12.5 and 20.7 ± 5.2 L, respectively. Milk lactose (4.3 vs. 4.2%) and solids-non-fat (SNF; 8.2 vs. 8.0) differed (p < 0.05) between MP and LP goats; milk protein content tended to differ (p = 0.08) between MP and LP goats with no difference for milk fat content (p > 0.05). Maternal body weight was positively associated with milk yield, milk lactose, and SNF content (p < 0.05 to p < 0.001). Goats giving birth to males produce more milk than goats giving birth to females, but milk fat percentage was higher in goats bearing females (p < 0.001). Milk yield and composition throughout lactation did not influence body weight gain (47.8 vs. 48.7 g/day for kids from MP and LP goats) and weaning weight (6.7 vs. 6.7 kg from MP and LP goats) of the offspring (p > 0.05). Birth weight and weaning weight of the progeny were positively related to maternal body weight (p ≤ 0.05). The postnatal growth of the kids was reduced, extending the time to reach market weight. Nevertheless, non-supplemented mixed-breed goats reared on semi-arid rangeland of northern Mexico have the potential for moderate milk production. Therefore, due to the limited nutrients ingested by grazing goats during the dry season, a nutritional supplement is necessary to keep up milk production and adequate growth of kids.

We tested whether periconceptional nutrition with cladodes from the cactus, Opuntia ficus-indica, with or without protein-enrichment, improved the metabolomic profile and reproductive outcomes of adult female sheep. Sixty Rambouillet ewes of similar body weight were randomly allocated among three nutritional treatments that were fed during the breeding period (34 days): Control (Control; n = 20), Opuntia (Opuntia; n = 20) and protein-enriched Opuntia (E-Opuntia; n = 20). There were no effects of treatment on body weight but assessment of urine samples indicated that, for 76 metabolites, the Control and Opuntia groups differed completely (P < 0.05), whereas there was overlap between the Control and E-Opuntia groups. It appears that, in Opuntia-fed and Control-fed sheep, different functional groups are activated leading to changes in the metabolism of glucose, tyrosine, methane, and glycerolipids. Fertility and reproductive rate tended to be higher in the Opuntia (70% and 95%) and E-Opuntia (90% and 110%) groups than in the Control (55% and 65%), and an orthogonal contrast revealed the difference between Control and Opuntia to be significant for both reproductive variables (P < 0.05). We conclude that nutritional supplementation with Opuntia cladodes, with or without protein enrichment, increased fertility rate and reproductive rate of female sheep, without any accompanying increases in body weight. Our observations suggest that the reproductive responses to Opuntia do not simply reflect a response to good nutrition, but might be caused by specific metabolites/metabolomic pathways, perhaps by an activation of the metabolism of glucose, methane, tyrosine and glycerolipids. There are few reports relating these metabolomic compounds with the metabolism of the sheep, let alone with reproductive efficiency. The novelty of these discoveries suggests that we need further research into the mechanisms through which nutrition affects the reproductive system.

We tested whether maternal energy supplementation during the last third of gestation improves birth weight, neonatal wellbeing, and mother-young bonding. Thirty-six pregnant French Alpine goats were randomly allocated among three nutritional treatments for the last third of pregnancy: (i) Control, fed alfalfa (T-0; n = 12); (ii) alfalfa + 150 g/head daily energy concentrate (T-150; n = 12); (iii) alfalfa + 300 g/head daily energy concentrate (T-300; n = 12). At birth, we collected progeny data on birth weight, birth type, sex, rectal temperature, heart rate, respiratory rate, time to standing, time to udder connection, and time to first feeding. For the dams, we collected data on the duration of labor, time to clean the progeny, and time to allow first suckling. At birth, body weight, rectal temperature, heart rate, and the respiratory rate did not differ among treatments (p > 0.05). In the dams, labor duration was not affected by the treatments (p > 0.05). The T-150 dams were faster to clean the newborn and allow first suckling (p < 0.05). The T-150 progeny were faster to stand and the T-300 progeny were faster to connect to the udder (p < 0.05). We conclude that energy supplementation of the dam during the last third of gestation does not affect the birth weight of the progeny, but enhances the mother-young bonding.

BACKGROUND: A growing body of evidence relates restless legs syndrome (RLS) to an increased risk of mortality attributable to both cerebrovascular and cardiovascular events. The aim was to investigate survival in patients with RLS. METHODS: This was an observational, retrospective longitudinal study of a cohort of patients followed up for 11 years. RLS was diagnosed by a physician using the International RLS Study Group criteria. Mortality was analyzed using age-standardized mortality ratios (SMR: observed/expected deaths) and Cox regression analysis. RESULTS: Vital status was studied in a cohort of 232 patients: 181 women (78%), 96 with RLS (41.4%) with a mean age at baseline of 49.8 ± 15.0 years and a mean RLS duration of 14.1 ± 1.9 years, and 136 non-RLS (58.6%) with a mean age of 51.3 ± 14.9 years. This RLS cohort was followed up for a period of 10.4 ± 2.0 years. As of September 2019, 17 (7.3%) patients died (6 with RLS, 6.3%), and the most frequent cause was oncological (66.7%). A total of 944 person-years of observations were available for survival analysis. RLS was not associated with increased mortality in adjusted Cox regression analysis (HR = 1.12, 95% CI: 0.40-3.15), and survival was similar to that expected for the general population (SMR = 0.61, 95% CI: 0.27-1.36). CONCLUSIONS: RLS seems not to be associated with increased mortality compared to the general population. Still, studies with prospective data collection with large samples are needed to study the long-term mortality risk factors in RLS cohorts.
