RESUMEN

Ornamental fish keeping is the second most preferred hobby in the world and it provides a great opportunity for entrepreneurship development and income generation. Controlling the environment in ornamental fish farm is a considerable challenge because it is affected by a variety of parameters like water temperature, dissolved oxygen, pH, and disease occurrences. One particular interesting ornamental fish species is goldfish (Carassius auratus). Machine learning (ML) and deep learning technique have significant potential in analysing voluminous data collected from fish farm. Through this technique, the fish farmers can get insight on feeding behaviour, fish growth patterns, predict diseases/stress, and environmental factors affecting fish health. The aim of the study is to analyze the behavioural changes in goldfish due to alterations in environmental parameters (water temperature and dissolved oxygen). Decision tree, Naïve Bayes classifier, K-nearest neighbour (KNN), and linear discriminant analysis (LDA) were used to analyse the behavioural change data. To compare the performance between all four classifiers, cross validation and confusion matrix used. The cross-validation error of LDA, Naïve Bayes classification, KNN and decision tree was 19.86, 28.08, 30.14 and 13.78 respectively. Decision tree was proved to be the most accurate and effective classifier. Different temperature and DO range were taken to predict fish behaviour. Some findings are, the behaviour of fish was rest between temperature 37.85 °C and 40.535 °C, erratic when temperature was greater than or equal to 40.535 °C, gasping when temperature was between 37.85 and 40.535 °C and when DO concentration was less than 6.58 mg/L. Blood parameter analysis has been done to validate the change in external behaviours with change in physiological parameters.

Thermostable direct haemolysin (TDH) is the key virulence factor secreted by the human gastroenteric bacterial pathogen Vibrio parahaemolyticus. TDH is a membrane-damaging pore-forming toxin. It evokes potent cytotoxicity, the mechanism of which still remains under-explored. Here, we have elucidated the mechanistic details of cell death response elicited by TDH. Employing Caco-2 intestinal epithelial cells and THP-1 monocytic cells, we show that TDH induces some of the hallmark features of apoptosis-like programmed cell death. TDH triggers caspase-3 and 7 activations in the THP-1 cells, while caspase-7 activation is observed in the Caco-2 cells. Interestingly, TDH appears to induce caspase-independent cell death. Higher XIAP level and lower Smac/Diablo level upon TDH intoxication provide plausible explanation for the functional inability of caspases in the THP-1 cells, in particular. Further exploration reveals that mitochondria play a central role in the TDH-induced cell death. TDH triggers mitochondrial damage, resulting in the release of AIF and endonuclease G, responsible for the execution of caspase-independent cell death. Among the other critical mediators of cell death, ROS is found to play an important role in the THP-1 cells, while PARP-1 appears to play a critical role in the Caco-2 cells. Altogether, our work provides critical new insights into the mechanism of cell death induction by TDH, showing a common central theme of non-classical programmed cell death. Our study also unravels the interplay of crucial molecules in the underlying signalling processes. Our findings add valuable insights into the role of TDH in the context of the host-pathogen interaction processes.

Polymers are known to adsorb spontaneously from liquid solutions in contact with high-energy substrates to form configurationally complex, but robust phases that often exhibit higher durability than might be expected from the individual physical bonds formed with the substrate. Rational control of the physical, chemical, and transport properties of such interphases has emerged as a fundamental opportunity for scientific and technological advances in energy storage technology but requires in-depth understanding of the conformation states and electrochemical effect of the adsorbed polymers. Here, we analyze the interfacial adsorption of oligomeric polyethylene glycol (PEG) chains of moderate sizes dissolved in protic and aprotic liquid electrolytes and find that there is an optimum polymer molecular weight of approximately 400 Da at which the highest columbic efficiency is achieved for both Zn and Li deposition. These findings point to a simple, versatile approach for extending the lifetime of batteries.

OBJECTIVE: To identify occupational hazards and hazard control strategies adopted by inland fishers of Odisha state in India. METHODS: Information on occupational hazards faced by fishers was collected by personal interviews with 90 riverine and reservoir fishers of Odisha. Through qualitative discussions with key informants and review of literature, a list of occupational hazards were prepared and classified into physical, ergonomics, psychosocial, and natural. Responses were quantified, and descriptive statistics were used for data analysis. RESULTS: Occupational hazards faced by fishers were physical (94%), ergonomic (75%), psychosocial (50%), and natural (34%) in nature. Physical hazards include sharp fishing instruments, uneven surfaces, high temperatures, and wet/slippery areas on the vessel along with the presence of snake/leech/crocodile/fish fin rays. About 97% of reservoir and 90% of the riverine fishers reported physical hazards leading to cuts, fractures, dehydration, headache, sunburn, snakebite, leech bite, crocodile bite, and injuries due to fish fin rays. Heavy nature of work and force exerting activities caused pain in the upper back (50%), shoulders (45%), and lower back (41%). Regarding psychosocial hazard, stress (31%), and anxiety (23%) was reported. Common reasons for this were uncertainty in fish catch and extended working hours, causing tiredness, headache, dizziness, and fatigue. Natural hazards like cyclones, floods, and excessive rain caused financial loss and damage to fishing nets and boats. Regarding personal protective equipment (PPE), floating buoys were used by 26% of riverine and 43% of reservoir fishers to avoid drowning. CONCLUSION: Occupational hazards in the inland fishing sector were identified as physical, ergonomic, psychosocial, and natural. No particular hazard control strategy was reported to be used. Integrating the occupational safety measures in the fisheries policies is thus needed.

Metal electrodeposition in batteries is fundamentally unstable and affected by different instabilities depending on operating conditions and electrolyte chemistry. Particularly, at high charging rates, a hydrodynamic instability loosely termed electroconvection sets in, which complicates all electrochemical processes by creating a nonuniform ion flux and preferential deposition at the electrode. Here, we isolate and study electroconvection by experimentally investigating how oligomer additives in liquid electrolytes interact with the hydrodynamic instability at a cation selective interface. From electrochemical measurements and direct visualization experiments, we find that electroconvection is delayed and suppressed at all voltages in the presence of oligomers. The underlying mechanism is revealed to involve formation of an oligomer ad-layer at the interface, which in response to perturbation is believed to exert an opposing body force on the surrounding fluid to preserve the ad-layer structure and in so doing suppresses electroconvection. Our results therefore reveal that in battery electrolytes without obvious sources of bulk elasticity, surface forces produced by adsorbed polymers can be used to advantage for suppressing instability.

BACKGROUND: Immune checkpoint inhibitor-mediated colitis (IMC) is a common adverse event following immune checkpoint inhibitor (ICI) therapy for cancer. IMC has been associated with improved overall survival (OS) and progression-free survival (PFS), but data are limited to a single site and predominantly for melanoma patients. AIM: To determine the association of IMC with OS and PFS and identify clinical predictors of IMC. METHODS: We performed a retrospective case-control study including 64 ICI users who developed IMC matched according to age, sex, ICI class, and malignancy to a cohort of ICI users without IMC, from May 2011 to May 2020. Using univariate and multivariate logistic regression, we determined association of presence of IMC on OS, PFS, and clinical predictors of IMC. Kaplan-Meier curves were generated to compare OS and PFS between ICI users with and without IMC. RESULTS: IMC was significantly associated with a higher OS (mean 24.3 mo vs 17.7 mo, P = 0.05) but not PFS (mean 13.7 mo vs 11.9 mo, P = 0.524). IMC was significantly associated with OS greater than 12 mo [Odds ratio (OR) 2.81, 95% confidence interval (CI) 1.17-6.77]. Vitamin D supplementation was significantly associated with increased risk of IMC (OR 2.48, 95%CI 1.01-6.07). CONCLUSION: IMC was significantly associated with OS greater than 12 mo. In contrast to prior work, we found that vitamin D use may be a risk factor for IMC.

Vibrio cholerae cytolysin (VCC) is a ß-barrel pore-forming toxin (ß-PFT). It exhibits potent hemolytic activity against erythrocytes that appears to be a direct outcome of its pore-forming functionality. However, VCC-mediated cell-killing mechanism is more complicated in the case of nucleated mammalian cells. It induces apoptosis in the target nucleated cells, mechanistic details of which are still unclear. Furthermore, it has never been explored whether the ability of VCC to trigger programmed cell death is stringently dependent on its pore-forming activity. Here, we show that VCC can evoke hallmark features of the caspase-dependent apoptotic cell death even in the absence of the pore-forming ability. Our study demonstrates that VCC mutants with abortive pore-forming hemolytic activity can trigger apoptotic cell death responses and cytotoxicity, similar to those elicited by the wild-type toxin. VCC as well as its pore formation-deficient mutants display prominent propensity to translocate to the target cell mitochondria and cause mitochondrial membrane damage. Therefore, our results for the first time reveal that VCC, despite being an archetypical ß-PFT, can kill target nucleated cells independent of its pore-forming functionality. These findings are intriguing for a ß-PFT, whose destination is generally expected to remain limited on the target cell membranes, and whose mode of action is commonly attributed to the membrane-damaging pore-forming ability. Taken together, our study provides critical new insights regarding distinct implications of the two important virulence functionalities of VCC for the V. cholerae pathogenesis process: hemolytic activity for iron acquisition and cytotoxicity for tissue damage by the bacteria.

Aqueous zinc batteries are attracting interest because of their potential for cost-effective and safe electricity storage. However, metallic zinc exhibits only moderate reversibility in aqueous electrolytes. To circumvent this issue, we study aqueous Zn batteries able to form nanometric interphases at the Zn metal/liquid electrolyte interface, composed of an ion-oligomer complex. In Zn||Zn symmetric cell studies, we report highly reversible cycling at high current densities and capacities (e.g., 160 mA cm-2; 2.6 mAh cm-2). By means of quartz-crystal microbalance, nuclear magnetic resonance, and voltammetry measurements we show that the interphase film exists in a dynamic equilibrium with oligomers dissolved in the electrolyte. The interphase strategy is applied to aqueous Zn||I2 and Zn||MnO2 cells that are charged/discharged for 12,000 cycles and 1000 cycles, respectively, at a current density of 160 mA cm-2 and capacity of approximately 0.85 mAh cm-2. Finally, we demonstrate that Zn||I2-carbon pouch cells (9 cm2 area) cycle stably and deliver a specific energy of 151 Wh/kg (based on the total mass of active materials in the electrode) at a charge current density of 56 mA cm-2.

The outer membrane of a gram-negative bacteria encapsulates the plasma membrane thereby protecting it from the harsh external environment. This membrane acts as a sieving barrier due to the presence of special membrane-spanning proteins called "porins." These porins are ß-barrel channel proteins that allow the passive transport of hydrophilic molecules and are impermeable to large and charged molecules. Many porins form trimers in the outer membrane. They are abundantly present on the bacterial surface and therefore play various significant roles in the host-bacteria interactions. These include the roles of porins in the adhesion and virulence mechanisms necessary for the pathogenesis, along with providing resistance to the bacteria against the antimicrobial substances. They also act as the receptors for phage and complement proteins and are involved in modulating the host cellular responses. In addition, the potential use of porins as adjuvants, vaccine candidates, therapeutic targets, and biomarkers is now being exploited. In this review, we focus briefly on the structure of the porins along with their important functions and roles in the host-bacteria interactions.

Reversible electrodeposition of metals at liquid-solid interfaces is a requirement for long cycle life in rechargeable batteries that utilize metals as anodes. The process has been studied extensively from the perspective of the electrochemical transformations that impact reversibility, however, the fundamental challenges associated with maintaining morphological control when a intrinsically crystalline solid metal phase emerges from an electrolyte solution have been less studied, but provide important opportunities for progress. A crystal growth stabilization method to reshape the initial growth and orientation of crystalline metal electrodeposits is proposed here. The method takes advantage of polymer-salt complexes (PEG-Zn2+ -aX- ) (a = 1,2,3) formed spontaneously in aqueous electrolytes containing zinc (Zn2+ ) and halide (X- ) ions to regulate electro-crystallization of Zn. It is shown that when X = Iodine (I), the complexes facilitate electrodeposition of Zn in a hexagonal closest packed morphology with preferential orientation of the (002) plane parallel to the electrode surface. This facilitates exceptional morphological control of Zn electrodeposition at planar substrates and leads to high anode reversibility and unprecedented cycle life. Preliminary studies of the practical benefits of the approach are demonstrated in Zn-I2 full battery cells, designed in both coin cell and single-flow battery cell configurations.

Precise development of the dendritic architecture is a critical determinant of mature neuronal circuitry. MicroRNA (miRNA)-mediated regulation of protein synthesis plays a crucial role in dendritic morphogenesis, but the role of miRNA-induced silencing complex (miRISC) protein components in this process is less studied. Here, we show an important role of a key miRISC protein, the GW182 paralog TNRC6A, in the regulation of dendritic growth. We identified a distinct brain region-specific spatiotemporal expression pattern of GW182 during rat postnatal development. We found that the window of peak GW182 expression coincides with the period of extensive dendritic growth, both in the hippocampus and cerebellum. Perturbation of GW182 function during a specific temporal window resulted in reduced dendritic growth of cultured hippocampal neurons. Mechanistically, we show that GW182 modulates dendritic growth by regulating global somatodendritic translation and actin cytoskeletal dynamics of developing neurons. Furthermore, we found that GW182 affects dendritic architecture by regulating the expression of actin modulator LIMK1. Taken together, our data reveal a previously undescribed neurodevelopmental expression pattern of GW182 and its role in dendritic morphogenesis, which involves both translational control and actin cytoskeletal rearrangement. BACKGROUND AND AIMS: Vitamin D downregulates the in vitro expression of the gut-tropic integrin α4ß7 on immune cells. The clinical relevance of this finding in patients with inflammatory bowel disease [IBD] is unclear. We tested the hypothesis that vitamin D is associated with α4ß7 immunophenotypes and risk of vedolizumab [anti-α4ß7] failure in IBD. METHODS: We performed single-cell immunophenotyping of peripheral and intestinal immune cells using mass cytometry [CyTOF] in vedolizumab-naïve patients with IBD [N = 48]. We analysed whole-genome mucosal gene expression [GSE73661] from GEMINI I and GEMINI long-term safety [LTS] to determine the association between vitamin D receptor [VDR] and integrin alpha-4 [ITGA4] and beta-7 [ITGB7] genes. We estimated the odds of vedolizumab failure with low pre-treatment vitamin D in a combined retrospective and prospective IBD cohort [N = 252] with logistic regression. RESULTS: Immunophenotyping revealed that higher 25[OH]D was associated with decreased α4ß7+ peripheral blood mononuclear cells [R = -0.400, p <0.01] and α4ß7+ intestinal leukocytes [R = -0.538, p = 0.03]. Serum 25[OH]D was inversely associated with α4ß7+ peripheral B cells and natural killer [NK] cells and α4ß7+ intestinal B cells, NK cells, monocytes, and macrophages. Mucosal expression of VDR was inversely associated with ITGA4 and ITGB7 expression. In multivariate analysis, 25[OH]D <25 ng/mL was associated with increased vedolizumab primary non-response during induction (odds ratio [OR] 26.10, 95% confidence interval [CI] 14.30-48.90, p <0.001) and failure at 1-year follow-up [OR 6.10, 95% CI 3.06-12.17, p <0.001]. CONCLUSIONS: Low serum 25[OH]D is associated with α4ß7+ immunophenotypes and predicts future vedolizumab failure in patients with IBD. At voltages above a certain critical value, Vc ≈ 20 kT/e, a space charge layer forms near ion-selective interfaces in liquid electrolytes. Interactions between the space charge and an imposed electric field drives a hydrodynamic instability known as electroconvection. Through particle tracking velocimetry we experimentally study the structure and dynamics of the resultant electroconvective flow. Consistent with previous numerical simulations, we report that, following imposition of a sufficiently large voltage, electroconvection develops gradually as pairs of counter-rotating vortices, which nucleate at the interface between an ion-selective substrate and a liquid electrolyte. Depending on the imposed voltage and cell geometry, the vorticies grow to length scales of hundreds of micrometers. Electroconvective flows are also reported to be structured and multiscale, with the size ratio of the largest to the smallest observable vortices inversely proportional to the Debye screening length.

Kerala, a maritime state on the west coast of peninsular India, constitutes approximately 10% of India's total coastline and claims a 20% share in national marine fish production. The registered fishing fleet consists of about 29,969 motorized, 4248 mechanized, and 2515 non-motorized crafts. This study was done to assess occupational hazards faced by fishers as well as the usage of sea safety devices (SSDs) in Kerala. Information was collected from 180 mechanized, motorized, and traditional boat owners. The study revealed that capsizing, sinking, grounding, burning, collision, mishaps, man overboard, and injuries were common occupational hazards. SSDs used were life buoy, life jacket, first aid kit, emergency rations, fire extinguisher, fire bucket, global positioning system, magnetic compass, emergency position indicating radio beacon (EPIRB), sonar, echo sounder, oil lamp, signaling torch, and batteries. Fishers with mechanized boats had high knowledge and used most SSDs. Fishers with motorized boats had adequate knowledge but less SSD usage. Traditional and small-scale fishers did not use most SSDs. Non-parametric Kruskal-Wallis statistical test revealed a significant difference in regards to usage between mechanized, motorized, and traditional fishers, indicating there are gaps in usage of SSDs between the three groups of fishers. It is necessary to educate and motivate fishers with motorized and traditional boats to use SSDs.

INTRODUCTION: The global burden of cancer incidence and mortality are rising continuously worldwide. As per the GLOBOCAN 2012 estimates, about 14.1 million cancer cases and 8.2 million cancer deaths occurred and 32.6 million people living with cancer (within 5 years of diagnosis) in 2012 worldwide. Reliable data on the magnitude and the pattern of cancer are essential for monitoring the health of the community, assessing the performance of the health care system. Cancer registries should pay great attention to quality of their data. The completeness of cancer registry data- the extent to which all of the incident cancers occurring in the population are included in the registry database- is an extremely important attribute of a cancer registry. There are mainly four aspects influencing the quality of data namely, comparability, completeness, validity and timeliness. MATERIALS AND METHODS: Data regarding incidence and mortality with methods of diagnosis for individual years were obtained from the National Cancer Registry Program database of the Indian Council of Medical Research for 2009 to 2014 periods and recalculated for combined years (2009-2014). RESULTS: In males in 2009-11, 77.1% were microscopically confirmed cases which are improved in the later years and for the year 2012-2014, it is 81.4%. In females also the percentage of microscopically confirmed cases were increased from 80.2% to 82.9%. An improvement in mortality to incidence ratio was observed over the years. MI ratio in males was improved to 32.9%. for the year 2012-14 as compared to 28.6% for the year 2009-11 while in female MI ratio is also increased from 18.8% to 21.8% over the period from 2009-11 to 2012-14. Whereas DCO was decreased from 12% to 10.7% in males and 7.3% to 6.6% in females respectively from the period 2009-11 to 2012-14. CONCLUSION: Although there is a slight improvement in data quality till date, there is an enormous scope for population based cancer registry Guwahati to improve the data quality.

Astrocytes are most abundant glial cell type in the brain and play a main defensive role in central nervous system against glutamate-induced toxicity by virtue of numerous transporters residing in their membranes and an astrocyte-specific enzyme glutamine synthetase (GS). In view of that, a dysregulation in the astrocytic activity following an insult may result in glutamate-mediated toxicity accompanied with astrocyte and microglial activation. The present study suggests that the lipopolysaccharide (LPS)-induced inflammation results in significant astrocytic apoptosis compared to other cell types in hippocampus and minocycline could not efficiently restrict the glutamate-mediated toxicity and apoptosis of astrocytes. Upon LPS exposure 76 % astrocytes undergo degeneration followed by 44 % oligodendrocytes, 26 % neurons and 10 % microglia. The pronounced astrocytic apoptosis resulted from the LPS-induced glutamate excitotoxicity leading to their hyperactivation as evident from their hypertrophied morphology, glutamate transporter 1 upregulation and downregulation of GS. Therapeutic minocycline treatment to LPS-infused rats efficiently restricted the inflammatory response and degeneration of other cell types but could not significantly combat with the apoptosis of astrocytes. Our study demonstrates a novel finding on cellular degeneration in the hippocampus revealing more of astrocytic death and suggests a more careful consideration on the protective efficacy of minocycline.

RESUMEN

PURPOSE: The aim of this study was to report cancer statistics in Kamrup Urban District, including incidence and, mortality. INTRODUCTION: In the last five year PBCR-Guwahati witnessed a remarkable growth in cancer incidence cases. The number of new cases of all cancer was increased from 155.3 to 188.5 and 102.7 to 165.3 per 100,000 men and women respectively from the year 2007 to 2011 in KUD. The data from KUD also have shown that for some of the specific types of cancer are highest or some of the highest incidence in rates in the world; particularly cancers of upper aero-digestive tract consist of anatomical sites such as oral cavity, hypopharynx, larynx gallbladder, stomach, lung, prostate and oesophageal cancer. MATERIALS AND METHODS: Age-standardized rates (ASR) (per 100,000 person-years) for incidence, mortality were calculated using the World Standard Population as proposed by Segi and modified by Doll et al. Descriptive statistics were presented by tables and figures. RESULTS: A total of 6623 number of cases (male = 3809, female = 2814) were diagnosed with cancer in the last five years (2007-2011) period of time. The overall age standardized cancer incidence rate is almost 21% higher in men than in women. The pooled ASR for the five year period is 175.2 and 144.7 per 100,000 men and women. CONCLUSION: Overall cancer incidence and mortality rates have increased since 2007.
