RESUMEN

Replacing lung cancer as the most commonly diagnosed cancer globally, breast cancer (BC) today accounts for 1 in 8 cancer diagnoses and a total of 2.3 million new cases in both sexes combined. An estimated 685,000 women died from BC in 2020, corresponding to 16% or 1 in every 6 cancer deaths in women. BC represents a quarter of a total of cancer cases in females and by far the most commonly diagnosed cancer in women in 2020. However, when detected in the early stages of the disease, treatment methods have proven to be very effective in increasing life expectancy and, in many cases, patients fully recover. Several medical imaging modalities, such as X-rays Mammography (MG), Ultrasound (US), Computer Tomography (CT), Magnetic Resonance Imaging (MRI), and Digital Tomosynthesis (DT) have been explored to support radiologists/physicians in clinical decision-making workflows for the detection and diagnosis of BC. In this work, we propose a novel Faster R-CNN-based framework to automate the detection of BC pathological Lesions in MRI. As a main contribution, we have developed and experimentally (statistically) validated an innovative method improving the "breast MRI preprocessing phase" to select the patient's slices (images) and associated bounding boxes representing pathological lesions. In this way, it is possible to create a more robust training (benchmarking) dataset to feed Deep Learning (DL) models, reducing the computation time and the dimension of the dataset, and more importantly, to identify with high accuracy the specific regions (bounding boxes) for each of the patient's images, in which a possible pathological lesion (tumor) has been identified. As a result, in an experimental setting using a fully annotated dataset (released to the public domain) comprising a total of 922 MRI-based BC patient cases, we have achieved, as the most accurate trained model, an accuracy rate of 97.83%, and subsequently, applying a ten-fold cross-validation method, a mean accuracy on the trained models of 94.46% and an associated standard deviation of 2.43%.

The comparison of closely related taxa is cornerstone in biology, as understanding mechanisms leading up to differentiation in relation to extant shared characters are powerful tools in interpreting the evolutionary process. Hotspots of biodiversity such as the west-Mediterranean, where many lineages meet are ideal grounds to study these processes. We set to explore the interesting example of Sooty Copper butterflies: widespread Eurasian Lycaena tityrus (Poda, 1761) comes into contact in Iberia with closely related and local endemic, L. bleusei (Oberthür, 1884), which hasn't always been considered a distinct species. An integrative analysis was designed, combining the use of extensive molecular data (five genes), geometric morphometrics analyses, verified and up-to-date distribution data, and environmental niche modelling, aimed at deciphering their true relationship, their placement within European Lycaena and trace their evolutionary history. We revealed several levels of differentiation: L. bleusei and L. tityrus appear to be reciprocally monophyletic independent gene-pools, distinct in all genes analysed, having mutually diverged 4.8 Ma ago. L. tityrus but not L. bleusei, further displays a genetic structure compatible with several glacial refugia, where populations assignable to infraspecific taxa surface. Conversely, L. bleusei shows a loss in mtDNA diversity in relation to nuDNA. Morphological analyses differentiate both species according to size and shape but also discriminate strong seasonal and sexual traits and a geographical phenotype segregation in L. tityrus. Finally, updated distribution and its modelling for current and glacial timeframes reveal both species respond differently to environmental variables, defining a mostly parapatric distribution and an overlapping belt where sympatry was recovered. During the last glacial maximum, a wider expansion in L. bleusei distribution explains current isolated populations. Our study highlights the importance of gathering several lines of evidence when deciphering the relationships between closely related populations in the fringe of cryptic species realm.

Introduction: Oxygen consumption (VO2 ) is indicative of cardiorespiratory fitness (CRF) and lower levels are related to a higher risk of total mortality among individuals with cancer whose therapy can have adverse consequences on the cardiovascular system. Objective: To examine the associations of patient-reported sociodemographic, clinical, anthropometric outcomes and functional variables with CRF in 69 women (55±10 years) and to identify whether walking is a predictor of peak VO2 variation in this population with breast cancer (BC). Method: Female BC survivors receiving hormone therapy after two types of surgery (breast-conserving and mastectomy) underwent a CRF test on a cycle ergometer to measure peak VO2 . A questionnaire containing sociodemographic, clinical data, patient reported outcomes (PROs) (depressive symptoms, sleep quality, fatigue, body image) and self-reported walking and tests to measure body fat percentage, waist circumference, flexibility and shoulder range of motion (RoM) were performed. Results: Unemployment and retirement were associated with low CRF, as was the use of aromatase inhibitors instead of tamoxifen. Depressive symptoms, worse body image, greater waist circumference, less flexibility and shoulder RoM were also associated with low CRF. Walking duration, controlled for age and body mass index (BMI), is a 13% predictor of peak VO2 variance in this sample. Conclusion: These factors must be considered in understanding the CRF profile of BC survivors. As walking was a predictor of peak VO2 variance, it should be recommended as a type of physical activity for patients with BC using hormone therapy.


Introdução: O consumo de oxigênio (VO2 ) é indicativo de aptidão cardiorrespiratória (ACR), e níveis mais baixos estão relacionados a um maior risco de mortalidade total entre indivíduos com câncer cuja terapia pode ter consequências adversas no sistema cardiovascular. Objetivo: Examinar as associações de desfechos sociodemográficos, clínicos, antropométricos relatados pelo paciente e variáveis funcionais com a ACR de 69 mulheres (55±10 anos), e identificar se a caminhada é um preditor de variação do pico de VO2 nessa população com câncer de mama (CM). Método: Mulheres sobreviventes de CM recebendo terapia hormonal após dois tipos de cirurgias (conservadora e mastectomia) realizaram um teste de ACR em cicloergômetro para medir o VO2 pico. Um questionário contendo dados sociodemográficos e clínicos; resultados relatados pelos pacientes (RRP) (sintomas depressivos, qualidade do sono, fadiga, imagem corporal); caminhada autorreferida; e testes para medir o percentual de gordura, circunferência da cintura, flexibilidade e amplitude do movimento ADM do ombro foram realizados. Resultados: Desemprego e aposentadoria foram associados à baixa ACR, assim como o uso de inibidores de aromatase ao invés de tamoxifeno. Sintomas depressivos, pior imagem corporal, maior circunferência da cintura, menor flexibilidade e ADM do ombro também foram associados à baixa ACR. A duração da caminhada, controlada por idade e índice de massa corporal (IMC), é um preditor de 13% da variância do VO2 pico nesta amostra. Conclusão: Tais fatores devem ser considerados na compreensão do perfil de ACR de sobreviventes de CM. Como a caminhada foi um preditor da variância do VO2 pico, deve ser recomendada atividade física para pacientes com CM em uso de hormonioterapia.


Introducción: El consumo de oxígeno (VO2 ) es indicativo de aptitud cardiorrespiratoria (ACR) y los niveles más bajos se relacionan con un mayor riesgo de mortalidad total entre las personas con cáncer cuya terapia puede tener consecuencias adversas sobre el sistema cardiovascular. Objetivo: Examinar las asociaciones de los resultados sociodemográficos, clínicos, antropométricos y variables funcionales informados por las pacientes con la ACR en 69 mujeres (55±10 años) e identificar si caminar es un predictor de la variación del VO2 máximo en esta población con cáncer de mama (CM). Método: Mujeres sobrevivientes de CM que recibieron terapia hormonal después de dos tipos de cirugía (conservadora y mastectomía) se sometieron a una prueba de ACR en un cicloergómetro para medir el VO2 máximo. Se aplicó un cuestionario que contenía datos sociodemográficos, clínicos, resultados informados por los pacientes (RIP) (síntomas depresivos, calidad del sueño, fatiga, imagen corporal) y caminata autoinformada y pruebas para medir el porcentaje de grasa corporal, la circunferencia de la cintura, la flexibilidad y el rango de movimiento (RoM) del hombro. Resultados: El desempleo y la jubilación se asociaron con una baja ACR, al igual que el uso de inhibidores de la aromatasa en lugar de tamoxifeno. Síntomas depresivos, peor imagen corporal, mayor perímetro de cintura, menor flexibilidad y RoM de los hombros también se asociaron con una baja ACR. La duración de la caminata, controlada por edad e índice de masa corporal (IMC), es un predictor del 13% de la variación del VO2 pico en esta muestra. Conclusión: Estos factores deben ser considerados para comprender el perfil de ACR de los sobrevivientes de CM. Como la caminata fue un predictor de la variación del VO2 pico, debe recomendarse actividad física para pacientes con CM en terapia hormonal.

In swimming, the speed-time relationship provides the critical speed (CS) and the maximum distance that can be performed above CS (D'). During intermittent severe intensity exercise, a complete D' depletion coincides with task failure, while a sub-CS intensity is required for D' reconstitution. Therefore, determining the balance D' remaining at any time during intermittent exercise (D'BAL) could improve training prescription. This study aimed to 1) test the D'BAL model for swimming; 2) determine an equation to estimate the time constant of the reconstitution of D' (τD'); and 3) verify if τD' is constant during two interval training sessions with the same work intensity and duration and recovery intensity, but different recovery duration. Thirteen swimmers determined CS and D' and performed two high-intensity interval sessions at a constant speed, with repetitions fixed at 50 m. The duration of passive recovery was based on the work/relief ratio of 2:1 (T2:1) and 4:1 (T4:1). There was a high variability between sessions for τD' (coefficient of variation of 306%). When τD' determined for T2:1 was applied in T4:1 and vice versa, the D'BAL model was inconsistent to predict the time to exhaustion (coefficient of variation of 29 and 28%). No linear or nonlinear relationships were found between τD' and CS, possibly due to the high within-subject variability of τD'. These findings suggest that τD' is not constant during two high-intensity interval sessions with the same recovery intensity. Therefore, the current D'BAL model was inconsistent to track D' responses for swimming sessions tested herein.

The prevailing view of metazoan gene regulation is that individual genes are independently regulated by their own dedicated sets of transcriptional enhancers. Past studies have reported long-range gene-gene associations1-3, but their functional importance in regulating transcription remains unclear. Here we used quantitative single-cell live imaging methods to provide a demonstration of co-dependent transcriptional dynamics of genes separated by large genomic distances in living Drosophila embryos. We find extensive physical and functional associations of distant paralogous genes, including co-regulation by shared enhancers and co-transcriptional initiation over distances of nearly 250 kilobases. Regulatory interconnectivity depends on promoter-proximal tethering elements, and perturbations in these elements uncouple transcription and alter the bursting dynamics of distant genes, suggesting a role of genome topology in the formation and stability of co-transcriptional hubs. Transcriptional coupling is detected throughout the fly genome and encompasses a broad spectrum of conserved developmental processes, suggesting a general strategy for long-range integration of gene activity.

NEW FINDINGS: What is the central question of this study? What is the reliability of near-infrared spectroscopy-derived muscle oxygen uptake ( mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ ) kinetics following running exercise and what is the relationship between the time constant of mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ off-kinetics and parameters of aerobic fitness? What is the main finding and its importance? The time constant of mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ kinetics in gastrocnemius following moderate running exercise presents good to excellent reliability. In addition, it was well correlated with parameters of aerobic fitness, such as maximal speed of an incremental test, ventilatory threshold and pulmonary V̇O2${\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ on-kinetics. Therefore, near-infrared spectroscopy-derived muscle oxidative capacity together with other physiological measurements may allow a concomitant local and systemic analysis of the components of the oxidative system. ABSTRACT: Near-infrared spectroscopy-derived muscle oxygen uptake ( mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ ) kinetics following single-joint exercise has been used to assess muscle oxidative capacity. However, little evidence is available on the use of this technique following whole-body exercise. Therefore, this study aimed to assess the reliability of the NIRS-derived mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ kinetics following running exercise and to investigate the relationship between the time constant of mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ off-kinetics ( τmV̇O2$\tau {\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ ) and parameters of aerobic fitness. After an incremental test to determine V̇O2max${\dot{V}_{{{\rm{O}}_{\rm{2}}}{\rm{max}}}}$ , first (VT1 ) and second (VT2 ) ventilatory thresholds, and maximal speed (Smax ), 13 males (age = 21 ± 4 years; V̇O2max${\dot{V}_{{{\rm{O}}_{\rm{2}}}{\rm{max}}}}$  = 55.9 ± 3.4 ml kg-1  min-1 ) performed three sets (two on the first day and one on a subsequent day) of two repetitions of 6-min running exercise at 90%VT1 . The pulmonary V̇O2${\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ ( pV̇O2${\rm{p}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ ) on-kinetics and mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ off-kinetics in gastrocnemius were assessed. τmV̇O2$\tau {\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ presented no systematic change and satisfactory reliability (the standard error of the measurement (SEM) and intraclass correlation coefficient of 4.21 s and 0.49 for between transitions; and 2.65 s and 0.74 averaging τmV̇O2$\tau {\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ within each time set), with no difference (P > 0.3) between the within- (SEM = 2.92 s) and between-day variability (SEM = 2.78 s and 2.19 s between first vs. third set, and second vs. third set, respectively). τmV̇O2$\tau {\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ (28.5 ± 4.17 s) correlated significantly (P < 0.05) with Smax (r = -0.66), VT1 (r = -0.64) and time constant of the p V̇O2${\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ on-kinetics (r = 0.69). These findings indicate that NIRS-derived mV̇O2${\rm{m}}{\dot{V}_{{{\rm{O}}_{\rm{2}}}}}$ kinetics in the gastrocnemius following moderate running exercise is a useful and reliable parameter to assess muscle oxidative capacity.

Developmental patterning networks are regulated by multiple inputs and feedback connections that rapidly reshape gene expression, limiting the information that can be gained solely from slow genetic perturbations. Here we show that fast optogenetic stimuli, real-time transcriptional reporters, and a simplified genetic background can be combined to reveal the kinetics of gene expression downstream of a developmental transcription factor in vivo. We engineer light-controlled versions of the Bicoid transcription factor and study their effects on downstream gap genes in embryos. Our results recapitulate known relationships, including rapid Bicoid-dependent transcription of giant and hunchback and delayed repression of Krüppel. In addition, we find that the posterior pattern of knirps exhibits a quick but inverted response to Bicoid perturbation, suggesting a noncanonical role for Bicoid in directly suppressing knirps transcription. Acute modulation of transcription factor concentration while recording output gene activity represents a powerful approach for studying developmental gene networks in vivo.

Past studies offer contradictory claims for the role of genome organization in the regulation of gene activity. Here, we show through high-resolution chromosome conformation analysis that the Drosophila genome is organized by two independent classes of regulatory sequences, tethering elements and insulators. Quantitative live imaging and targeted genome editing demonstrate that this two-tiered organization is critical for the precise temporal dynamics of Hox gene transcription during development. Tethering elements mediate long-range enhancer-promoter interactions and foster fast activation kinetics. Conversely, the boundaries of topologically associating domains (TADs) prevent spurious interactions with enhancers and silencers located in neighboring TADs. These two levels of genome organization operate independently of one another to ensure precision of transcriptional dynamics and the reliability of complex patterning processes.

J Strength Cond Res 36(6): 1610-1617, 2022-This study aimed to verify the reliability of the BodyMetrix portable A-mode ultrasound in measuring fat and muscle tissue thickness. Thirty physically active men participated in daily body composition evaluations. The evaluations comprised 2 techniques: (a) graphic technique (GTBM), which measured the fat thickness at 9 body sites (abdomen, axillary, biceps brachii, calf, chest, subscapular, suprailiac, thigh, and triceps brachii), and (b) imaging technique (ITBM), which simultaneously measured the fat and muscle thickness of 6 body surfaces (abdomen, biceps brachii, chest, thigh, trapezius, and triceps brachii). Regarding GTBM, relative reliability was moderate to excellent (intraclass correlation coefficient [ICC]: 0.81-0.98), whereas absolute reliability was acceptable for abdomen, calf, chest, subscapular, suprailiac, and triceps brachii (coefficient of variation [CV]: 6.9-8.8%) but high for axillary, biceps brachii, and thigh (CV: 12.0-17.4%) in measuring fat thicknesses. Concerning ITBM, relative reliability was good to excellent (ICC: 0.93-0.99 and 0.90-0.98), whereas absolute reliability was acceptable (CV: 3.0-9.2% and 3.5-5.9%) in measuring fat and muscle thickness, respectively. These findings suggest that the, GTBM was only reliable in measuring fat thickness of abdomen, calf, chest, subscapular, suprailiac, and triceps brachii, whereas ITBM was reliable in measuring both fat and muscle thickness in all regions, but showed better reliability values in measuring muscle than fat thickness.

Early embryos must rapidly generate large numbers of cells to form an organism. Many species accomplish this through a series of rapid, reductive, and transcriptionally silent cleavage divisions. Previous work has demonstrated that the number of divisions before both cell cycle elongation and zygotic genome activation (ZGA) is regulated by the ratio of nuclear content to cytoplasm (N/C). To understand how the N/C ratio affects the timing of ZGA, we directly assayed the behavior of several previously identified N/C ratio-dependent genes using the MS2-MCP reporter system in living Drosophila embryos with altered ploidy and cell cycle durations. For every gene that we examined, we found that nascent RNA output per cycle is delayed in haploid embryos. Moreover, we found that the N/C ratio influences transcription through three overlapping modes of action. For some genes (knirps, fushi tarazu, and snail), the effect of ploidy can be primarily attributed to changes in cell cycle duration. However, additional N/C ratio-mediated mechanisms contribute significantly to transcription delays for other genes. For giant and bottleneck, the kinetics of transcription activation are significantly disrupted in haploids, while for frühstart and Krüppel, the N/C ratio controls the probability of transcription initiation. Our data demonstrate that the regulatory elements of N/C ratio-dependent genes respond directly to the N/C ratio through multiple modes of regulation.

To enable large-scale analyses of transcription regulation in model species, we developed DeepArk, a set of deep learning models of the cis-regulatory activities for four widely studied species: Caenorhabditis elegans, Danio rerio, Drosophila melanogaster, and Mus musculus DeepArk accurately predicts the presence of thousands of different context-specific regulatory features, including chromatin states, histone marks, and transcription factors. In vivo studies show that DeepArk can predict the regulatory impact of any genomic variant (including rare or not previously observed) and enables the regulatory annotation of understudied model species.

Eleven competitive male swimmers (20 ± 3 years, 182 ± 5 cm, and 77 ± 5 kg) performed 2 successive 50-m front crawl trials in a 50-m swimming pool at 10 am and 5 pm. Block time (tB), 15-m performance (t.15-m), and 50-m performance (t.50-m) were measured. Velocity (V), stroke rate (SR), stroke length (SL), and stroke index (SI) were measured at 3 time points during the trials. Pre-trial and post-trial blood samples were taken to determine blood lactate accumulation (Δ[Lac]). For t.50-m, the relative difference between 10 am and 5 pm reached 0.1% (p = 0.7; effect size [ES] = 0.02). Furthermore, no differences in tB (p = 0.12; ES = -0.28) and t.15-m (p = 0.39; ES = -0.16) were observed between periods. Both V (p = 0.11; ES = -0.14) and SI (p = 0.16; ES = 0.15) were also similar. Higher values of SR were recorded at 10 am (p = 0.03; ES = -0.32), whereas the morning values of SL were lower (p = 0.04; ES = 0.3). Δ[Lac] was not significantly different between periods (p = 0.07; ES = -0.27). Although time of the day did not impact performance in 2 successive 50-m front crawl performances, different stroke parameters profiles were observed during these trials. This may help coaches design specific warm-up exercises to enhance performance at different times of the day.

BACKGROUND: Critical swimming speed (CSS) and critical stroke rate (CSR) have important practical applications in evaluating endurance capacity and stroke parameters. The CSS and CSR are determined from the linear regression between two or more performance times with the respective predictive distance or "number of stroke cycles," respectively. It is already known that CSS is dependent on the number and duration of the predictive trials chosen, and performance times ranging from 2 to 12 min have been recommended. However, the effects of predictive trials on the CSR have not been reported. It was hypothesized that CSS and CSR determined by different predictive trials lasting 2 to 12 min would elicit similar values. Therefore, the purpose of the present study was to determine the impact of different combinations of predictive trials lasting 2 to 12 min on both CSR and CSS. METHODS: Thirteen swimmers performed three fixed-distance (200, 400, and 800 m) performances. All possible combinations of CSR and CSS with two (CSR200-400/CSS200-400, CSR200-800/CSS200-800, CSR400-800/CSS400-800) and three (CSR200-400-800/CSS200-400-800) trials were determined. RESULTS: No significant differences were found between CSR and CSS determined with different predictive distance tests. In addition, CSR200-800 and CSS200-800 showed the lowest coefficient of variation and highest intraclass correlation coefficients with CSR200-400-800 and CSS200-400-800, respectively. CONCLUSIONS: This study demonstrated that CSR and CSS were not statistically different when determined with different predictive trials located within the recommended durations of 2-12 min. Nevertheless, CSR200-800 and CSS200-800 exhibited the best consistency with CSR200-400-800 and CSS200-400-800, respectively.

PURPOSE: Metabolic perturbation and V˙O2 on-kinetics are potential modifiers of fatigue and vary in importance depending on the exercise task. Thus, performance fatigability during high-intensity exercise seems to be exercise mode dependent, affecting tolerance in the severe domain. However, the effects of exercise mode on severe domain amplitude are still unknown. The aims of this study were to compare the severe domain amplitude in endurance runners and cyclists and to verify its possible determinants. METHODS: Ten runners and eleven cyclists were tested to determine V˙O2 max, maximal velocity/power output of incremental test (v V˙O2 max/p V˙O2 max), critical velocity/power (CV/CP), distance/work above CV/CP (D'/W'), and the highest velocity/power output which V˙O2 max is attained during constant exercise (VHIGH/PHIGH). The severe domain amplitude was considered as VHIGH/PHIGH relative to CV/CP. RESULTS: When normalized by v V˙O2 max/p V˙O2 max, although VHIGH and PHIGH were similar, CV (89.0 ± 2.2% v V˙O2 max) was higher than CP (84.0 ± 4.1% p V˙O2 max; p < .05; ES = 1.51). Consequently, the severe domain amplitude was higher in cyclists (153.6 ± 14.4% CP vs. 137.2 ± 14.6% CV; p < .05; ES = 1.13). Runners presented faster V˙O2 on-kinetics than cyclists at VHIGH/PHIGH. The severe domain amplitude was correlated with D' (r = .65) and W' (r = .71), but not with V˙O2 on-kinetics. CONCLUSIONS: Cyclists have a lower CP (%p V˙O2 max) and a greater severe domain amplitude than runners, providing a greater range of intensities for attainment of V˙O2 max. Furthermore, the severe domain amplitude appears to be linked to finite energy reserves, but unrelated to V˙O2 on-kinetics.

J Strength Cond Res 33(2): 408-416, 2019-The purpose of this study was to compare cardiopulmonary, neuromuscular, and metabolic responses to severe-intensity intermittent exercises with variable or constant work rate (CWR). Eleven cyclists (28 ± 5 years; 74 ± 7 kg; 175 ± 5 cm; 63 ± 4 ml·kg·min) performed the following tests until exhaustion on separate days: (a) an incremental test; (b) in random order, 2 CWR tests at 95 and 110% of the peak power for the determination of critical power (CP); (c) 2-4 tests for the determination of the highest power that still permits the achievement of maximal oxygen uptake (PHIGH); and (d) 2 random severe-intensity intermittent exercises. The last 2 sessions consisted of a CWR exercise performed at PHIGH or a decreasing work rate (DWR) exercise from PHIGH until 105% of CP. Compared with CWR, DWR presented higher time to exhaustion (635 ± 223 vs. 274 ± 65 seconds), time spent above 95% of V[Combining Dot Above]O2max (t95% V[Combining Dot Above]O2max) (323 ± 227 vs. 98 ± 65 seconds), and O2 consumed (0.97 ± 0.41 vs. 0.41 ± 0.11 L). Electromyography amplitude (root mean square [RMS]) decreased for DWR but increased for CWR during each repetition. However, RMS and V[Combining Dot Above]O2 divided by power output (RMS/PO and V[Combining Dot Above]O2/PO ratio) increased in every repetition for both protocols, but to a higher extent and slope for DWR. These findings suggest that the higher RMS/PO and V[Combining Dot Above]O2/PO ratio in association with the longer exercise duration seemed to have been responsible for the higher t95% V[Combining Dot Above]O2max observed during severe DWR exercise.

The establishment of new interactions between transcriptional regulators increases the regulatory diversity that drives phenotypic novelty. To understand how such interactions evolve, we have studied a regulatory module (DDR) composed by three MYB-like proteins: DIVARICATA (DIV), RADIALIS (RAD), and DIV-and-RAD-Interacting Factor (DRIF). The DIV and DRIF proteins form a transcriptional complex that is disrupted in the presence of RAD, a small interfering peptide, due to the formation of RAD-DRIF dimers. This dynamic interaction result in a molecular switch mechanism responsible for the control of distinct developmental processes in plants. Here, we have determined how the DDR regulatory module was established by analyzing the origin and evolution of the DIV, DRIF, and RAD protein families and the evolutionary history of their interactions. We show that duplications of a pre-existing MYB domain originated the DIV and DRIF protein families in the ancestral lineage of green algae, and, later, the RAD family in seed plants. Intraspecies interactions between the MYB domains of DIV and DRIF proteins are detected in green algae, whereas the earliest evidence of an interaction between DRIF and RAD proteins occurs in the gymnosperms, coincident with the establishment of the RAD family. Therefore, the DDR module evolved in a stepwise progression with the DIV-DRIF transcription complex evolving prior to the antagonistic RAD-DRIF interaction that established the molecular switch mechanism. Our results suggest that the successive rearrangement and divergence of a single protein domain can be an effective evolutionary mechanism driving new protein interactions and the establishment of novel regulatory modules.

OBJECTIVE: The aims of this study are to investigate the association between: (i) forward head posture (FHP) and pressure pain thresholds (PPTs); (ii) FHP and maladaptive cognitive processes; and (iii) FHP and neck pain characteristics in university students with subclinical neck pain. MATERIALS/METHODS: A total of 140 university students, 90 asymptomatic and 50 with subclinical neck pain, entered the study. Demographic data, anthropometric data, FHP, and PPTs were collected for both groups. In addition, pain characteristics, pain catastrophizing, and fear of movement were assessed for participants with neck pain. FHP was characterized by the angle between C7, the tragus of the ear, and the horizontal line. Correlation analysis and multivariate regression analysis were conducted. RESULTS: Participants with subclinical neck pain showed significantly lower PPTs than participants without neck pain (p < .05), but similar FHP (p > .05). No significant association was found between FHP and PPTs in the asymptomatic group. In the group of participants with subclinical neck pain, PPTs at the right trapezius and neck pain duration explained 19% of the variance of FHP (R2 = 0.23; adjusted R2 = 0.19; p < .05). CONCLUSION: This study suggests that FHP is not associated with PPTs in asymptomatic university students. In university students with subclinical neck pain, increased FHP was associated with right trapezius hypoalgesia and with neck pain of shorter duration. These findings are in contrast with current assumptions on the association between neck pain and FHP.

PURPOSE: Although ischaemic preconditioning (IPC), induced by cycles of transient limb ischaemia and reperfusion, seems to improve exercise performance, the optimal duration of ischaemia-reperfusion cycles is not established. The present study investigated the effect of ischaemia-reperfusion duration within each IPC cycle on performance in a 2000-m rowing ergometer test. METHODS: After incremental and familiarization tests, 16 trained rowers (mean ± SD: age, 24 ± 11 years; weight, 74.1 ± 5.9 kg; [Formula: see text] peak, 67.2 ± 7.4 mL·kg-1·min-1) were randomly submitted to a 2000-m rowing test preceded by intermittent bilateral cuff inflation of the lower limbs with three cycles of ischaemia-reperfusion, lasting 5 min (IPC-5) or 10 min (IPC-10) at 220 or 20 mmHg (control). Power output, [Formula: see text], heart rate, blood lactate concentration, pH, ratings of perceived exertion (RPE), and near-infrared spectroscopy-derived measurements of the vastus lateralis muscle were continuously recorded. RESULTS: No differences among treatments were found in the 2000-m test (control: 424 ± 17; IPC-5: 425 ± 16; IPC-10: 424 ± 17 s; P = 0.772). IPC-10 reduced the tissue saturation index and oxy-haemoglobin concentration during exercise compared with control. The power output during the last 100-m segment was significantly lower with IPC-10. The IPC treatments increased the heart rate over the first 500 m and decreased the pH after exercise. No alterations were observed in [Formula: see text], blood lactate, or RPE among the trials. CONCLUSION: In conclusion, IPC does not improve the 2000-m rowing ergometer performance of trained athletes regardless of the length of ischaemia-reperfusion cycles.
