RESUMEN

Objective To study the effects of Lactobacillus casei ATCC334 on the secretion of inflammatory factors and joint destruction in collagen-induced arthritis (CIA) rats and explore the possible mechanism. Methods Female Wistar rats were randomly divided into normal control group, CIA group and ATCC334 group with 7 rats each. The CIA model was established by intradermal injection of bovine type 2 collagen into the tail root of rats in the CIA group and ATCC334 group. The rats in the control group and CIA group were intragastrically perfused with 500 µL normal saline every day, and simultaneously the rats in the ATCC334 group were intragastrically perfused with Lactobacillus casei ATCC334 solution [2×108 colony forming unit (CFU)/mL] for continuous 28 days. Arthritis index and increased hind paw volume were used to evaluate the severity of arthritis. HE staining was used to observe the joint lesions. Levels of tumor necrosis factor α (TNF-α), interleukin-6 (IL-6) and IL-17 in the serum of rats were detected by ELISA. Results Compared with the CIA group, the indices of arthritis index, increased posterior paw volume, joint lesions and TNF-α, IL-6 and IL-17 levels were significantly reduced in the serum of the ATCC334 group. Conclusion Lactobacillus casei ATCC334 can significantly reduce the secretion of inflammatory factors and joint bone damage in CIA rats.

@#Objective: To investigate the expression of lncRNA LINC00886 in human esophageal squamous cell carcinoma (ESCC) tissues and cell lines, and its effects on proliferation, migration and invasion of Eca109 cells. Methods: The cancer tissues and corresponding para-cancerous tissues of 69 ESCC patients were collected in the biological specimen bank of the Fourth Hospital of Hebei Medical University from June 2014 to December 2016; the ESCC cell lines Eca109, TE13, TE1, Kyse150, Yes-2 and Kyse170 were also collected. LINC00886 gene expression in ESCC tissues and cell lines was detected by qPCR. Eca109 cells were transfected with pIRES2-LINC00886 and pIRES2-NC, respectively, and the overexpression efficiency of LINC00886 gene in Eca109 cells was detected by qPCR; MTS, clone formation assay, wound-healing assay and Transwell invasion assay were respectively used to detect the effect of LINC00886 over-expression on proliferation, migration and invasion ability of Eca109 cells. Results: The expression of LINC00886 gene in ESCC tissues was significantly lower than that in para-cancerous tissues (P<0.01), and its expression level was associated with tumor TNM stage and lymph node metastasis (both P<0.05). The expression level of LINC00886 gene in ESCC cell lines was also lower than that of the control group (all P<0.01). Compared with control group, the expression level of LINC00886 gene was significantly higher in Eca109 cells transfected with pIRES2-LINC00886 (both P<0.05). Compared with the control group, LINC00886 overexpression significantly inhibited the proliferation, migration and invasion abilities of Eca109 cells (all P<0.01). Conclusion: The decreased expression of LINC00886 gene may be related to the occurrence and development of ESCC. Over-expression of LINC00886 gene inhibits the proliferation, migration and invasion abilities of ESCC cells.

@# Objective: To investigate the expression of long non-coding RNA NUP50-AS1 (lncRNA NUP50-AS1) in esophageal squamous cell carcinomas (ESCC) tissues and cell lines, and to explore its effect on proliferation, migration and invasion of human esophageal cancer Eca109 cells. Methods: 49 pairs of ESCC tissues and corresponding para-cancerous tissues obtained from the Biological Specimen Base of the Fourth Hospital of Hebei Medical University during Jan. 2015 to Jan. 2016 were used in this study. qRT-PCR method was applied to detect the expression of NUP50-AS1 in collected tissues samples and five esophageal cancer cell lines (TE1, TE13, Eca109, Kyse150 and Kyse170). ShRNAs were transiently transfected into Eca109 cells to interfere the expression of NUP50AS1 gene, and finally, sh2-NUP50-AS1 was used for the following experiments. The effect of NUP50-AS1 gene knockdown on the proliferation of Eca109 cells was detected by MTS and colony formation assay; the effect of NUP50-AS1 gene knockdown on the migration of Eca109 cells was detected by scratch test, and the effect on cell invasion was detected by Transwell assay. Results: The expression of NUP50-AS1 in ESCC was correlated with the lymphnode metastasis and TNM stage (all P<0.01). The expression of NUP50AS1 in ESCC tissues was significantly higher than that in corresponding normal tissues (2.003±0.870 vs 1.000±0.000, P<0.05). The expression of NUP50-AS1 in five esophageal cancer cell lines was significantly up-regulated (P<0.05), and it had the highest expression in Eca109 cell line. After transfection, sh2-NUP50-AS1 had the highest transfection efficiency, and knocking down NUP50-AS1 gene significantly inhibited the proliferation, invasion and migration of the Eca109 cells. Conclusion: The expression of lncRNA NUP50AS1 in ESCC tissues was significantly higher than that in the para-cancerous tissues, and correlated with the TNM stage and lymphnode metastasis. The down-regulation of NUP50-AS1 inhibited the proliferation, invasion and migration of esophageal cancer cells. The high expression of NUP50-AS1 gene may be closely related to the occurrence and development of ESCC.

PURPOSE: KLF17 belongs to the Sp/KLF zinc-finger protein family as a regulator in tumor development. However, its expression and biologic function has remained unclear in EC. METHODS: The esophageal carcinoma tissue samples and adjacent normal tissues were obtained from the Second Hospital of Hebei Medical University. Immunohistochemistry, Western blot, and transfection were applied to evaluate the expression and clinical significance of KLF17 in esophageal cancer. RESULTS: In this study, we showed that KLF17 was overexpressed in esophageal normal samples compared to the cancer. Moreover, KLF17 was upregulated at lymph node non-metastatic cancer tissues when compared to metastatic cancer tissues. KLF17 overexpression decreased EC cell proliferation, migration and invasion ability. In contrast, the knockdown of KLF17 increased EC cell proliferation, migration and invasion ability. CONCLUSION: These results suggest that KLF17 inhibits tumor development and may serve as a potential therapeutic target.

RESUMEN

The Classical Least Square regression (CLS) is one of the most popular regression methods in FTIR quantitative estimation. However, CLS is the best unbiased estimator only under the assumption that error (noise) in the spectrum has equal variance, which usually is not the case in FTIR. This paper proposed a noise calibration method for FTIR spectrum analysis. Based on measured variance of noise in the FTIR spectrum by computer, the Weighted Least Square regression (WLS) method is used in quantitative estimation. The experiment results showed that the WLS performs much better than CLS in quantitative estimation of VOCs pollution.

Femtosecond optical frequency comb, with large spectral range, narrow pulse width, high stability of frequency and many other remarkable properties, has a significant impact on optical frequency metrology, absolute distance measurement and high-precision spectroscopy. The time-domain and frequency-domain properties of femtosecond optical frequency comb is traced back to the microwave frequency standard, making it possible to open up the door to high-precision gas absorption spectrum detection. Femtosecond optical frequency comb spectroscopy has some excellent performances, such as fast measurement, high sensitivity, high resolution, high signal-to-noise ratio and so on. Therefore, more investments on femtosecond optical frequency comb spectroscopy will better make contribution to the environmental protection, industrial production, biological medicine, scientific research and other social fields. High-precision gas absorption spectroscopy with femtosecond optical frequency comb mainly includes frequency comb based cavity ring-down spectroscopy, cavity-enhanced frequency comb spectroscopy and dual-comb multi-heterodyne spectroscopy. Among them, according to the data collection, cavity-enhanced frequency comb spectroscopy can be divided into comb vernier method, virtually imaged phased array method and Fourier transform method. At present, related research has been widely carried out abroad, and domestic research is still in its infancy. This review summarizes main techniques in the high-precision gas absorption spectrum detection based on optical frequency comb, and demonstrates typical experimental schemes for different methods. It also analyzes the advantages and disadvantages of each method, and tracks frontier achievements of main research groups.

This paper presents a method for measuring five-degrees-of-freedom errors of a moving stage with a monolithic prism and phase-sensitive detection technique. It consists of a pigtailed laser diode, three position-sensitive detectors (PSDs), a monolithic prism, and additional optical and electronic components. The monolithic prism mounted on the moving stage generates three beams that are detected by three PSDs, respectively, so that the straightness, pitch, yaw, and roll errors can be simultaneously measured. Theoretical analysis of each error measurement process is presented. To reduce the influence of disturbing light, the laser diode is modulated by a sinusoidal wave current, and a phase-sensitive detection technique is developed to demodulate the signals. Compared with a laser interferometer, the deviation errors when measuring the horizontal and vertical straightness errors are better than ±0.25 and ±0.4 µm, respectively. The deviation errors for the pitch, yaw, and roll are better than ±0.5, ±0.3, and ±2 arc sec, respectively, in comparison with an autocollimator. The system can be assembled to measure five error components of machine tools in an industrial environment.

In view of the complexity of the conventional control of scanning mirror based on a digital-analog hybrid design, this paper focuses on a concise and efficient control method around the completely digital signal. The quadrate-encoder pulses A and B generated by the scanning mirror were sent into CPLD pins where an algorithm on position control and velocity control had been built. As a result, the CPLD will generate two PWM signals. These two signals are regenerated by a digital power chip to drive the voice of motor bundled with the moving mirror. Experiments validated that the stability of velocity of moving mirror is better than 97.2% and the method can improve the SNR of the measured spectrum and guarantee the accuracy of spectral qualitative and quantitative analyses.

We present a frequency-sweeping heterodyne interferometer to measure an absolute distance based on a frequency-tunable diode laser calibrated by an optical frequency comb (OFC) and an interferometric phase measurement system. The laser frequency-sweeping process is calibrated by the OFC within a range of 200 GHz and an accuracy of 1.3 kHz, which brings about a precise temporal synthetic wavelength of 1.499 mm. The interferometric phase measurement system consisting of the analog signal processing circuit and the digital phase meter achieves a phase difference resolution better than 0.1 deg. As the laser frequency is sweeping, the absolute distance can be determined by measuring the phase difference variation of the interference signals. In the laboratory condition, our experimental scheme realizes micrometer accuracy over meter distance.

A method of principal component interactive validation in multi-region is presented. Based on the characteristic peaks of target gas the full spectral region is divided into multiple characteristic regions. In succession the principal component analysis around the pure components spectral matrix in divided characteristic regions is applied. Thus the potential components included in target gas are identified according to the main components number according to 99% larger cumulated contribution and the descending sorting of maximum value of pure component spectrums. After that, the characteristic regions of potential components are compared with the ones of target gas one by one. In this way the real components of target gas are confirmed. Because the principal component analysis was applied in multiple characteristic regions, this method can avoid omitted identification. Owning to the matching validation between the characteristic absorption regions of target gas and the ones of pure component gas this method can avoid additional identification. The simulated and the measured spectra both validate the qualitative ability of this method.

Field of view (FOV) is an important contributor for instrumental line shape (ILS) function of high-resolution Fourier transform infrared spectrometer (FTS) only next to maximum optical difference. For the reason of optical design and layout, commonly, the measured FOV located in detector is not regularly rounded as original one. There exits more or less difference between the measured vertical FOV and measured horizontal one. In view of this case, the present paper replaces the generally circular area light source with an elliptical one, which is probably more suitable for actual FTS. At the same time, the factor maximum optical difference was considered. After these, the mathematic and graphic description about ILS function with unequal field of view (UFOV) was given Finally, comparison between measured spectrum with standard monoxide gas and calculated one with equal field of view (EFOV) and UFOV respectively was taken Experimental results show that the spectral residual from UFOV is less fluctuant than those from EFOV and its root mean square value is the smallest. All these indicate that the ILS function from UFOV is more accurate and suited to reflecting the response of high-resolution FTS than the one from EFOV.
