RESUMEN

The traditional method for sampling for lead on surfaces uses inductively coupled plasma atomic emission spectroscopy (ICP-AES) to analyze the concentration of lead and other metals on surfaces. This type of analysis is time consuming and costly. Field portable X-ray fluorescence (FP XRF) is another analysis method that is not as accurate as traditional laboratory methods but is more cost efficient and has a turnaround time of less than an hour. The primary goal of this study is to find the best method to increase the level of agreement between the ICP-AES concentrations and the FP XRF concentrations when analyzing lead concentrations on surface wipes. Inverse regression and ratio of the means correction factors were analyzed to try to improve the prediction of ICP-AES concentrations using FP XRF results. Fifty-seven dust wipe samples were analyzed using a split-half design. Half of the samples were used to create the correction factor and the other half were used to test the level of agreement. Linear regression and Bland -Altman plots were used to determine the correction factor that provided the highest level of agreement. A ratio of the means correction factor was determined to be the most appropriate.

OBJECTIVE: Refractory idiopathic generalised epilepsy (IGE; also known as genetic generalised epilepsy) is a clinical challenge due to limited available therapeutic options. While vagus nerve stimulation (VNS) is approved as an adjunctive treatment for drug-resistant focal epilepsy, there is limited evidence supporting its efficacy for refractory IGE. METHODS: We conducted a single-centre retrospective analysis of adult IGE patients treated with VNS between January 2003 and January 2022. We analysed the efficacy, safety, tolerability, stimulation parameters and potential clinical features of VNS response in this IGE cohort. RESULTS: Twenty-three IGE patients were implanted with VNS between January 2003 and January 2022. Twenty-two patients (95.65%) were female. The median baseline seizure frequency was 30 per month (interquartile range [IQR]= 140), including generalised tonic-clonic seizures (GTCS), absences, myoclonus, and eyelid myoclonia with/without absences. The median number of baseline anti-seizure medications (ASM) was three (IQR= 2). Patients had previously failed a median of six ASM (IQR= 5). At the end of the study period, VNS therapy remained active in 17 patients (73.9%). amongst patients who continued VNS, thirteen (56.5% of the overall cohort) were considered responders (≥50% seizure frequency reduction). Amongst the clinical variables analysed, only psychiatric comorbidity correlated with poorer seizure outcomes, but was non-significant after applying the Bonferroni correction. Although 16 patients reported side-effects, none resulted in the discontinuation of VNS therapy. SIGNIFICANCE: Over half of the patients with refractory IGE experienced a positive response to VNS therapy. VNS represents a viable treatment option for patients with refractory IGE, particularly for females, when other therapeutic options have been exhausted.

The United States Environmental Protection Agency and the Houlton Band of Maliseet Indians (HBMI) built a stream temperature spatial statistical network (SSN) model for the Meduxnekeag Watershed. The headwaters of the Meduxnekeag Watershed are in Maine, United States of America and the outlet is in New Brunswick, Canada, creating an additional challenge because many datasets are constrained to political boundaries. The release of the High-Resolution National Hydrology Dataset Plus included transboundary watersheds and enabled creation of fine resolution (1:24,000) SSN temperature models consistent with management scales for riparian buffers. SSN models were developed for July, August, and September median stream temperatures and the growing season maximum (GSM). Fitted SSN models had relatively high R2 values (0.88-0.96) and all final models included significant parameters for shade-attenuated solar radiation, reference flow, air temperature, and bankfull depth or width. Fitted models predicted stream temperatures during a dry (2010) and wet (2011) year. Monthly models predicted the fewest cold water (<19.0 °C) reaches in July with 28% in the dry and 68% in the wet year. September had >99% cold water reaches, and August results were intermediate between July and September. GSM predictions found 81% of stream reaches could not support salmonid survival (>27.0 °C) in the dry year and 59% of the reaches were warmwater (22.5-27.0 °C) in the wet year. The model was used to predict stream temperatures following restoration scenarios of a forested 30-m or 90-m buffer of stream segments bordered by agricultural or developed land. The restoration scenarios expanded cold water habitat based on monthly median temperatures and decreased the habitat area with GSM above survival thresholds, with little difference in effectiveness of the two buffer widths. These results will guide riparian restoration projects by the HBMI to expand habitat for cold water fishes.

Besides functioning as a chemotactic factor, CCL5 has been associated with progression of disease in women with breast cancer, immune modulation and metastasis. Here we asked whether CCL5 produced by tumor cells contributed to growth or metastasis of breast cancer. For this purpose, we used two murine mammary carcinomas, the 4T1 tumor which is metastatic and constitutively expresses CCL5, and the 168 tumor which is not metastatic and does not constitutively express CCL5. RNA interference was used to inhibit CCL5 expression from the 4T1 tumor, and a CCL5 transgene was used to express CCL5 by the 168 tumor. Six different clones of 4T1 that exhibited stable reduction in CCL5 expression, and three different clones of 168 that exhibited stable CCL5 expression were compared to the parental tumors and vector transfected controls. Significantly, in both models, tumor-derived CCL5 expression did not correlate with MHC expression, growth rate, or metastatic ability of the tumors. These results show that tumor-derived CCL5 expression alone does not make a significant contribution to breast cancer progression.
