RESUMEN

A new morphologically distinct species of cowry (family Cypraeidae Rafinesque, 1815) is described from the Late Pliocene Roe Calcarenite of the Roe Plains, Western Australia. Previously assigned to Umbiliahesitata (Iredale, 1916), the new species differs morphometrically from related taxa and is differentiated from U.hesitata by a number of shell features including a prominent, projecting protoconch, less extended posterior and anterior terminals, coarser columellar teeth extending onto the base, and well-developed, thickened anterior flanges, supporting a rounded anterior extremity with blunt anterior tips. Umbiliatomdarraghi sp. nov. is the third Umbilia species to be described from the Pliocene.

Multivariate approaches to morphological study of shell form have rarely been applied to cowries (Gastropoda: Cypraeidae) with preference, instead, for comparing formulaic notations of shell form that report averages (i.e., means) for key morphometrics such as shell dimensions, their ratios, and counts of apertural teeth. Although widely applied, the "shell formula" does not account for variation among individuals or support statistical comparison between taxa. This study applied a multivariate approach to analyse shell form within the four accepted subspecies of the cowrie, Umbiliaarmeniaca (Verco, 1912) and included a previously unstudied, and most northerly, population of U.armeniaca from Lancelin, Western Australia. Multivariate analyses readily separated the recognised subspecies of U.armeniaca (U.a.armeniaca, U.a.diprotodon, U.a.clarksoni and U.a.andreyi), but did not separate the Lancelin population from U.a.andreyi, indicating that the former represents a northward extension of U.a.andreyi that is not morphometrically distinguishable. These results provide improved understanding of infraspecific differences in shell form of U.armeniaca across its broad distribution, and demonstrate the utility of multivariate morphometric methods for statistical comparison of shell form between taxa. This approach is complimentary to existing research practices and has broad potential application in future morphometric studies of both extant and fossil taxa within the family Cypraeidae.

The sea cucumber Holothuria (Metriatyla) scabra, known as sandfish, is a high-value tropical echinoderm central to the global bêche-de-mer (BDM) trade. This species has been heavily exploited across its natural range, with overharvesting and ineffective fishery management leaving stocks in the Pacific region heavily depleted. In Fiji, sandfish stocks have not recovered since a 1988 harvest ban, with surveys reporting declining populations and recruitment failure. Therefore, to inform fishery management policy for the wild sandfish resource and to guide hatchery-based restocking efforts, a high-resolution genomic audit of Fijian populations was carried out. A total of 6,896 selectively-neutral and 186 putatively-adaptive genome-wide SNPs (DArTseq) together with an independent oceanographic particle dispersal model were used to investigate genetic structure, diversity, signatures of selection, relatedness and connectivity in six wild populations. Three genetically distinct populations were identified with shallow but significant differentiation (average Fst = 0.034, p≤0.05), comprising (1) Lakeba island (Lau archipelago), (2) Macuata (Vanua Levu), and (3) individuals from Yasawa, Ra, Serua island and Kadavu comprising the final unit. Small reductions in allelic diversity were observed in marginal populations in eastern Fiji (overall mean A = 1.956 vs. Lau, A = 1.912 and Macuata, A = 1.939). Signatures of putative local adaptation were also discovered in individuals from Lakeba island, suggesting that they be managed as a discrete unit. An isolation-by-distance model of genetic structure for Fijian sandfish is apparent, with population fragmentation occurring towards the east. Hatchery-based production of juveniles is promising for stock replenishment, however great care is required during broodstock source population selection and juvenile releases into source areas only. The successful use of genomic data here has the potential to be applied to other sea cucumber species in Fiji, and other regions involved in the global BDM trade. While preliminary insights into the genetic structure and connectivity of sandfish in Fiji have been obtained, further local, regional and distribution-wide investigations are required to better inform conservation efforts, wild stock management and hatchery-based restocking interventions for this valuable invertebrate.

Stage I phyllosoma larvae of the spiny lobster Panulirus ornatus hatch in tropical oceanic waters with limited and variable food resources. To better understand how these larvae cope with food deprivation, this study examined specific (mU mg-1 protein) and total (mU larva-1) activities of major digestive enzymes (i.e., α-amylase, non-specific esterase, trypsin-like protease) during a series of ex situ experiments. Specifically, temporal changes in enzyme profiles were examined in three experiments simulating scenarios in which phyllosoma hatch and begin development in an environment where (1) prey is either continuously present or absent; (2) prey is initially present, but subsequently absent; (3) prey is initially absent, but subsequently present. Results indicated that the accessibility of suitable prey provides an overarching influence on digestive enzyme activities and substrate utilisation in Stage I phyllosoma of P. ornatus, with enzymatic responses to both intermittent prey availability and food deprivation being influenced by their nutritional history. In the absence of prey, larval digestive enzyme activities (mU larva-1) initially remained static from hatch but eventually declined with extended food deprivation. When prey became available, enzyme activities increased, with delayed access to prey having minimal impact on this enzymatic response. Furthermore, phyllosoma were able to adjust α-amylase and non-specific esterase activities (within 8 h) in response to the disappearance of prey, demonstrating adaptive changes to endure periods of food deprivation that were based on their nutritional history. The ability of phyllosoma to regulate enzyme activities from hatch is an important physiological strategy allowing them to survive in an environment characterised by highly variable zooplankton biomass and abundance, and explains why diets that differ greatly in nutrient composition have consistently been found effective for rearing phyllosoma in captivity.

Seagrasses provide vital ecosystem services which include the accumulation and storage of carbon and nutrients in sediments and biomass. Despite their importance in climate change mitigation and adaptation, seagrass ecosystems have been poorly studied, particularly in the Pacific. Therefore, the present study assessed variability in sedimentary and vegetative C, N and P storage in three monospecific seagrass meadows (Halophila ovalis, Halodule pinifolia and Halodule uninervis), reporting baseline data for the first time. Sediment Corg stocks ranged from 31 to 47 Mg C ha-1 and varied (p < 0.001) between seagrass meadows, unvegetated areas and locations. Comparison of N and P storage between vegetated meadows and unvegetated areas revealed differences (p < 0.05); implying seagrass meadows function as C, N and P sinks. Differences in species composition and environmental conditions, may play a key role in estimating C, N and P stocks, which are valuable data for conservation and monitoring of seagrass ecosystems.

RESUMEN

A new morphologically distinct species of cowry (family Cypraeidae Rafinesque, 1815) is described from the Pliocene of Flinders Island, Tasmania. Austrocypraeajimgracei sp. nov. differs morphologically from other members of the genus and is particularly characterised by the development of a heavily callused labral margin, with a distinct marginal edge that bends up towards the dorsum centrally. This feature is unique within the genus. The new taxon is only the second known Austrocypraea from the Pliocene. A revised key to the known Austrocypraea fossil species is presented.

A unique feature of seagrass among other ecosystem services is to have high phytoremediation potential that is a cost-effective plant-based approach and environmentally friendly solution for metal contamination in coastal areas. The goal of this study was to assess the phytoremediation prospective of seagrass for Cu, Fe, Mn and Zn in Fiji Islands. Heavy metal content was measured in sediments and tissues of the seagrasses Halophila ovalis, Halodule pinifolia and Halodule uninervis to test for local-scale differences. The local study shows that metal concentration in sediment and seagrass tissue was significantly variable, regardless of species and sediment type. Sedimentary concentration of Cu, Fe, Mn and Zn obtained in the present study seemed to be lower than that of previous studies. The results support that H. ovalis is a good bioindicator species since it accumulated up to 5-fold more of these metals compared to the Halodule species.

Mortalities of bivalve larvae and spat linked with Vibrio spp. infection have been described in hatcheries since 1959, causing potential development of resistant bacteria. A reliable and sustainable solution to this problem is yet to be developed. Potential treatment of bacterial infection with bacteriophages is gaining interest in aquaculture as a more sustainable option for managing Vibrio spp. infection. This study assessed the effectiveness of bacteriophages (Φ-5, Φ-6, and Φ-7) against pathogenic Vibrio isolates (USC-26004 and USC-26005). These phage isolates were found to belong to the Myoviridae viral family. A total of 212 ORFs of Φ-5 were identified and annotated. The genome of this phage contained putative thymidine kinase and lysin enzyme. During infections with phages, the OD values of the isolates USC-26005 and USC-26004 remained stable at a much lower reading compared to the control after 9 h of incubation. Mortality rate of oyster (Saccostrea glomerata) larvae was 28.2 ± 3.5% in the bacteriophage treatment group, compared to 77.9 ± 9.1% in the bacterial treatment group after 24 h incubation. Findings of this study indicate that lytic phages might be utilized as potential bio-control agents of luminescent bacterial disease in oyster hatcheries.

Fisheries and aquaculture industries worldwide remain reliant on seed supply from wild populations, with their success and sustainability dependent on consistent larval recruitment. Larval dispersal and recruitment in the marine environment are complex processes, influenced by a multitude of physical and biological factors. Biophysical modelling has increasingly been used to investigate dispersal and recruitment dynamics, for optimising management of fisheries and aquaculture resources. In the Fiji Islands, culture of the black-lip pearl oyster (Pinctada margaritifera) is almost exclusively reliant on wild-caught juvenile oysters (spat), through a national spat collection programme. This study used a simple Lagrangian particle dispersal model to investigate current-driven larval dispersal patterns, identify potential larval settlement areas and compare simulated with physical spat-fall, to inform targeted spat collection efforts. Simulations successfully identified country-wide patterns of potential larval dispersal and settlement from 2012-2015, with east-west variations between bi-annual spawning peaks and circulation associated with El Niño Southern Oscillation. Localised regions of larval aggregation were also identified and compared to physical spat-fall recorded at 28 spat collector deployment locations. Significant and positive correlations at these sites across three separate spawning seasons (r(26) = 0.435; r(26) = 0.438; r(26) = 0.428 respectively, p = 0.02), suggest high utility of the model despite its simplicity, for informing future spat collector deployment. Simulation results will further optimise black-lip pearl oyster spat collection activity in Fiji by informing targeted collector deployments, while the model provides a versatile and highly informative toolset for the fishery management and aquaculture of other marine taxa with similar life histories.

Cultured microalgae are the primary food source for oyster larvae during hatchery culture and are a potential vector for Vibrio spp. infection of larval cultures. Bacteriophages have shown potential for controlling contamination of Vibrio spp. in aquaculture systems and their application could be an effective biological control method to eliminate such bacterial contamination of microalgae. This study investigated whether Vibrio-free microalgae sources could be ensured via the application of Vibrio specific phages. As a first step, four different Vibrio bacteriophages (belonging to the Myoviridae viral family) were isolated from marine waters in Queensland, Australia and used in challenge tests against a Vibrio host species, previously isolated from New South Wales oyster hatchery and found to be closely related to V. alginolyticus (ATCC 17749). The genome sequence of one of the four isolated bacteriophages, Vibrio Φ-2, that displayed strongest virulence against the host was determined. The 242446 bp genome of this bacteriophage was predicted to encode 217 proteins with an average GC content of 43.91%, containing putative thymidine kinases and a lysin enzyme. Application of these bacteriophages to pathogenic Vibrio spp. contaminating microalgae suspensions resulted in significant decreases in their numbers within 2 h. Findings indicated that direct application of bacteriophages to microalgae suspensions could be an effective method of reducing the occurrence of vibriosis in oyster hatcheries.

BACKGROUND: The black-lip rock oyster (Saccostrea echinata) has considerable potential for aquaculture throughout the tropics. Previous attempts to farm S. echinata failed due to an insufficient supply of wild spat; however, the prospect of hatchery-based aquaculture has stimulated renewed interest, and small-scale farming is underway across northern Australia and in New Caledonia. The absence of knowledge surrounding the population genetic structure of this species has raised concerns about the genetic impacts of this emerging aquaculture industry. This study is the first to examine population genetics of S. echinata and employs both mitochondrial cytochrome c oxidase subunit I gene (COI) and single nucleotide polymorphism (SNP) markers. RESULTS: The mitochondrial COI data set included 273 sequences of 594 base pair length, which comprised 74 haplotypes. The SNP data set included 27,887 filtered SNPs for 272 oysters and of these 31 SNPs were identified as candidate adaptive loci. Data from the mitochondrial COI analyses, supports a broad tropical Indo-Pacific distribution of S. echinata, and showed high haplotype and nucleotide diversities (0.887-1.000 and 0.005-0.008, respectively). Mitochondrial COI analyses also revealed a 'star-like' haplotype network, and significant and negative neutrality tests (Tajima's D = - 2.030, Fu's Fs = - 25.638, P < 0.001) support a recent population expansion after a bottleneck. The SNP analyses showed significant levels of population subdivision and four genetic clusters were identified: (1) the Noumea (New Caledonia) sample location; (2) the Bowen (north Queensland, Australia) sample location, and remaining sample locations in the Northern Territory, Australia (n = 8) were differentiated into two genetic clusters. These occurred at either side of the Wessel Islands and were termed (3) 'west' and (4) 'east' clusters, and two migrant individuals were detected between them. The SNP data showed a significant positive correlation between genetic and geographic distance (Mantel test, P < 0.001, R2 = 0.798) and supported isolation by distance. Three candidate adaptive SNPs were identified as occurring within known genes and gene ontology was well described for the sex peptide receptor gene. CONCLUSIONS: Data supports the existence of genetically distinct populations of S. echinata, suggesting that management of wild and farmed stocks should be based upon multiple management units. This research has made information on population genetic structure and connectivity available for a new aquaculture species.

A thorough understanding of livelihoods is necessary to ensure development policies are compatible with both resource conservation and the social and economic goals of development. Few studies, however, focus on value-adding activities occurring post-harvest in artisanal fisheries. The transformation of mollusc shells and skeletal remains of other marine taxa into artistic jewellery and decorative items is becoming an increasingly important livelihood activity for rural, coastal communities across the Pacific. We examine the potential challenges facing the shell-based handicraft sector and opportunities for overcoming these challenges using a quantitative study of artisans among the Tigak Islands of Papua New Guinea. The major challenges facing this livelihood sector are perceptions of marine resource declines and a lack of livelihood flexibility, attributed to the specialisation of material assets and skills. Improving market heterogeneity and developing coastal aquaculture may facilitate sustainable development of this livelihood sector.

To determine the effectivity of bacteriophages in controlling the mass mortality of striped catfish (Pangasianodonhypophthalmus) due to infections caused by Aeromonas spp. in Vietnamese fish farms, bacteriophages against pathogenic Aeromonashydrophila were isolated. A.hydrophila-phage 2 and A.hydrophila-phage 5 were successfully isolated from water samples from the Saigon River of Ho Chi Minh City, Vietnam. These phages, belonging to the Myoviridae family, were found to have broad activity spectra, even against the tested multiple-antibiotic-resistant Aeromonas isolates. The latent periods and burst size of phage 2 were 10 min and 213 PFU per infected host cell, respectively. The bacteriophages proved to be effective in inhibiting the growth of the Aeromonas spp. under laboratory conditions. Phage treatments applied to the pathogenic strains during infestation of catfish resulted in a significant improvement in the survival rates of the tested fishes, with up to 100% survival with MOI 100, compared to 18.3% survival observed in control experiments. These findings illustrate the potential for using phages as an effective bio-treatment method to control Motile Aeromonas Septicemia (MAS) in fish farms. This study provides further evidence towards the use of bacteriophages to effectively control disease in aquaculture operations.

A major difficulty in managing live organism wildlife trade is often the reliance on trade data to monitor exploitation of wild populations. Harvested organisms that die or are discarded before a point of sale are regularly not reported. For the global marine aquarium trade, identifying supply-chain losses is necessary to more accurately assess exploitation from trade data. We examined quality control rejections and mortality of marine invertebrates (Asteroidea, Gastropoda, Malacostraca, Ophiuroidea) moving through the Papua New Guinea marine aquarium supply-chain, from fisher to importer. Utilizing catch invoices and exporter mortality records we determined that, over a 160 day period, 38.6% of the total invertebrate catch (n = 13,299 individuals) was lost before export. Supply-chain losses were divided among invertebrates rejected in the quality control process (11.5%) and mortality of the accepted catch in transit to, and during holding at, an export facility (30.6%). A further 0.3% died during international transit to importers. We quantified supply-chain losses for the ten most fished species which accounted for 96.4% of the catch. Quality control rejections (n = 1533) were primarily explained by rejections of oversized invertebrates (83.2% of rejections). We suggest that enforceable size limits on species prone to size-based rejections and elimination of village-based holding of invertebrates would reduce losses along the Papua New Guinea supply-chain. This case study underscores that low mortality during international transit may mask large losses along supply-chains prior to export and exemplifies the limitations of trade data to accurately monitor exploitation.

Evolutionary relationships in the black-lip pearl oyster Pinctada margaritifera which is highly valued for pearl production remain poorly understood. This species possesses an 18,000 km Indo-Pacific natural distribution, and its current description includes six subspecies defined exclusively on morphological characters. To evaluate its taxonomic identity using molecular data, 14 populations in both the Indian and Pacific Oceans (n = 69), and the congeneric taxa P. maxima and P. mazatlanica (n = 29 and n = 10, respectively) were sampled. Phylogenomic reconstruction was carried out using both 8,308 genome-wide SNPs and 10,000 dominant loci (DArTseq PAVs). Reconstructions using neighbour-joining (Nei's 1972 distances), maximum likelihood and Bayesian approaches all indicate that the taxonomy of P. margaritifera is quite complex, with distinct evolutionary significant units (ESUs) identified within Tanzanian and Iranian populations. Contrastingly, phylogenies generated for Pacific Ocean oysters resolved a large monophyletic clade, suggesting little support for two current morphological subspecies classifications. Furthermore, P. mazatlanica formed a basal clade closest to French Polynesian P. margaritifera, suggesting it may be conspecific. Collectively, these findings provide evidence that P. margaritifera comprises a species complex, perhaps as a result of population fragmentation and increased divergence at range limits.

Bacterial contamination on seafood resulting from unhygienic food-handling practices causes foodborne diseases and significant revenue losses. Moreover, control measures are complicated by a high prevalence of antibiotic-resistant bacteria. Alternative measures such as the phage therapy, therefore, is considered as an environmental and consumer-friendly biological control strategy for controlling such bacterial contamination. In this study, we determined the effectiveness of a bacteriophage cocktail in controlling E. coli strains [JM 109, ATCC 13706 and the, extended spectrum beta-lactamase resistant strain (ATCC BAA 196)] and S. enterica subsp. enterica (ATCC 13311) as single and combined contaminants of the edible oysters. Five different E. coli-specific phages (belonging to the Siphoviridae family) and a Salmonella phage (belonging to the Tectiviridae family) were successfully isolated from sewage water samples taken from a local sewage treatment plan in the Sunshine Coast region of Australia. Phage treatments applied to the pathogens when they were presented on the oysters as either single or combined hosts, resulted in significant decrease of the number of these bacteria on edible oysters. Results obtained indicated that bacteriophages could have beneficial applications in oyster-processing plants in controlling pathogenic bacterial infestations. This study thus contributes towards ongoing international efforts into the effective use of bacteriophages for biological control purposes.

BACKGROUND: Genetic structure in many widely-distributed broadcast spawning marine invertebrates remains poorly understood, posing substantial challenges for their fishery management, conservation and aquaculture. Under the Core-Periphery Hypothesis (CPH), genetic diversity is expected to be highest at the centre of a species' distribution, progressively decreasing with increased differentiation towards outer range limits, as populations become increasingly isolated, fragmented and locally adapted. The unique life history characteristics of many marine invertebrates such as high dispersal rates, stochastic survival and variable recruitment are also likely to influence how populations are organised. To examine the microevolutionary forces influencing population structure, connectivity and adaptive variation in a highly-dispersive bivalve, populations of the black-lip pearl oyster Pinctada margaritifera were examined across its ~18,000 km Indo-Pacific distribution. RESULTS: Analyses utilising 9,624 genome-wide SNPs and 580 oysters, discovered differing patterns of significant and substantial broad-scale genetic structure between the Indian and Pacific Ocean basins. Indian Ocean populations were markedly divergent (F st = 0.2534-0.4177, p < 0.001), compared to Pacific Ocean oysters, where basin-wide gene flow was much higher (F st = 0.0007-0.1090, p < 0.001). Partitioning of genetic diversity (hierarchical AMOVA) attributed 18.1% of variance between ocean basins, whereas greater proportions were resolved within samples and populations (45.8% and 35.7% respectively). Visualisation of population structure at selectively neutral loci resolved three and five discrete genetic clusters for the Indian and Pacific Oceans respectively. Evaluation of genetic structure at adaptive loci for Pacific populations (89 SNPs under directional selection; F st = 0.1012-0.4371, FDR = 0.05), revealed five clusters identical to those detected at neutral SNPs, suggesting environmental heterogeneity within the Pacific. Patterns of structure and connectivity were supported by Mantel tests of isolation by distance (IBD) and independent hydrodynamic particle dispersal simulations. CONCLUSIONS: It is evident that genetic structure and connectivity across the natural range of P. margaritifera is highly complex, and produced by the interaction of ocean currents, IBD and seascape features at a broad scale, together with habitat geomorphology and local adaptation at regional levels. Overall population organisation is far more elaborate than generalised CPH predictions, however valuable insights for regional fishery management, and a greater understanding of range-wide genetic structure in a highly-dispersive marine invertebrate have been gained.

Fishery management and conservation of marine species increasingly relies on genetic data to delineate biologically relevant stock boundaries. Unfortunately for high gene flow species which may display low, but statistically significant population structure, there is no clear consensus on the level of differentiation required to resolve distinct stocks. The use of fine-scale neutral and adaptive variation, considered together with environmental data can offer additional insights to this problem. Genome-wide genetic data (4,123 SNPs), together with an independent hydrodynamic particle dispersal model were used to inform farm and fishery management in the Fijian black-lip pearl oyster Pinctada margaritifera, where comprehensive fishery management is lacking, and the sustainability of exploitation uncertain. Weak fine-scale patterns of population structure were detected, indicative of broad-scale panmixia among wild oysters, while a hatchery-sourced farmed population exhibited a higher degree of genetic divergence (Fst = 0.0850-0.102). This hatchery-produced population had also experienced a bottleneck (NeLD = 5.1; 95% C.I. = [5.1-5.3]); compared to infinite NeLD estimates for all wild oysters. Simulation of larval transport pathways confirmed the existence of broad-scale mixture by surface ocean currents, correlating well with fine-scale patterns of population structuring. Fst outlier tests failed to detect large numbers of loci supportive of selection, with 2-5 directional outlier SNPs identified (average Fst = 0.116). The lack of biologically significant population genetic structure, absence of evidence for local adaptation and larval dispersal simulation, all indicate the existence of a single genetic stock of P. margaritifera in the Fiji Islands. This approach using independent genomic and oceanographic tools has allowed fundamental insights into stock structure in this species, with transferability to other highly-dispersive marine taxa for their conservation and management.

A major difficulty in managing wildlife trade is the reliance on trade data (rather than capture data) to monitor exploitation of wild populations. Collected organisms that die or are rejected before a point of sale often go unreported. For the global marine aquarium trade, identifying the loss of collected fish from rejection, prior to export, is a first step in assessing true collection levels. This study takes a detailed look at fish rejections by buyers before export using the Papua New Guinea marine aquarium fishery as a case study. Utilizing collection invoices detailing the species and quantity of fish (Actinopteri and Elasmobranchii) accepted or rejected by the exporting company it was determined that, over a six month period, 24.2% of the total fish catch reported (n = 13,886) was rejected. Of the ten most collected fish families, rejection frequency was highest for the Apogonidae (54.2%), Chaetodontidae (26.3%), and Acanthuridae (18.2%) and lowest for Labridae (6.6%) and Hemiscylliidae (0.7%). The most frequently cited reasons for rejection were fin damage (45.6% of cases), undersized fish (21.8%), and fish deemed too thin (11.1%). Despite fishers receiving feedback on invoices explaining rejections, there was no improvement in rejection frequencies over time (r = -0.33, P = 0.15) with weekly rejection frequencies being highly inconsistent (range: 2.8% to 79.4%; s = 16.3%). These findings suggest that export/import statistics can greatly underestimate collection for the marine aquarium trade as additional factors such as fisher discards, escapees, post-collection mortalities, and unregulated domestic trade would further contribute to this disparity.
