Leptospirosis is a zoonosis of great importance for One Health. In this context, the Amazonian biome may harbor numerous hosts for Leptospira spp. that contribute to the maintenance of the pathogen in the environment. Some reptiles, such as chelonians, have been little studied in terms of their involvement with leptospires. The objective of this study was to detect Leptospira spp. DNA in Kinosternon scorpioides turtles kept in captivity in a region of the Brazilian Amazon. A total of 147 samples of blood (n = 40), cloacal fluid (n = 27), cloacal lavage (n = 40), and stomach (n = 40) were collected from 40 chelonians. After DNA extraction, the samples were subjected to amplification of a 331 base pair product of the 16S rRNA gene using the Lep1 and Lep2 primers. PCR products were Sanger sequenced, assembled, and subjected to online blast search and phylogenetic analysis. Of the animals tested, 40% (16/40, 95% confidence interval [CI]: 25-55) had at least one or two samples positive for Leptospira spp. Considering the total number of samples collected, 12.93% (19/147) were positive, being blood clots (27.5%; 11/40), followed by cloacal washings (10%; 4/40), cloacal fluid (11.11%; 3/27) and gastric washings (2.5%; 1/40). Of these, 11 samples were sequenced and showed 99% to 100% identity with Leptospira interrogans sequences, which was confirmed by phylogenetic analysis. This is the first study to detect pathogenic Leptospira DNA in chelonians in a region of the Brazilian Amazon. It has been concluded that K. scorpioides turtles in captivity have been exposed to pathogenic Leptospira.

In Brazil, there have been few leptospires isolated from cattle, especially in the Amazon, implying that the epidemiology of the disease in this region is still largely unclear. In a previous study, 52 Leptospira isolates were obtained from urine of cattle raised in the Brazilian Amazon and, to achieve a greater understanding of Leptospira infection in cattle of this region, the present study aimed to serologically and molecularly characterizes all these isolates. The laboratory assays used were the microscopic agglutination test (MAT) adopting a panel of polyclonal antisera against Leptospira spp. for serogrouping the isolates, DNA sequencing (secY) and multiple locus variable number tandem repeat analysis (MLVA). The isolates belonged to five species: 20/52 were identified as L. borgpetersenii (38.5 %); 18/52 as L. kirschneri (34.6 %); 9/52 as L. santarosai (17.3 %); 3/52 as L. noguchii (5.8 %) and 2/52 as L. interrogans (3.8 %). With serogrouping, nine different serogroups were detected, with a high frequency of the Sejroe serogroup. MLVA showed that all L. borgpetersenii isolates had a profile compatible with serovar Hardjo; moreover, the other isolates demonstrated a diversity of patterns, and some of them may represent strains not yet characterized. In the Brazilian Amazon, the leptospires circulating in cattle revealed the unique aspects of infections in this area which, in addition to a variety of strains, were characterized by a high frequency of the Sejroe serogroup, highlighting the serovar Hardjo, which has not been reported in other regions of Brazil.

We evaluated the prevalence of anti-Toxoplasma gondii antibodies in the serum samples collected from domestic cats in Belém, Pará, Brazil. We also correlated the presence of T. gondii antibodies with environmental variables and cat-owner habits. Four-hundred and forty-seven serum samples from domestic cats were analyzed. The sera were tested using an indirect immunofluorescence assay. Among the animals analyzed, 21.92% (98/447) were seropositive. A statistically significant association was found in relation to age and serology among the animals over 1 year old (p<0.01): in the group up to 1 year old, 12.82% (20/156) of the animals were positive, and in the group over 1 year old, 26.80% (78/291) were positive. Our results show that the cats in Belém, Pará region have anti-T. gondii antibodies, and their owners are not aware of toxoplasmosis or how to prevent its transmission.

We describe anti-Leptospira spp. agglutinin in yellow-spotted river turtles (Podocnemis unifilis)in the Amazon region. Ninety-eight serum samples from individuals housed at the Bosque Rodrigues Alves Zoobotanical Garden of Amazonia, Belém, PA, Brazil, were subject to the microscopic agglutination test (MAT) using 19 different Leptospira spp. antigen serogroups. Thirty-four of the 98 samples (35%) were reactive, with titers ranging from 100 to 3200, and eight 8 reactive samples (23.5%) co-agglutinated under two or more serovars.The most common serogroup was Hebdomadis (26.9%, 7/26), followed by Semaranga (23%, 6/26), Shermani (19.2%, 5/26), Djasiman (11.5%, 3/26), and Australis (7.7%, 2/26); Bataviae, Javanica, and Sejroewere represented by a single sample each (3.9%). The presence of turtles reactive to anti-Leptospira spp. antibodies implies exposure to the pathogen.(AU)
Este trabalho descreve a detecção de aglutininas anti-Leptospira spp. em tracajás (Podocnemis unifilis) na região Amazônica. Obtivemos 98 amostras de soro, as quais foram submetidas ao teste de aglutinação microscópica (MAT), utilizando 31 antígenos de diferentes sorogrupos de Leptospira spp. Das 98 amostras de soro coletadas, 35% (34/98) reagiram apresentando titulações de 100 a 3200; 23,53% dessas amostras (8/34) coaglutinaram na presença de dois ou mais sorovares. O sorogrupo mais comum foi Hebdomadis com 26,9% (7/26), seguido de Semaranga com 23% (6/26), Shermani com 19,2% (5/26), Djasiman com 11,5% (3/26), e Australis com 7,7% (2/26). Bataviae, Javanica, e Sejroe foram representados com apenas uma amostra (3,9%). A presença de tracajás reagentes a anticorpos contra Leptospira spp. implica na exposição ao patógeno.(AU)

We analyzed the presence of Leptospira spp. in liver and kidney tissue of wild marsupials and rodents trapped in a periurban forest in the eastern Brazilian Amazon. We examined 25 individuals of four marsupial and seven rodent species for the presence of the 16S rRNA gene of Leptospira in the DNA extracted from 47 liver and kidney tissue samples using PCR. We detected positive samples in 12% (3/25) of the individuals, in kidney fragments of two marsupial species (Didelphis marsupialis and Marmosops pinheiroi) and in a liver fragment of one rodent species (Echimys chrysurus). These are the first records of Leptospira spp. in M. pinheiroi and E. chrysurus and it is the first molecular survey of marsupials and rodents in the Brazilian Amazon.(AU)
Analisamos a presença de Leptospira spp. em tecido hepático e renal de marsupiais e roedores silvestres capturados em uma floresta periurbana no leste da Amazônia brasileira. Foram examinados 25 indivíduos de quatro espécies de marsupial e sete de roedor quanto à presença do gene 16S rRNA de Leptospira no DNA extraído de 47 amostras de tecido hepático e renal usando PCR. Detectamos amostras positivas em 12% (3/25) dos indivíduos, em fragmentos renais de duas espécies de marsupial (Didelphis marsupialis e Marmosops pinheiroi) e em um fragmento hepático de uma espécie de roedor (Echimys chrysurus). Estes são os primeiros registros de Leptospira spp. em M. pinheiroi e E. chrysurus, e é o primeiro levantamento molecular de marsupiais e roedores na Amazônia brasileira.(AU)

Leptospirosis is a zoonosis transmitted by contact with infected urine or water contaminated with the agent. Searches for Leptospira spp. in reptiles are scarce although most species have contact with aquatic environments. We evaluated the presence of anti-Leptospira spp. antibodies in Podocnemis expansa housed at the Amazonian Zoobotanical Garden, in Belém, Pará state, Brazil. We analyzed 74 serum samples through the microscopic agglutination test using 31 live antigens from different Leptospira spp. serogroups. Thirty samples (40.5%) were positive against Leptospira spp., with titrations between 100 and 3,200 for one or more serogroups. The Hebdomadis serogroup was the most prevalent, with 26 (87%) out of the 30 positive samples, followed by Djasiman, with two (7%) and Celledoni and Bataviae with one (3%) sample each. The detection of anti-Leptospira spp. agglutinins in P. expansa suggests that the aquatic environment is a transmission route for this pathogen among chelonians.(AU)

Estudos de Leptospira spp. em répteis são escassos, apesar da maioria das espécies terem contato com ambientes aquáticos. Avaliamos a presença de anticorpos anti-Leptospira spp. em Podocnemis expansa mantidas no Jardim Zoobotânico da Amazônia, em Belém, Pará, Brasil. Analisamos 74 amostras de soro por meio do teste de aglutinação microscópica usando 31 antígenos vivos de diferentes sorogrupos de Leptospira spp. Trinta amostras (40,5%) foram reagentes contra Leptospira spp., com titulações entre 100 e 3.200 para um ou mais sorogrupos. O sorogrupo Hebdomadis foi o mais prevalente, com 26 (87%) das 30 amostras positivas, seguido por Djasiman, com duas (7%) e Celledoni e Bataviae com uma (3%) amostra cada. A detecção de aglutininas anti-Leptospira spp. em P. expansa sugere que o ambiente aquático é uma via de transmissão para esse patógeno entre quelônios.(AU)

Utilizou-se a reação de imunofluorescência indireta (RIFI), com lâminas sensibilizadas de acordo com os seus respectivos antígenos. Das 263 amostras, 100 foram reagentes, com 38,0% de positividade para Toxoplasma gondii, sendo os títulos prevalentes 16UI e 64UI, 15,97% de sororreagentes para anticorpos anti-N. caninum com maior soroprevalência para os títulos 25UI e 50UI. Na pesquisa de anticorpos anti-L. major, 3,42% foram sororreagentes com o título 40UI, sendo o mais prevalente. A pesquisa de anticorpos anti-T. cruzi revelou negatividade em todas as amostras. As variáveis procedência e faixa etária mostraram ser um fator de risco para os animais. Os animais de Belém apresentaram 2,6 vezes mais chances de infecção do que os animais de Castanhal. Os animais na faixa etária de três a sete anos apresentaram 2,27 vezes mais chances de serem sororreagentes do que os cães com até três anos de idade. A serological inquiry was performed to Toxoplasma gondii, Neospora caninum, Leishmania spp. and Trypanosoma cruzi in dogs from the city of Castanhal and Belém, State of Pará, and the association between seropositive animals and factors such as sex, age, access to the streets and provenance of the animals was studied. 269 healthy dogs with different ages, coming from animal shelters, commercial kennels, Zoonosis Control Centre, and domiciles were used. The Indirect immunofluorescence (IIF) reaction was used with plates sensitized with their respective antigens. Among the 263 samples, 100 were reactant, with 38.0% of positivity for Toxplasma gondii, being 16UI and 64UI the prevalent titers, 15.97% of reagent animals for anti-N. caninum antibodies with higher seroprevalence to titers 25UI and 50UI.At the anti-L. major antibodies research, 3.42% were reagent with the titer 40UI being the prevalent. The research of anti-T.cruzi antibodies revealed negativity in all the samples. The provenance and age range variables have been shown as a risk factor to the animals. The animals from the city of Belém presented 2.6 times more changes of infection than the animals from the city of Castanhal. The animals at the age range from three to seven years old presented 2.27 times more changes of being reagent than the dogs with up to three years old. INTRODUCTION: Serological surveys are important to assess the health status of wild animals. In this study, antibodies against Leptospira spp, causal agents of leptospirosis, were detected in free-living marsupials in the State of Pará, Brazil. METHODS: Nineteen blood samples collected from marsupials in the municipalities of Peixe-Boi, Viseu, and Castanhal were subjected to microscopic agglutination tests. RESULTS: In total, 36.8% (7/19) of samples were positive, and two exhibited co-agglutination. The most frequent serovars were Icterohaemorrhagiae (60%; 3/5), Panama (20%; 1/5), and Nupezo (20%; 1/5). CONCLUSIONS: Anti-Leptospira spp antibodies currently circulate in free-living marsupials in Northeastern Pará.

RESUMO: Embora estudos sorológicos para leptospirose em iguanas já tenham sido realizados, não a relatos na região amazônica. Portanto, o presente trabalho teve como objetivo pesquisar a presença de anticorpos anti-Leptospira sp. em Iguanas-verdes (Iguana iguana) de vida livre do Parque Mangal das Garças, região metropolitana de Belém, PA, e Norte do Brasil. Foram coletadas 29 amostras de sangue de Iguana-verde (I. iguana) que frequentaram o Parque Mangal das Garças. Para o diagnóstico sorológico da infecção por Leptospira sp. foi empregado o teste de aglutinação microscópica (MAT) com 25 antígenos vivos de Leptospira sp. como antígenos. Observou-se que todas as amostras foram não reagentes, o que sugere não exposição ao agente.

Portanto, o presente trabalho teve como objetivo pesquisar a presença de anticorpos anti-Leptospira sp. em Iguanas-verdes (Iguana iguana) de vida livre do Parque Mangal das Garças, região metropolitana de Belém, PA, e Norte do Brasil. Foram coletadas 29 amostras de sangue de Iguana-verde (I. iguana) que frequentaram o Parque Mangal das Garças. Para o diagnóstico sorológico da infecção por Leptospira sp. foi empregado o teste de aglutinação microscópica (MAT) com 25 antígenos vivos de Leptospira sp. como antígenos. Observou-se que todas as amostras foram não reagentes, o que sugere não exposição ao agente.
RESUMEN
Although, serological studies for leptospirosis in iguanas have already been performed, there is no report in the Amazon region. Therefore, the present study aimed to investigate the presence of anti-Leptospira sp. in free living Iguanas-verdes (Iguana iguana) of Mangal das Garças Park, metropolitan region of Belém, Pará, Northern region of Brazil. Twenty-nine blood samples of Iguana-verde (I. iguana) were collected from Mangal das Garças Park. For the serological diagnosis of Leptospira sp. it was employed microscopic agglutination test (MAT) with 25 live antigens of Leptospira sp. as antigens. It was observed that all samples were non-reactive, suggesting no exposure to the agent.(AU)

Brucellosis and leptospirosis are widely spread bacterial infections and dogs are the most important source of infection and reservoir for diseases. Dogs can disseminate the agents in the environment and transmit them to humans and/or other animals. The objective of this study was assess the occurrence of reactive to antibodies anti-Leptospira spp., Brucella canis and B. abortus in Belém and Castanhal, State of Pará, Amazon, Brazil. A total of 156 samples were randomly collected in the city of Belém and 158 samples in Castanhal. The anti-B. canis antibodies research was performed by Agar Gel Immunodiffusion (AGID) with and without 2-mercaptoethanol serum treatment (AGID-2ME). To assess the anti-B. abortus antibodies, the technique of Fast Seroagglutination with buffered acidified plate antigen (BAPAT) was used. For anti-Leptospira spp. antibodies research, the Microscopic Agglutination Technique (MAT) was used. No animal reacted to Brucella abortus and one animal was reactive to B. canis at the AGID, but it was negative to the AGID-2ME test. Seventeen percent of dogs (47/274) presented anti-Leptospira spp. antibodies, with prevalence of serovar Canicola. The dogs from Belém and Castanhal are not source of infection for B. abortus and B. canis, however, they are reservoirs for different serovars of Leptospira spp.

Em dois períodos distintos, foram colhidas amostras sanguíneas de 52 animais, de ambos os sexos e diferentes idades (2 a 17 anos), totalizando 104amostras. Para a pesquisa de anticorpo anti-Brucella lisa foi utilizado o teste de soroaglutinação rápidaem placa. Na primeira colheita, nenhum animal foi reagente, entretanto na segunda colheita houve trêsanimais sororeagentes. A pesquisa de anticorpos anti-Leptospira spp. foi efetuada com o emprego datécnica de soroaglutinação microscópica (SAM), na primeira colheita houve 23,07% animais reagentese 15,38% na segunda colheita, para um ou mais sorovares de Leptospira spp. com títulos variando de100 a 200. O sorovar predominante na primeira e segunda colheita foi o Autumnalis com 40% e 37,5% respectivamente. De acordo com a idade, observou-se no grupo 1 (2 a 7 anos) 27,78% e 13,89% nas duas colheitas respectivamente e no grupo 2 (> 7 anos) encontrou-se 12,50% e 18,75% de sororeagentes. Os resultados observados no presente estudo demonstraram que na ilha de Maiandeua, estado doPará, existe a presença de infecção por Leptospira spp, com o sorovar Autumnalis, o mais frequente, e possível exposição dos animais a Brucela lisa, sugerindo risco baixo de infecção na população de equinos examinada.(AU)

In two distinct periods, blood samples were collectedfrom 52 animals of both sexes and different ages (2 to 17 years), totaling 104 samples. For the research of antibodies anti-Brucella abortus were used in rapid agglutination test in plate. In the first harvest, none animal was positive, however in the second harvest there were three animals reactive serum. The research of antibodies anti-Leptospira spp. was performed with the use of the microscopic agglutination test (MAT), the first harvest was 23.07% reacting animals and 15.38% at the second harvest, for one or more Leptospira spp. with titers ranging from 100 to 200. The predominant serotype at first and second harvest was the Autumnalis 40% and 37.5% respectively. According to age, it was observed in group1 (2 to 7 years) 27.78% and 13.89% respectively in the two samples and the second group (> 7 years) was found 12.50% and 18 75% of reactive serum. The results observed in this study demonstratedthat the island of Maiandeua, state of Pará, there is the presence of Leptospira spp, with the most frequent serovar autumnalis and possible exposure of animals to brucella smooth, suggesting low risk of infection in the population of horses examined.(AU)
