RESUMEN

In dairy ruminants, the combination of milking parameters must ensure good milking performance without harming udder conditions. Commonly, milking conditions for goats are established without having checked the admissible limits for optimal and fast milking. The aim of this study was to establish a limit combination of machine milking parameters that improves machine milking performance without altering milkability or udder status. To this end, we studied the effect of 2 combinations (42 kPa, 120 cpm, 60% vs. 44 kPa, 120 cpm, 60% in terms of kilopascals of vacuum level, cycles per minute of pulsator rate, and percentage of pulsator ratio, respectively) on milk production and composition, milk fractioning during milking, SCC, teat tissue thickness variation after milking, and the milk emission kinetics parameters throughout 1 lactation period (6 mo). The 42 and 44 kPa measured at the vacuum gauge level became average values of 37.5 and 39.3 kPa, respectively, measured at the teat sphincter level during milking. Milk flow significantly increased and total milking time decreased 25 s with the elevation of the vacuum level from 42 to 44 kPa without any adverse effect on milk fractioning at milking. However, the use of 44 kPa also showed an increase in tissue thickness above 5%, and we observed a tendency of average conductivity of milk to increase, although without any adverse effect on SCC. It seems that 44 kPa, 120 cpm, 60% is a possible limit combination of parameters to improve milking performance without altering milkability or udder conditions. We concluded that this combination can be used for milking Murciano-Granadina breed goats in conditions similar to those of this study (mid-level milking system and 1 milking/d), although further studies are necessary to verify its application in the case of 2 milkings/d.

In dairy goats, the kid rearing system can have critical importance in financial returns. Commonly used criteria for the choice of rearing system are not always clear due to the high number of factors involved. The aim of this study was to quantify all those factors to facilitate decision making. So, the effect of two different kid rearing systems, mixed rearing system (MRS) and artificial rearing system (ARS), on milk yield, milk composition and somatic cell count (SCC), milk yield loss at weaning for MRS, kid growth and costs of the different traits on the financial returns in Murciano-Granadina breed goats was studied. Twenty-four goats per group were used. In the MRS, goats reared only one kid, which had free access to goat milk 24 h a day and were weaned at week 6 of lactation, whereas kids in the ARS were separated from their mothers at kidding, colostrum and artificially reared. In both systems, dams were machine-milked once a day throughout lactation and the records took place weekly. Potential milk yield was estimated according to the oxytocin method up to week 12 of lactation, and was similar for both rearing systems, although a 12.3% drop in potential milk yield at weaning was observed for MRS. During the first 6 weeks of lactation, marketable milk was lower for dams in MRS compared to those in ARS (72.1 v. 113.0 l), but similar for the rest of the experiment (101.5 v. 99.4 l, respectively). Marketable milk composition and SCC throughout the 12 weeks of lactation were unaffected by the rearing system. Artificial rearing system entailed an increment in production cost of 22.2€ per kid compared to the rearing by MRS. A similar economic return per goat and kid was obtained from ARS and MRS in this experiment, although, due to one herd's prolificacy of 1.8, the actual results would be 16.2€ per goat in favour of MRS. The real interest of this experiment may be the possibility of extrapolation to different flocks with diverse levels of milk production, prolificacy and prices and costs for incomes and outputs, to estimate the production system that increases returns. In conclusion, the results showed an increase in the cost of €22.2 per kid bred in the ARS, compared to the MRS, and a final return of 16.2€ per goat in favour of the mixed system.

Milk flow during the first minute of milking was analyzed using data from 1,132 Murciano-Granadina breed goats belonging to 17 herds. During the individual lactations, 2 test days were scheduled for recording several milk flow traits, total milk, milk composition (fat and protein percentages), and somatic cell count. Average lag time from teatcup attachment to arrival of milk at the milk claw (T0) was 4.9 s and at the milk meter (T1) was 15.8 s. Average milk flow after 30 s (MF0.5) was 0.29kg/30 s (0 to 1.1kg/30 s) and milk flow at 60 s or milking speed (MF1) was 0.67kg/min (0.1 to 2.1kg/min). Repeatabilities of T0, T1, MF0.5, and MF1 were 0.45, 0.58, 0.62, and 0.68, respectively. The MF1 showed high phenotypic correlation withT1(-0.63) and MF0.5 (0.90), medium values withT0(-0.42) and total milk (0.22), and very low values (-0.04 to -0.12) with fat, protein, and somatic cell count. We found no differences between flows during the first 3 lactations, with a reduction as the lactation number increased. Months in milk since parturition affected MF1, being highest in the first 3mo (0.67-0.71kg/min) and decreasing until the end of lactation (0.58kg/min). The effect of herd-test day was significant for all traits. Inclusion of all these effects for the analysis of milk flow traits is considered necessary.

The aim of this work was to quantify, on a half-udder basis, the changes in ewe milk yield and composition caused by unilateral subclinical mastitis within the current lactation. Fluctuations due to production level, infection severity, time from the onset of infection, and lactation curves were also studied. Yield and composition of milk from half-udders of unilateral infected ewes were compared between them and with a set of healthy halves using a mixed model. The experiment was completed with a whole-udder approach on the same animals. To test the effect of intramammary infection (IMI) in the 7 wk following the onset of infection, 20 ewes that acquired unilateral subclinical mastitis during lactation and 40 healthy ewes were used. Another group of 20 unilaterally infected ewes from wk 1 of lactation and other 40 healthy ewes were studied to test the effect of IMI on lactational milk yield and composition. The individual milk loss in ewes infected during lactation was 15% for the 7 wk following the onset of infection, and 6.6% more milk was produced by the uninfected half to compensate milk lost by the infected half. Lactational milk yield loss in ewes infected from wk 1 postpartum was 17%. The changes in milk yield were noticed from the week of infection diagnosis. The production level of animals influenced the milk yield changes caused by IMI in such a way that the more productive ewes lost more milk, although these losses were proportional to their production level. On the other hand, infection severity affected milk loss between glands, being more pronounced as somatic cell count increased. A clear decrease of lactose content and casein:protein ratio due to subclinical IMI was observed and it remained throughout the postinfection period. Improving udder health status is necessary to maintain milk production and quality in dairy ewes during lactation.

Two repeated experiments were carried out in 2 different years to study the effect of estrus on somatic cell count (SCC) in dairy goats. In the first year, 36 Murciano-Granadina goats were used [12 primiparous and 24 multiparous; 22 healthy and 14 with an intramammary infection (IMI)] and, after a 6-d pre-experimental period, were divided into 2 groups according to lactation number, udder health status, SCC, and milk production. One group was kept as a control, whereas the other received an estrus synchronization hormonal treatment lasting 11d. At 24, 48, and 72h after cessation of the hormone treatment, goats were placed in contact with a buck to confirm that they were in estrus. For 32 consecutive days (6 pre-experimental, 11 in hormone treatment, and 15 post-treatment) the SCC per gland and udder were monitored in all animals. In the second year, we repeated the same experimental design using a total of 38 Murciano-Granadina breed goats (12 primiparous and 26 multiparous; 26 healthy and 12 with IMI). Throughout this experiment, milk yield and composition were also recorded daily for each goat. Upon termination of the hormonal treatment, the SCC in udder milk increased significantly in the treatment group compared with the control group over 3 consecutive days. This increase was observed for year (1 and 2), parity (primiparous and multiparous), and udder health status (healthy and IMI). The log10 SCC (cells/mL) increased from 5.5±0.09 before estrus to 6.04±0.09 during treatment; therefore, the geometric mean of the SCC increased 3.5 times during treatment. The maximum values obtained in healthy glands of primiparous goats (geometric mean=0.37 million cells/mL) were lower than in healthy glands (1.1 million cells/mL) or infected glands (1.7 million cells/mL) of multiparous goats. The increase in SCC observed during estrus (200% increase in geometric means) could not be explained by the changes in milk production, which only fell by 13%. During estrus, the percentage of protein and dry matter in the milk also increased significantly. We concluded that it is necessary to consider the presence of estrus to correctly interpret milk SCC, as an indirect method for detecting IMI or as a commercial milk quality parameter.

Fifty-seven goats were included in an experiment designed to study the effect of lactation stage, parity number, and mammary gland health status on milk cortisol concentration as a method to assess the welfare of Murciano-Granadina goats. The relationships of milk cortisol concentration with different production parameters (milk yield, milk composition, and mechanical milking ability: milk fractioning during milking and milking time) were also studied. The experiment lasted 8 mo and monthly samplings were carried out to determine total milk yield (MY), fractioning during milking (machine milk, MM; machine stripping milk, MSM), and milking time (MT), and a sample was taken from the total milk yield to determine milk cortisol concentration, somatic cell count, and milk composition (fat, protein, and lactose). To determine the infection status of the gland, an aseptic sample was taken for bacteriological analysis before each monthly sampling. Third-parity goats presented higher concentrations of milk cortisol than those of 1, 2, or ≥ 4 parities. Intramammary infection had no effect on milk cortisol concentration, and somatic cell count did not correlate with cortisol concentration. Cortisol presented a significant correlation with MY and MM, but showed no significant correlation with MSM, MT, or milk composition parameters. Variations in milk cortisol concentration in goats may be associated with different physiological factors in the animal (e.g., milk production level, lactation stage, and parity number) and therefore need not always indicate stress for the animal.

Measurements of electrical conductivity (EC) of milk are used in mastitis detection in cows due to the low cost, possibility of automation, and rapid diagnosis, but the literature about EC measurement in goats is scarce. In this study, we studied the effect of the establishment of intramammary infection (IMI) on EC of goat milk by gland using daily measurements. Additionally, the effects on milk yield, somatic cell count (SCC), and mineral content were analyzed. Eight primiparous and 10 multiparous Murciano-Granadina goats free from IMI were included in the study. Health conditions of the participating animals were monitored for 16 d and then various unfavorable health situations that may arise on commercial farms were simulated to increase the chances of IMI. Once the IMI was confirmed, the experiment continued for another 16 d. Statistical analysis was conducted using a linear mixed model considering several periods regarding the establishment of the infection and whether it affected one or both glands in the animal. The establishment of IMI caused a significant increase of EC, SCC, and chlorides in the infected glands, whereas the sodium:potassium ratio and the ratio of EC between collateral glands showed significant increases only in bilaterally infected animals. The microorganisms that caused greater increases of EC were Staphylococcus aureus and a gram-negative bacterium. Changes due to other isolated microorganisms (coagulase-negative staphylococci and streptococci) were small. No significant differences in milk yield were determined. The significant effect of infection on EC in the affected glands suggests that the use of a system based on daily readings of EC could be useful in IMI detection of goats.

OBJECTIVE: To examine whether patients with Eating Disorders (ED) who restore menses differ from those who remain amenorrheic after treatment and to provide longitudinal data about this debated criterion of Anorexia Nervosa (AN). METHODS: 184 outpatients were recruited: 50 patients with AN Restrictive type, 75 amenorrheic Eating Disorder Not Otherwise Specified (EDNOS) patients, 24 patients who recovered from AN with persistent amenorrhea, and 35 amenorrheic patients without ED. All participants were clinically assessed by psychiatrists and gynaecologists at the beginning of treatment and at the one-year follow-up. They also completed several psychometric tests: Eating Disorder Inventory-2, Temperament and Character Inventory, and Beck Depression Inventory. RESULTS: After treatment, a large portion of the sample resumed menses but very few recovered completely from the ED. No psychopathological variables could clearly predict the recovery of menses in the different groups. However, Body Mass Index and some biological variables were associated with restoration of menses in ED patients. CONCLUSION: Resumption of menses was not associated with a less severe eating symptomatology at the beginning of treatment nor with unequivocal psychopathological changes after treatment. It is important that clinicians consider not only the presence or absence of regular menses but also that they improve both ED screening and assessment in amenorrheic patients. Amenorrhea does not seem to represent a specific predictor of severity of illness or to show prognostic value.

The aim of this work was to evaluate the effect of intramammary infection (IMI) on the endogenous proteolysis of milk. Four control checks were carried out in the half-udder milk of 10 ewes that acquired unilateral subclinical mastitis. Two of these checks were conducted before the infection was established and 2 after. Ten healthy ewes were tested as a control group. The presence of a subclinical IMI involved an increase of the products of casein hydrolysis, the proteose-peptone (p-p) fraction and minor (m) caseins, and a decrease of ß-casein. As a result, a significant increase in the proteolysis index (PI), calculated as the ratio of m-casein to the sum of caseins (α + ß + κ), took place. α-Casein and κ-casein were not significantly affected by IMI. Correlations confirmed the scenario: log(10) of somatic cell count (SCC) was positively correlated with p-p content and negatively with ß-casein, whereas log(10) SCC was not correlated with α-casein or κ-casein. On the other hand, p-p content was positively correlated with m-casein and PI and negatively with ß-casein, but no correlation was detected between p-p content and α- or κ-casein. Furthermore, between casein fractions, m-casein was only significantly correlated with ß-casein. These results suggest that use of indices of proteolysis of caseins such as p-p, m-casein, and PI, could be applied together with SCC to evaluate the cheese-making quality of milk.

The objectives were to determine the effect of administering recombinant bovine ST (bST) every 14 d on milk yield and milk composition in dairy ewes and to assess the possible effects of treatment on milk yield loss that occurs at lamb weaning. Seventy-two lactating dairy ewes were separated into 4 groups at lambing, and each group received no bST (control) or 40, 80, or 120 mg of bST every 14 d until wk 20 of lactation. During the first 5 wk of lactation, ewes suckled their lambs during the night and were milked once daily (period 1). After weaning, from 6 to 22 wk of lactation, ewes were exclusively milked twice daily (period 2). Actual milk yield, potential milk yield, and milk component percentages were recorded weekly throughout lactation. In period 1, bST-treated groups increased yields of potential milk (P = 0.04) and the corresponding 6% fat-corrected milk (FCM; P = 0.04) but not actual milk yield (P = 0.42) compared with the untreated group. In period 2, treated groups increased potential (P < 0.01) and actual (P < 0.01) milk yields as well as their corresponding 6% FCM (potential, P < 0.01; actual, P < 0.01), in comparison with the untreated group. The group treated with 120 mg of bST every 14 d yielded 39% more actual milk and 44% more 6% FCM than the control group for period 2. At weaning, between the last week of period 1 and the first week of period 2, treated groups showed similar absolute (P = 0.15) and relative (P = 0.33) values for the potential milk losses compared with the control group. Treatments increased (P = 0.04) actual milk fat percentages during period 1, but did not affect the other milk components or somatic cell count. In summary, bST increased potential milk yield throughout lactation and actual milk yield only after weaning in dairy ewes. In dairy ewes, bST was not useful for reducing the milk yield loss that occurred at weaning.

This experiment studied the effect of milking pipeline height (mid- vs. low-level milking system) on milking efficiency and milk composition. The experiment was of 8 wk duration: 2 wk preexperimental period and 6 wk experimental, in crossover design (2 x 2). Ewes were milked in a 2 x 12 milking parlor with 2 milking pipelines set at a milking vacuum of 36 kPa with a pulsation rate of 180 cycle/min and ratio of 50%. Height of the milkline had no effect on yield of milk at the time of milking, yield after stripping, milk composition, SCC, and number of teatcup fall-offs. Nor did milkline height have any effect on milk lipolysis or on the distribution of fatty acids. The level of free fatty acids was higher in evening than in morning milk (60.5 vs. 25.6 mg/L). Likewise, the increase in the degree of lipolysis between the receiver (40.4 mg/L) and the refrigeration tank (45.8 mg/L) underlines the importance of the milk delivery line design. The parameters (time and flow rate) that define the first peak in the milk emission kinetics were statistically different between lines, so care must be taken when comparing milk emission curves from both types of pipeline.

In this work on machine milking of ewes, pulsation rates of 120 and 180 cycles per min were compared, both with a pulsation ratio of 50:50 and a vacuum level of 36 kPa, comparing intramammary infection (IMI), somatic cell count (SCC) and teat end thickness changes. To this end, two groups of 20 Manchega ewes were used in a crossover experimental design with two experimental periods of 24 d for each. Bacterial exposure of all teats was increased by dipping them in a suspension of Staphylococcus simulans at four consecutive milkings of each period. Pulsation rate of 180 cycles per min, compared with 120 cycles per min, had no negative effect upon new IMI (11 and 16% of ewes infected, respectively) and SCC. No teat end lesions were observed in those animals milked with the two pulsations assayed. Also, teat thickness changes (-0.38 and -0.36 mm at 120 and 180 cycles per min, respectively) were not affected significantly. Finally, in absence of IMI, the two pulsation rates assayed did not affect the SCC.

The effects of vacuum level and overmilking on udder health were studied in ewes. Vacuum levels of 36 and 42 kPa were assigned to two groups of 23 Manchega ewes in a crossover study design with two experimental periods of 5 wk for each. Moreover, for each ewe, one teat was overmilked 1.5 to 2 min at all milkings during these 10 wk. The milking machine used had a midlevel milkline and pulsation was fixed at 180 cycles per min and a pulsation ratio of 50:50. Bacterial exposure of all teats was increased by dipping them in a suspension of Staphylococcus simulans at eight milkings of each period. New intramammary infections (IMI) were not significantly affected by the vacuum level used (18 and 23% of ewes infected, at 36 and 42 kPa, respectively) or application of overmilking (9 and 11% of half udders infected without and with overmilking, respectively). Likewise, neither factor significantly affected the somatic cell count (SCC) of the milk. Teat thickness changes after milking varied significantly due to the presence of overmilking (-13.6 and -7.4%, in teats not overmilked and overmilked, respectively) but were not affected by vacuum level. At no time were any lesions or variations visibly noted in the teat walls or orifice. So, in this work we were unable to demonstrate that the vacuum and overmilking levels assayed, both used with a pulsation rate of 180 cycles/min, have an important effect on the state of udder health in the short term. Furthermore, it was also observed that, in absence of IMI, the two factors studied did not cause irritation of any kind in the gland that might influence the SCC of the milk.

RESUMEN

OBJECTIVE: The purpose of this study is to determine the prevalence rate of estrogen and progesterone receptors and HER 2/neu in the breast cancer biopsies analyzed in the Laboratory of Immunohistochemistry of the University of Puerto Rico School of Medicine in the year 2000. This data may serve as a reference point for future studies of the epidemiological aspects of breast cancer among women living in Puerto Rico. BACKGROUND: Determination of estrogen receptor (ER) and progesterone receptor (PR) on biopsy specimens of breast carcinoma prior to treatment is standard practice in the management of breast carcinoma. ER and PR are used to identify patients who are likely to respond to endocrine therapy. The prevalence of ER, PR and Her2/neu among USA women is 77 per cent, 55 per cent and 10-34 per cent, respectively. One of the major clinical roles for testing HER 2/neu expression is to determine eligibility for treatment with Trastuzumab. METHODS: Retrospective analysis of 309 breast cancer biopsies was done. Paraffin embedded blocks of breast cancer tissue biopsies were received from different hospitals and Pathology Laboratories located throughout the island specifically for routine analysis of steroid receptor (ER/PR) and/or HER 2/neu expression. Immunostaining was performed in a Ventana Medical Systems automated instrument. RESULTS: Positive nuclear staining for ER and PR were seen in 65.9 per cent (203/308) and 51.8 per cent (159/307), respectively. In the HER2/neu test, 27.8 per cent (46/165) gave a strong and complete membranous staining (score 3+). CONCLUSIONS: There is a lower prevalence of estrogen receptor in the breast cancer biopsies of women living in Puerto Rico than their USA counterparts, but similar prevalence of progesterone receptorstatus and HER 2/neu protein over expression.

OBJECTIVE: To determine (1) the prevalence of Helicobacter pylori infection in male and female patients with reproductive disorders and controls; (2) the presence of anti-H. pylori antibodies in samples of follicular fluid, vaginal secretions and sperm; and (3) the existence of a structural homology between a major spermatozoa protein, tubulin, and H. pylori proteins. PATIENTS AND METHODS: Serum samples from 167 patients with infertility and 837 age- and gender-matched controls (blood donors) were examined by enzyme-linked immunosorbent assay (ELISA) and Western blotting to determine the seropositivity for H. pylori infection. The presence of anti-H. pylori antibodies in samples of follicular fluid, vaginal secretions and sperm was determined using the same techniques. The possible cross-reactivity with spermatozoa of anti-H. pylori hyperimmune sera and human antibodies was studied by immunofluorescence. The N-acid homology of human tubulin with the principal H. pylori proteins was assayed by the WU-blastp program available on the Internet. RESULTS: The prevalence of infection was significantly higher in patients than controls (49.1% v. 33.5%, P < 0.001). Follicular fluids from infected patients contained specific antibodies in all cases, sperm samples in about 50% of cases, and vaginal secretions in a minority of cases. Sera to H. pylori whole antigens and VacA reacted with the tails and the pericentriolar area of human spermatozoa (which are rich in tubulin); sera to urease and heat-shock protein (Hsp) did not. Follicular fluids with anti-H. pylori antibodies immune reacted with spermatozoa. A linear homology was found between beta-tubulin and three H. pylori proteins, flagellin, VacA and CagA. CONCLUSIONS: H. pylori infection may increase the risk of developing reproductive disorders or worsen the clinical expression of this syndrome.

It is well known that gemfibrozil increases the biliary output of cholesterol and phospholipids, but we have little knowledge about the impact these changes have on liver cholesterol and phospholipid biosynthetic pathways. In the present study, no changes were detected in liver lipids and CTP:phosphocholine cytidylyltransferase after gemfibrozil administration to rats. On the contrary, 3-hydroxy-3-methylglutaryl-CoA reductase mRNA (9.9-fold) and Rd activity (16.7-fold) and phosphatidate phosphohydrolase activity (1.7-fold) increased, while plasma apo B-cholesterol (40%) and triglyceride (43%) levels decreased. As a part of a compensatory homeostatic response, we report for the first time that gemfibrozil administration to rats increased the hepatic sterol regulatory element binding protein-2 (SREBP-2) mRNA (2.9-fold) and mature protein (2.2-fold) levels. An early increase in the transcriptional activity of SREBP-2 elicited by gemfibrozil administration might be responsible for the observed changes in HMG-CoA reductase, phosphatidate phosphohydrolase, and SREBP-2 expression.

Immediate transfusion reactions were characterized in recipients of 15,990 RBCs, 18,013 platelets, 409 single donor platelets, 3,451 FFP and 1,507 units of cryoprecipitate. The overall incidence of immediate reactions was 0.2%. Allergic reactions occurred in 40 patients, febrile non-hemolytic in 36 patients, bacterial contamination in 2 patients, circulatory overload in 3 patients and mechanical hemolysis in one patient. Three patients had nonspecific reactions. The incidence of immediate transfusion reactions was low when compared with similar studies. Possible causes are under-reporting transfusion reactions and the use of leukoreduced components.

The effects of atorvastatin (3 mg kg(-1)) and simvastatin (3 mg kg(-1)) on hepatic enzyme activities involved in very low density lipoprotein metabolism were studied in coconut oil/cholesterol fed rabbits. Plasma cholesterol and triglyceride levels increased 19 and 4 fold, respectively, after 7 weeks of feeding. Treatment with statins during the last 4 weeks of feeding abolished the progression of hypercholesterolaemia and reduced plasma triglyceride levels. 3-Hydroxy-3-methyl-glutaryl Coenzyme A reductase, acylcoenzyme A:cholesterol acyltransferase, phosphatidate phosphohydrolase and diacylglycerol acyltransferase activities were not affected by drug treatment. Accordingly, hepatic free cholesterol, cholesteryl ester and triglyceride content were not modified. Simvastatin treatment caused an increase (72%) in lipoprotein lipase activity without affecting hepatic lipase activity. Atorvastatin caused a reduction in hepatic phospholipid content and a compensatory increase in CTP:phosphocholine cytidylyl transferase activity. The results presented in this study suggest that, besides the inhibitory effect on 3-hydroxy-3-methyl-glutaryl Coenzyme A reductase, simvastatin and atorvastatin may have additional effects that contribute to their triglyceride-lowering ability.
