RESUMEN

OBJECTIVE: To investigate the change trends of sperm quality with seasonal variations among the volunteers of sperm donors in Beijing area, as well as the relationship between two parameters. METHODS: Semen data from the volunteers of sperm donors in Human Sperm Bank of Peking University Third Hospital were collected using a retrospective study method. The subjects were divided into 4 seasonal groups based on the lunar solar terms and the time of sperm donation. The data were assessed to find whether there were differences in semen parameters among different seasonal groups, and to analyze the change trends and the influence of seasonal factors on semen parameters. RESULTS: A total of 21 174 semen parameter data were analyzed. Firstly, to analyze all data as a whole, in spring, summer, autumn and winter groups, sperm concentration was (106.04±59.67)×106/mL, (97.61±47.41)×106/mL, (100.18±47.17)×106/mL, (100.59±38.68)×106/mL, respectively, and the spring group was significantly higher than the other 3 seasonal groups (P < 0.001); proportion of progressive motility sperm (PR) was 56.49%±12.76%, 58.02%±13.65%, 58.05%±12.36%, and 57.66%±12.61%, respectively, spring group was lower than the other three seasonal groups, and summer group was better among the latter (P < 0.001). There was no difference in normal rate of sperm morphology among the four seasonal groups. The qualified rate of sperm donors in the winter group was higher than that in the other three seasons groups (P < 0.01), while the qualified rate in the summer group was lower than that in the other three seasons groups. In addition, the semen parameters of the volunteers during the screening period and the official sperm donation period were analyzed respectively, which revealed that sperm concentration of spring group was higher than that of summer and winter groups, and PR was lower than that of summer and autumn groups. On account of the semen parameters of official sperm donation period, multiple linear regression analysis found that season was the main factor affecting sperm concentration, the average sperm concentration in spring group was about 6×106/mL higher than in winter group, but PR was 2.9% lower in spring group compared with autumn group (all P < 0.05). CONCLUSION: Season was the influencing factor of semen quality of sperm donors in Beijing area. We recommend spring and winter may be the preferred seasons for sperm donation.

It is known that the expression of the deubiquitinating enzyme BRCA1-BRCA2-containing complex subunit 3 (BRCC3) and cyclin-dependent protein kinase 5 (Cdk5) is increased in Parkinson's disease (both are involved in neuroinflammatory response). However, the regulatory mechanism of Cdk5 on the post-translational modification of BRCC3 remains unclear. Here we studied whether Cdk5 phosphorylates BRCC3. Phosphorylation of BRCC3 by Cdk5 was predicted by GPS 5.0 software. His-BRCC3 plasmid was constructed by cloning the BRCC3 gene into pGEX-6P-1 vector, and then His-BRCC3 fusion protein was induced with isopropyl ß-d-1-thiogalactopyranoside and purified using His-Tag affinity chromatography purification agarose. Phosphorylation of BRCC3 fusion protein by Cdk5 in vitro was detected by mass spectrometry and Western blotting. The results showed that multiple phosphorylation sites were predicted by GPS 5.0, and the His-BRCC3 fusion protein was successfully induced and purified. In vitro kinase assay, Western blotting, and mass spectrometry showed that Cdk5 can phosphorylate BRCC3. It has been demonstrated that protein kinase Cdk5 can phosphorylate the deubiquitinating enzyme BRCC3 in vitro, which provides new data for further study on the mechanism of neurodegeneration.

The present study provides the first report on the molecular epidemiological data regarding infection by hemoplasma and piroplasma species in wild Rattus edwardsi, from China. In the current study, blood samples were investigated from 32 wild Rattus edwardsi from Hunan (23) and Guangxi (9) provinces, China. The prevalence of hemoplasma and piroplasma was 65.63% (21/32) and 6.25% (2/32), respectively. Phylogenetic analyses indicated that hemoplasmas (HQ183731, HQ183732) derived from wild Rattus edwardsi in China, can be grouped into a solitary clade closely related to H. muris (HMU82963) and M. haemomuris (AB758435). In addition, it was shown that piroplasmas from this study have very close genetic distance to other unidentified piroplasma species isolated from China (AB242140) and Japan (AB188086). The results suggested that hemoplasmas isolated in this study should be represented as a new genotype. Piroplasmas on the other hand needs more sequenced samples in its life-cycle and evidence to check its taxonomic status. These data may have important implications for researching on the epidemiology and population biology as well as for studying the taxonomy status of hemoplasmas and piroplasmids of wild rodents.

We attempted to create a new germplasm of cucumber cultivar Chinese long (9930) using different doses of ethyl methyl sulfonate (EMS) to induce variability. We tested EMS concentration (0, 0.5, 1.0, 1.5, 2, 3% v/v) with post-treatment (0.1 M Na2S2O3 and water), EMS concentration (0, 0.5, 1.0, 1.5% v/v) over different treatment times (8, 16, 24 h), and EMS concentration (0, 0.5, 1.0, 1.5% v/v) with different treatment temperatures (20 and 28°C). In all experiments with increasing EMS concentration, germination percent, index, and rate were decreased. After addition of stop solution (0.1 M Na2S2O3), post-treatment mutated seeds showed higher germination (84.44%) and rate (37.5%) than seeds treated with water (80 and 34.07%, respectively), while the germination index was high in seeds treated with water. At 20°C, the germination index (4.13) and rate (56.25%) were affected to a greater extent than at 28°C (7.68 and 91.31%, respectively). Treatment times of 16 and 24 h showed similar results for germination percent and rate, while the germination index was decreased over time. There were significant differences in seedling height, fresh true leaf weight, seedling weight, and plant survival with increasing EMS concentration and time. Higher variations in the form of dwarf seedlings were recorded after treatment with 1.5% EMS for 24 h. Based on germination and morphological data, an EMS concentration of 1.5% for 24 h at 20°C and post-treatment with stop solution (0.1 M Na2S2O3) efficiently caused mutation.

RESUMEN

In order to search for new compounds with higher anti-cancer activities and lower toxicities, 18 dehydrogenated carboncyclic analogs of norcantharidin, of which 17 are unknown compounds, were designed and synthesized. Preliminary screening results revealed that compound V6, VI1 and VI4 exhibited fairly apparent inhibitory activity against the growth of human-hepatoma cells, Bel-7402, in vitro. The inhibitory rate of VI4 is 52%, almost the same as that of norcantharidin, in the concentration of 0.05 mumol/ml.

In order to search for new compounds with higher anti-cancer activities and lower toxicities, 19 amino acid derivatives of norcantharidin, of which 16 are unknown compounds, were designed and synthesized. Preliminary screening results revealed that 2-(syn-exo-7-oxabicyclo [2.2.1] heptane-2,3-dicarboxylic imido)-N-phenyl glutaramic acid exhibited a fairly apparent inhibitory activity against human-hepatoma cells in vitro (inhibitory rate 39.4% at 0.025 mumol/ml).
