RESUMEN

OBJECTIVE: Orexin A (OXA) is a hypothalamic neuropeptide involved in regulation of food intake and nutritional status. There are multiple disturbances of neuropeptide signaling described in girls with anorexia nervosa (AN), but OXA levels have not been addressed in this population to date. Therefore, we analyzed OXA levels of AN girls in this study. METHOD: OXA (radioimmunoassay/RIA/method), leptin, insulinlike growth factor-1 (IGF-1), and insulinlike growth factor-1 binding protein-3 (IGFBP-3) levels were measured before and after 8 weeks of realimentation in 36 girls with AN and in 14 healthy controls (control group: CG). RESULTS: Average weight increased significantly in AN during the study (p < .0001), while plasma levels of OXA decreased (before realimentation: 56.2 ± 2.4 pg/ml; after realimentation: 47.5 ± 1.4 pg/ml; p = .0025). OXA levels before realimentation differed from levels in the CG (47.15 ± 2.6 pg/ml, p = .034), but not afterward. We did not find any correlation between OXA and age, height, weight, BMI; or IGF-1, IGFBP-3, and leptin levels. DISCUSSION: OXA levels in untreated AN patients differ significantly from healthy subjects and decrease during realimentation. These findings indicate that OXA may be involved in the nutritional regulation of malnourished children and adolescents.

Several different organic materials, typical of those used in passive treatment systems for mine influenced water, were tested for their ability to support sulfate-reducing bacteria and sulfate reduction in an anaerobic biological reactor (ABR). The quantity of sulfate-reducing bacteria (SRB) in each organic material, as determined using quantitative polymerase chain reaction (q-PCR) of the dissimilatory sulfite reductase (dsr) gene, correlated with the initial C/N ratio of each material. Potential sulfate reduction rates measured in the laboratory ranked silage > compost = molasses/hay > cattails > pulp mill biosolids and correlated with the q-PCR estimates of SRB in the submerged materials. A comparison of bacterial communities using 16S rRNA gene clone library sequencing revealed similar distribution of clones among the phyla Bacteroidetes, Firmicutes and Proteobacteria for silage, compost and molasses/hay after 174 days of exposure in the seepage water. Silage, the most successful material tested, contained more δ-Proteobacteria-related sequences than the other materials and Spirochaetes-related clones were more abundant in silage than in compost or molasses/hay. According to sequenced dsr clones, the SRB community in silage differed from that for compost and molasses/hay, with fewer Desulfovibrio- and more Desulfomicrobium-related sequences in the silage. Pulp mill biosolids used in the ABR since 2004 contained an overall bacterial community that was more diverse than those for the freshly submerged organics, but only Desulfovibrio desulfuricans-related sequences were found in the dsr library.

At two stations surveyed in Nitinat Lake, a approximately 200-m-deep anoxic tidal fjord, sulfide was detected as close as 15 m from the surface. Biological characterization, determined from small subunit ribosomal RNA gene sequencing, of the chemocline and anaerobic zone revealed many sequences related to sulfur-oxidizing bacteria, suggesting that sulfur cycling is a dominant process. gamma- and epsilon-Proteobacteria related to thiotrophic symbionts, as well as Chlorobium sp., dominated the transition zone. These are expected to play a role in dark and phototrophic CO(2) fixation, respectively. epsilon-Proteobacteria phylotype abundance increased with depth, eventually comprising 69-97% of all sequences recovered from the anoxic zone. The vast majority (74%) of these phylotypes were affiliated with a novel Acrobacter sp. group (NITEP5). Quantification of NITEP5 revealed that up to 2.8 x 10(5) cells ml(-1) were present in the anoxic zone. Surprisingly, although sequences related to known sulfate-reducing bacteria were recovered from the transition zone, quantification of the dsr gene and (35)SO(4)(2-) uptake tests suggest that sulfate-reduction within the water column is negligible. Overall, sequence diversity between different vertical zones was high, although the spatial segregation of gamma-Proteobacteria, Chlorobi, and epsilon-Proteobacteria did not appear to vary significantly between seasons.
