RESUMEN

Thermoresponsive polymer brushes have attracted considerable research attention owing to their unique properties. Herein, we developed silica beads grafted with poly(N-isopropylacrylamide (NIPAAm)-co-3-acrylamidopropyl trimethylammonium chloride (APTAC)-co-tert-butyl acrylamide (tBAAm) and P(NIPAAm-co-APTAC-co-n-butyl methacrylate(nBMA)) brushes. The carbon, hydrogen, and nitrogen elemental analysis of the copolymer-grated silica beads revealed the presence of a large amount of the grafted copolymer on the silica beads. The electrostatic and hydrophobic interactions between biomolecules and prepared copolymer brushes were analyzed by observing their elution behaviors via high-performance liquid chromatography using the copolymer-brush-modified beads as the stationary phase. Adenosine nucleotides were retained in the bead-packed columns, which was attributed to the electrostatic interaction between the copolymers and adenosine nucleotides. Insulin was adsorbed on the copolymer brushes at high temperatures, which was attributed to its electrostatic and hydrophobic interactions with the copolymer. Similar adsorption behavior was observed in case of albumin. Further, at a low concentration of the phosphate buffer solution, albumin was adsorbed onto the copolymer brushes even at relatively low temperatures owing to its enhanced electrostatic interaction with the copolymer. These results indicated that the developed thermoresponsive strong cationic copolymer brushes can interact with peptides and proteins through a combination of electrostatic and temperature-modulated hydrophobic interactions. Thus, the developed copolymer brushes exhibits substantial potential for application in chromatographic matrices for the analysis and purification of peptides and proteins.

Background: We explore an optimized approach for increasing lesion size using a novel ablation catheter with a surface thermocouple and efficient irrigation in a temperature-control setting. Methods: We conducted radiofrequency applications at various power levels (35 W, 40 W, and 45 W), contact forces (CFs, 10 g/20 g), and durations (60 s/120 s/180 s) in perpendicular/parallel catheter orientations, with normal saline irrigation (NS-irrigation) and Half NS-irrigation (HNS-irrigation) in an ex-vivo model (Step 1). In addition, we performed applications (35 W/40 W/45 W for 60 s/120 s/180 s in NS-irrigation and 35 W/40 W for 60 s/120 s/180 s in HNS-irrigation) in four swine (Step 2), evaluating lesion characteristics and the occurrence of steam pops. Results: In Step 1, out of 288 lesions, we observed 47 (16.3%) steam pops, with 13 in NS-irrigation and 34 in HNS-irrigation (p = .001). Although steam pops were mostly observed with the most aggressive setting (45 W/180 s, 54%) with NS-irrigation, they happened in less aggressive settings with HNS irrigation. Lesion size significantly increased with longer-duration ablation but not with HNS-irrigation. The optimal %impedance-drop cutoff to predict steam pops was 20% with a negative-predictive-value (NPV) = 95.1% including NS- and HNS-irrigation groups, and 22% with an NPV = 96.1% in NS-irrigation group. In Step 2, similar to the ex-vivo model, lesion size significantly increased with longer-duration ablation but not with HNS-irrigation. Steam pops were absent with NS-irrigation (0/35) even with the largest %impedance-drop reaching 31% at 45 W/180 s. All steam pops were observed with HNS-irrigation (6/21, 29%). The optimal %impedance-drop cutoff predicting steam pops was 24% with an NPV = 96.3% including both NS- and HNS-irrigation groups. Conclusions: Rather than using HNS-irrigation, very long-duration of radiofrequency applications up to 45 W/180 s may be recommended to safely and effectively increase lesion dimensions using this catheter with NS-irrigation.

Leptospirosis is a zoonotic disease caused by the pathogenic spirochaetes of the genus Leptospira. It is a public health concern in the Pacific Islands and is considered endemic in Palau. However, information on the genotypes and serotypes of causative Leptospira spp. in the country is limited. In this study, we isolated leptospires and detected antileptospiral antibodies in dogs and pigs. The isolates were characterized using a serological method and whole-genome sequencing. Leptospira interrogans was isolated from five of the 20 symptomatic dogs and one of the 58 healthy pigs. Their serogroups were identified as Icterohaemorrhagiae and Pyrogenes; however, the serogroup of one isolate could not be determined. Anti-Leptospira antibodies were detected in 14.4% (26/181) of the dogs and 20% (10/50) of the pigs. The reactive serogroups in dogs and pigs were almost identical, except for the Panama serogroup. Core genome multilocus sequence typing revealed that five of the six core genome sequence types (cgSTs) were newly identified in this study. The cgSTs from the serogroup Icterohaemorrhagiae isolates belonged to the same group as the Copenhageni and Icterohaemorrhagiae serovars isolated in other countries, whereas no similar cgSTs were identified in the Pyrogenes or unidentified serogroup strains. We demonstrated a high incidence of canine and porcine leptospirosis and identified new L. interrogans genotypes (cgSTs) circulating in Palau. Further investigations are needed to determine whether dogs and pigs serve as maintenance hosts for newly identified L. interrogans genotypes and whether they pose a risk of leptospirosis transmission to humans.

OBJECTIVE: Previous studies show that the COVID-19 pandemic affected the number of surgeries performed. However, data on the association between the COVID-19 pandemic and otolaryngologic surgeries according to subspecialties are lacking. This study was performed to evaluate the impact of the COVID-19 pandemic on various types of otolaryngologic surgeries. METHODS: We retrospectively identified patients who underwent otolaryngologic surgeries from April 2018 to February 2021 using a Japanese national inpatient database. We performed interrupted time-series analyses before and after April 2020 to evaluate the number of otolaryngologic surgeries performed. The Japanese government declared its first state of emergency during the COVID-19 pandemic in April 2020. RESULTS: We obtained data on 348,351 otolaryngologic surgeries. Interrupted time-series analysis showed a significant decrease in the number of overall otolaryngologic surgeries in April 2020 (-3619 surgeries per month; 95% confidence interval, -5555 to -1683; p < 0.001). Removal of foreign bodies and head and neck cancer surgery were not affected by the COVID-19 pandemic. In the post-COVID-19 period, the number of otolaryngologic surgeries, except for ear and upper airway surgeries, increased significantly. The number of tracheostomies and peritonsillar abscess incisions did not significantly decrease during the COVID-19 pandemic. CONCLUSION: The COVID-19 pandemic was associated with a decrease in the overall number of otolaryngologic surgeries, but the trend differed among subspecialties.

The architecture of species interaction networks is a key factor determining the stability of ecological communities. However, the fact that ecological network architecture can change through time is often overlooked in discussions on community-level processes, despite its theoretical importance. By compiling a time-series community dataset involving 50 spider species and 974 Hexapoda prey species/strains, we quantified the extent to which the architecture of predator-prey interaction networks could shift across time points. We then developed a framework for finding species that could increase the flexibility of the interaction network architecture. Those "network coordinator" species are expected to promote the persistence of species-rich ecological communities by buffering perturbations in communities. Although spiders are often considered as generalist predators, their contributions to network flexibility vary greatly among species. We also found that detritivorous prey species can be cores of interaction rewiring, dynamically interlinking below-ground and above-ground community dynamics. We further found that the predator-prey interactions between those network coordinators differed from those highlighted in the standard network-analytical framework assuming static topology. Analyses of network coordinators will add a new dimension to our understanding of species coexistence mechanisms and provide platforms for systematically prioritizing species in terms of their potential contributions in ecosystem conservation and restoration.

Various types of hemostatic agents are used to manage bleeding in surgery. Many such agents are animal products, which carry the risk of secondary infection. The aim of this study is to develop a novel hemostatic agent from a non-animal source that quickly stops bleeding, is easy to use, and has no risk of infection. In this study, we synthesized calcium ion-crosslinked sodium alginate (Alg-Na/Ca) by partial substitution of Ca ions for Na ions in sodium alginate. We prepared 12 kinds of Alg-Na/Ca powders with different Ca mass ratios, molecular weights, M/G ratios and particle size distributions and measured their swelling ratio and the burst pressure generated. We found that Alg-Na/Ca began to swell immediately after contact with saline, especially Alg-Na/Ca at Ca mass ratios of 74.1-77.0 % showed a high swelling ratio after 2 min and a high burst pressure, over 200 % and 500 mmHg respectively. Also, there is a correlation between the swelling ratio after 2 min and the burst pressure. Our results suggest that, by optimizing the composition conditions, Alg-Na/Ca may be an effective hemostatic agent that could act as a tamponade by absorbing and swelling at a bleeding site to quickly achieve primary hemostasis.

RESUMEN

A fire started in Shurijo Seiden, or the main hall of Shurijo Castle, Naha-city, Okinawa, Japan on the morning of October 30th, 2019. The fire resulted in loss of 8 structures and many important Okinawan cultural assets. The original Shurijo Castle was destroyed many years ago and a replica was constructed and rebuilt to be as close as possible to the original building. The replica was fabricated mainly from wood (Chamaecyparis taiwanensis). Firebrands were reported during the fire. In this study, firebrands from Shurijo Castle was collected and analyzed. The data was compared with those from other investigation as well as experimental data.

Over the past few years, the large outdoor fire problem has been a growing concern throughout the world. It is recommended to clear the combustibles around homes and within communities to avoid potential loss of properties, as firebrand shower ignition is a dangerous threat. One of the common combustibles around homes is mulching materials. A reduced-scale experimental protocol was developed to study ignition of mulching materials by firebrands and resulting impact to adjunct wall assemblies. Reduced-scale experimental results were compared with full-scale experimental results. Specifically, two trends were of interest in the comparisons. First, the ranking of the ease of ignition for various mulch types from exposure to firebrand showers. Second, if a given mulch type ignited from exposure to firebrand showers, was the resulting mulch bed fire able to ignite the adjacent wall assembly. The reduced-scale experimental results captured some of these trends observed from full-scale experiments but not completely. The findings still suggest that the reduced-scale experiments may give insights into how easily different mulch beds may be ignited by firebrands, as compared to much more costly and time-consuming full-scale experiments. While it is interesting to conduct full-scale experiments, this is very expensive and not always practical, so the authors are devising far cheaper reduced-scale experiments to provide more in-depth scientific understanding of firebrand shower ignition of construction components.
