RESUMEN

The development of fluorine-free firefighting foams has been proposed as a way to reduce the adverse environmental consequences of foams containing per- and poly-fluoroalkyl substances. While there are likely fewer environmental and ecological concerns with these new fluorine-free foams in terms of persistence and bioaccumulation, it is prudent to evaluate the ecotoxicity of these fluorine-free foam products given the absence of data. Oral chronic drinking water exposure studies on adult pairs of northern bobwhite quail (Colinus virginianus) were conducted with a short-chain fluorinated and a fluorine-free foam: Buckeye Platinum Plus C6 and National Foam Avio Green KHC, respectively, at three exposure concentrations (0.01%, 0.1%, and 0.25%). Adults were monitored for survival, growth, and reproductive output; and chicks were monitored for survival and growth. Growth parameters in adult quail were not affected by exposure to the Buckeye or Avio foam. However, liver lipid content was higher in adult males exposed to the Buckeye foam or the Avio foam at the highest exposure concentrations. Chicks were heavier and had higher growth rates after adult exposure to Avio at the highest exposure level (0.25%) and to Buckeye at the two lowest exposure levels but not at the highest exposure level. The two adverse reproductive effects observed from avian exposure to Buckeye were an increased percentage of cracked eggs and earlier arrested embryonic development. Similarly, chronic exposure to Avio also induced earlier arrested embryonic development. These results show that the fluorine-free foams tested did cause toxicity to bobwhite quail, but whether they pose a risk at contaminated sites requires further laboratory and field study and additional exposure data. Per- and polyfluoroalkyl substances (PFAS) are ubiquitous in the environment. Locations where PFAS-containing aqueous film-forming foam (AFFF) has been used or accidentally released have resulted in persistently high concentrations of PFAS, including in surface water that may be adjacent to release sites. Perfluorooctane sulfonic acid (PFOS) is most frequently measured near AFFF release sites; however, other PFAS are being quantified more frequently and, of those, perfluorononanoic acid (PFNA) is common. The goal of our study was to fill data gaps on PFNA toxicity to freshwater fish using the fathead minnow (Pimephales promelas). We aimed to understand how PFNA may impact apical endpoints following a 42-day exposure to mature fish and a 21-day exposure to second-generation larval fish. Exposure concentrations were 0, 124, 250, 500, and 1000 µg/L for both adult (F0) and larval (F1) generations. The most sensitive endpoint measured was development in the F1 generation at concentrations of ≥250 µg/L. The 10% and 20% effective concentration of the tested population for the F1 biomass endpoint was 100.3 and 129.5 µg/L, respectively. These data were collated with toxicity values from the primary literature on aquatic organisms exposed to PFNA for subchronic or chronic durations. A species sensitivity distribution was developed to estimate a screening-level threshold for PFNA. The resulting hazard concentration protective of 95% of the freshwater aquatic species was 55 µg PFNA/L. Although this value is likely protective of aquatic organisms exposed to PFNA, it is prudent to consider that organisms experience multiple stressors (including many PFAS) simultaneously; an approach to understand screening-level thresholds for PFAS mixtures remains an uncertainty within the field of ecological risk assessment. Researchers have developed numerous per- and polyfluoroalkyl substances (PFAS)-free aqueous film-forming foam (AFFF) formulations to replace PFAS-containing AFFF used for fire suppression. As part of the Department of Defense's Strategic Environmental Research and Development Program (SERDP), we examined the direct lethal effects of seven PFAS-free AFFF and a PFAS-containing AFFF on 14 aquatic species using a series of lethal concentration (LC50) tests. We assessed the LC10, LC50, and LC90 values using log-logistic and logit analyses. Across all aquatic species tested, we discovered that exposure to at least one PFAS-free AFFF was more or as toxic as exposure to the PFAS-containing AFFF. For most cases, National Foam Avio F3 Green KHC 3% and Buckeye Platinum Plus C6MILSPEC 3% were the most and least toxic formulations, respectively. Moreover, we found consistency among results from multiple experiments using the same minnow species (Pimephales promelas) and among closely related taxa (e.g., daphnids, amphibians). Lastly, the LC50 values for AFFF formulations trended lower for tested marine species as compared to those of freshwater species. These results dramatically increase the current knowledge on the potentially toxic effects of AFFF but also highlight the need for additional research and the development of new PFAS-free AFFF that are more "ecologically friendly" than those containing persistent PFAS.

As urban areas continue expanding, major cities become connected forming megacities. Urban encroachment into natural areas transforms the landscape into a built environment with heterogeneously distributed patches of novel habitat. Community structure within novel habitats is influenced by anthropogenic factors including fragmentation and species interactions. Alterations in complex biodiversity patterns may be used to assess how urban stressors impact community assemblages which, ultimately, may inform sustainable management decisions. To manage algal blooms, Aquashade® is applied directly to ponds. We investigated the effects of Aquashade®, nutrient loading and dispersal on local species diversity and compositional turnover of zooplankton communities from suburban ponds in Columbia, MD, USA using a mesocosm approach. We found that Aquashade® acted as an environmental filter by increasing local species diversity and decreasing compositional turnover. This ultimately could have an overall homogenizing effect on the regional species pool (or γ-diversity). The same pattern was observed in mesocosms that received simulated dispersal events of zooplankton. Nutrients, overall, increased autotrophic biomass and while Aquashade® had no effect on autotrophic biomass, the interaction of nutrients and Aquashade® similarly caused a homogenization of the zooplankton community. Additionally, there was an overall increase in cladoceran ephippia in mesocosms receiving Aquashade® compared to those not, suggesting there is a 'trigger' switching cladocerans from parthenogenic to sexual reproduction. Taken together, our results show the application of Aquashade®, nutrient loading and dispersal shift biodiversity patterns in urban zooplankton communities. We hypothesize these shifts originate at the resource level through alterations in the phytoplankton community either through composition or nutritive value in ponds receiving Aquashade®. Our study illustrates the importance of investigating multiple-scales of community assemblages when assessing impacts of anthropogenic stressors. Consideration of how to best manage green spaces may include planned un-managed areas to maintain regional biodiversity and heterogenous communities within the urban environment.

Freshwater salinization is a global concern partly attributable to anthropogenic salt contamination. The authors examined the effects of increased salinity (as NaCl, 250-4,000 µS/cm, specific conductance) on two sympatric freshwater gastropods (Helisoma trivolvis and Physa pomillia). Life stage sensitivities were determined by exposing naive eggs or naive juveniles (through adulthood and reproduction). Additionally, progeny eggs from the juvenile-adult exposures were maintained at their respective parental salinities to examine transgenerational effects. Naive H. trivolvis eggs experienced delayed development at specific conductance > 250 µS/cm; reduced survivorship and reproduction were also seen in juvenile H. trivolvis at 4,000 µS/cm. Survival and growth of P. pomilia were not affected by increased salinity following egg or juvenile exposures. Interestingly, the progeny of H. trivolvis exposed to higher salinity may have gained tolerance to increased salinity whereas P. pomilia progeny may have experienced negative transgenerational effects. The present study demonstrates that freshwater snail species vary in their tolerance to salinization and also highlights the importance of multigenerational studies, as stressor impacts may not be readily apparent from shorter term exposures.

A large data gap for reptile ecotoxicology still persists; therefore, ecological risk assessments of reptiles usually incorporate the use of surrogate species. This necessitates that (1) the surrogate is at least as sensitive as the target taxon and/or (2) exposures to the surrogate are greater than that of the target taxon. We evaluated these assumptions for the use of birds as surrogates for reptiles. Based on a survey of the literature, birds were more sensitive than reptiles in less than 1/4 of the chemicals investigated. Dietary and dermal exposure modeling indicated that exposure to reptiles was relatively high, particularly when the dermal route was considered. We conclude that caution is warranted in the use of avian receptors as surrogates for reptiles in ecological risk assessment and emphasize the need to better understand the magnitude and mechanism of contaminant exposure in reptiles to improve exposure and risk estimation.

Although anthropogenic pollutants are thought to threaten reptilian species, there are few toxicity studies on reptiles. We evaluated the toxicity of Pb as lead acetate to the Western fence lizard (Sceloporus occidentalis). The acute lethal dose and sub-acute (14-day) toxicity studies were used to narrow exposure concentrations for a sub-chronic (60-day) study. In the sub-chronic study, adult and juvenile male lizards were dosed via gavage with 0, 1, 10 and 20 mg Pb/kg-bw/day. Mortality was limited and occurred only at the highest dose (20 mg Pb/kg-bw/d). There were statistically significant sub-lethal effects of 10 and 20 mg Pb/kg-bw/d on body weight, cricket consumption, organ weight, hematological parameters and post-dose behaviors. Of these, Pb-induced changes in body weight are most useful for ecological risk assessment because it is linked to fitness in wild lizard populations. The Western fence lizard is a useful model for reptilian toxicity studies.

2,4-dintitrotoluene (2,4-DNT) is an explosive frequently found in the soil of military installations. Because reptiles can be common on these sites, ecological risk assessments for compounds such as 2,4-DNT could be improved with toxicity data specific to reptiles. Western fence lizards, Sceloporus occidentalis, were used to develop a laboratory toxicity model for reptiles. A hierarchical approach was used; acute to subchronic studies were conducted to provide toxicity data relevant to short- and long-term exposures. First, a modified median lethal dose (LD50) study was conducted on male and female lizards using a stage-wise probit model. The LD50 was 577 mg/kg for female and 380 mg/kg for male lizards. Subsequently, a subacute experiment was conducted to further assess 2,4-DNT toxicity to male lizards and to define exposure levels for a longer term, subchronic study. The subchronic study was conducted for 60 consecutive days; male lizards were exposed to 0, 9, 15, 25, 42, 70 mg/kg/d. Dose-dependent mortality was observed in the three highest dose groups (25, 42, and 70 mg/kg/d); all other animals survived the study duration. Benchmark dose model calculations based on mortality indicated a 5% effect level of 15.8 mg/kg/d. At study termination, a gross necropsy was performed, organ weights were taken, and blood was collected for clinical and hematological analysis. Body weight, kidney weight, food consumption, postdose observations, and blood chemistries all were found to be significantly different from controls at doses above 9 mg/kg/d. Also, preliminary results suggest behavioral observations, and reduced food consumption may be a sensitive indicator of toxicity. The present study indicates Sceloporus occidentalis is suitable for evaluating toxicity of compounds to reptilian species.
