RESUMEN

The purpose was to study consequences of long-duration (105-day) isolation and confinement on the rate of chromosomal aberrations in human blood lymphocytes in vivo. Blood samples for cytogenetic analysis were drawn from 6 test-subjects before and on completion of the experiment. Analysis of unstable chromosomal aberrations showed a statistically reliable increase (p < 0.05) in the rate of chromatid aberrations (virtually, 25-fold) in the form of single fragments as compared with baseline values. Rate of chromosome aberrations did not differ from baseline values except 1 test-subject with a dicentric and associated fragment found after the experiment.

The paper deals with a comparative study of two regimens of Macaca mulatta continuous 2-week gamma- and double fractionated acute irradiation by the total absorbed doses of 250 and 132 cGy, respectively; data about chromosomal aberration rates in peripheral lymphocytes were correlated. Based on calculations it was hypothesized that, regardless of regimen, by day-12 of irradiation the effective residual dose would be same, i.e. approximately 100 cGy. Analysis of instable aberration chromosomes showed similarity of the effect of gamma-irradiation by 137Cs at earlier time points (on days 2-21). In primates with the gamma-irradiation profile close to chronic (Gamma 25) the total rate of chromosomal aberrations and rate of radiation markers remain high, whereas animals exposed to fractionated radiation (Gamma 66) displayed a statistically significant (p < 0.05) reduction in cytogenetic parameters on days 36, 50 and 85. Cytogenetic analysis of peripheral lymphocytes permits the conclusion that though markedly different in dose values and rates, both regimens produce an essentially equal early damaging effect.

RESUMEN

The probability of losses of different chromosome aberrations during the dicentric chromosome assay of metaphase cells with incomplete sets of chromosome centromeres was estimated using a mathematical model for low doses of ionizing radiation. A dicentric assay of human blood lymphocytes without determination of the total amount of chromosome centromeres in cells without chromosome aberrations (rapid dicentric assay) has been proposed. The rapid dicentric analysis allows to register chromosome aberrations in full compliance with the conventional classification. The experimental data have shown no statistically significant difference between the frequencies of dicentric chromosomes detected by rapid and classical dicentric chromosome assays of human lymphocytes exposed to 0.5 Gy of 60Co gamma-rays. The rate of the rapid dicentric assay was almost twice as high as that of the classical dicentric assay.

Cytogenetic assay of the chromosomal apparatus of human blood lymphocytes was carried out after in vitro irradiation by heavy charged particles with high LET values. Blood plasm samples enriched with lymphocytes were irradiated by accelerated ions of carbon 12C (290 MeV/nucleon and LET = 70 keV/microm), neon 20Ne (400 MeV/nucleon and LET = 70 keV/microm), and iron 56Fe (500 MeV/nucleon and LET = 200 keV/microm) in the dose range from 0.25 to 1 Gy. Rate of chromosome aberrations showed a linear dependence on doses from the densely ionizing radiations with high LET values. Frequency of dicentrics and centric rings in human lymphocytes irradiated by 12C with the energy of 290 MeV/nucleon was maximal at 1 Gy (p < 0.05) relative to the other heavy particles. It was found that relative biological effectiveness of heavy nuclei is several times higher than of 60Co gamma-radiation throughout the range of doses in this investigation.

RESUMEN

Effects of 120-d bed rest on chromosomal aberration rate were studied in lymphocytes of human peripheral blood in vivo. Chromosomal analysis revealed statistically significant increases in dicentrics and paired acentric fragments in blood lymphocytes of human test-subjects.

RESUMEN

Cytogenetic disorders in peripheral blood lymphocytes of humans were studied in culture following single irradiation by protons with energy of 1 GeV. Doses ranged from 0.05 to 2.0 Gy. A large number of structural damages were observed. Nature of dependence of frequency of such irradiation markers as dicentrics and centric ring was established. Relative biological coefficient of relative energy protons was calculated as 1.2.

RESUMEN

The investigation had the goal to fulfill in vivo cytogenetic studies of blood lymphocytes of human subjects in a 7-d experiment with "dry immersion". Blood was sampled from 10 subjects prior to and on completion of the experiment. Analysis of chromosomal aberrations in blood lymphocytes after exposure to simulated microgravity did not reveal statistically reliable chromosomal alterations in comparison with spontaneous levels.

The aim of the investigation was the study of cytogenetic effects in human blood lymphocytes of low doses of ionizing radiation in vitro. The analysis of unstable chromosome aberrations in human lymphocytes after irradiation by the accelerated ions 12C with the energy 500 MeV/nucleon and LET 10.7 keV/microm was carried out. Blood samples were irradiated on Nuclotron of the High Energy Laboratory of the Joint Institute for Nuclear Research. The doses of irradiation were in the range from 0.05 up to 1.0 Gy. Was shown that the frequency of unstable chromosome aberrations depends from the dose of ionizing radiation and can be described by linear function. At the doses 0.25-0.50 Gy the dose-independent curve was obtained for dicentrics and centric rings. The frequencies of dicentrics and centric rings as markers of the radiation action were slightly different for different donors that could be explained by different radiosensitivity. Using the calibration curve obtained earlier for gamma-rays coefficients of relative biological efficiency of accelerated 12C with the energy 500 MeV/nucleon were defined: they varied from 1.0 at the doses (0.5-1.0 Gy) up to 3.2 at the lower doses (0.05-0.25 Gy).

An analysis was performed of unstable chromosomal aberrations in peripheral blood of 36 cosmonauts after long-term space missions on "Mir" orbital station. 25 cosmonauts were examined before their flights to score spontaneous yields of cytogenetical damage. In all cases the doses absorbed by crews during space flights did not exceed permissible levels of irradiation, adopted for cosmonauts. The frequencies of chromosomal-type aberrations after space missions were found to increase significantly compared to the pre-flight levels. The yields of dicentrics and centric rings on the average were as high as 0.12 +/- 0.02 and 0.47 +/- 0.06% before and after the 1st flight, 0.18 +/- 0.05 and 0.71 +/- 0.11% before and after the 2nd flight respectively. During the inter-flight periods, usually lasted 1.5-2 years, the yields of chromosome damage lowered, but did not reach their spontaneous values. After each next flight the yields of chromosome aberrations increased again. The cytogenetical damage detected in cosmonauts' peripheral blood lymphocytes after chronic action of low doses of space radiation points out a possible increase in risks of stochastic effects in distant future for crews after long-term space missions.
