RESUMEN

The Global Atmospheric Passive Sampling (GAPS) network, initiated in 2005 across 55 global sites, supports the global monitoring plan (GMP) of the Stockholm Convention on Persistent Organic Pollutants (POPs) by providing information on POP concentrations in air on a global scale. These data inform assessments of the long-range transport potential of POPs and the effectiveness evaluation of chemical regulation efforts, by observing changes in concentrations over time. Currently, measurements spanning 5-10 sampling years are available for 40 sites from the GAPS Network. This study was the first time that POP concentrations in air were reported on a global scale for an extended time period and the first to evaluate worldwide trends with an internally consistent sample set. For consistency between sampling years, site- and sample specific sampling rates were calculated with a new, public online model, which accounts for the effects of wind speed variability. Concentrations for legacy POPs in air between 2005 and 2014 show different trends for different organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs). The POPs discussed in this study were chosen due to being the most frequently detected, with detection at the majority of sites. PCB, endosulfan, and hexachlorocyclohexane (HCH) concentrations in air are decreasing at most sites. The global trends reflect global sources and recycling of HCH, ongoing emissions from old stockpiles for PCBs, and recent use restrictions for endosulfan. These chlorinated OCPs continue to present exposure threat to humans and ecosystems worldwide. Concentrations of other OCPs, such as chlordanes, heptachlor and dieldrin, are steady and/or declining slowly at the majority of sites, reflecting a transition from primary to secondary sources (i.e., re-emission from reservoirs where these POPs have accumulated historically) which now control ambient air burdens.

A retrospective analysis of a comprehensive series of high-volume air samples (n = 70) collected during 2010-2011 in Toronto (Canada) was performed. Seven UV compounds were analyzed by gas chromatography-tandem mass spectrometry (GC-MS/MS) with sum of concentrations (gas + particle phase) ranging from 80 to 2030 pg/m3. Homosalate (HMS) was the most prevalent organic UV-filter in air (47% of the total concentration), followed by 2-ethylhexyl salicylate (EHS, ∼29%), E- and Z-2-ethylhexyl 4-methoxycinnamate (EHMC, ∼17%). Ambient air (gas + particle phase) concentrations of organic UV-filters showed a strong seasonality, with peak levels during the summer. An analysis of Clausius-Clapeyron slopes indicated that much of the ambient burden of organic UV-filters are explained by volatilization from terrestrial and aquatic surfaces and supplemented with human activities and use of lotions and sunscreens, containing organic UV-filters, in addition to its use in plastics, textiles, paints, and pesticides. The results showed that organic UV-filters exist mainly in the gas phase with some exceptions, for instance, octocrylene (OCR), which was associated with both gas and particle phases, and avobenzone (AVB), which was predominantly in the particle phase. Lastly, this study revealed the need for basic physical chemical property data for organic UV-filters, including information on transformation rates and products, for better evaluating their environmental fate and effects.

Passive air sampling was conducted in Toronto and the Greater Toronto Area from 2016 to 2017 for 6 periods, in order to investigate ambient levels of polycyclic aromatic compounds (PACs) associated with different source types. The selected sampling sites (n = 8) cover geographical areas with varying source emissions including background, traffic, urban, industrial and residential sites. Passive air samples were analyzed for PACs which include PAHs, alkylated PAHs (alk-PAHs), dibenzothiophene and alkylated dibenzothiophenes (DBTs) and results for PAHs were used to calculate inhalation cancer risks using different approaches. The samples were also characterized for PAH derivatives including nitrated PAHs (NPAHs) and oxygenated PAHs (OPAHs). Concentrations of Σalk-PAHs and DBTs, which are known to be enriched in fossil fuels, as well as ΣNPAHs, were highest at a traffic site (MECP) located adjacent to the 18-lane Highway 401 that runs across Toronto. Except for an industrial site (HH/BU), PAC compositions were similar across the sampling sites with Σalk-PAHs being the most abundant class of PACs suggesting traffic emission was a major contributor to PACs in the atmosphere of Toronto. The industrial site exhibited a distinct chemical composition with ΣPAHs dominating over Σalk-PAHs and with elevated levels of fluoranthene, 9-nitroanthracene, and 9,10-anthraquinone, which likely reflects emissions from nearby industrial sources. MECP and HH/BU exhibited higher lifetime excess inhalation cancer risks indicating an association with traffic and industrial sources. The importance of the traffic sector as a source of PACs to ambient air is further supported by strong correlations of the ΣPAHs, Σalk-PAHs, DBTs, and ΣOPAHs with NOx. This study highlights the importance of traffic as an emission source of PACs to urban air and the relevance of PAC classes other than just unsubstituted PAHs that are important but currently not included in air quality guidelines or for assessing inhalation cancer risks.

Organic pollutants have been monitored in the atmosphere of the Great Lake Basin (GLB) since the 1990s in support of the Canada-US Great Lakes Water Quality Agreement and to determine the effectiveness of source reduction measures and factors influencing air concentrations. Air samples were collected between 2005 and 2014 at three sites with different geographical characteristics (Burnt Island, Egbert and Point Petre) in the Canadian GLB using high-volume air samplers and the air samples were analyzed for polybrominated diphenyl ethers (PBDEs) and several other non-PBDE halogenated flame retardants (HFRs). Spatial and temporal trends of total concentrations of HFRs were examined. BDE-47, BDE-99, and BDE-209 were the dominant PBDE congeners found at the three sites. For the non-PBDE HFRs, allyl 2,4,6-tribromophenyl ether (TBP-AE), hexabromobenzene (HBBz), pentabromotoluene (PBT), anti-dechlorane plus (anti-DDC-CO) and syn-dechlorane plus (syn-DDC-CO) were frequently detected. High atmospheric concentrations of PBDEs were found at the Egbert site with a larger population, while lower levels of PBDEs were detected at Point Petre, which is close to urban centers where control measures are in place. The strong temperature dependence of air concentrations indicates that volatilization from local sources influences atmospheric concentrations of BDE-28 and BDE-47 at Point Petre and Burnt Island, while long-range atmospheric transport (LRAT) was important for BDE-99. However, a weaker correlation was observed between air concentrations and ambient temperature for non-PBDE HFRs such as TBP-AE and HBBz. Atmospheric PBDE concentrations are decreasing slowly, with half-lives in the range of 2-16 years. Faster declining trends of PBDEs were observed at Point Petre rather than at Burnt Island. As Point Petre is closer to urban centers, faster declining trends may reflect the phase out of technical BDE mixtures in urban centers while LRAT influences the air concentrations at Burnt Island. The levels of syn-DDC-CO and anti-DDC-CO are decreasing at Point Petre and the levels of other non-PBDE HFRs such as TBP-AE, PBT and HBBz are increasing. Long-term declining trends of PBDEs suggest that regulatory efforts to reduce emissions to the GLB environment have been effective but that continuous measurements are required to gain a better understanding of the trends of emerging chemicals in the atmosphere of the GLB.

Extracts of passive air samples collected from 15 passive sampling network sites across the Athabasca Oil Sands region were used to explore the application of in vitro assays for mutagenicity (Salmonella mutation assays) and cytotoxicity (lactate dehydrogenase assay) to assess the toxicity of the air mixture. The air monitoring of polycyclic aromatic compounds (PACs) and PAC transformation products, including nitrated polycyclic aromatic hydrocarbons (NPAHs) and oxygenated polycyclic aromatic hydrocarbons (OPAHs) was then linked to the potential toxicity of air. The PACs in air during April to May 2014 were elevated near mining activities and declined with distance from the source region, whereas NPAHs and OPAHs exhibited a more variable spatial distribution with the highest levels in Fort McMurray. Overall, the air samples exhibited a weak mutagenicity. The highest indirect-acting mutagenicity was observed for sites closest to mining activities; however, the indirect-acting mutagenicity did not decline sharply with distance from mining areas. Indirect-acting mutagenicity was strongly correlated with levels of total PACs, benzo(a)pyrene equivalent mass, and OPAHs. Most of the samples exhibited cytotoxic potential, but the magnitude of the response was variable across the sample region and did not correlate with levels of target analytes. This indicates that PACs and PAC derivatives were not a major contributor to the cytotoxicity observed in the air samples.

The potential of wastewater treatment plants (WWTPs) to act as sources of poly and perfluoroalkyl substances (PFASs), volatile methyl siloxanes (VMSs) and organic UV-filters to the atmosphere was investigated. Target compounds included: PFASs (fluorotelomer alcohols (FTOHs), perfluorooctane sulfonamides/sulfonamidoethanols (FOSAs/FOSEs), perfluroalkyl sulfonic acids (PFSAs) and perfluroalkyl carboxylic acids (PFCAs)), cyclic VMSs (D3 to D6), linear VMSs (L3 to L5) and eight UV-filters. Emissions to air were assessed at eight WWTPs using paired sorbent-impregnated polyurethane foam passive air samplers, deployed during summer 2013 and winter 2014. Samplers were deployed on-site above the active tank and off-site as a reference. Several types of WWTPs were investigated: secondary activated sludge in urban areas (UR-AS), secondary extended aeration in towns (TW-EA) and facultative lagoons in rural areas (RU-LG). The concentrations of target compounds in air were ∼1.7-35 times higher on-site compared to the corresponding off-site location. Highest concentrations in air were observed at UR-AS sites while the lowest were at RU-LG. Higher air concentrations (∼2-9 times) were observed on-site during summer compared to winter, possibly reflecting enhanced volatilization due to higher wastewater temperatures or differences in influent wastewater concentrations. A significant positive correlation was obtained between concentrations in air and WWTP characteristics (influent flow rate and population in the catchment of the WWTP); whereas a weak negative correlation was obtained with hydraulic retention time. Emissions to air were estimated using a simplified dispersion model. Highest emissions to air were seen at the UR-AS locations. Emissions to air (g/year/tank) were highest for VMSs (5000-112,000) followed by UV-filters (16-2000) then ΣPFASs (10-110).

The occurrence of persistent organic pollutants (POPs) in the atmosphere of the near-coast South Atlantic Ocean was studied. Air samples were collected using a high-volume air sampler (filter and polyurethane foam) on board the Argentinean research cruise R/V Puerto Deseado (CONICET). Samples were analyzed for 50 polychlorinated biphenyl (PCBs), 22 organochlorine pesticides (OCPs) and 14 polybrominated diphenyl ethers (PBDEs). These POPs classes showed a trend of decreasing levels from near-shore to open ocean sites. OCPs and PCBs were in the same order of magnitude (2.71-87.1pg/m(3) and 9.56-130pg/m(3), respectively) while PBDEs levels were significantly lower (0.69-2.58pg/m(3)). Dichlorodiphenyltrichloroethanes (DDTs), endosulfans, chlordanes, hexachlorocyclohexanes (HCHs) and heptachlors were between 0.20 and 17.8pg/m(3), while drines and hexachlorobenzene (HCB) were at lower levels (0.28-3.71pg/m(3)). The most frequently detected PCBs were congeners 32, 28, 44, 52, 95, representing >50% of the total. The PBDEs congener pattern was dominated by congener 209 (70%), followed by 47 and 99 (16% and 7%, respectively). Air parcel back trajectories for the study period provided few insights as trajectories mainly stemmed from the open ocean with limited inputs from continental sources. These results indicate that the concentration of POPs (namely PCBs, OCPs, and PBDEs) in air remain elevated in the near-shore environment and then drop-off substantially beyond a distance of about 400km. This has implications for the loading of POPs and delivery to the marine environment in the near coastal zone.

HLJ1 (DNAJB4), a DNAJ/Hsp40 chaperone, has emerged as a novel prognostic marker in lung cancers; however, the molecular contribution and functionality in neoplastic diseases remain to be established. This study demonstrated that HLJ1 inhibits epithelial-mesenchymal transition in vitro and reduces lung cancer metastasis in vivo. Using shRNA silencing and ectopic expression of HLJ1, we found that HLJ1 not only suppresses catalytic activity of Src but also downregulates the formation of oncogenic complexes associated with the EGFR, FAK and STAT3 signaling pathways. A screen of specimens from HLJ1-knockout mice and lung cancer patients validated that HLJ1 expression is inversely correlated with Src activity. Mechanistically, HLJ1 protein directly bound to catalytic and protein-binding domains of Src through its amino acid Y172 and the P301/P304 motif. Following Src-induced HLJ1 phosphorylation at Y172, HLJ1-Src interaction was elevated, resulting in Src inhibition and malignancy suppression. Interestingly, both Src-binding regions also occurred in other DNAJB family members and contributed to anti-invasive activities of DNAJB proteins. We conclude that HLJ1 is an endogenous Src inhibitor that can suppress cancer metastasis through complex interacting mechanisms. This HLJ1-Src complex might provide a promising molecular model for developing new anticancer strategies.

Long-term air monitoring data for POPs are required to determine the effectiveness of source reduction measures and factors controlling air concentrations. Air samples were collected between 1992 and 2012 at three sites with different geographical characteristics (Burnt Island, Egbert and Point Petre) in the Canadian Great Lakes Basin (GLB) using high-volume samplers and analyzed for organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs). Spatial and temporal trends of gas-phase concentrations of OCPs, selected PCB congeners and Æ©PCBs (84 congeners) were assessed. Egbert had the highest concentrations of some OCPs due to historical [dichlorodiphenyltrichloroethanes (DDTs), dieldrin, γ-hexachlorocyclohexane (γ-HCH)] and current (endosulfan) applications of these pesticides in the surrounding agricultural cropland. This shows that agricultural areas are a source of OCPs to the GLB. High o,p'-/p,p'-DDT ratios were determined and an increasing trend was observed at Point Petre and Burnt Island up to 2004; indicating that the GLB is influenced by dicofol-type DDT sources, which have higher o,p'-/p,p'-DDT ratios than technical DDT. Atmospheric PCB concentrations at Egbert and Point Petre are higher than those measured at Burnt Island, likely due to urban influence and greater populations. Loadings calculations suggest that the atmosphere is a source of α-endosulfan and p,p'-DDT to the lakes and the opposite is true for p,p'-DDE. Long-term decreasing trends were observed for both OCPs and PCBs; consistent with control measures implemented in North America. Atmospheric PCB concentrations are decreasing relatively slowly, with halflives in the range of 9-39 years. Chlordane, α-endosulfan, ß-endosulfan, dieldrin, and DDT-related substances showed halflives in the range of 7-13 years. α-HCH and γ-HCH were decreasing rapidly in air, with halflives of 5 years. Long-term declining trends of PCBs and OCPs suggest that regulatory efforts to reduce emissions to the GLB environment have been effective, but emissions from primary and secondary sources might limit future declines.

Results are reported from an ongoing passive air monitoring study for polycyclic aromatic compounds (PACs) in the Athabasca oil sands region in Alberta, Canada. Polyurethane foam (PUF) disk passive air samplers were deployed for consecutive 2-month periods from November 2010 to June 2012 at 17 sites. Samples were analyzed for polycyclic aromatic hydrocarbons (PAHs), alkylated PAHs, dibenzothiophene and its alkylated derivatives (DBTs). Relative to parent PAHs, alkylated PAHs and DBTs are enriched in bitumen and therefore considered to be petrogenic markers. Concentrations in air were in the range 0.03-210 ng/m(3), 0.15-230 ng/m(3) and 0.01-61 ng/m(3) for ∑PAHs, ∑alkylated PAHs and ΣDBTs, respectively. An exponential decline of the PAC concentrations in air with distance from mining areas and related petrogenic sources was observed. The most significant exponential declines were for the alkylated PAHs and DBTs and attributed to their association with mining-related emissions and near-source deposition, due to their lower volatility and greater association with depositing particles. Seasonal trends in concentrations in air for PACs were not observed for any of the compound classes. However, a forest fire episode during April to July 2011 resulted in greatly elevated PAH levels at all passive sampling locations. Alkylated PAHs and DBTs were not elevated during the forest fire period, supporting their association with petrogenic sources. Based on the results of this study, an "Athabasca PAC profile" is proposed as a potential source marker for the oil sands region. The profile is characterized by ∑PAHs/∑Alkylated PAHs = ∼0.2 and ∑PAHs/∑DBTs = ∼5.

Malignant pleural effusion (MPE) is a useful specimen allowing for the evaluation of EGFR status in nonsmall cell lung cancer (NSCLC). However, direct sequencing of genomic DNA from MPE samples was found not to be sensitive for EGFR mutation detection. To test whether EGFR analysis from RNA is less prone to interference from nontumour cells that have no or lower EGFR expression, we compared three methods (sequencing from cell-derived RNA versus sequencing and mass-spectrometric analysis from genomic DNA), in parallel, for EGFR mutation detection from MPE samples in 150 lung adenocarcinoma patients receiving first-line tyrosine kinase inhibitors (TKIs). Among these MPE samples, EGFR mutations were much more frequently identified by sequencing using RNA than by sequencing and mass-spectrometric analysis from genomic DNA (for all mutations, 67.3 versus 44.7 and 46.7%; for L858R or exon 19 deletions, 61.3 versus 41.3 and 46.7%, respectively). The better mutation detection yield of sequencing from RNA was coupled with the superior prediction of clinical efficacy of first-line TKIs. In patients with acquired resistance, EGFR sequencing from RNA provided satisfactory detection of T790M (54.2%). These results demonstrated that EGFR sequencing using RNA as template greatly improves sensitivity for EGFR mutation detection from samples of MPE, highlighting RNA as the favourable source for analysing EGFR mutations from heterogeneous MPE specimens in NSCLC.

Systemic lupus erythematosus (SLE) is a prototypic autoimmune disease characterized by the expression of antibodies to DNA. These antibodies form immune complexes that can stimulate cytokine production as well as deposit in the tissues to incite inflammation and damage. For the formation of immune complexes, the availability of extracellular DNA in an immunologically relevant form is essential. While apoptosis has been implicated as the source of this nuclear material in SLE, as shown with in vitro or in vivo systems, extracellular DNA can originate from apoptotic as well as necrotic cells. In experimental models, the release of DNA occurs with the administration of cells induced to die, in vitro as well as the administration of agents to induce cell death in situ. This release can be influenced by the presence of inflammatory cells such as macrophages that can interact with dead and dying cells to modulate the translocation of DNA from the inside to the outside of cells. In vivo, both glucocorticoids and oestrogens can modify the extent of DNA release from the administration of dead and dying cells. Together, these findings indicate that the generation of extracellular DNA in SLE can result from cell death and that steps in this process represent potential targets for new therapies.

Rheumatoid arthritis (RA) is a common rheumatic disease in Caucasians and in other ethnic groups. Diagnosis is mainly based on clinical features. Before 1998, the only serological laboratory test that could contribute to the diagnosis was that for rheumatoid factor (RF). The disease activity markers for the evaluation of clinical symptoms or treatment outcome were the erythrocyte sedimentation rate (ESR) and C-reactive protein (CRP). As a matter of fact, the diagnosis of early RA is quite impossible, as the clinical criteria are insufficient at the beginning stage of the disease. In 1998, Schelleken reported that a high percentage of RA patients had a specific antibody that could interact with a synthetic peptide which contained the amino acid citrulline. The high specificity (98%) for RA of this new serological marker, anti-cyclic citrullinated antibody (anti-CCP antibody), can be detected early in RA, before the typical clinical features appear. The presence or absence of this antibody can easily distinguish other rheumatic diseases from RA. Additionally, the titer of anti-CCP can be used to predict the prognosis and treatment outcome after DMARDs or biological therapy. Therefore, with improvement of sensitivity, the anti-CCP antibody will be widely used as a routine laboratory test in the clinical practice for RA.

To assess the efficacy of sibutramine 15 mg once daily as weight reduction in overweight and obese (body mass index > 25 kg/m2) Chinese female type 2 diabetic patients and to evaluate the influence of weight loss on diabetic control, a randomised, double-blind, placebo-control, 12-week study was conducted. Chinese female type 2 diabetic patients, poorly controlled glucose levels and HbA(1C) > 8% were randomly assigned to two groups. In addition to their hypoglycaemic agents (maximal doses of sulphonylureas and metformin), one group (n = 30) received a sibutramine 15 mg once daily for 12 weeks, and the other (n = 30) received placebo for the same period. Comparing the changes that occurred after 12 weeks in the sibutramine and placebo groups, the former showed significantly greater reduction in body weight (2.5 vs. 0.1 kg, p < 0.05), fasting plasma insulin level (28.8 vs. 2.4 pmol/l, p < 0.01), 2-h postprandial blood glucose after standard test meal (3.2 vs. 1.1 mmol/l, p < 0.01), insulin resistance (5.1 vs. 0.2, p < 0.01), HbA1C (1.7% vs. 0.2%, p < 0.05), triglyceride (0.43 vs. 0.12 mmol/l, p < 0.05) and total cholesterol (0.52 vs. 0.08 mmol/l, p < 0.05). No significant differences were found between treatment groups in blood pressure and heart rate. The addition of sibutramine to diet and oral hypoglycaemic therapy resulted in significant weight loss and improvement in metabolic parameters in the treatment group. Sibutramine should be considered for use alongside diet and oral hypoglycaemic therapy in Chinese overweight and obese women with poorly controlled type 2 diabetes.
