RESUMEN

Alopecia areata (AA) is a tissue-specific autoimmune disease characterized by non-scarring and rapid onset of hair loss. Interleukin (IL)-17A is mainly produced by T helper 17 (Th17) cells and may play a crucial role in the pathogenesis of various autoimmune diseases including AA. We conducted this research to measure serum level of IL-17A in patients with AA and investigated its relationship with the clinical manifestations in patients with AA. We assessed 36 patients with AA and 20 healthy control subjects. Demographic information and clinical characteristics were determined by physical examination and via the review of medical history. Serum IL-17A was measured by using enzyme-linked immunosorbent assay. Serum IL-17A concentration was significantly higher in patients with AA than in the control group (P=0.004). The AA patients with severe presentation, personal atopy, nail abnormalities, or active phase had significantly higher serum IL- 17A levels compared to others without these signs. Increased serum IL-17A levels in patients with AA correlate with severity and indicate an active disease state. These findings suggest that IL-17A may play an important role in determining the pathogenesis of AA and may serve as a valuable clinical biomarker of this disease.

A series of schiartane C29 nortriterpenoids with 5/5/7/6/5 membered consecutive rings (1‒5) with an unique schinortriterpenoid skeleton including a new, kadcoccilactone V (1), together with four known ones (2‒5) and three known triterpenoids (6‒8) were identified from stems of Kadsura coccinea (Lem.) A. C. Smith. The structures of 1 and known compounds were elucidated by interpretation of 1D and 2D NMR, and HR-ESI-MS data as well as comparing those data in the literature. All the isolated compounds were examined for cytotoxic effects against six human cancer cell lines [(HCT-15 (colon), NUGC-3 (stomach), NCI-H23 (lung), ACHN (renal), PC-3 (prostate), and MDA-MB-231 (breast)]. Among them, compound 6 showed potent cytotoxicity against NCI-H23 (GI50 1.28 µM) and NUGC-3 (GI50 1.28 µM), and significantly inhibited on PC-3, MDA-MB-231, ACHN, HCT-15 with GI50 values around 2.33 to 2.67 µM.

Three-dimensional (3D) culture dish patterned with a microwell structure demonstrates a great application potential in biotechnology. This study reports on the easy fabrication of an ordered customizable honeycomb microwell array on the surface of polymer substrates including the commercial Petri dish to create a biological platform for cell culture. The fabrication method is based on a very simple solvent dip-coating technique and the methanol accumulation-induced phase separation in which a binary mixture of chloroform and methanol is used to induce a ternary solution and to guarantee the formation of the ordered pore array on the substrate. The surface topology of the honeycomb substrate is manipulated through varying the experimental conditions; notably, the obtained honeycomb structure is part of the substrate, which reveals an increase in the structure's stability for the practical applications. Honeycomb-structured Petri dish fabricated using this method is applied as a scaffold for cell growth to demonstrate its potential in biomedical applications.
