INTRODUCTION: We report, herein, in vitro, and in vivo toxicity evaluation of silver nanoparticles stabilized with gum arabic protein (AgNP-GP) in Daphnia similis, Danio rerio embryos and in Sprague Dawley rats. PURPOSE: The objective of this investigation was to evaluate in vitro and in vivo toxicity of silver nanoparticles stabilized with gum arabic protein (AgNP-GP), in multispecies due to the recognition that toxicity evaluations beyond a single species reflect the environmental realism. In the present study, AgNP-GP was synthesized through the reduction of silver salt using the tri-alanine-phosphine peptide (commonly referred to as "Katti Peptide") and stabilized using gum arabic protein. METHODS: In vitro cytotoxicity tests were performed according to ISO 10993-5 protocols to assess cytotoxicity index (IC50) values. Acute ecotoxicity (EC50) studies were performed using Daphnia similis, according to the ABNT NBR 15088 protocols. In vivo toxicity also included evaluation of acute embryotoxicity using Danio rerio (zebrafish) embryos following the OECD No. 236 guidelines. We also used Sprague Dawley rats to assess the toxicity of AgNP-GP in doses from 2.5 to 10.0 mg kg-1 body weight. RESULTS: AgNP-GP nanoparticles were characterized through UV (405 nm), core size (20±5 nm through TEM), hydrodynamic size (70-80 nm), Zeta (ζ) potential (- 26 mV) using DLS and Powder X ray diffraction (PXRD) and EDS. PXRD showed pattern consistent with the Ag (1 1 1) peak. EC50 in Daphnia similis was 4.40 (3.59-5.40) µg L-1. In the zebrafish species, LC50 was 177 µg L-1. Oral administration of AgNP-GP in Sprague Dawley rats for a period of 28 days revealed no adverse effects in doses of up to 10.0 mg kg-1 b.w. in both male and female animals. CONCLUSION: The non-toxicity of AgNP-GP in rats offers a myriad of applications of AgNP-GP in health and hygiene for use as antibiotics, antimicrobial and antifungal agents.

Mucoepidermoid lung carcinoma is frequently manifested by obstructive trachea symptoms. It is necessary to develop effective curative or palliative therapeutic strategies for treating mucoepidermoid lung carcinoma through administration of compounds that improve ionizing radiation treatment, thereby increasing the effects of the treatment on tumor cells while minimizing the effects on normal tissues surrounding the tumor cells. Resveratrol is a polyphenolic phytoalexin present in wines and several plants and has a broad spectrum of biological activities, including antioxidant and anticarcinogenic effects. The biological effects of ionizing radiation plus resveratrol have been examined in different types of cell in many studies. Here, we aimed to verify the effects of resveratrol on mucoepidermoid lung carcinoma cells NCI-H292 exposed to ionizing radiation. In vitro studies in NCI-H292 cell culture using neutral red uptake assays showed that the cytotoxicity index (IC50) of resveratrol was 401.5 µM, and the lethal dose 50 % (LD50) of ionizing radiation in the absence of resveratrol was 693 Gy. The fascicular composition and organisation of the inferior alveolar nerve (IAN) were determined to confirm the microarchitecture of the IAN bundles into each of the mandibular teeth, including the composition of the mental nerve. The aim of this study was to evaluate peripheral nerve repair after the application of an antioxidant compound to the damaged nerve tissue to elevate the concentration and bioavailability of elements capable of favouring tissue repair. Twenty-five Wistar rats were divided into groups: The Control 1 (Ctl 1) (n = 5) animals had the ischiatic nerve exposed with no suture injury and were sacrificed at 30 days post-operatively. The Control 2 (Ctl 2) (n = 10) animals had the ischiatic nerve exposed, and the nerve was injured using suture in three distinct regions. In the experimental (Exp) animals (n = 10), an antioxidant organic compound was applied to the nerve injury site. The animals with nerve injury (Ctl2 and Exp group) were sacrificed at 15 and 30 days post-operatively. The histological analysis showed less degeneration in the Exp group at 15 and 30 days post-operatively. Nerve neoformation forming a connection between the distal and proximal suture sites was observed in the experimental group. This study presented an alternative to nerve repair using an antioxidant compound.

The mechanical strength of bioactive glasses can be improved by controlled crystallization, turning its use as bulk bone implants viable. However, crystallization may affect the bioactivity of the material. The aim of this study was to develop glass-ceramics of the nominal composition (wt%) 52.75(3CaO·P2O5)-30SiO2-17.25MgO, with different crystallized fractions and to evaluate their in vitro cytotoxicity and bioactivity. Specimens were heat-treated at 700, 775 and 975 °C, for 4 h. The major crystalline phase identified was whitlockite, an Mg-substituted tricalcium phosphate. The evaluation of the cytotoxicity was carried out by the neutral red uptake methodology. Ionic exchanges with the simulated body fluid SBF-K9 acellular solution during the in vitro bioactivity tests highlight the differences in terms of chemical reactivity between the glass and the glass-ceramics. The effect of crystallinity on the rates of hydroxycarbonate apatite (HCA) formation was followed by Fourier transformed infrared spectroscopy. Although all glass-ceramics can be considered bioactive, the glass-ceramic heat-treated at 775 °C (V775-4) presented the most interesting result, because the onset for HCA formation is at about 24 h and after 7 days the HCA layer dominates completely the spectrum. This occurs probably due to the presence of the whitlockite phase (3(Ca,Mg)O·P2O5). All samples were considered not cytotoxic.

RESUMEN

Polymeric hydrogels are used as wound dressing material since these materials show advantages such as pain relief, exudates absorption, barrier to microorganisms, permeability, and others. This article shows the results obtained in a study aiming to know the biological performance of different polymeric materials to be used in contact with skin: PVP hydrogels and acrylate adhesive. The biocompatibility was determined by in vitro assay of cytotoxicity and in vivo assay by using the contact test of irritability in rabbits. All the tested samples presented no toxicity and no dermal irritation.

Hidrogéis constituídos de poli N-vinil-2-porrolidona (PVP), ágar e polietilenoglicol (PEG) säo utilizados como membranas para uso tópico, no tratamento de lesöes de pele. Com o objetivo de melhorar algumas das propriedades mecânicas da membrana, como tensäo de ruptura e elongaçäo, substituiu-se o PEG pelo copolímero poli (dimetilsiloxano)-co-poli (óxido de etileno) (SEO). A biocompatibilidade "in vitro" das membranas de PVP-SEO com diferentes concetraçöes de SEO foi testada em cultura de células. O teste de citotoxicidade foi realizado pelo estudo comparativo de dois métodos: 1) difusäo em ágar, utilizando-se as linhagens celulares RC-IAL e NCTC Clone L-929 e 2) inibiçäo da formaçäo de colônias utilizando-se a linhagem celular CHO-K1. Os resultados obtidos nos dois métodos foram análogos e demonstraram que as membranas do PVP-SEO apresentaram toxicidade nas concentraçöes testadas e nas linhagens celulares utilizadas. Foi possível também concluir que a adiçäo de SEO ocasiona caráter citotóxico às membranas de PVP


Hydrogels composed by PVP, agar and PEG are used as wound dressings in the treatmentof skin ulcers. In order to improve the mechanical properties of the membrane, such as tensile strengthand elongation, PEG was replaced by copolymer poly (dimethilsiloxane)-co-polyoxiethylene (SEO).The "in vitro" biocompatibility evaluation of PVP-SEO membranes with different concentrations of SEOwas carried out in cell culture. A comparative study of cytotoxicity assays was performed by twomethods: 1) diffusion agar assay, using RC-IAL and NCTC Clone L-929 cells and 2) colony suppressionassay, using CHO-K1 cells. In the cytotoxic evaluation of PVP-SEO membranes by both methods,analogous results were obtained. It was also possible to conclude that the addition of SEO promotescytotoxicity in PVP membranes
