RESUMEN

Accurate estimations of the biomass and value of livestock in Indonesia are of great use in supporting investment decisions by the public and private sector and as a basis for estimating the losses due to animal disease. Biomass and the partial direct use value for key livestock species (cattle, buffalo, sheep, goats, pigs, chickens) for all provinces of Indonesia were derived from secondary data using a novel spreadsheet-based model. Using beef cattle as an example, we also explored the use of a herd dynamics model to validate base data on populations and productivity used to generate biomass values, and these were found to be generally robust. Total partial direct use value of livestock is estimated to be almost USD54 billion in 2021, comprising almost USD33 billion of population value and almost USD21 billion of production value. Beef cattle account for 44% of total value and chicken (broiler, layer and native chickens) account for a further 36% of the total. Breaking the data down by province reveals the regional importance of some livestock types that are of relatively minor importance nationally (pigs in East Nusa Tenggara and sheep in West Java). It also reveals the importance of livestock in the poorest provinces of Indonesia, where livestock acts as a store of wealth and serves socio-cultural purposes.

Background and Aim: African swine fever (ASF) is a disease that emerged in Indonesia in 2019 in the North Sumatra province and spread rapidly to other areas, such as South Sumatra and Lampung, in 2020. This study aimed to identify the phylogenetics of the ASF virus (ASFV) in the provinces of South Sumatra and Lampung. Materials and Methods: Nine ASFV isolates collected from the Disease Investigation Center in Lampung were used in this study. The isolates were from ASF cases in South Sumatra and Lampung in 2020-2022. The isolates were sequenced and compared with other ASFV isolates to establish the virus genotype. Sequencing was performed using the complete E183L gene target encoding the p54 protein. Results: This study showed that ASFV from South Sumatera and Lampung Province belongs to genotype II. Conclusion: Based on the analysis of the E183L gene, all nine ASFV isolates that originated from South Sumatra and Lampung were identical to other genotype II ASFV isolates from Georgia, China, Vietnam, and Timor Leste.

Background and Aim: Ruminant slaughterhouse is one of the food-producing units to meet the protein demand of the people in Central Java. This study aimed to evaluate the implementation of sanitation and hygiene in ruminant slaughterhouses in Central Java based on their veterinary control number(NKV) certification and the microbiological quality of the meat produced. Materials and Methods: This study was conducted from September 2021 to December 2021. Thirty-three priority slaughterhouses, representing 33 districts/cities in Central Java Province, were assessed for their hygiene and sanitation practices according to the NKV criteria mandated by The Minister of Agriculture Regulation No. 11/2020 on NKV Certification for Animal Production Unit. Sixty-six meat samples from these slaughterhouses were obtained for microbiological analysis. The total plate count (TPC), counts of Escherichia coli and Staphylococcus aureus, and the presence of Salmonella spp. were determined. The microbiological tests followed the standard national testing procedure according to the Indonesian National Standard 2897:2008 on Method of Analysis for Microbiological Contaminants in Meat, Eggs, Milk, and its derived products. Results: The sanitation hygiene assessment of the 33 slaughterhouses showed that seven (21.2%) met the NKV criteria level 3, while the others did not. The average TPC of the meat samples was 1.57 × 105 CFU/g (4.93 log10), the S. aureus count was 7.6 CFU/g, and the E. coli count was 9.2 most probable number/g. Only one sample (1.50%) tested positive for Salmonella spp. A comprehensive assessment comparing the NKV criteria with the level of meat contamination showed that the ruminant slaughterhouses that satisfied the NKV criteria had more meat samples (85.71%), on average, that complied with the Indonesian National Standard for microbial contamination compared with those that did not satisfy the NKV criteria (69.23%). The odds ratio was 2.67. Conclusion: Most of the priority ruminant slaughterhouses in Central Java did not meet the NKV standards. The research only looks at the level of hygiene sanitation according to NKV standards in slaughterhouses, the level of contamination produced does not reflect the level of the consumer; therefore, the level of contamination should continue to be investigated at the post-production stage.

Background and Aim: Subclinical mastitis (SCM) in Etawah-grade (PE) goats in Yogyakarta, Indonesia, is commonly due to Staphylococcus aureus. At present, S. aureus from SCM in PE goats in Yogyakarta has not been characterized. Therefore, this study aimed to phenotypically characterize S. aureus, which has been isolated from SCM of PE goats. Materials and Methods: A total of 314 lactating PE goats were collected from 60 PE goat farms (e.g., Sleman, Bantul, and Kulonprogo) located in parts of Yogyakarta with an average age of 3-4 years old, three of which showed SCM based on the California mastitis test (CMT). Subclinical mastitis is confirmed in PE goats if CMT shows ++ or +++. Furthermore, S. aureus was detected by biochemical assays. Staphylococcus aureus could determine hemolysin (Hae), coagulase (Coa), clumping factor (Cf), and antibiotic susceptibility. Hemolytic bacteria were detected by culturing on blood agar plate, and Cf was detected by slide agglutination. The production of Coa was detected by tube coagulation. Staphylococcus aureus susceptibility was determined by antimicrobial agar diffusion using a paper disc. Results: Phenotypically characterized S. aureus from PE goats with SCM in Yogyakarta, Indonesia, Coa-, Cf-, and Hae- were found to be resistant to erythromycin (ERYTHRO), ampicillin (AMP), penicillin (PEN-G), and sulfamethoxazole (SULFA). Conclusion: The phenotypic characteristic of S. aureus, which was obtained from SCM in PE goats in Yogyakarta, consists of Coa, and Cf-. S. aureus cannot perform hemolysis of red blood cells. This phenotypic characteristic can prevent and control SCM in PE goats. Several antibiotics such as ERYTHRO, AMP, PEN-G, and SULFA were no longer effective for treating SCM in PE goats because S. aureus has developed its resistance to these antibiotics.

Mesenchymal stem cells-conditioned medium (MSC-CM) is the extraction from stem cell medium containing biological substances, including growth factors and cytokines. These substances play roles in the various functions of body regulatory, including bone formation. However, the effect of MSC-CM derived from human umbilical cord injection in femur fracture healing of rats has not been reported previously. This study aims to see the effect of MSC-CM derived from human umbilical cord injection on the callus formation of bone fracture healing in Wistar rats (Rattus norvegicus). A femur fracture in 54 Wistar rats was made by surgery according to the procedure under sterile conditions. After the surgery, rats were divided into 2 groups of 27, respectively. Injection in the control (0.1 mL/kg body weight NaCl) and MSC-CM group (0.1 mL/kg body weight MSC-CM) was performed on weeks 0, 1, 2, 3, 4, 5, 6, 7, and 8 after surgery. Radiographic images and the femur bone samples were taken and collected on days 1, 7, 14, 21, 28, 35, and 60 after surgery. Bone samples were then fixed in Bouin solution. Histologic preparations were done by the paraffin method, by cutting the tissue blocks into 5 µm thickness and then staining with Mallory aniline blue staining. The results were analyzed descriptively and quantitatively. The result showed that the soft callus formation occurred rapidly and got wider in the MSC-CM group than that of the control group. The administration of MSC-CM injection postfracture surgery to femur fracture cases in rats was capable to accelerate the callus formation.

BACKGROUND AND AIM: Feline lower urinary tract disease (FLUTD) is one of the common cat diseases. The aim of this study was to detect feline idiopathic cystitis (FIC) as a cause of FLUTD in Sleman Regency, which is a problem in the population. MATERIALS AND METHODS: Seventy-three cats with FLUTD symptoms were used from seven veterinary practices in Sleman Regency. The logging of each cat's medical history, clinical examination, urinalysis, routine blood screening, and ultrasonography was conducted to diagnose the cause of FLUTD. RESULTS: The percentages of diseases causing FLUTD included FIC 21.9%, urolithiasis 57.5%, urinary tract infection (UTI) 16.4%, neoplasia 1.4%, trauma 1.4%, and nervous disorders 1.4%. CONCLUSION: FIC, one of the causes of FLUTD, is found in cats and has become a problem among the cat population in Sleman Regency, Yogyakarta, Indonesia. Various handling and preventive efforts should be undertaken against the disease.

BACKGROUND AND AIM: In Indonesia, highly pathogenic avian influenza (HPAI) H5N1 outbreaks in poultry are still reported. The disease causes a decrease in egg production and an increase in mortality; this has an impact on the economic losses of farmers. Several studies have considered that ducks play a role in the HPAI endemicity in the country; however, little is known about whether or not the type of duck farming is associated with HPAI H5 virus infection, particularly within clade 2.3.2.1c, which has been predominantly found in poultry since 2014. A cross-sectional study was conducted to determine the HPAI seroprevalence for H5 subtype clade 2.3.2.1c in laying ducks that are kept intensively and nomadically and to determine the associated risk factors. MATERIALS AND METHODS: Forty-nine duck farmers were randomly selected from ten sub-districts in Purbalingga District, Central Java, Indonesia; a cross-sectional study was implemented to collect field data. Based on an expected HPAI prevalence level of 10%, estimated accuracy of ± 5%, and 95% confidence interval (CI), the total sample size was calculated at 36 individuals. Samples must be multiplied by 7 to reduce bias; thus, 252 ducks were taken as samples in this study. Considering that the maintenance and duck handling were uniform and farmers complained that the effect of activity to take duck samples would reduce egg production, this study only took samples from 245 ducks (oropharyngeal swabs and serum). Those samples were taken from five birds on each farm. Hemagglutination inhibition tests examined the serum samples for HPAI H5 Clade 2.3.2.1c, and pool swab samples (five swabs in one viral media transport) were examined by real-time reverse transcription-polymerase chain reaction (qRT-PCR) test for influenza Type A and H5 subtype virus. Information regarding farm management was obtained using a questionnaire; face-to-face interviews were conducted with the duck farmers using native Javanese language. RESULTS: Serum and swabs from 245 ducks were collected in total. For individual birds, 54.69% (134/245) of serum samples were H5 seropositive. Seroprevalence among nomadic ducks was 59.28% (95% CI: 0.48-0.61), which was higher than among intensively farmed ducks (48.57%, 95% CI: 0.38-0.58). Farm-level seroprevalence was 50% (95% CI: 0.30-0.69) for nomadic ducks but only 28.57% (95% CI: 0.11-0.51) for intensively farmed ducks. The farm-level virus prevalence (proportion of flocks with at least one bird positive for influenza Type A) was 17.85% (95% CI: 0.07-0.35) for nomadic ducks and 4.76% (1/21) for intensively farmed ducks (95% CI: 0.008-0.23). All influenza Type A positive samples were negative for the H5 subtype, indicating that another HA subtype AI viruses might have been circulating in ducks in the study area. A relationship between duck farms that were H5 seropositive and their maintenance system was present; however, this relationship was not significant, the nomadic duck system detected 2 times higher H5-seropositive ducks than the intensive farming system (OR: 2.16, 95% CI: 0.33-14.31). CONCLUSION: This study found that the seroprevalence of HPAI in the duck population level in Purbalingga was 54.69% and demonstrated that the nomadic duck farming system was more likely to acquire HPAI H5 infection than the intensive farming duck system. Other risk factors should be further investigated as the diversity of the farming system is partially related to HPAI H5 infection.

Cattle are known as the main reservoir of zoonotic agents verocytotoxin-producing Escherichia coli. These bacteria are usually isolated from calves with diarrhea and/or mucus and blood. Tolerance of these agents to the environmental conditions will strengthen of their transmission among livestock. A total of 238 cattle fecal samples from four sub-districts in Badung, Bali were used in this study. Epidemiological data observed include cattle age, sex, cattle rearing system, the source of drinking water, weather, altitude, and type of cage floor, the cleanliness of cage floor, the slope of cage floor, and the level of cattle cleanliness. The study was initiated by culturing of samples onto eosin methylene blue agar, then Gram stained, and tested for indole, methyl-red, voges proskauer, and citrate, Potential E.coli isolates were then cultured onto sorbitol MacConkey agar, and further tested using O157 latex agglutination test and H7 antisera. Molecular identification was performed by analysis of the 16S rRNA gene, and epidemiological data was analyzed using STATA 12.0 software. The results showed, the prevalence of E. coli O157:H7 in cattle at Badung regency was 6.30% (15/238) covering four sub districts i.e. Petang, Abiansemal, Mengwi, and Kuta which their prevalence was 8.62%(5/58), 10%(6/60), 3.33%(2/60), and 3.33(2/60)%, respectively. The analysis of 16S rRNA gene confirmed of isolates as an E. coli O157:H7 strain with 99% similarities. Furthermore, the risk factors analysis showed that the slope of the cage floor has a highly significant effect (P<0.05) to the distribution of infection. Consequently, implementing this factor must be concerned in order to decrease of infection.

AIM: This research was conducted to clarify the capability of human umbilical mesenchymal stem cells conditioned medium (HU-MSCM) to promote regenerations of primary wound healing on the incision skin injury. MATERIALS AND METHODS: In this study, two approaches in vitro and in vivo already done. On in vitro analysis, tube formation was performed using HU vein endothelial cells in the presence of HU-MSCM, in some experiments cells line was incubated prior the presence of lipopolysaccharide and HU-MSCM then apoptosis assay was performed. Furthermore, in vivo experiments 12 female rats (Rattus norvegicus) were used after rats anesthetized, 7 mm wound was made by incision on the left side of the body. The wound was treated with HU-MSCM containing cream, povidone iodine was run as a control. Wound healing regenerations on the skin samples were visualized by hematoxylin-eosin staining. RESULTS: In vitro models elucidate HU-MSCM may decreasing inflammation at the beginning of wound healing, promote cell migration and angiogenesis. In addition in vivo models show that the incision length on the skin is decreasing and more smaller, HE staining describe decreasing of inflammation phase, increasing of angiogenesis, accelerate fibroplasia, and maturation phase. CONCLUSIONS: Taken together our observation indicates that HU-MSCM could promote the acceleration of skin tissue regenerations in primary wound healing process.

AIM: Various studies have shown that secreted factors alone in culture medium without stem cell are capable of repairing tissues by itself in various conditions involving damaged tissue/organ. Therefore, this study was aimed to investigate the role of human umbilical cord mesenchymal stem cell-derived conditioned medium (CM) on the recovery of pancreatic ß-cells in Wistar rats (Rattus norvegicus) with type 1 diabetes mellitus. MATERIALS AND METHODS: The 0.05 ml CM induction was applied to the diabetic group of rats in weeks 1, 2, 3, and 4. 1 week after each CM induction, insulin concentration was analyzed using ELISA. The pancreas was divided into 3 regions, processed by paraffin method, stained with hematoxylin-eosin, and immunohistochemical method for insulin. RESULTS: This study indicated the decrease in the total number of islets and insulin concentration after the injection of single dose of alloxan. The exocrine acini were also damaged. Microscopic observation detected the presence of small islets in the diabetic group 1 week after the first 0.05 ml CM induction. The number and size of the islets increased in line with the CM doses and time of inductions. Immunohistochemically, the presence of low intensity of insulin-positive cells could be recognized at the splenic and duodenal regions of the pancreas, but not gastric region, 1 week after the first and second 0.05 ml CM induction. The intensity of staining and the number of insulin-positive cells increased dramatically in 1 week after the third and fourth 0.05 ml of CM induction in all regions of the pancreas. The data of insulin blood concentration showed clear differences between the second and the fourth induction of 0.05 ml CM induction. CONCLUSIONS: This study showed very strong evidence on the role of human umbilical cord mesenchymal stem cell-derived CM in recovering the pancreatic ß-cells damage in Wistar rats (R. norvegicus) with type 1 diabetes mellitus, structurally and functionally.
