RESUMEN

BACKGROUND: Dengue virus (and Chikungunya and Zika viruses) is transmitted by Aedes aegypti and Aedes albopictus mosquitoes and causes considerable human morbidity and mortality. As there is currently no vaccine or chemoprophylaxis to protect people from dengue virus infection, vector control is the only viable option for disease prevention. The purpose of this paper is to illustrate the design and placement process for an attractive lethal ovitrap to reduce vector populations and to describe lessons learned in the development of the trap. METHODS: This study was conducted in 2010 in Iquitos, Peru and Lopburi Province, Thailand and used an iterative community-based participatory approach to adjust design specifications of the trap, based on community members' perceptions and feedback, entomological findings in the lab, and design and research team observations. Multiple focus group discussions (FGD) were held over a 6 month period, stratified by age, sex and motherhood status, to inform the design process. Trap testing transitioned from the lab to within households. RESULTS: Through an iterative process of working with specifications from the research team, findings from the laboratory testing, and feedback from FGD, the design team narrowed trap design options from 22 to 6. Comments from the FGD centered on safety for children and pets interacting with traps, durability, maintenance issues, and aesthetics. Testing in the laboratory involved releasing groups of 50 gravid Ae. aegypti in walk-in rooms and assessing what percentage were caught in traps of different colors, with different trap cover sizes, and placed under lighter or darker locations. Two final trap models were mocked up and tested in homes for a week; one model was the top choice in both Iquitos and Lopburi. DISCUSSION: The community-based participatory process was essential for the development of novel traps that provided effective vector control, but also met the needs and concerns of community members.

Until recently, facultative automictic parthenogenesis within the squamate reptiles exhibiting ZZ:ZW genetic sex determination has resulted in single reproductive events producing male (ZZ) or female (ZW) offspring. With the recent discovery of viable parthenogenetically produced female (WW) Boa constrictors, the existence of further parthenogenetic events resulting in WW females was questioned. Here, we provide genetic evidence for consecutive virgin births by a female Colombian rainbow boa (Epicrates maurus), resulting in the production of WW females likely through terminal fusion automixis. Samples were screened at 22 microsatellite loci with 12 amplifying unambiguous products. Of these, maternal heterozygosity was observed in 4, with the offspring differentially homozygous at each locus. This study documents the first record of parthenogenesis within the genus Epicrates, a second within the serpent lineage Boidae, and the third genetically confirmed case of consecutive virgin births of viable offspring within any vertebrate lineage. Unlike the recent record in Boa constrictors, the female described here was isolated from conspecifics from birth, demonstrating that males are not required to stimulate parthenogenetic reproduction in this species and possibly other Boas.

The nesting behaviors of 10 females of Tachysphex inconspicuus (Kirby) (Hymenoptera: Crabronidae) were studied on a sandy, mowed lawn at the La Selva Biological Station in northeastern Costa Rica on 27-29 April 1980. Twenty-four completed nests were observed, excavated, and measured. The nests had oblique, short burrows leading to one or two shallow cells. Prey cockroaches belonging to 11 species of Chorisoneura and Riatia fulgida (Saussure) (Blattaria: Blattellidae), all tropical wet forest canopy indicator species, were removed from the cells, weighed, and identified. The cockroaches consisted mainly of adult females, selectively preyed upon over adult males and nymphs due to their larger sizes. The aggregate prey mass in cells was separable into prospective larger (heavier) female and smaller (lighter) male cells. Wasps usually oviposited on the heaviest cockroach in a cell, in most cases an adult female. Atypical genus behavior included (1) prey being carried to one side of the wasp and perhaps grasped by a hindleg during removal of the temporary entrance closure and nest entry and (2) wasp's egg being laid affixed to a forecoxal corium and extending backward in a longitudinally posteriad position across the prey's ventral thorax. A comparison with the nesting behavior of other species in the Tachysphex obscuripennis species group is made.

In lowland Amazonian rainforests, specific ants collect seeds of several plant species and cultivate them in arboreal carton nests, forming species-specific symbioses called ant-gardens (AGs). In this obligate mutualism, ants depend on the plants for nest stability and the plants depend on ant nests for substrate and nutrients. AG ants and plants are abundant, dominant members of lowland Amazonian ecosystems, but the cues ants use to recognize the seeds are poorly understood. To address the chemical basis of the ant-seed interaction, we surveyed seed chemistry in nine AG species and eight non-AG congeners. We detected seven phenolic and terpenoid volatiles common to seeds of all or most of the AG species, but a blend of the shared compounds was not attractive to the AG ant Camponotus femoratus. We also analyzed seeds of three AG species (Anthurium gracile, Codonanthe uleana, and Peperomia macrostachya) using behavior-guided fractionation. At least one chromatographic fraction of each seed extract elicited retrieval behavior in C. femoratus, but the active fractions of the three plant species differed in polarity and chemical composition, indicating that shared compounds alone did not explain seed-carrying behavior. We suggest that the various AG seed species must elicit seed-carrying with different chemical cues.

Introduced populations of the Argentine ant, Linepithema humile, have experienced moderate to severe losses of genetic diversity, which may have affected nestmate recognition to various degrees. We hypothesized that cuticular hydrocarbons (CHC) serve as nestmate recognition cues, and facilitate colony fusion of unrelated L. humile colonies that share similar CHC profiles. In this study, we paired six southeastern U.S. L. humile colonies in a 6-month laboratory fusion assay, and determined if worker and queen CHC profile similarity between colonies was associated with colony fusion and intercolony genetic similarity. We also compared worker and queen CHC profiles between fused colony pairs and unpaired controls to determine if worker and queen chemical profiles changed after fusion. We found that colony fusion correlated with the CHC similarity of workers and queens, with the frequency of fusion increasing with greater CHC profile similarity between colonies. Worker and queen CHC profile similarity between colonies also was associated with genetic similarity between colonies. Queen CHC profiles in fused colonies appeared to be a mix of the two colony phenotypes. In contrast, when only one of the paired colonies survived, the CHC profile of the surviving queens did not diverge from that of the colony of origin. Similarly, workers in non-fused colonies maintained their colony-specific CHC, whereas in fused colonies the worker CHC profiles were intermediate between those of the two colonies. These results suggest a role for CHC in regulating interactions among mutually aggressive L. humile colonies, and demonstrate that colony fusion correlates with both genetic and CHC similarities. Further, changes in worker and queen chemical profiles in fused colonies suggest that CHC plasticity may sustain the cohesion of unrelated L. humile colonies that had fused.

In moth pheromone communication signals, both quantitative and qualitative intraspecific differences have been found across geographic regions. Such variation has generally been hypothesized to be due to selection, but evidence of genetic control of these differences is largely lacking. To explore the patterns of variation in pheromone signals, we quantified variation in the female sex pheromone blend and male responses of two closely related noctuid moth species in five different geographic regions for 2-3 consecutive years. We found significant variation in the ratios of sex pheromone blend components as well as in male response, not only between geographic regions but also within a region between consecutive years. The temporal variation was of a similar magnitude as the geographic variation. As far as we know, this is the first study reporting such temporal variation in moth chemical communication systems. The geographic variation seems to at least partly be controlled by genetic factors, and to be correlated with the quality of the local chemical environment. However, the pattern of temporal variation within populations suggests that optimization of the pheromonal signal also may be driven by within-generation physiological adjustments by the moths in response to their experience of the local chemical environment.

Throughout lowland Amazonia, arboreal ants collect seeds of specific plants and cultivate them in nutrient-rich nests, forming diverse yet obligate and species-specific symbioses called Neotropical ant-gardens (AGs). The ants depend on their symbiotic plants for nest stability, and the plants depend on AGs for substrate and nutrients. Although the AGs are limited to specific participants, it is unknown at what stage specificity arises, and seed fate pathways in AG epiphytes are undocumented. Here we examine the specificity of the ant-seed interaction by comparing the ant community observed at general food baits to ants attracted to and removing seeds of the AG plant Peperomia macrostachya. We also compare seed removal rates under treatments that excluded vertebrates, arthropods, or both. In the bait study, only three of 70 ant species collected P. macrostachya seeds, and 84% of observed seed removal by ants was attributed to the AG ant Camponotus femoratus. In the exclusion experiment, arthropod exclusion significantly reduced seed removal rates, but vertebrate exclusion did not. We provide the most extensive empirical evidence of species specificity in the AG mutualism and begin to quantify factors that affect seed fate in order to understand conditions that favor its departure from the typical diffuse model of plant-animal mutualism.

In social insects, individuals typically recognize and behave aggressively towards alien conspecifics, thereby maintaining colony integrity. This is presumably achieved via a nestmate recognition system in which cuticular compounds, usually cuticular hydrocarbons (CHC), of genetic and/or environmental origin serve as recognition cues. Most invasive populations of the Argentine ant, Linepithema humile (Mayr), display minimal nestmate-non-nestmate discrimination, resulting in low levels of intraspecific aggression allowing free movement of workers and queens among nests. However, invasive L. humile in the southeastern United States show relatively high levels of intraspecific aggression, and selectively adopt non-nestmate queens. Using behavioral assays and gas chromatography, we found an association between non-nestmate queen adoption and similarity of the CHC profiles of adopted and host colony queens. Also, nestmate and non-nestmate queen CHC profiles became more similar after adoption by queenless colonies. Furthermore, queens treated with non-nestmate queen CHC had distinct CHC profiles and were generally attacked by nestmate workers. We suggest that in L. humile, CHC are used as queen recognition cues, and that queen recognition errors are more likely to occur when the CHC profiles of non-nestmate and host colony queens are similar. Our findings provide further evidence for the complex and dynamic nature of L. humile nestmate discrimination, which may in part underlie the success of introduced populations of this invasive ant.

Seed dispersal mutualisms are essential for the survival of diverse plant species and communities worldwide. Among invertebrates, only ants have a major role in seed dispersal, and thousands of plant species produce seeds specialized for ant dispersal in "diffuse" multispecies interactions. An outstanding but poorly understood ant-seed mutualism occurs in the Amazonian rainforest, where arboreal ants collect seeds of several epiphyte species and cultivate them in nutrient-rich nests, forming abundant and conspicuous hanging gardens known as ant-gardens (AGs). AG ants and plants are dominant members of lowland Amazonian ecosystems, and their interaction is both specific and obligate, but the means by which ants locate, recognize, and accept their mutualist seeds while rejecting other seeds is unknown. Here we address the chemical and behavioral basis of the AG interaction. We show that workers of the AG ant Camponotus femoratus are attracted to odorants emanating from seeds of the AG plant Peperomia macrostachya, and that chemical cues also elicit seed-carrying behavior. We identify five compounds from P. macrostachya seeds that, as a blend, attract C. femoratus workers. This report of attractive odorants from ant-dispersed seeds illustrates the intimacy and complexity of the AG mutualism and begins to illuminate the chemical basis of this important and enigmatic interaction.

The mate attraction signal of Heliothis subflexa (Hs) females consists of a multicomponent sex pheromone blend. In this study, we assessed the intraspecific importance of three groups of compounds found in Hs pheromone glands: three acetate esters (Z7-16:OAc, Z9-16:OAc, and Z11-16:OAc), two 14-carbon aldehydes (14:Ald and Z9-14:Ald), and one 16-carbon alcohol (Z11-16:OH). Because the relative importance of pheromone components may vary in different regions, we conducted experiments in Eastern US (North Carolina) and Western Mexico (Jalisco). Our experiments in Eastern US showed that when the acetates were omitted from a 7-component blend in rubber septa, fewer males were caught in cone traps. Subsequent experiments conducted both in Eastern US and Western Mexico indicated that the addition of Z9-16:OAc alone does not increase attraction of male Hs, while Z11-16:OAc does. The Hs male response to Z7-16:OAc differed between the two regions. In Eastern US, significantly more males were attracted to a minimal three-component blend to which Z7-16:OAc was added, but this was not the case in Western Mexico. The two 14-carbon aldehydes also showed differential attraction between the two regions. 14:Ald and Z9-14:Ald appeared not to play any role in the sexual communication of Hs in Eastern US, but reduced trap catches in Western Mexico. The alcohol Z11-16:OH was tested in two concurrent dose-response studies with Hs males in Western Mexico, one using a minimal blend and one using a complete blend. The minimal three-component blend provided a more discriminating tool for delineating dose-response effects of Z11-16:OH than the seven-component blend. In the minimal blend, the optimal dose of Z11-16:OH was 1%, while in the complete blend similar numbers of males were caught when the alcohol ranged from 1 to 25%.
