RESUMEN

The elasmoid scales in teleost fish serve as exemplary models for natural fibre composites with integrated flexibility and protection. Yet, limited research has been focused on the potential structural, chemical, and mechanical heterogeneity within individual scales. This study presents systematic characterizations of the elasmoid scales from black drum fish (Pogonias cromis) at different zones within individual scales as a natural fibre composite, focusing on the microscopic structural heterogeneities and corresponding mechanical effects. The focus field at the centre of the scales exhibits a classical tri-layered collagen-based composite design, consisting of the mineralized outermost limiting layer, external elasmodine layer in the middle, and the unmineralized internal elasmodine layer. In comparison, the rostral field at the anterior end of the scales exhibits a two-layered design: the mineralized outermost limiting layer exhibits radii sections on the outer surface, and the inner elasmodine layer consists of collagen fibre-based sublayers with alternating mineralization levels. Chemical and nanoindentation analysis suggests a close correlation between the mineralization levels and the local nanomechanical properties. Comparative finite element modelling shows that the rostral-field scales achieve increased flexibility under both concave and convex bending. Moreover, the evolving geometries of isolated Mandle's corpuscles in the internal elasmodine layer, transitioning from irregular shapes to faceted octahedrons, suggest the mechanisms of mineral growth and space-filling to thicken the mineralized layers in scales during growth, which enhances the bonding strength between the adjacent collagen fibre layers. This work offers new insights into the structural variations in individual elasmoid scales, providing strategies for bioinspired fibre composite designs with local-adapted functional requirements.

OBJECTIVE: The tyrosine-protein kinase inhibitor, genistein, can inhibit cell malignant transformation and has an antitumor effect on various types of cancer. It has been shown that both genistein and KNCK9 can inhibit colon cancer. This research aimed to investigate the suppressive effects of genistein on colon cancer cells and the association between the application of genistein and KCNK9 expression level. METHODS: The Cancer Genome Atlas (TCGA) database was used to study the correlation between the KCNK9 expression level and the prognosis of colon cancer patients. HT29 and SW480 colon cancer cell lines were cultured to examine the inhibitory effects of KCNK9 and genistein on colon cancer in vitro, and a mouse model of colon cancer with liver metastasis was established to verify the inhibitory effect of genistein in vivo. RESULTS: KCNK9 was overexpressed in colon cancer cells and was associated with a shorter Overall Survival (OS), a shorter Disease-Specific Survival (DFS), and a shorter Progression-Free Interval (PFI) of colon cancer patients. In vitro experiments showed that downregulation of KCNK9 or genistein application could suppress cell proliferation, migration, and invasion abilities, induce cell cycle quiescence, promote cell apoptosis, and reduce epithelial-mesenchymal transition of the colon cancer cell line. In vivo experiments revealed that silencing of KCNK9 or application of genistein could inhibit hepatic metastasis from colon cancer. Additionally, genistein could inhibit KCNK9 expression, thereby attenuating Wnt/ß-catenin signaling pathway. CONCLUSION: Genistein inhibited the occurrence and progression of colon cancer through Wnt/ß-catenin signaling pathway that could be mediated by KCNK9.

An increasing number of studies have observed that ignoring individual exposures to non-residential environments in people's daily life may result in misleading findings in research on environmental exposure. This issue was recognized as the neighborhood effect averaging problem (NEAP). This study examines ethnic segregation and exposure through the perspective of NEAP. Focusing on Xining, China, it compares the Hui ethnic minorities and the Han majorities. Using 2010 census data and activity diary data collected in 2013, the study found that NEAP exists when examining ethnic exposure. Respondents who live in highly mixed neighborhoods (with high exposures to the other ethnic group) experience lower activity-space exposures because they tend to conduct their daily activities in ethnically less mixed areas outside their home neighborhoods (which are more segregated). By contrast, respondents who live in highly segregated neighborhoods (with low exposures to the other ethnic group) tend to have higher exposures in their activity locations outside their home neighborhoods (which are less segregated). Therefore, taking into account individuals' daily activities in non-residential contexts in the assessment of environmental exposure will likely lead to an overall tendency towards the mean exposure. Using Tobit models, we further found that specific types of activity places, especially workplaces and parks, contribute to NEAP. Ignoring individual exposures in people's activity places will most likely result in misleading findings in the measurement of environmental exposure, including ethnic exposure.

Acute lung injury (ALI) is a common clinical disease with high morbidity in both humans and animals. Studies have shown that intestinal microbiota affect the pathology and immune function of respiratory diseases through the "gut-lung axis". The authors investigated the therapeutic effect of fecal microbiota transplantation (FMT) in rats with ALI induced by lipopolysaccharide (LPS). Rats were treated with FMT, and then measured lung wet/dry ratio, PaO2 in artery, proinflammatory marker, and TGF-ß1, Smad3, Smad7, and phosphorylated ERK (p-ERK) protein levels, as well as a histopathologic analysis and high-throughput sequencing of intestinal microbiota. FMT significantly reduced lung wet/dry ratio and TNF-α, IL-1ß, and IL-6 levels, but increased the levels of PaO2 in artery. In addition, FMT significantly decreased the expression of TGF-ß1, Smad3, and p-ERK, while increased the levels of Smad7. Lung histopathological analyses showed that FMT reduced the inflammatory cell infiltration and interstitial lung exudates. High-throughput sequencing of intestinal microbiota analyses showed that FMT reconstructed the structure of intestinal microbiota, and increased the gene abundance of the bacterial community. Therefore, FMT may act on the TGF-ß1/Smads/ERK pathway by regulating intestinal microbiota, inhibiting immune inflammation, reducing the production of inflammatory markers in the body and release, and reducing alveolar epithelial damage and repair, thereby improving the endotoxic ALI in rats induced by LPS.

CoCrFeNiPdMnx (x = 0, 0.2, 0.4, 0.6, 0.8) high entropy alloys (HEAs) were prepared and characterized. With an increase in Mn addition, the microstructures changed from dendrites (CoCrFeNiPd with a single face-centered-cubic (FCC) phase) to divorced eutectics (CoCrFeNiPdMn0.2 and CoCrFeNiPdMn0.4), to hypoeutectic microstructures (CoCrFeNiPdMn0.6), and finally to seaweed eutectic dendrites (CoCrFeNiPdMn0.8). The addition of Mn might change the interface energy anisotropy of both the FCC/liquid and MnPd-rich intermetallic compound/liquid interfaces, thus forming the seaweed eutectic dendrites. The hardness of the FCC phase was found to be highly related to the solute strengthening effect, the formation of nanotwins and the transition from CoCrFeNiPd-rich to CoCrFeNi-rich FCC phase. Hierarchical nanotwins were found in the MnPd-rich intermetallic compound and a decrease in either the spacing of primary twins or secondary twins led to an increase in hardness. The designing rules of EHEAs were discussed and the pseudo binary method was revised accordingly.

In traditional long bone fracture reduction surgery, there are some drawbacks such as low accuracy, high radiation for surgeons and a risk of infection. To overcome these disadvantages, a removable hybrid robot system is developed, which integrates a removable series-parallel mechanism with a motor-double cylinder (MDC) driven mode. This paper describes the mechanism in detail, analyses the principle and the method of the fracture reduction, presents the surgical procedure, and verifies the reduction accuracy by experiments with bone models. The results are shown as follows. The mean deviations of the axial displacement and lateral displacement are 1.60mm and 1.26 mm respectively. The standard deviations are 0.69 mm and 0.30 mm. The mean deviations of the side angle and turn inward are 2.06° and 2.22° respectively. The standard deviations are 0.50° and 0.99°. This minimally invasive robot features high accuracy and zero radiation for surgeons, and is able to conduct fracture reduction for long bones.
