RESUMEN

Global warming and glacier retreat have significant impacts on the structure and function of natural ecosystems. However, little is known about how glacier retreat affects the long-term evolution of ecosystems at high-altitude regions. In this study, we explored the possible effects of glacier retreat on catchment vegetation and lake productivity in Lake Puma Yumco, southeastern Tibetan Plateau, based on detailed organic molecular compositions determined by an ultra-high-resolution Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS), and combined with various sedimentary geochemical indicators. The glaciers in the catchment keep retreating since 1870 CE, as inferred from the multiple indices of total organic carbon content (TOC), total nitrogen content (TN), C/N ratios, and carbonate contents. Accompanying modern global warming and glacier shrinkage, the relative abundance of soil- and vegetation-derived large molecular compounds (e.g., vascular plant-derived polyphenols, highly unsaturated and phenolic compounds, and condensed aromatics) increased gradually in lake sediments, suggesting that ice-covered land was exposed under warming condition, and gradually revegetation occurred. Both increases in relative abundance of nitrogen-containing compounds (e.g., CHNO) and chlorophyll derivative contents in the lake sediments were observed since 1870 CE, suggesting that stronger catchment weathering and increasing terrestrial nutrient loads enhanced the downstream lake productivity after glacier retreat. Our results imply that continued global warming and alpine glacier retreat in the future may further promote vegetation expansion and increases in lake productivity on the Tibetan Plateau.

BACKGROUND: Cripto-1 (CR-1) has been reported to be involved in the development of several human cancers. The potential role of CR-1 in clear cell renal cell carcinoma (ccRCC) is still not clear. METHODS: CR-1 expression was evaluated in ccRCC tissues by Real-time quantitative PCR, Western blot and immunohistochemistry. Serum levels of CR-1 were tested by enzyme-linked immunosorbent assay (ELISA). The clinical significance of CR-1 was analyzed. The effects of CR-1 on cell proliferation, migration, invasion and angiogenesis were investigated in ccRCC cell lines in vitro and in vivo, and markers of the epithelial -mesenchymal transition (EMT) were analyzed. The impact of CR-1 on Wnt/ß-catenin signaling pathway was also evaluated in vitro and in vivo. RESULTS: CR-1 expression was elevated in ccRCC tumor tissues and serum samples. CR-1 expression was correlated with aggressive tumor phenotype and poor survival. Ectopic expression of CR-1 significantly promoted cell proliferation, migration, invasion and angiogenesis whereas knockdown of CR-1 inhibited these activities both in vitro and in vivo. Moreover, we found that CR-1 induced EMT and activated Wnt/ß-catenin signaling pathway both in vitro and in vivo. CONCLUSIONS: These results suggest that CR-1 is likely to play important roles in ccRCC development and progression, and that CR-1 is a prognostic biomarker and a promising therapeutic target for ccRCC.

Based on a quantitative analysis of nitrate concentrations, the nitrate sources and temporal variability of the rivers, lakes, and wetlands of Tibet were assessed for the first time using dual isotope technology. Water samples were collected once in July 2017 for analysis of nitrate concentration and isotopic composition. The overall values of δ15N-NO3- and δ18O-NO3- ranged from +1.8‰ to +23.0‰, and from -6.3‰ to +22.2‰ respectively. Duel isotopic composition suggested that nitrification of soil organic nitrogen was the main source of nitrate in the Yalu Tsangpo River. Furthermore, anthropogenic nitrogen inputs become more important in downstream than upstream because of intensive agricultural activities and urban input. In the rivers of the Ngari District, nitrate is mainly derived from desert deposits, manure and sewage, and chemical fertilisers. Different rivers show different characteristics of nitrate sources depending on the location, topography, landform, and climate of the river basins. Animal manure, nitrification of soil organic matter, and desert deposits are mainly responsible for the shifting of nitrate isotopic signatures in lakes, which are minimally affected by human activities. In wetlands, biological nitrification and denitrification could be the main processes of nitrogen migration and transformation. These results provide useful information in revealing the fate of nitrate in different aquatic ecosystems and different areas of Tibet.

OBJECTIVE: To biomechanical analysis of the correlation between sacral tilt displacement and L5-S1 disc degeneration. METHODS: From July 2011 to July 2013, 81 patients with lumbar disc herniation and sacroiliac joint disorder including 45 males and 36 females with an average age of (45.39±1.30) years ranging from 18 to 65 years old were selected. The course of the disease ranged from 1 to 144 months with an average of (12.64±2.19) months. All patients were taken lumbar spine lateral X-ray films, the lumbar curvature angle, L4-L5 or L5-S1 intervertebral gap distance between points, and the lumbosacral angle was measured and correlated analyzed. RESULTS: The lumbar curvature of female patients with L5S1DH were significantly larger than male patients [(22.18±8.62)° vs (16.17±4.97)°, P<0.05]. Lumbar curvature and lumbosacral angle showed a positive correlation in LDH (R=0.48, P<0.01,y=7.25+0.38x, P<0.01); Male patients with L4-5DH were more obvious (R=0.55, P<0.05, y=5.80+0.43x, P<0.01); Female patients with L5S1DH were particularly evident(R=0.74, P<0.01,y=0.91x-5.30, P<0.01). The lumbosacral angle and L4-5 intervertebral gap was a positive correlation in L4-5DH(R=0.27, P<0.05); While L5-S1 intervertebral gap and lumbosacral angle were not correlated(P>0.05) in L5S1DH. CONCLUSIONS: The sacral tilt displacement and L5-S1 disc degeneration were closely related to provide a new understanding philosophy and therapeutic approach for clinical treatment of intractable lumbar L5S1DH.

OBJECTIVE: To study the biomechanical relationship between iliac rotation displacement and L(4,5) disc degeneration, and to provide clinical evidences for the prevention and treatment of L(4,5) disc degeneration and herniation. METHODS: From March 2012 to February 2014,68 patients with lumbar disc herniation combined with sacroiliac joint disorders were selected. Among them, 42 patients with L(4,5) disc herniation combined with sacroiliac joint disorders included 22 males and 20 females, ranging in age from 19 to 63 years old, with an average of (51.78 +/- 20.18) years old, and the duration of the disease ranged from 1 to 126 months with an average of (11.18 +/- 9.23) months. Twenty-six patients with L5S1 disc herniation combined with sacroiliac joint disorders included 11 males and 15 females, ranging in age from18 to 65 years old with an average of (45.53 +/- 27.23) years old, and the duration of the disease ranged from 0.5 to 103 months with an average of (11.99 +/- 12.56) months. Sixty-eight anteroposterior lumbar radiographs, 68 lateral lumbar radiographs,and 68 pelvic plain films were taken. The degree of lumbar scoliosis, pelvic tilt,and disc thickness were measured. The correlation between pelvic tilt and lumbar scoliosis ,lumbar scoliosis and disc thickness were studied by using linear and regression methods. The hiomechanical analysis was performed. RESULTS: There was a positive correlation between pelvic tilt and lumbar scoliosis in patients with L(4,5) disk herniation (R=0.49, P=0.00). There was a causal relationship and good linear proportional relationship (Y=3.05+1.07X, P=0.00) in the two variables. There was a negative correlation between lumbar scoliosis and intervertebral space in male patients with L (4,5) disk herniation (R = -0.50, P=0.01). There was a causal relationship and good linear proportional relationship in the two variables (Y=13.09-0.27X, P=0.02). But there was a positive correlation between lumbar scoliosis and intervertebral space in male patients with L5S1 disk herniation (R=0.46, P=0.04). CONCLUSION: Iliac rotational displacement are closely related with L(4,5) disc degeneration and herniation in biomechanics. A new concepts and therapeutic approach is provided for clinical treatment of chronic and refractory herniation of L(4,5) disc in patients

Stability of short-cut nitrification nitrogen removal performance was studied in a step-feeding, intermittently aerated sequencing batch reactor (IASBR) at 30°C to treat digested piggery wastewater. Results showed that the nitrogen removal was greatly influenced by the ratio of chemical oxygen demand (COD) to total nitrogen (TN) in the influent. Nitrite nitrogen kept accumulating up to 800 mg · L⁻1 when the influent COD/TN ratio was 0.8 ± 0.2, and the removal rates of TN, ammonium nitrogen and total organic carbon (TOC) were only 18.3% ± 12.2%, 84.2% ± 10.3% and 60.7% ± 10.7%, respectively. By contrast, as the influent COD/ TN ratio was increased to 2.4 ± 0.5, the accumulated concentration of nitrite nitrogen sharply decreased from 800 mg · L⁻¹ to below 10 mg-L⁻¹, and the removal rates of TN, ammonium nitrogen and TOC were increased to over 90%, 95% and 85%, respectively. Gradually shortened hydraulic retention time ( HRT) reveales that the ammonia load is a restricting factor for nitrogen removal. The ammonia load should be controlled at no more than 0.30 kg · (m³ · d) ⁻¹, or else, the removal rates of TN, ammonium and TOC would be greatly decreased. The nitrite accumulation rate over the whole run was 74.6%-97.8% and the TN removal rate in the stable phase was over 90%. With efficient and stable short-cut nitrification-denitrification in a low COD/TN, moreover, and unnecessary for addition of alkaline, IASBR shows great advantage for treating wastewater with high concentration of ammonia while low COD/TN ratio.

RESUMEN

Network alignment is a computational technique to identify topological similarity of graph data by mapping link patterns. In bioinformatics, network alignment algorithms have been applied to protein-protein interaction (PPI) networks to discover evolutionarily conserved substructures at the system level. In particular, local network alignment of PPI networks searches for conserved functional components between species and predicts unknown protein complexes and signaling pathways. In this article, we present a novel approach of local network alignment by semantic mapping. While most previous methods find protein matches between species by sequence homology, our approach uses semantic similarity. Given Gene Ontology (GO) and its annotation data, we estimate functional closeness between two proteins by measuring their semantic similarity. We adopted a new semantic similarity measure, simVICD, which has the best performance for PPI validation and functional match. We tested alignment between the PPI networks of well-studied yeast protein complexes and the genome-wide PPI network of human in order to predict human protein complexes. The experimental results demonstrate that our approach has higher accuracy in protein complex prediction than graph clustering algorithms, and higher efficiency than previous network alignment algorithms.

With high concentrations of chemical oxygen demand (COD) and ammonium while low ratio of COD to total nitrogen (TN), digested piggery wastewater is difficult to treat using conventional biological methods. In this study, a biofilm membrane bioreactor (BF-MBR) and a traditional type of membrane bioreactor (MBR) were parallel operated to treat digested piggery wastewater, and the pollutant removal performance were compared at influent COD/TN ratios of 1. 0 ± 0. 2 and 2. 3 ± 0. 4, respectively. The results showed that the effluent quality in both reactors was poor and unstable when the influent COD/TN ratio was 1. 0 ± 0. 2. The effluent quality and stability were greatly improved as the influent COD/TN ratio was increased to 2. 3 ± 0. 4. The removal rates of COD and ammonium were respectively 92. 3% ± 2. 4% and 97. 5% ± 4. 1% in BF-MBR, slightly higher than 91. 9% ± 1. 5% and 91. 2% ± 14. 0% in MBR. Benefited from the biofilm, 36. 7% ± 19. 5% of TN and 54. 0% ± 18. 9% of TP were removed by BF-MBR, significantly higher than the respective values of 19. 2% ± 12. 4% and 29. 0% ± 18. 1% by MBR. Moreover, BF-MBR consumed less than 40% of the alkaline chemicals as MBR. BF-MBR was considered more suitable for treatment of digested piggery wastewater due to its better pollutant removal performance and low consumption of alkaline.

METHODS: From August 2009 to October 2010,129 cases with lumbar disc herniation were studied with epidemiological methods. Among them, 61 patients with L4, disc herniation included 37 males and 24 females, ranging in aged from 20 to 75 years old, duration of the disease ranged from 1 to 144 months; The other 68 patients with L5S1 disc herniation included 32 males and 36 females,ranging in aged froml8 to 76 years old,duration of the disease ranged from 0.5 to 240 months. The clinical data, symptoms and signs,X-ray characteristics of lumbar spine and pelvis of the patients were investigated by epidemiological. The risk of lumbar disc herniation was calculated with case-control study; independent variables were screened with single factor analysis; the risk factors for lumbar disc herniation were determined with logistic regression analysis, and biomechanics analyses were taken. RESULTS: Among 129 patients with lumbar disc herniation, 88 cases associated with sacroiliac joint disorders, sacroiliac joint disorder was a risk factor of lumbar disc herniation (OR = 4.61, P = 0.00); 47 cases associated with sacroiliac joint disorders in 61 patients with L4,5 disc herniation, iliac crest uneven caused by iliac rotational displacement was a high risk factor of L4,5 disc herniation (OR = 11.27, P = 0.00); 41 cases associated with sacroiliac joint disorders in 68 patients with L5S1 disc herniation, lumbar sacral angle abnormalities caused by sacral tilt shift was a high risk factor L5S1 disc herniation (OR = 2.31, P = 0.03). CONCLUSION: Lumbar disc herniation and sacroiliac joint disorder are two of fallot, the two factors affect each other and there is a causal relationship. They are common exists in low back pain.

OBJECTIVE: To observe the influence of electroacupuncture (EA) therapy on the expression of tyrosine hydroxylase (TH) and cyclooxygenase-2 (COX-2) proteins of Substantia Nigra cells(SNc) in the rotenone-induced Parkinson's disease(PD) rats, so as to explore the mechanism of EA underlying improvement of PD. METHODS: Forty rats were randomly divided into normal, sham-operation (sham), model and EA groups (n = 10/group). The PD model was established by successive subcutaneous injection of rotenone (highly selective lesions of nigrostriatal dopaminergic neurons) for 28 days. EA (2 Hz, 1 mA) was applied to bilateral "Fengfu" (GV 16) and "Taichong" (LR 3) for 20 min, once daily for 14 days. The expression levels of TH and COX-2 proteins in the Substantia Nigra of midbrain were detected with Western blotting. RESULTS: Compared with the normal group, the expression level of TH protein in the model group was significantly decreased (P < 0.01), and that of COX-2 protein in the model group was significantly increased (P < 0.01). After the EA treatment, the expression level of TH in the EA group was obviously upregulated (P < 0.01), and that of COX-2 protein in the EA group was considerably down-regulated (P < 0.01). No significant differences were found between the normal and sham groups in the expression levels of TH and COX-2 proteins (P > 0.05). CONCLUSION: EA therapy can decrease inflammation mediator COX-2 protein expression and upregulate TH protein expression in the Substantia Nigra of midbrain in PD rats, which may contribute to its effect in relieving PD in clinic.

OBJECTIVE: To study the X-ray characteristics of sacroiliac joint disorders and its clinical significance,so as to provide clinical diagnosis basis for Tuina treatment of sacroiliac joint disorder. METHODS: From July 2009 to March 2011,104 patients with sacroiliac joint disorder were reviewed,including 64 males and 40 females,ranging in age from 18 to 81 years, with an average of (45.39 +/- 1.30) years. The duration of the disease ranged from 1 to 144 months,with an average of (12.64 +/- 2.19) months. One hundred and four pelvic plain films and 97 lumbar spine lateral films of the patients with sacroiliac joint disorder were taken. On the lateral X-ray of lumbar,the sacral horizontal angles (lumbosacral angle) were measured; and on the X-ray of pelvis,the vertical distance of two side iliac crest (iliac crest difference), the distance from lateral border to medial margin of two hips (hip width),the clip angle between sacral spin connection and vertical axis were measured,and then the data were analyzed. RESULTS: The mean difference of iliac crest was (10.34+/-0.73) mm; the mean width difference of hip'was (6.73+/-1.01) mm; and the mean difference of the iliac crest was larger than that of mean difference of hip (P<0.01). The occurrence rate of inequal width of hip was higher(P<0.01). The mean abnormal lumbosacral angle was (7.29 +/- 1.86) degrees,and the mean angle of sacral crest tilting to left or right was (3.18 +/- 0.47) degrees; the mean abnormal lumbosacral angle was larger than that of angle of sacral crest tilting to left or right (P<0.01), and the occurrence rate of sacral crest tilting to left or right was higher

OBJECTIVE: To investigate the expressions of matrix metalloprotein-2 (MMP-2) and tissue inhibitor of metallopeptidase inhibitor-1 (TIMP-1) in the renal allografts of patients with chronic active antibody-mediated rejection (ABMR), and explore their role in the pathogenesis of ABMR. METHODS: Immunohistochemistry and computer-assisted image analysis were used to detect the expression of MMP-2 and TIMP-1 in the renal allografts of 46 patients with interstitial fibrosis and tubular atrophy (IF/TA), with 15 normal renal tissue specimens as the control. The association of MMP-2 and TIMP-1 with the pathological grade of IF/TA in ABMR was analyzed. RESULTS: The expressions of MMP-2 and TIMP-1 significantly increased in the renal tissues of the patients as compared with the normal renal tissues (P<0.05). MMP-2 expression tended to decrease, while TIMP-1 and serum creatinine increased with the pathological grades of IF/TA (P<0.05). In IF/TA group, the expression of TIMP-1 was positively correlated to serum creatinine level (r=0.718, P=0.00<0.05). CONCLUSION: Abnormal expressions of MMP-2 and TIMP-1 can promote the development of renal fibrosis in chronic ABMR.

OBJECTIVE: To explore the mechanism of electroacupuncture therapy on Parkinson's disease (PD). METHODS: Fifty Wistar rats were randomly divided into a normal group, a sham-operation group, a model group, a Fengfu-Taichong group and a Shuanggu Yitong group. PD model was duplicated by microinjection of 6-Hydroxyl-Dopamine into right corpora striata, and by microinjection of normal saline in sham-operation group. Rats in normal group, sham-operation group and model group were not treated. In Fengfu-Taichong group, the rats were treated by electroacupuncture at "Fengfu" (GV 16) and "Taichong" (LR 3) on the basis of the PD model, and by electroacupuncture at "Fengfu" (GV 16), "Taichong" (LR 3), "Guanyuan" (CV 4) and "Zusanli" (ST 36) in Shuanggu Yitong group, once daily for 2 weeks. GDNF and Ret expression were detected by immunohistochemistry and western blotting, respectively. RESULTS: The number of GDNF positive cells and the content of Ret receptor increased significantly in the two electroacupuncture groups compared with those in the other groups (all P < 0.01), and the expression of GDNF increased significantly in Shuanggu Yitong group compared with that in Fengfu-Taichong group (P < 0.01). CONCLUSION: Electroacupuncture can not only increase the expression of GDNF, but also enhance its effect. "Shuanggu Yitong" method is better than simple acupuncture at "Fengfu" (GV 16) and "Taichong" (LR 3) in increasing expression of GDNF.

OBJECTIVE: To observe the influence of electroacupuncture (EA) of "Fengfu" (GV16) and "Taichong" (LR3) on morphological changes and apoptotic percentage of substantia nigra cells so as to explore its underlying mechanism in relieving Parkinson's disease. METHODS: Forty Wistar rats were randomly divided into normal control, sham-operation (sham), model and EA groups, with 10 rats in each. The Parkinson's disease model was established by micro-injection of 6-hydroxyl-dopamine (6-OHDA) into the right striatum. EA (2 Hz, 1 mA) was applied to "Fengfu"(GV16) and "Taichong" (LR3) for 30 min, once daily for 2 weeks. HE staining, Nissl staining and immunohistochemical staining were conducted for observing the morphological changes of substantia nigra pars compacta (SNc) and striatum, and the tyrosine hydroxylase(TH) immuno-reaction (IR) positive neurons and nerve fibers. The apoptosis was detected by flow cytometry. RESULTS: Compared to the normal control and sham groups, the total number of TH-IR positive neurons and Nissl-stained cells, and OD value of TH-IR positive nerve fibers in the SNc on the 6-OHDA-lesioned side were significantly lower in the model group (P < 0.01). Compared with the model group, the aforementioned 3 indexes of the EA group were increased obviously (P < 0.01). The percentage of apoptosis of SNc on the 6-OHDA-lesioned side was obviously lower in the EA group than in the model group (P < 0.01), but still obviously higher than in the normal group and sham group (P < 0.01). CONCLUSION: Electroacupuncture therapy can significantly increase the number of neurons of substantia nigra and the density of striatum nerve fibers,and reduce the apoptotic percentage of substantia nigra cells in the Parkinsonian rats, which may contribute to its effect in relieving Parkinson's disease.
