RESUMEN

The present study was designed to assess the phytochemical content of Ferulago angulata (FA) and possible in vivo nephroprotective effect of FA administration on trace elements, minerals, MDA and GSH in kidney and liver tissue samples, serum vitamin (α-tocopherol, retinol, cholecalciferol, phylloquinone), TSA, and LSA in a rat model of DMN-induced nephrotoxicity. In the study, Wistar albino rats were assigned to six groups: Control (0.9% NaCl), (DMN 10 mg/kg), (FA 150 mg/kg), (DMN + FA 150 mg/kg), (FA 300 mg/kg), and (DMN + FA 300 mg/kg). Rats were intraperitoneally given DMN for the first 7 days. Renal injury caused by DMN was proved by the histopathological alterations. The FA (300 mg/kg) treatment significantly normalized Se, Cr, Ca levels in liver and Co level in kidney tissue samples. These observed positive effects are due to the phytochemical content of the plant. The flower extract of FA (300 mg/kg) can be used for the prevention of kidney damage. PRACTICAL APPLICATIONS: Ferulago angulata flowers are used in traditional medicine for treat kidney and liver digestive system diseases. This species is endemic taxa of the family Apiaceae, which has been used both as food and therapeutics because of their phytochemical composition. In this study, the phenolic characterization of FA flower was used to a new RP-HPLC method, as well as the biological activity of FA flower and possible in vivo nephroprotective effect of FA flowers on trace elements, minerals, MDA and GSH in kidney and liver tissue samples and vitamins, TSA, and LSA in serum samples a rat model of DMN-induced nephrotoxicity. It was found that high level of phenolic compounds (chlorogenic acid, vanillic acid, 2-hydroxycinnamic acid) present in the flower extract of F. angulata has positive effects and antioxidant properties. Due to its phenolic content, FA flower extract could protect for kidney damage and can be used as antioxidants in the food additive and pharmaceutical industry.

ETHNOPHARMACOLOGICAL RELEVANCE: This study serves as the first investigation of the knowledge of traditional medicinal plants used in Agri Province. The mountainous and strongly fragmented area, diversity of local flora, limited access to modern medicines because of the people's seminomadic lifestyle, and harsh climatic conditions have prompted the locals to benefit from natural resources, principally plants. AIM OF THE STUDY: Although the use of medicinal plant is common in the area, documentation and inventory analysis are not available on the plant taxa used in traditional medicine in Agri Province. This study focused on the answer of the following question: What are the most medicinally valuable plant taxa in the province? Therefore, this study aimed to (i) document medicinal plant taxa used in Agri Province; (ii) determine the endemic plant species and their risk categories; (iii) determine the most widely used plant taxa for specific ailments to further contribute to drug discovery; (iv) determine the local names, plant part(s) used, preparation, and utilization methods of these local plant medicines to assist with pharmaceutical studies; and (v) reveal the origin(s) of traditional medicinal knowledge of Agri Province. MATERIALS AND METHODS: Ethnopharmacological field studies were conducted between 2014 and 2016 in 638 locations within Agri Province. Plant samples used in folk medicine were determined through face-to-face interviews with 120 local healers. Traditional medicinal plant samples were collected from wild areas with the help of the local healers and transported to Van Pharmaceutical Herbarium (Van, Turkey) for botanical identification. The statistical analyses of data were conducted using the informant consensus factor (ICF) method. RESULTS: In this study, 118 medicinal taxa belonging to 35 families of plants were determined to be used in Agri Province. Ninety-three different local names were recorded, which mainly described the plant's morphological characteristics. Data analysis (ICF) revealed that the traditional medicinal plants used in Agri Province might offer promising information for new pharmaceuticals. CONCLUSIONS: The local population in the study area has a preserved and comprehensive knowledge of traditional medicine because of its limited access to modern medicine, seminomadic lifestyle, isolated geographical position, traditional taste habits, climatic conditions, diversity of local flora, and mountainous and rugged geography. The medicinal plants presented in this study might provide valuable leads for the identification of bioactive compounds for pharmaceutical use and represent a vast and underutilized resource for the development of newer and more efficient pharmacological treatments.

Previously tested n-hexane extracts of the Scorzonera latifolia showed promising bioactivity in vivo. Because triterpenes could account for this activity, n-hexane extracts were analyzed by HPLC to identify and quantify the triterpenes as the most abundant constituents. Other Scorzonera and Podospermum species, potentially containing triterpenic aglycones, were included in the study. An HPLC method for simultaneous determination of triterpene aglycones was therefore developed for analysis of Podospermum and Scorzonera species. n-Hexane extracts of root and aerial parts of S. latifolia, ten other Scorzonera species and two Podospermum species were studied to compare the content of triterpenes. HPLC was used for the qualitative and quantitative analysis of α-amyrin, lupeol, lupeol acetate, taraxasteryl acetate, 3-ß-hydroxy-fern-7-en-6-one acetate, urs-12-en-11-one-3-acetyl, 3-ß-hydroxy-fern-8-en-7-one acetate, and olean-12-en-11-one-3-acetyl. Limits of detection and quantification were determined for each compound. HPLC fingerprinting of n-hexane extracts of Podospermum and Scorzonera species revealed relatively large amounts of triterpenes in a majority of investigated taxa. Lupeol, lupeol acetate, and taraxasteryl acetate were found in a majority of the species, except S. acuminata. The presence of α-amyrin, 3ß-hydroxy-fern-7-en-6-one-acetate, urs-12-en-11-one-3-acetyl, 3ß-hydroxy-fern-8-en-7-one-acetate, and olean-12-en-11-one-3-acetyl was detected in varying amounts. The triterpene content could correlate with the analgesic and anti-inflammatory activity of Scorzonera, which was previously observed and Scorzonera species that have been determined to contain triterpenes in large amounts and have not yet been tested for their analgesic activity should be tested for their potential analgesic and anti-inflammatory potential. The presented HPLC method can be used for analysis of triterpene aglycones, for example dedicated to chemosystematic studies of the Scorzonerinae.

We investigated the effects of Leontice leontopetalum and Bongardia chrysogonum on apoptosis, gamma-aminobutyric acid (GABAA) receptor positive cell number, cyclin-B1 and bcl-2 levels and oxidative stress in pentylenetetrazol (PTZ) kindling in rats. Kindling was produced by subconvulsant doses of PTZ treatments in rats. Wistar albino rats were divided into 4 groups; Control, PTZ treated (PTZ), PTZ+L. leontopetalum extract treated (PTZ+LLE) and PTZ+B. chrysogonum extract treated (PTZ+BCE) groups. Extracts were given a dose (200 mg/kg) 2h before each PTZ injection. PTZ treatment significantly decreased the glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities and bcl-2 levels and increased the total oxidant status (TOS), malondialdehyde (MDA), cyclin B1, oxidative stress index (OSI) and number of neurons that expressed GABAA receptors when compared to the control. LLE and BCE possessed antioxidant activity in the brain and ameliorated PTZ induced oxidative stress, decreased cyclin-B1, increased bcl-2 levels, and kept the GABAA receptor number similar to that of the control despite the PTZ application.

CONTEXT: Ferulago angulata (Schlecht.) Boiss. (Apiaceae) (FASB) is used to treat liver diseases and has been used both as food and therapeutics by many cultures for thousands of years because of the natural antioxidant compounds. OBJECTIVE: This study determines antioxidant properties of FASB flowers, the levels of minerals and vitamins, and also, evaluates the hepatoprotective effect of flowers against N-nitrosodimethylamine (NDMA) induced on liver tissue by assessing antioxidant enzymes and histopathological parameters in Wistar albino rats. MATERIALS AND METHODS: In the study, the rats were divided into six groups of ten. Control, untreated animals were given 0.9% NaCl. Rats were intraperitoneally given NDMA (10 mg/kg) for the first 7 days. FASB methanol extract (150 and 300 mg/kg) was administered orally for 21 days. RESULTS: α-Tocopherol, retinol, ascorbic acid, total antioxidant activity, phenolic and flavonoid contents of FASB were 0.70 ± 0.13, 0.29 ± 0.03 µg/g, 139.32 ± 7.06 µg/100 g, 171.61 ± 6.05 mM ascorbic acid/g, 90.47 ± 4.11 mg GA/g and 37.39 ± 2.85 mg QE/g. DPPH and hydroxyl radical scavenging activity was obtained IC50 67.34 ± 4.14 and 64.87 ± 4.68 µg/mL, respectively. DISCUSSION AND CONCLUSION: The results of the study indicated that FASB flowers contain high levels of vitamins, minerals, total antioxidant activity, phenolics and flavonoids. Due to the positive effect on significant changes in antioxidant enzymes of liver tissue and histopathological examination, it is thought that the plant could be used as a hepatoprotective.

Eight known secondary metabolites were isolated from the methanolic extract of the whole plant of Astragalus lycius Boiss. They were identified as 5,5'-dihydoxy-3'-methoxy-isoflavone-7-0-ß-D-glucoside (1), genistin (2), sissotrin (3), 5,4'-dimethoxy-isoflavone-7-0-ß-D-glucopyranoside (4), (7S,8R)-5-methoxydehydrodiconiferyl alcohol-4-0-ß-D-glucopyranoside (5), 4-0-lariciresinol-glucoside (6), 2-phenylethyl-ß-D-glucopyranoside (7) and ß-sitosterol-3-O-ß-D-glucopyranoside (8) by spectroscopic methods including (1)H- and (13)C-NMR and HR-MS experiments, and by comparison with literature values. Compounds 1-7 are reported for the first time from Astragalus taxa. All of the compounds were tested for their cytotoxic activities against a number of cancer cell lines. Among them, only 6 exhibited significant activity against human colon carcinoma (HT-29) at 2.69 µM concentration.

CONTEXT: Scorzonera L. species (Asteraceae) are edible and as medicinal plants are used for various purposed in folk medicine. OBJECTIVE: The methanol extracts of the aerial parts and roots from 27 Scorzonera taxa were investigated for their possible neurobiological effects. MATERIALS AND METHODS: Inhibitory potential of the Scorzonera species was tested against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and tyrosinase (TYRO) at 100 µg mL(-1) using ELISA microtiter assay. Antioxidant activity of the extracts was tested with radical scavenging activity, metal-chelation capacity, ferric- (FRAP), and phosphomolibdenum-reducing antioxidant power (PRAP) assays. Chlorogenic acid, hyperoside, rutin, and scorzotomentosin-4-O-ß-glucoside were also screened in the same manner. Total phenol and flavonoid quantification in the extracts were determined spectrophotometrically. RESULTS: The aerial parts of Scorzonera pisidica (40.25 ± 0.74%) and chlorogenic acid (46.97 ± 0.82%) displayed the highest TYRO inhibition, while the remaining samples showed only trivial inhibition against cholinesterases (2.08 ± 1.35%-25.32 ± 1.37%). The same extract of S. pisidica was revealed to be the most potent in scavenging of all three radicals and FRAP assay. DISCUSSION AND CONCLUSION: Out of 27 taxa, S. pisidica, in particular, may deserve further investigation for its neuroprotective potential.

Four cycloartane- (hareftosides A-D) and oleanane-type triterpenoids (hareftoside E) were isolated from Astragalus hareftae along with fifteen known compounds. Structures of the compounds were established as 3,6-di-O-ß-D-xylopyranosyl-3ß,6α,16ß,24(S),25-pentahydroxycycloartane, 3,6,24-tri-O-ß-D-xylopyranosyl-3ß,6α,16ß,24(S),25-pentahydroxycycloartane, 3-O-ß-D-xylopyranosyl-3ß,6α,16ß,25-tetrahydroxy-20(R),25(S)-epoxycycloartane, 16-O-ß-D-glucopyranosyl-3ß,6α,16ß,25-tetrahydroxy-20(R),24(S)-epoxycycloartane, 3-O-[ß-D-xylopyranosyl-(1→2)-O-ß-D-glucopyranosyl-(1→2)-O-ß-D-glucuronopyranosyl]-soyasapogenol B by the extensive use of 1D- and 2D-NMR experiments along with ESI-MS and HR-MS analyses.

The study was designed to assess the effect of Hypericum perforatum L. (H.P) on serum and hair trace elements and mineral levels, oral administration of 7,12-dimethylbenz[a]anthracene (DMBA) induced oxidative stress in Sprague-Dawley female rats. Analysis of the trace element has been carried out using atomic absorption spectrophotometer method at end of 60th day. It has been found out that the DMBA group contained statistically lower Zn and Cr compared to the control group (p<0.01) and (p<0.05), Cu, Mg and Na contained higher than control group (p<0.05), (p<0.05) and (p<0.05). In DMBA+H.P group, Zn higher and Na lower than DMBA group (p<0.05), (p<0.05), in hair samples Cd, K and Zn contained lower DMBA compared to the control group (p<0.05), (p<0.05) and (p<0.05). In group DMBA+H.P, Cd was higher than DMBA group and Cr lowered accordance with control group (p<0.05). The present study demonstrated significantly positive and beneficial effect of H.P on the concentration levels of Zn and Na in serum, also on Cd levels in hair between DMBA and DMBA+H.P groups.

Van Herby Cheese is a traditional milk product including local herb species in eastern Turkey. This special milk product was previously produced only for the local market, but industrial scale production and marketing have recently started in the region. However, some quality characteristics such as microbial flora and heavy metal concentrations of this novel product need to be investigated. In this study, lead concentrations of 28 different herbs mostly used in Van Herby Cheese were analyzed by AAS. The highest lead concentration of 1.69 mg kg(-1) of the analyzed herbs was found in Mentha longifolia (L.) Hudson subsp. longifolia.

Eight cycloartane-type triterpene glycosides (1-8) were isolated from Astragalus aureus Willd along with ten known cycloartane-type glycosides (9-18). Their structures were established by the extensive use of 1D and 2D-NMR experiments along with ESIMS and HRMS analyses. Compounds 1-5 are cyclocanthogenin glycosides, whereas compounds 6-8 are based on cyclocephalogenin as aglycon, more unusual in the plant kingdom, so far reported only from Astragalus spp. Moreover, for the first time monoglycosides of cyclocanthogenin (5) and cyclocephalogenin (7, 8) are reported. All of the compounds tested for their cytotoxic activities against a number of cancer cell lines. Among the compounds, only 8 exhibited activity versus human breast cancer (MCF7) at 45 µM concentration.

Six cycloartane-type triterpene glycosides were isolated from Astragalus icmadophilus along with two known cycloartane-type glycosides, five known oleanane-type triterpene glycosides and one known flavonol glycoside. From the roots of Delphinium linearilobum (Trautv.) N. Busch two new norditerpene alkaloids linearilobin and linearilin, and the known alkaloids lycoctonine, 14-acetyltalatizamine, browniine, cammaconine, talatizamine, and cochlearenine were isolated. Spectroscopic techniques were used for structure determination. Antioxidant activity was performed by DPPH and metal chelating activity assays.

RESUMEN

A survey of the flavonoid glycosides of selected taxa in the genus Veronica yielded two new acylated 5,6,7,3',4'-pentahydroxyflavone (6-hydroxyluteolin) glycosides and two unusual allose-containing acylated 5,7,8,4'-tetrahydroxyflavone (isoscutellarein) glycosides. The new compounds were isolated from V. liwanensis and V. longifolia and identified using NMR spectroscopy as 6-hydroxyluteolin 4'-methyl ether 7-O-alpha-rhamnopyranosyl(1"'-->2")[6"-O-acetyl-beta-glucopyranoside] and 6-hydroxyluteolin 7-O-(6"-O-(E)-caffeoyl)-beta-glucopyranoside, respectively. Isoscutellarein 7-O-(6"'-O-acetyl)-beta-allopyranosyl(1"'-->2")-beta-glucopyranoside was obtained from both V. intercedens and V. orientalis and its 4'-methyl ether from V. orientalis only. Complete 1H and 13C NMR spectral assignments are presented for both isoscutellarein glycosides. Two iridoid glucosides new to the genus Veronica (melittoside and globularifolin) were also isolated from V. intercedens.
