RESUMEN

A new UGT1A1*28 detection method combining PCR and high-resolution agarose gel electrophoresis was developed. The viability of this method was demonstrated on 15 healthy adult volunteers. Subjects included 13 wild type homozygotes (86.7 %), 2 heterozygotes (13.3 %), and no mutant type homozygotes (0 %). The new UGT1A1*28 detection method results were fully consistent with DNA sequencing. PCR and agarose gel electrophoresis are common techniques with high-resolution agarose gels available commercially. These results support the clinical viability of this method potentially reducing UGT1A1*28 diagnosis complexity and cost.

This study ventures into the exploration of potential poly-3-hydroxybutyrate (PHB) degradation in alpine environments. PHB-degrading bacteria were identified in both campus soil, representing a residential area, and Mt. Kurodake soil, an alpine region in Hokkaido, Japan. Next-generation sequencing analysis indicated that the campus soil exhibited higher microbial diversity, while Ralstonia insidiosa C1, isolated from Mt. Kurodake soil, displayed the highest proficiency in PHB degradation. R. insidiosa C1 efficiently degraded up to 3% (w/v) of PHB and various films composed of other biopolymers at 14 °C. This bacterium synthesized homopolymers using substrates such as 3-hydroxybutyric acid, sugars, and acetic acid, while also produced copolymers using a mixture of fatty acids. The analysis results confirmed that the biopolymer synthesized by strain C1 using glucose was PHB, with physical properties comparable to commercial products. The unique capabilities of R. insidiosa C1, encompassing both the production and degradation of bioplastics, highlight its potential to establish a novel material circulation model.

BACKGROUND: Rice bran a by-product of the rice milling process is currently underutilized. Recent studies have shown that plant-derived nanoparticles (pdNPs) can be mass-produced at a low cost and exhibit biological and therapeutic activities. Rice bran contains various anti-cancer compounds, including γ-oryzanol and γ-tocotrienol, and rice bran-derived nanoparticles (rbNPs) can be employed as novel therapeutic agents for cancer treatment. RESULTS: Koshihikari rice bran was suspended in water, and the suspension was centrifuged and filtered through a 0.45-µm-pore size syringe filter. The filtrate was ultracentrifuged, and the precipitates were suspended to obtain rbNPs. The rbNPs were negatively charged exosome-like nanoparticles with an average diameter of approximately 130 nm. The rbNPs exhibited cytotoxic activities against cancer cells but not against normal cells. The cytotoxic activity of rbNPs to murine colon adenocarcinoma colon26 cells was significantly greater than DOXIL® or other pdNPs. The rbNPs induced cell cycle arrest and apoptosis, and reduced the expression of proliferative proteins, including ß-catenin and cyclin D1. Intraperitoneal injections of rbNPs into mice bearing peritoneal dissemination of colon26 cells significantly suppressed tumor growth with no significant adverse effects. CONCLUSION: These results indicated that rbNPs are promising nanoparticles, hold significant potential for anti-cancer applications, and are expected to play a vital role in cancer treatment.

RESUMEN

In a windowless poultry house raising layer chickens in Kanagawa prefecture, Japan, a slight increase in the mortality of chickens and a decrease in egg production were observed. Necropsy revealed numerous tapeworms and proglottids in chicken intestines. Histopathologically, gut-associated lymphoid tissues were observed in the lamina propria of the jejunum; however, no significant changes were observed in the other organs. Numerous hide beetles, Dermestes maculatus DeGeer, intermediate hosts of Raillietina cesticillus, were observed in the poultry house. Following a decline in beetle numbers, egg production increased and chicken mortality decreased. The life cycle of a tapeworm was easily established in a closed space, such as a windowless house, which led to severe infections.

Systemic lupus erythematosus is known to cause autoimmune hyperlipidemia. We present a case in which hypertriglyceridemia was exacerbated by dermatomyositis. A 53-year-old woman with a medical history of undertreated hypertriglyceridemia complained of dyspnea and arthralgia. Despite the treatment, her triglyceride levels increased concurrently with the onset of arthralgia. She had characteristic skin manifestations and tested positive for anti-Jo-1 autoantibodies, leading to a diagnosis of dermatomyositis. Chronic inflammation may result in elevated triglyceride levels. When dermatomyositis is diagnosed, evaluating lipid abnormalities is important.

Among the various elements that facilitate the movement of the lower limbs, the anterior cruciate ligament (ACL) is prone to injury. An adequate joint control of the lower limb can prevent ACL injury. Balancing activities between the agonist and the antagonist muscles is vital for joint control. However, prior studies on muscle activities were limited since they could not determine passive muscle activities. In this study, we develop a muscle model considering the passive properties to analyze the movement mechanism of the ACL under heavy loads, such as those produced during jump landing. We estimated the muscle activities occurring during a drop vertical jump (DVJ) by applying to the proposed method the physiological constraint that muscle activities are constant during a short time around landing. In addition, the knee joint torque and muscle forces were calculated from the estimated muscle activities, which were thereafter compared with those obtained using the conventional method. The results revealed that this passive muscle model appropriately represented the knee joint torque at DVJ landing by decreasing the passive muscle strain and increasing the isometric maximum muscle force. Moreover, the estimated muscle activities were larger than those obtained using the conventional method, which may be caused by the co-contraction between agonist and antagonist muscles that cannot be represented by the conventional method. This muscle co-contraction estimation algorithm would estimate the muscle load under heavy loads, and applying this knowledge to training would help to prevent ACL injuries.

In this paper, we propose the analysis method for finding out the similarity of the muscle force patterns to mine the risk factor of the anterior cruciate ligament (ACL) injury. Akaike information criteria (AIC) under the assumption of the auto-regression model is adapted to analyze the similarities of muscle force patterns in time-series. The difference of AIC values between 2 muscles is considered to be the distance between 2 muscle force patterns and the dexterity of the maneuver is expected to be discussed. We measured drop vertical jump (DVJ) and use the data around the contact timing of whom hadn't had ACL injury experiments. The results showed that we could successfully calculate AIC distance according to the similarity of the time-series data pattern and it can be useful to discuss one's dexterity of controlling body maneuvers soon after contact timing of DVJ motion.

Eucalyptol is one of the most popular volatile components. It is used in many essential oils for relieving sinus and lung congestion caused by a variety of conditions. This pilot study sought to analyze clinical evidence for the effect of the scent of eucalyptol on the cognitive function of elderly people. Seventy nursing-home residents with cognitive impairment were recruited. Three one-week experiments were performed: eucalyptol scent was diffused in bedrooms with a diffuser only at wake-up time in the first experiment, and at wake-up time and bedtime in the second and third experiments. Results showed that although an improvement was not seen when using Mini Mental State Examination (MMSE) and Cohen-Mansfild Agitation Inventory (CMAI) measures, Dementia Behavior Disturbance Scale (DBD) scores improved significantly, even though no subject reported perceiving the scent. The significant improvements of the behaviour were found not only among the subjects whose room had a diffuser but also among the subjects who were exposed to an unperceivable level of eucalyptol drifted in the living room.
