RESUMEN

From the reaction of ceftriaxone 1 antibiotic with 2,6-diaminopyridine 2 a ceftriaxone-based Schiff base (H2L,3) was obtained and its transition metal complexes were synthesized. Spectroscopic and physicochemical techniques, namely, UV-visible, FT-IR, 1H NMR, EPR, mass spectrometry, molar conductance, magnetic susceptibility and density functional theory (DFT) calculations, together with elemental and thermal analyses, were used to find out the binding mode and composition of these complexes. The ceftriaxone-based Schiff base 3 behaves as a monoanionic tridentate N,N,O ligand. Spectral and magnetic data suggest an octahedral geometry for all complexes and the general formulae [M(HL)(OAc)(H2O)2] (M(II) = Mn2+4, Co2+5, Ni2+6, Cu2+7, Zn2+8), are proposed for them. All compounds were screened for antibacterial activity using both the agar disc diffusion method and the minimal inhibitory concentration (MIC). It was found that complex 8 exhibited the most promising bactericidal activity against S. aureus (MIC = 0.0048 µmol/ml) and E. coli (MIC = 0.0024 µmol/ml). It is more active than the free ligand 1 (MIC = 0.0560 µmol/ml for S. aureus and 0.0140 µmol/ml for E. coli). These MIC results were compared with those obtained using similar zinc(II) Schiff base complexes, and with the values obtained using ceftriaxone conjugated with silver and gold nanoparticles (NPs), using earlier published data. Synthesized metal complexes exhibited LC50 values >1000 ppm indicating their nontoxicity against brine shrimp nauplii (Artemia Salina).

In recent years, high-throughput lipid profiling has contributed to understand the biological, physiological and pathological roles of lipids in living organisms. Across all kingdoms of life, important cell and systemic processes are mediated by lipids including compartmentalization, signaling and energy homeostasis. Despite important advances in liquid chromatography and mass spectrometry, sample extraction procedures remain a bottleneck in lipidomic studies, since the wide structural diversity of lipids imposes a constrain in the type and amount of lipids extracted. Differences in extraction yield across lipid classes can induce a bias on down-stream analysis and outcomes. This review aims to summarize current lipid extraction techniques used for untargeted and targeted studies based on mass spectrometry. Considerations, applications, and limitations of these techniques are discussed when used to extract lipids in complex biological matrices, such as tissues, biofluids, foods, and microorganisms.

Actinobacteria are the major source of bioactive secondary metabolites and are featured in the search for antimicrobial compounds. We used nuclear magnetic resonance (RMN)-metabolic profiling and multivariate data analysis (MVDA) to correlate the metabolites' production of Streptomyces sp. PNM-9 from the algae Dictyota sp. and their biological activity against the rice phytopathogenic bacteria Burkholderia spp. The compounds 2-methyl-N-(2'-phenylethyl)-butanamide (1) and 3-methyl-N-(2'-phenylethyl)-butanamide (2) were identified through MVDA and 2D NMR experiments in the organic extract of a 15-days LB media culture of Streptomyces sp. PNM-9. Compounds 1 and 2 were isolated and their structures confirmed by one- and two-dimensional NMR and mass spectrometry (MS) data. Compounds 1 and 2 were active against the rice pathogenic bacteria Burkholderia glumae (ATCC 33,617) displaying minimal inhibitory concentration (MIC) values of 2.43 mM and 1.21 mM, respectively. The metabolomics-guided approach employing NMR-metabolic profiling was useful for marine microbial bioprospecting and suggested Streptomyces sp. PNM-9 strain and its compounds as a potential control against phytopathogenic bacteria.

Two cyclotetrapeptides, henceforth named Provipeptides A (1) and B (2), along with five known diketopiperazines (3-7) were isolated from the liquid culture of marine Streptomyces sp. 161a recovered from a sample of sea grass Bryopsis sp. The structures of cyclotetrapeptides and diketopiperazines (DKPs) were established by 1D and 2D NMR data, MS, and by comparison with literature data. The absolute stereochemistry of compounds cyclo-(L-Pro-L-Leu-D-Pro-L-Phe) 1 and cyclo-(-Pro-Ile-Pro-Phe) 2 was established by the Marfey's method. Compound 1 showed antibacterial activity against rice phytopathogenic strains Burkholderia glumae (MIC = 1.1 mM) and Burkholderia gladioli (MIC = 0.068 mM), compound 2 was active only against B. glumae (MIC = 1.1 mM), and DKP cyclo-[L-Pro-L-Leu] 5 showed to be active against B. gladioli (MIC = 0.3 mM) and B. glumae (MIC = 2.4 mM). Compounds 1 and 2 showed 65% and 50% inhibition of Colletotrichum gloeosporioides (yam pathogen) conidia germination, respectively at a concentration of 1.1 mM.

The aim of this study was to describe the ontogeny of thyroid follicles in cobia Rachycentron canadum. Larvae were sampled daily (n=15 - 20) from hatching until 15 dah (days after hatching). Following, larvae were sampled every two days by 28 dah; a new sample was taken at 53 dah. The samples were dehydrated, embedded in Paraplast, and sections of 3 µm were dewaxed, rehydrated and stained with HE and PAS. A single follicle was already present 1 dah and three follicles were found 8 dah. The number of follicles increased up to 19 on 53 dah. The diameter of follicles and follicular cell height were lower 1 dah (6.83 ± 1.00 and 4.6 ± 0.01 µm), but increased from 8 dah (24.03 ± 0.46 µm e 6.43 ± 0.46 µm). From 8 dah, the presence of reabsorption vesicles was observed in the colloid and from the 19 dah some follicles did not present colloid. The early thyroid follicle appearance in cobia larvae as well as the high quantity of follicles without colloid and/or with vesicles even after the metamorphosis, might be the explanation of the fast growth of the cobia.

Se realizó un estudio descriptivo y transversal de 108 ancianos con 60 años y más, pertenecientes al Policlínico Universitario 28 de Septiembre de Santiago de Cuba, desde abril hasta octubre del 2011, a fin de evaluar los estados funcional y afectivo en esta población geriátrica. En la casuística prevalecieron las edades de 70-79 años (35,2 por ciento), el sexo femenino (60,2 por ciento) y las que tenían dependencia funcional en la realización de 1 o 2 tareas básicas de la vida diaria, así como en 3 o 4 actividades instrumentadas. También primaron los trastornos afectivos con predominio de la depresión y la ansiedad(AU)
