RESUMEN

We studied the effects of a dual-vector DYSF gene delivery system based on adeno-associated virus serotype 9 capsids on pathological manifestations of dysferlinopathy in skeletal muscles of Bla/J mice lacking DYSF expression. The mice received intravenous injection of 3×1013 genomic copies of the virus containing the dual-vector system. M. gastrocnemius, m. psoas major, m. vastus lateralis, and m. gluteus superficialis were isolated for histological examination in 3, 6, and 12 weeks after treatment. Healthy wild-type (C57BL/6) mice served as positive control and were sacrificed 3 weeks after injection of 150 µl of 0.9% NaCl into the caudal vein. To detect dysferlin in muscle cryosections, immunohistochemical analysis with diagnostic antibodies was performed; paraffin sections were stained with hematoxylin and eosin for morphometric analysis. After administration of gene-therapeutic constructs, muscle fibers with membrane or cytoplasmic dysferlin location were detected in all examined muscles. The proportion of necrotic muscle fibers decreased, the number of muscle fibers with central location of the nucleus increased, and the mean cross-section area of the muscle fibers decreased.

In this computational work applicability of IR spectral parameters for evaluations of OH···N hydrogen bond length is discussed. For a set of 124 complexes with OH···N hydrogen bond formed by combinations of methanol/acetic acid and pyridine (and their fluorine substituted versions) geometries, energies and IR parameters were calculated at MP2/def2-TZVP level of theory. For a number of IR parameters (the shift of proton donor group stretching vibration Δνs, increase of its intensity I, the low-frequency hydrogen bond stretching vibration νσ, bending in-plane δ and out-of-plane γ vibrations) equations linking them with interatomic distances are proposed, the robustness and accuracy of such equations are discussed. The enthalpy of OH···N hydrogen bond formation ΔH was also linked with electron density parameters in (3; -1) critical point.

An adequate experimental model is important to understand pathophysiological processes ongoing in the pancreas with diabetes mellitus. Our study was aimed to describe early ultra- and microstructural changes in the rat pancreas in 12-48 h after alloxan administration in a dose of 180 mg/kg. A histopathological examination of the endocrine pancreas revealed the loss of borders between endocrine cells, granular dystrophy and degranulation, sings of necrosis in central cells of the Langerhans islets and apoptosis of their peripheral ones manifested as DNA fragmentation and an increased expression of apoptosis markers. There was a gradual increase of a Langerhans islet area, a decreased percentage of insulin+ cells and an increased one of glucagon+ cells, as well as the presence of proliferating islet cells were found. Structural changes of the exocrine pancreas included fatty degeneration, signs of exocrine cell mitochondrial damage, increased acini, which are located mainly around the Langerhans islets, as well as perivascular edema and leukocytic infiltration. Described ultra- and microstructural alterations suggest a significant contribution of apoptosis to death of endocrine cells exposed to alloxan. Coexisting damage of the exocrine pancreas with its stroma involvement is for the first time described.

The paper presents data on the hygienic assessment of the content of heavy metals and petroleum products in the soil on the territory of oil-producing areas of the Republic of Tatarstan. The average total content of lead (10,1 mg/kg), cadmium (0.073 mg/kg), manganese (792.4 mg/kg), cobalt (14.2 mg/kg), chromium (87.6 mg/kg), nickel (65.2 mg/ kg), arsenic (8.0 mg/kg), copper (36.6 mg/kg) and zinc (521 mg/kg) in soil for agricultural purposes near objects oil production was shown not to exceed the established hygienic standards. Soil contamination with oil products leads to an increase in its toxicity and mutagenicity. Soil contamination with oil products at the level of as low as 30 mg/ kg leads to the gain in its toxicity (low toxicity) and mutagenicity (average) (specific gravity of germinated seeds was 70.7±0.67%, aberrations - 1.78±0.19%).

RESUMEN

In the paper there are presented data on the hygienic evaluation of the air pollution in the city of Kazan as a risk factor for the public health. The largest contribution to the air pollution in the city of Kazan was shown to be endowed by vehicles. The proportion of vehicle emissions in total emissions in the city in 2012 amounted to 71.4%. According to monitoring data the average annual concentrations of pollutant substances in 2012 exceeded the hygienic standards for benzo (a) pyrene--in 2.5 times, soot--2.2 times, nitrogen dioxide--1.8 times, formaldehyde--1.7 times. The risk of inhalation exposure is assessed as high, the greatest contribution to the risk is contributed by suspended matter PM2.5, soot and nitrogen dioxide.

RESUMEN

Patients with lymphomas were shown to have authentically a higher frequency of fragile sites, both spontaneous and induced by aphidikoline expression, in comparison with the control group. Differential sensitivity of chromosomes caused by the action of aphidikolin was revealed. The fragility of chromosomes 9 and 11 in patients with lymphomas on sites 9q31-32 and 11p13-14 was authentically higher (alpha = 0.05) than that in healthy individuals. Site 9q31-32 appeared to be strictly specific for lymphoma disease.

RESUMEN

The article reviews foreign and domestic literature on the problem of maxillary fibrous dysplasia (MFD) and reports 10 cases of disseminated monoosseous MFD in children aged 3-14 years treated surgically. The scope of the intervention depended on the process location, age of the patients and function-sparing principle. All the operations provided satisfactory cosmetic and functional effect. Their results advocate an individual approach to choice of the scope and kind of surgical intervention.
