RESUMEN

The veterinarian is responsible for monitoring and inspecting rodeos, thus ensuring the welfare of the animals. Clinical follow-up is of great importance in animal athletes, and ultrasound has become an important, widely used complementary diagnostic method in the evaluation of these animals. In equine medicine and dairy cattle, there are several studies that describe tendon injuries through ultrasonography. However, in athlete bulls, there are still no studies using this tool as a diagnostic aid for these injuries. The first objective of the present study is to describe the ultrasound anatomy of the distal flexor structures of the thoracic limbs of healthy rodeo bulls and their measurements. The second objective is to report sonographic findings referring to changes found in the studied region of these animals. Forty-two bulls from a farm specialized in breeding and training high-performance animals were evaluated. Physical evaluation and measurements of external parameters were performed to verify possible correlations. For the ultrasound examination, a 10 MHz linear probe was used. Measurements were taken of the tendons of the flexor digit muscles, branch of the interosseous muscle for the tendon of the superficial digital flexor muscle and interosseous muscle. 42 animals were evaluated, but two were excluded from the statistical analysis because they had a history of lameness or excessive effort in the preceding six months, and their cases are described in this study. Based on knowledge of ultrasound and anatomy, greater reliability is obtained in the diagnosis and prognosis of these animals.

This study aimed to describe an animal model for studying equine visceral pain using minimally invasive and video-assisted cecum and ileum instrumentation. The access to the cecum and ileum was affected because of a previous typhlostomy. For video-assisted distention of the cecum and ileum, a distention device, which we developed using an endotracheal Rusch probe, was used, adapted, and coupled to a cuffometer to inflate and measure the pressure of the cuff attached to its distal portion. In a video-assisted manner, the distal portion of the device was introduced into the cecum and ileum, which contained the cuff in its distal portion, properly positioning it in the lumen. The cuff of the distension device was insufflated after the measurement of baseline physiological parameters of the animals and video-assisted confirmation of its right placement in the cecum and ileum lumen (M0). Was performed in one moment through two simultaneous cuff compressions and 1 minute of animal observation to evaluate the degree of abdominal discomfort manifestations (M1). To cease these stimuli, the cuff was deflated by disconnecting the extensor of the distension device attached to its proximal portion (M2). The procedure was easily performed in most cases. Slow and progressive insufflation allowed subjective adjustment of the intensity of the pain stimulus based on behavioral manifestations. Even with a low rate of complications, the model is feasible and reproducible for studies on visceral pain and the evaluation of analgesic effects.

This study was conducted to evaluate and validate the efficacy and safety of videoceloscopy and gonadal biopsy as sexing methods for the A. ocellatus. A total of 31 adult individuals were used. Florfenicol (50 mg/kg) and morphine (5 mg/kg) were administered intramuscularly during the pre-surgical period. Animals were maintained in a supine position preceding a ventral midline incision and endoscope optics were then utilized for gonad visualization and sex identification. A gonadal fragment was collected using laparoscopic forceps and conditioned in 10 % formalin. To suture the cavity, polyamide yarn was used in a simple and continuous pattern. At 15 days subsequent to surgery, healing was evaluated, and the stitches were removed. Videoceloscopy accuracy and gonadal biopsy effectiveness were 97 % and 83 %, respectively. Total time devoted in the videoceloscopy, gonadal biopsy and surgery was longer for animals identified as males compared to females The survival rate was 100 %. There were differences regarding food consumption at 24 and 36 h post-surgery when compared to control specimens (pre-surgical) Regarding position in the water column, differences were observed at 24 and 72 h after surgery when compared individually to the control specimens. There were differences for interaction behavior at 24, 36 and 60 h, and regarding search for hiding places at 12 and 24 h after surgery in relation to the control specimens. The applied videoceloscopy and gonadal biopsy surgical techniques are, therefore, effective and safe for A. ocellatus sexing procedures.

A Catuama® é a associação de quatro extratos hidroalcoolicos obtidos de plantas brasileiras (Paullinia cupana, Trichilia catigua, Ptychopetalum olacoides e Zingiber officinale) com conhecida ação neuroprotetora, anti-inflamatória, antioxidante e antidepressiva. O bilobalide é um componente extraído das folhas do Ginkgo biloba, que tem comprovada ação neuroprotetora nos sistemas nervosos central e periférico. O presente estudo avaliou os efeitos da Catuama® e do bilobalide na regeneração nervosa periférica de ratos submetidos à secção do nervo isquiático. Foram utilizados 40 ratos com implante de tubo de silicone preenchido por colágeno líquido, deixando-se um intervalo entre os segmentos nervosos de 10mm. Os animais foram divididos em 4 grupos: o grupo controle (A); os grupos que receberam a Catuama® administrada por via oral nos primeiros 28 dias de pós-operatório, nas doses de 100 (B) e 400mg.kg-1 (C); e o grupo que recebeu o bilobalide na dose de 200µM (D), este, adicionado ao colágeno líquido utilizado no implante de silicone. Os animais foram avaliados na primeira, quinta e décima semanas de pós-operatório pelo teste de marcha. Na décima semana, foi realizada avaliação eletrofisiológica e análises quantitativa e qualitativa dos cortes histológicos de amostras do nervo isquiático e do músculo gastrocnêmio. Em todas as análises utilizadas observou-se excelente regeneração dos nervos, no entanto, não foi encontrada diferença significativa (P>0,05) entre os grupos experimentais e controle.


The Catuama® is composed of four Brazilian plants extracts (Paullinia cupana, Trichilia catigua, Ptychopetalum olacoides e Zingiber officinale). The Catuama® is known as having neuroprotector, anti-inflammatory, antioxidant and antidepressant effects. Bilobalide, extracted from leaves of Ginkgo biloba, is known by its neuroprotective effect in the central and peripheral nervous systems. The present study evaluates the effect of Catuama® and bilobalide on peripheral nerve regeneration in rats following a sciatic nerve section. Sciatic nerve of forty adult rats was transected with a 10-mm gap and the proximal and distal nerve stumps were fixed in a silicone tube filled with liquid collagen. The animals were divided into four groups: the control group (A), two groups treated with Catuama® by gavage along 28 days after the surgery in different doses of 100 (B) and 400mg.kg-1 (C) and the group using 200µM bilobalide (D) associated with the liquid collagen in the silicone tube. Evaluations were done by a walk test on the first, fifth and tenth week after the surgery. Electrophysiological stimulation and quantitative and qualitative histological analyses of the sciatic nerve and gastrocnemius muscle were also performed on the tenth week after the surgery. All groups showed good regeneration but no statistical difference was found between treatments and control groups (P>0.05).

Visando estudar os parâmetros do equilíbrio ácido-base de eqüinos submetidos a um modelo experimental de obstrução intestinal, 24 animais foram distribuídos em quatro grupos, controle instrumentado (GI), obstrução do duodeno (GII), íleo (GIII) e cólon maior (GIV). As amostras de sangue venoso foram coletadas antes das cirurgias (T0), durante as obstruções (T30ob-T180ob) e após as desobstruções (T60des-T180des). Os eqüinos do GIV, no T30ob, e os eqüinos do GII, nos T60ob, T90ob e T120ob, apresentaram aumento do pH(v), da cHCO-3(vP) e da cBase(v) que, acrescidos do aumento da pCO2(v) e da ctCO2(v), caracterizou a alcalose metabólica com compensação respiratória. Nos T90ob e T120ob, nos animais do GII, e no T180ob, nos animais do GIII, a pO2(v) e a sO2(v) tiveram comportamento semelhante. Os valores baixos apresentados pelos animais do GII foram associados à hipercapnia ou à hipoventilação, desencadeadas para a correção da alcalose metabólica. Entretanto, a hipoxemia apresentada pelos animais do GIII foi associada à hipovolemia presente neste período. As alterações ácido-base observadas constituem-se de alterações leves e temporárias, as quais não são capazes de predizer o diagnóstico das obstruções intestinais específicas em eqüinos com cólica. Entretanto, por relacionarem-se diretamente com a precocidade do distúrbio gastrintestinal, elas auxiliam o pesquisador no prognóstico.


This study aimed to evaluate parameters of acid-base balance in horses submitted to an experimental model of intestinal obstruction. Twenty-four animals were divided in four groups: instrumented control (GI), duodenum obstruction (GII), ileum obstruction (GIII) and large colon obstruction (GIV). Venous blood samples were collected before surgery (T0), during the obstruction (T30ob-T180ob) and after unblocking procedures (T60des-T180des). Animals from GIV, at T30ob and animals from GII, at T60ob, T90ob and T120ob, presented higher values for pH(v), cHCO-3(vP) and cBase(v) which, added to the increase of pCO2(v) and ctCO2(v), characterized the metabolic alkalosis with respiratory compensation. At T90ob and T120ob, in GII animals, and at T180ob, in GIII animals the pO2(v) and sO2(v) had similar response. Low values presented by GII animals were associated to hypercapnia or hypoventilation triggered for metabolic alkalosis correction. However, a hypoxemia presented by GIII animals was associated to hypovolemia at that period. There were light and temporary acid-base alterations that are not capable to predict specific intestinal obstruction diagnosis in equines with colic. However they help in prognosis since both have a direct relation with the precociousness of gastrointestinal disturbance.

A reconstituição cirúrgica de nervos em humanos, em cães e em diversos animais de laboratório é bastante utilizada e tem indicações clínicas e experimentais importantes. No entanto, em eqüinos há poucas informações sobre esta prática. Há relatos sobre a excessiva proliferação de tecido conjuntivo e a formação de neuromas à neurorrafia experimental, mesmo quando se utilizam tubos de silicone para condução do crescimento axonal. O presente estudo teve o objetivo de acrescentar informações sobre o processo de reparação de nervos periféricos em eqüinos por meio de implante de tubo de silicone preenchido ou não com colágeno. Para tanto, foram utilizados oito eqüinos, alocados em dois grupos: GI-13 semanas e GII-26 semanas de observação. Foi realizada secção dos nervos ulnares (NUs) e dos ramos cutâneos laterais dos 17º nervos torácicos (NTs), bilateralmente, seguindo tubulização, realizada em cada animal, alternando-se tubos de silicone vazios (TS), em um dos antímeros, ou preenchidos com solução de colágeno (TSC), no contralateral, deixando-se um espaço de 5mm entre os cotos. Nenhuma alteração foi encontrada ao exame do aparelho locomotor e as primeiras reações positivas ao teste de sensibilidade cutânea nos NUs e NTs com TS e TSC foram observadas a partir da 9ª semana, em ambos os grupos. Ao final do período de observação, verificou-se, macroscopicamente, que os nervos encontravam-se envolvidos por tecido conjuntivo e o interior da câmara estava preenchido por tecido de coloração esbranquiçada, de forma cilíndrica, interligando os cotos proximal e distal. Microscopicamente, constatou-se a presença de axônios mielinizados interligando os cotos, as células de Schwann e o processo de remielinização do coto distal, principalmente no TSC. Em ambos os grupos, não houve formação de neuromas e o processo inflamatório limitou-se às áreas perineurais. Concluiu-se que o implante de tubo de silicone conduz à regeneração de NUs e NTs de eqüinos...


Surgical nerve repair in humans, dogs and laboratory animals is widely utilized for important clinical and experimental purposes. However, in horses there is scarce information. Neuroma and excessive conjunctive proliferation are reported. The aim of the present study was to add information about the repair process utilizing implant of silicone tubes with or without collagen in nerve regeneration of horses. Eight horses were allocated in two groups: GI-13 weeks and GII-26 weeks of observation. A complete section of the ulnar nerves (UNs) and of the lateral cutaneous branch of the 17th thoracic nerves (TNs), bilaterally, was followed by repair with silicone tubes alternately filled with (STC) or without collagen (ST), with a 5mm gap between stumps. Clinical evaluation was performed weekly, by cutaneous sensitivity testing of the region innervated by the respective operated nerves and physical examination of the thoracic limbs. No alterations were found in the locomotor apparatus. The first positive reactions to the cutaneous test occurred at the 9th week post-surgery. At the end of the observation period it was verified, grossly, that the nerves were involved by conjunctive tissue and the lumen of the tube was filled by a whitish tissue, in a cylindrical shape. The ends nerves were interconnected. Microscopically, myelinated axons and Schwann cells were present in the gap between the stumps. In the GII the number of myelinated fibers was greater. The distal nerve showed remyelination process. There was no presence of neuroma and the inflammatory process was limited to the perineural areas. It was concluded that the implant of silicone tubes results in regeneration of UNs and TNs of horses without formation of neuromas and that de addition of collagen promotes an increase in the number of myelinated fibers.

RESUMEN

Sample preparation and DNA extraction protocols for DNA amplification by PCR, which can be applied in human fecal samples for taeniasis diagnosis, are described. DNA extracted from fecal specimens with phenol/chloroform/isoamilic alcohol and DNAzol reagent had to be first purified to generate fragments of 170 pb and 600 pb by HDP2-PCR. This purification step was not necessary with the use of QIAmp DNA stool mini kit. Best DNA extraction results were achieved after eggs disruption with glass beads, either with phenol/chloroform/isoamilic alcohol, DNAzol reagent or QIAmp DNA stool mini kit.

Com o intuito de utilizar a Reação em Cadeia pela Polimerase (PCR) como método de diagnóstico diferencial da teníase humana, avaliaram-se alguns protocolos de preparação e extração de DNA de ovos de Taenia saginata presentes em amostras de fezes de paciente naturalmente infectado. O DNA obtido após extração com fenol/clorofórmio/álcool isoamílico ou DNAzol® teve que ser purificado antes da PCR para que fosse possível a amplificação dos fragmentos de 170 pb e 600 pb desejados. Com o kit QIAmp DNA stool mini kit® tal purificação não foi necessária. Os melhores resultados foram observados após o tratamento prévio das amostras com pérolas de vidro, tanto quando da utilização de fenol/clorofórmio/álcool isoamílico, quando de DNAzol® ou QIAmp DNA stool mini kit®.

The taeniasis-cysticercosis complex is a zoonosis of great medical and economic importance where humans play an important role as the carrier of adult stage of Taenia solium and Taenia saginata. This paper describes PCR standardization that can be applied in human fecal samples for taeniasis diagnosis. DNA extraction was achieved with DNAzol reagent, after egg disruption with glass beads. DNA prepared from fecal specimens was first purified and PCR amplified generating fragments of 170 and 600 bp. The assay described herein provides an important tool for T. saginata identification in human fecal samples.
