RESUMEN

Bio-inspired conductive scaffolds composed of sodium hyaluronate containing a colloidal dispersion of water-miscible polyaniline or polypyrrole particles (concentrations of 0.108, 0.054 and 0.036% w/w) were manufactured. For this purpose, either crosslinking with N-(3-dimethylaminopropyl-N-ethylcarbodiimide hydrochloride and N-hydroxysuccinimid or a freeze-thawing process in the presence of poly(vinylalcohol) was used. The scaffolds comprised interconnected pores with prevailing porosity values of ~ 30% and pore sizes enabling the accommodation of cells. A swelling capacity of 92-97% without any sign of disintegration was typical for all samples. The elasticity modulus depended on the composition of the scaffolds, with the highest value of ~ 50 kPa obtained for the sample containing the highest content of polypyrrole particles. The scaffolds did not possess cytotoxicity and allowed cell adhesion and growth on the surface. Using the in vivo-mimicking conditions in a bioreactor, cells were also able to grow into the structure of the scaffolds. The technique of scaffold preparation used here thus overcomes the limitations of conductive polymers (e.g. poor solubility in an aqueous environment, and limited miscibility with other hydrophilic polymer matrices) and moreover leads to the preparation of cytocompatible scaffolds with potentially cell-instructive properties, which may be of advantage in the healing of damaged electro-sensitive tissues.

BACKGROUND: The aim of the study was to analyze the degree of obesity and its associations with age, gender, inflammation, an estimated glomerular filtration rate (eGFR), and liver function in type 2 diabetes mellitus (T2DM) patients. METHODS: A total of 874 consecutive adult Caucasian T2DM patients from outpatient diabetic clinic were included in the study. The relative fat mass (RFM) and body mass index (BMI) were used as obesity markers. Serum creatinine and cystatin C were used for the GFR estimation. Serum high-sensitive C-reactive protein (hsCRP) was used as the indicator of inflammation. RESULTS: The median, interquartile range (IQR) of RFM in females was higher than that in males (44.8 (42.3-47.2) % vs 31.3 (28.8-34.1) %, respectively; P < .0001). The median (IQR) of BMI in females was no higher than that in males (30 (27-34) kg/m2 vs 30 (27-34), respectively; P = .5152). The obesity prevalence was 99% in males and 98% in females according to RFM. BMI recognized obesity in 51% males and 53% females. RFM was positively associated with hsCRP in both males (rs  = .296, P < .0001) and females (rs  = .445, P < .0001). ALT was positively correlated with eGFRcys in both males (rs  = .379, P < .0001) and females (rs  = .308, P < .0001). CONCLUSION: The RFM equation leads to higher obesity prevalence compared to BMI. Women have higher RFM compared to men. The kidney function was positively correlated with ALT serum concentrations.

Post-process thermal treatment of electrospun fibers obtained from poly(ethylene oxide) (PEO) water and methanol solutions was examined. PEO fibers from methanol solution showed larger diameters as observed by scanning electron microscopy. Fibers both from water and methanol solutions exhibited a significant dimensional stability and surface cracking during the specific exposure time after thermal treatments at 40, 50, and 60 °C. Changes in crystallinity after the thermal treatment were studied by wide-angle X-ray diffraction. The kinetics of secondary crystallization were positively influenced by the as-processed level of the amorphous phase and temperature of thermal treatment. Samples treated at 60 °C were degraded by thermooxidation within the time.

Polyaniline cryogel is a new unique form of polyaniline combining intrinsic electrical conductivity and the material properties of hydrogels. It is prepared by the polymerization of aniline in frozen poly(vinyl alcohol) solutions. The biocompatibility of macroporous polyaniline cryogel was demonstrated by testing its cytotoxicity on mouse embryonic fibroblasts and via the test of embryotoxicity based on the formation of beating foci within spontaneous differentiating embryonic stem cells. Good biocompatibility was related to low contents of low-molecular-weight impurities in polyaniline cryogel, which was confirmed by liquid chromatography. The adhesion and growth of embryonic stem cells, embryoid bodies, cardiomyocytes, and neural progenitors prove that polyaniline cryogel has the potential to be used as a carrier for cells in tissue engineering or bio-sensing. The surface energy as well as the elasticity and porosity of cryogel mimic tissue properties. Polyaniline cryogel can therefore be applied in bio-sensing or regenerative medicine in general, and mainly in the tissue engineering of electrically excitable tissues.

Effects of the addition of water soluble glycinated Kraft lignin (WS/KL) on the mechanical stability and biocompatibility of hyaluronan (NaHy) hydrogels were evaluated in this work. Water soluble lignin was obtained by the modification of Kraft lignin via a Mannich reaction. It was found that WS/KL is highly compatible with hyaluronan due to its improved water solubility, which favours its use in designing new advanced composite hydrogels. The effects of the concentration of WS/KL on morphological, swelling and creep/recovery behaviours of hyaluronan hydrogels were investigated. It was detected that the creep resistance and creep recovery of NaHy hydrogels was improved by the incorporation of up to 3% (w/w) of WS/KL. In contrast, the swelling capacity of hydrogels was decreased. The cytotoxicity tests proved that glycinated KL lignin limits the viability of cells only slightly, and the final hyaluronan/lignin hydrogels were non-toxic materials.

AIM: The aim of this study is to compare markers of glomerular filtration rate (GFR), estimated GFR (eGFR), and metabolic parameters between admission and recovery in 13 patients of Tomas Bata hospital with methanol poisoning during methanol problems in the Czech Republic in 2012. The impact of methanol concentration and age on metabolic parameters were discovered at the time of admission to hospital. MATERIALS AND METHODS: The serum osmolality, methanol, ethanol, creatinine, cystatin C, Troponin I, ALT, plasma pH and lactate were measured in these 13 patients. The eGFR from serum creatinine (creatnine eGFR) and from cystatin C (cystatin C eGFR) were also determined. RESULTS: Increased serum osmolality and markers of metabolic acidosis are key indirect laboratory findings in patients with methanol poisoning. There were no significant changes in eGFR in our patients between admission and recovery. Increased serum troponin I concentration was confirmed as an indicator of myocardial necrosis in four patients. Two patients developed acute kidney injury (AKI) before admission. CONCLUSIONS: We found statistically significant differences in serum osmolality concentration, plasma pH and lactate between admission and recovery. We found no changes in eGFR between admission and recovery. One patient had vision problems due to damage to the occipital lobes. Methanol poisoning may cause increase in markers of cardiac damage.

RESUMEN

Two types of ionic liquids, 1-ethyl-3-methylimidazolim acetate and 1-ethyl-3-methylimidazolium lactate, were employed for the direct processing of pine wood into microfibers. The concentration of 5 wt.% of wood in ionic liquids was rated as the most appropriate for electrospinning. The fibers were electrospun into the collector water bath. The final structure varied from individual microfibers to fiber bundles. It was demonstrated that 1-ethyl-3-methylimidazolium lactate is a powerful solvent and provides the direct transformation of pristine pine wood into the non-wovens.
