RESUMEN

Nanoparticles (NP) are potentially reprotoxic, which may compromise the success of populations. However, the reprotoxicity of NP is still scarcely addressed in marine fish. Therefore, we evaluated the impacts of environmentally relevant and supra environmental concentrations of titanium dioxide (TiO2: 10 to 10,000 µg·L-1) and silver NP (Ag: 0.25 to 250 µg·L-1) on the sperm of gilthead seabream (Sparus aurata). We performed short-term direct exposures (ex vivo) and evaluated sperm motility, head morphometry, mitochondrial function, antioxidant responses and DNA integrity. No alteration in sperm motility (except for supra environmental Ag NP concentration), head morphometry, mitochondrial function, and DNA integrity occurred. However, depletion of all antioxidants occurred after exposure to TiO2 NP, whereas SOD decreased after exposure to Ag NP (lowest and intermediate concentration). Considering our results, the decrease in antioxidants did not indicate vulnerability towards oxidative stress. TiO2 NP and Ag NP induced low spermiotoxicity, without proven relevant ecological impacts.

RESUMEN

Tritia reticulata (L.) is a neogastropod ubiquitous in the coastal communities of the NE Atlantic. Its life cycle relies on the swimming performance of planktonic early life stages, whose sensitivity to the climate conditions projected for the near future, namely of ocean acidification (OA) and warming (W), is, to our best knowledge, unknown. To examine the resilience of larval stages to future environmental conditions, this work investigates the effect of OA-W on the swimming performance of T. reticulata veligers under a range of experimental conditions, based on the end-of-century projections of the Intergovernmental Panel on Climate Change. Veligers were exposed to six experimental scenarios for 14 days, employing a full factorial design with three temperatures (T°C: 18, 20 and 22 °C) and two pH levels (pHtarget: 8.1 and 7.8). Mortality was assessed throughout the trial, after which swimming behaviour - characterised by the activity, speed and the distance travelled by veligers - was analysed by automated video recordings in a Zebrabox® device. Mortality increased with OA-W and, although more active, larvae travelled shorter distances revealing reduced swimming speed under acidic and warmer conditions, with the interaction of the tested stressors - pH and T°C - being highly significant. Results motivated the morpho-histological analysis of larvae preserved at the end of the trial, to check for the integrity of the organs involved in veligers' motion: statocysts, velum and foot. Statocyst and velar morpho-structure were conserved but histological damage of metapodial epithelia was evident under acidity, namely an apparent hypertrophy and protrusion of the secretory cells, with dispersed pigmented granules and, at 22 °C, less cilia, with potential functional implications. Negative consequences of the OA-W scenarios tested on veligers' competence are unveiled, pointing towards the eminent threat these phenomena constitute to T. reticulata perpetuation in case no mitigation measures are taken, and projections become effective.

RESUMEN

BACKGROUND AND AIMS: Although many studies have reported the effects of AT1 receptor on dietary salt overload, the role of AT2 receptor in this model is far from completely elucidated. The present study aimed to better understand the role of AT2 receptor in cardiac structure alterations in response to chronic high salt intake in rats. METHODS AND RESULTS: Male Wistar rats were fed a normal or high salt diet from weaning until 18 weeks of age. Both groups were subdivided into two groups. Starting at 7 weeks of age, rats were treated with or without compound 21 (0.3 mg/kg/day, n = 16), an AT2 receptor agonist. Metabolics and structural parameters were measured. BP, transverse cardiomyocyte and intersticial fibrose was higher in animals fed with high salt diet compared with normal salt fed animals. CONCLUSION: Compound 21 prevented the development of cardiac hypertrophy and fibrosis, reduced the increase in blood pressure and prevented the lower weight gain in animals fed a high salt diet.

RESUMEN

INTRODUCTION: The high morbidity and mortality caused by chronic kidney disease, and consequently, the increase in the need for kidney transplants, makes the evaluation of the kidney donation process relevant to verifying the service's fragility with the goal of optimizing this process. OBJECTIVE: This study analyzed the process of kidney donation in the Northern Macroregional area of Paraná, Brazil. METHOD: This was a cross-sectional and retrospective evaluation study based on 586 reports of deaths provided by the Intra-Hospital Organ Donation and Transplant Tissue Commissions to the Organ Procurement Organization of the Northern Macroregional area of Paraná in Brazil between 2011 and 2015. Data were analyzed by descriptive statistics, prevalence ratio, and Poisson regression. RESULTS: The prevalence of kidney donation was higher among the deaths of individuals younger than 61 years of age (P = .018) due to traumatic brain injury and hemorrhagic cardiovascular accident (P < .001), assisted in private institutions (P = .037), and occurring in the second half of the year (P = .015). CONCLUSION: The process of kidney donation was mainly compromised due to the refusal of the family and was associated with clinical and organizational aspects.

RESUMEN

This work reports the chemical monitoring of saline water and sediments at Aratu port, All Saints Bay, Brazil, when subjected to a deep dragging process (DDP) over a 2-year period (2010-2012). The aim of this work was to detect which metals had possible deleterious effects on the biota. It analyzed percentage of silt + clay and TOC and metal concentrations (As, Cd, Cu, Cr, Ni, Pb, and Zn) before, during, and after the DDP. The determination of metal concentration in sediments used the following techniques: atomic absorption spectrometer with flame, also with hydrate generation, and cold vapor spectrophotometry. The DDP disturbed the concentrations of metals in sediments and water, but 15 months after the DDP, only Cu and Zn were detectable on the bottom saline water, and 20 months after the DDP, only Zn was detectable in water. This fact indicates that these metals were longer available for absorption by the marine biota with probable deleterious effect. Indeed, studies by others during the DDP in Aratu port, indicated the decrease from 50 to 40 of the number of microalgae species identified. On the other hand, in the sediments, 20 months after the DDP, the concentrations of As, Cr, Cu, Pb, and Hg, returned to the original values, but Ni and Zn did not return to their original values. Comparison with studies by others indicated that the sediments from dredging were inappropriate for disposal at sea. This work indicated that the metals Cu, Ni, and Zn are the most deleterious elements to the marine environment.

RESUMEN

In order to support further genetic, diversity, and phylogeny studies of Capsicum species, the transferability of a Capsicum annuum L. simple sequence repeat (SSR) microsatellite set was analyzed for C. frutescens L. ("malagueta" and "tabasco" peppers) and C. chinense Jacq. (smell peppers, among other types). A total of 185 SSR primers were evaluated in 12 accessions from 115 C. frutescens L. and 480 C. chinense Jacq, representing different types within each species. Transferability to C. frutescens L. and C. chinense Jacq. occurred for 116 primers (62.7%). Nineteen (16.37%) were polymorphic in C. frutescens L. and 36 (31.03%) in C. chinense Jacq., 17 of which were coincident and could be used to analyze samples obtained for the 2 species. Among these primers, CA49 showed a different amplitude range of alleles between the 2 species (130-132 base pairs for C. frutescens L. and 120-128 base pairs for C. chinense Jacq.), and could differentiate the species. A total of 55 alleles were identified among the 19 polymorphic SSR loci among accessions of C. frutescens L., with the number of alleles per locus ranging from 2 to 5, a mean of 2.89, and the polymorphic information content ranging from 0.30 to 0.65. The number of alleles identified in C. chinense Jacq. was 119, ranging from 2 to 5 alleles per locus, an average of 3.30, and polymorphic information content from 0.19 to 0.68. The C. annuum L. SSR primers were most often transfer-able and polymorphic for C. frutescens L. and C. chinense Jacq., and we present a set of SSR for each species.

RESUMEN

BACKGROUND AND AIM: To evaluate the effects of low or high salt intake during pregnancy on left ventricle of adult male offspring. METHODS AND RESULTS: Low- (LS, 0.15%), normal- (NS, 1.3%) or high-salt (HS, 8% NaCl) diet was given to Wistar rats during pregnancy. During lactation all dams received NS as well as the offspring after weaning. To evaluate cardiac response to salt overload, 50% of each offspring group was fed a high-salt (hs, 4% NaCl) diet from the 21st to the 36th week of age (LShs, NShs, HShs). The remaining 50% was maintained on NS (LSns, NSns and HSns). Echocardiography was done at 20 and 30 weeks of age. Mean blood pressure (MBP), histology and left ventricular angiotensin II content (AII) were analyzed at 36 weeks of age. Interventricular septum, left ventricular posterior wall and relative wall thickness increased from the 20th to the 30th week of age only in HShs, cardiomyocyte mean volume was higher in HShs compared to NShs, LShs and HSns. AII and left ventricular fibrosis were not different among groups. CONCLUSIONS: HS during pregnancy programs adult male offspring to a blood pressure and angiotensin II independent concentric left ventricular hypertrophy, with no fibrosis, in response to a chronic high-salt intake.

RESUMEN

BACKGROUND AND AIM: To study the influence of high salt intake during pregnancy and lactation on body weight, blood pressure, and the function of the renin-angiotensin system in adult rats. METHODS AND RESULTS: Female Wistar rats received a low (0.15 NaCl), normal (1.30), or high (8% diet) salt diet. Mating occurred on the 12th week of age. From weaning, the offspring received normal salt diet. Weekly tail-cuff blood pressure and body weight measurements were performed during pregnancy and in the offspring (body weight since weaning and tail-cuff blood pressure between the 8th and the 12th week of age). Salt sensitivity of the blood pressure was evaluated and plasma renin activity determinations were performed in the 12-week-old offspring. Immunohistochemistry for renal angiotensin II was performed in the adult offspring. Renal mass and the number of glomeruli were determined. Tail-cuff blood pressure was higher in salt overloaded dams than in normal and low salt ones. In the adult offspring from the high salt dams, lower body weight, higher tail-cuff blood pressure, lower salt sensitivity in females, and increased kidney angiotensin II were observed. Plasma renin activity did not change with changes in salt intake in the adult offspring submitted to high salt environment during the perinatal period. In the offspring, renal mass and the number of glomeruli were not influenced by the dams' salt intake. CONCLUSIONS: Salt overload during pregnancy and/or lactation has long-term effects on offspring's body weight and blood pressure. In addition, high salt diet during the perinatal period induced renin-angiotensin system functional disturbances in the offspring.
