RESUMEN

A solid-phase competitive enzyme immunoassay for measurements of barbiturates in biologic tissues of human body was developed. Antibodies to barbiturate derivatives, obtained by rabbit immunization with conjugated antigen (barbamyl conjugate with ovalbumin) were used as reagents in the assay. Together with the chromatographic procedures the developed EIA may be used in practical forensic medicine and narcology.

RESUMEN

Comparative analysis of ephedrine derivative content in 96 urine specimen obtained from people suspected in abuse with these drugs. Physical and chemical methods of the analysis (gas and thin-layer chromatography) as well as ELISA were used. It was stated that the latter is specific for determination of compounds related to ephedrine by structure and it allows one to detect its analogues in 87 cases. Gas chromatography made it possible to detect ephedrine derivatives in 94 specimen tested and thin-layer chromatography--in 64 specimen tested. The given ELISA may be recommended for medicolegal expert purposes and narcology practice.

RESUMEN

Concentration of PGF2 alpha were correlated to those of antiprostaglandin antibodies IgM and IgG to this mediator in donors and myocardial infarction patients. In donors correlation between PGF2 alpha concentration and IgG antibody levels is positive. In uneventful myocardial infarction relevant correlation partially recovered at the expense of IgM antibodies. This is not true for complicated myocardial infarction.

RESUMEN

The important role played by prostaglandins in the pathogenesis of coronary disease and essential hypertension is well known. The authors have attempted to reveal a relationship between blood levels of natural autoantibodies to PGF2 alpha and PGE2 and specific features of coronary disease and essential hypertension course and complications. A total of 87 subjects were examined, 23 of these--normal controls, 25 patients with myocardial infarction, 22 with angina pectoris, and 17 with essential hypertension. Solid-phase enzyme immunoassay was employed to detect anti-PGF2 alpha and anti-PGE2 antibodies. These antibodies were found in normal subjects, coronary patients and hypertensives. Blood levels of these antibodies correlated with some complications of coronary disease and essential hypertension. These results permit a hypothesis that the pathogenetic physiologic role of the detected natural antibodies to prostaglandins consists in their defense homeostatic function.

RESUMEN

A study was made of changes in the content of natural antibodies (N-AB) to thrombin and alpha 2-macroglobulin in patients with angina pectoris under the influence of plasma perfusion. The content of N-AB being increased, plasma perfusion makes it possible to reduce their content to normal. Provided the content of N-AB is initially normal, plasma perfusion does not produce any changes in their level. In 78% of the patients, a beneficial clinical effect was attained, being more pronounced in initially high content of N-AB.

RESUMEN

Thrombin and natural antithrombin antibody content was comparatively studied in donors and myocardial infarction patients. It was shown that the level of thrombin and antithrombin antibody was stable in the group of clinically normal subjects. In patients with myocardial infarction an increased thrombin level attended by a decrease in the level of natural antithrombin antibody was recorded. The presence of linear correlation between the levels of thrombin and antithrombin antibody, as well as impossibility of the diagnosis of disorders in the blood coagulation system using one of the parameters studied have permitted introduction of an additional diagnostic parameter--"antithrombin index."

RESUMEN

Competitive ELISA method for ephedrine assay in biological fluids is developed. Anti-ephedrine antibodies obtained by rabbit immunization with conjugated antigen ephedrine-protein and ephedrine complex with horseradish peroxidase were used as reagents. ELISA in combination with chromatographic methods may be used in medicolegal and narcological practice.

RESUMEN

Simultaneous levels of thrombin and its natural antibodies were comparatively analyzed in donors and patients with myocardial infarction. Thrombin and its antibody levels were demonstrated to remain unchanged in the healthy subjects. With myocardial infarction development, there was an increase in thrombin concentrations and a decrease in natural antithrombin antibodies at the same time. A linear correlation was established between the levels of thrombin and its antibodies and failure in diagnosing coagulative disorders by one of the parameters studied was also ascertained, which made it possible to introduce a new diagnostic criterion, namely an "antithrombin index" that enabled the groups of donors and patients to be differentiated.

RESUMEN

Elevated levels of autoantibodies to types I-V collagens were detected in local scleroderma patients. In 75 percent of cases the levels of autoantibodies to types I and III collagens were elevated, in 30 percent to type II collagens, in 25 percent to type IV collagens, and in 20 percent to type V collagens. Anticollagen autoantibody levels did not differ from the normal values in the patients with a prolonged (for more than a year) remission. A reduction of these antibody levels almost to the normal values was achieved in 10-12 months after tigason therapy during a prolonged clinical remission. The ratio between autoantibodies to types I and II collagens was found to be a constant both in the donors and patients with local scleroderma.

RESUMEN

An enzyme immunoassay technique was developed for the detection of natural IgG and IgM antibodies to thrombin and alpha 2-macroglobulin in normal donor plasma and serum. The optimal conditions for the detection of IgG antibodies to alpha 2-macroglobulin are the same for the blood plasma and serum. IgG antithrombin antibodies are detectable in the plasma within a small range of the sorbed antigen concentrations, and if the serum is analyzed, this range is lowered by two orders. Natural antibody levels to the studied antigens were found to be constant in a person.

RESUMEN

Natural IgG and IgM antibodies to thrombin and alpha 2-macroglobulin were measured in donor and coronary patients' plasma by enzyme immunoassay. The patients were divided into 3 groups with various types of changes in antibody levels. Increased level of antithrombin antibodies appears to be related to additional production of antibodies in response to elevation of thrombin concentration in the blood stream or to shift of the synchronous process between free antibodies and thrombin, bound in immune complexes. Further studies will clarify the mechanism of these processes.

The ELISA test was performed on sera of normal donors and patients with myocardial infarction (MI) and essential hypertension (EH) to detect autoantibodies against prostaglandin F2 alpha (a-PGF2 alpha). The a-PGF2 alpha level was much higher after immune complexes dissociation. Baseline a-PGF2 alpha was elevated in EH patients with a sudden rise in blood pressure, and in MI patients without hypertension and unstable angina. MI patients with bradyarrhythmias also had elevated a-PGF2 alpha levels. Baseline a-PGF2 alpha was rather low in the sera of donors, chronic EH patients with hypertension and unstable angina, suggesting that a-PGF2 alpha has a physiological role to play.

It was found that 2-3 weeks after a destructive damage of the rabbit hippocamp antibodies to noradrenaline appear in the blood. Their titers reach the maximum by the 6th-10th weeks and decrease by the 20th week.

RESUMEN

Two new methods are developed for synthesis of conjugated antigens of o-aminoazotoluene-bovine serum albumin (o-AAT--BSA): (1) from the microsomal fraction of the guinea pig liver which contains the cytochrome-P-450-dependent monooxygenase enzymic system and (2) from the m-chloroperbenzoic acid. A possible mechanism of the covalent binding of o-AAT with albumin under the effect of monooxigenases and of their chemical model is considered. Differences of antigenic determinants in conjugated antigens of o-AAT--BSA synthetized by the chemical and enzymic methods are detected.
